Mechanisms of Borrelia burgdorferi internalization and intracellular innate immune signaling.

Lyme disease is a long-term infection whose most severe pathology is characterized by inflammatory arthritis of the lower bearing joints, carditis, and neuropathy. The inflammatory cascades are initiated through the early recognition of invading Borrelia burgdorferi spirochetes by cells of the innate immune response, such as neutrophils and macrophage. B. burgdorferi does not have an intracellular niche and thus much research has focused on immune pathways activated by pathogen recognition molecules at the cell surface, such as the Toll-like receptors (TLRs). However, in recent years, studies

Schistosoma mansoni: assessment of effects of oleic acid, cercarial age and water temperature on parasite-host attraction.

Although the lifecycle of Schistosoma spp. and pathophysiology of schistosomiasis have been established, the mechanism by which cercariae find their host is not well understood. Speculatively, host infection by random and accidental host contact is not as biologically plausible as a biochemical mechanism of mammalian attraction. A few studies have indicated that biochemical cues and temperature gradients may play a role in host identification, attraction and attachment triggers. This study aimed to elucidate these mechanisms more specifically through evaluation of biochemical, age and temperat

Ulcerated yellow spot syndrome: implications of aquaculture-related pathogens associated with soft coral Sarcophyton ehrenbergi tissue lesions.

We introduce a new marine syndrome called ulcerated yellow spot, affecting the soft coral Sarcophyton ehrenbergi. To identify bacteria associated with tissue lesions, tissue and mucus samples were taken during a 2009 Indo-Pacific research expedition near the Wakatobi Island chain, Indonesia. Polymerase chain reaction targeting the 16S rDNA gene indicated associations with the known fish-disease-causing bacterium Photobacterium damselae, as well as multiple Vibrio species. Results indicate a shift toward decreasing diversity of bacteria in lesioned samples. Photobacterium damselae ssp. piscicid

Host response cytokine signatures in viral and nonviral acute exacerbations of chronic obstructive pulmonary disease.

Viruses are strongly associated with acute exacerbations of chronic obstructive pulmonary disease (AECOPD). Interferon-inducible protein-10 has been recently described as a biomarker of human rhinovirus infection, but there are no reports on the role of other immune mediators in AECOPD of viral origin. As an attempt to evaluate the differences in the systemic immune mediators profiles between AECOPD patients with presence/absence of viral infection, we measured 27 cytokines, chemokines, and cellular growth factors in the plasma of 40 patients with AECOPD needing of hospitalization by using a L

Occurrence of soil- and tick-borne fungi and related virulence tests for pathogenicity to Ixodes scapularis (Acari: Ixodidae).

Ixodes scapularis Say, the blacklegged tick, vectors Borrelia burgdorferi Johnson et al. 1984, the bacterium that causes Lyme disease, the most important vector-borne disease in the United States. Efforts to reduce I. scapularis populations are shifting toward the development of biological control methods. Currently, only a few entomopathogenic fungal species are considered virulent to ticks. We hypothesized that these species may not represent the most abundant local taxa that would be pathogenic to ticks in situ. To identify potential entomopathogenic fungi at a study site in Westchester Cou

Phosphatidylcholine biosynthesis and its significance in bacteria interacting with eukaryotic cells.

Phosphatidylcholine (PC), a typical eukaryotic membrane phospholipid, is present in only about 10% of all bacterial species, in particular in bacteria interacting with eukaryotes. A number of studies revealed that PC plays a fundamental role in symbiotic and pathogenic microbe-host interactions. Agrobacterium tumefaciens mutants lacking PC are unable to elicit plant tumors. The human pathogens Brucella abortus and Legionella pneumophila require PC for full virulence. The plant symbionts Bradyrhizobium japonicum and Sinorhizobium meliloti depend on wild-type levels of PC to establish an efficie

Shigella infection of intestinal epithelium and circumvention of the host innate defense system.

Shigella, Gram-negative bacteria closely related to Escherichia coli, are highly adapted human pathogens that cause bacillary dysentery. Although Shigella have neither adherence factors nor flagella required for attaching or accessing the intestinal epithelium, Shigella are capable of colonizing the intestinal epithelium by exploiting epithelial-cell functions and circumventing the host innate immune response. During Shigella infection, they deliver many numbers of effectors through the type III secretion system into the surrounding space and directly into the host-cell cytoplasm. The effector

Temperature-mediated patterns of local adaptation in a natural plant-pathogen metapopulation.

There have been numerous investigations of parasite local adaptation, a phenomenon important from the perspectives of both basic and applied evolutionary ecology. Recent work has demonstrated that temperature has striking effects on parasite performance by mediating trade-offs in parasite life history and through genotype x environment interactions. To test whether parasite local adaptation is mediated by temperature, I measured the performance of sympatric populations against allopatric populations of a fungal pathogen, Podosphaera plantaginis, on its host Plantago lanceolata, across a temper

First record of myiasis in Aplastodiscus arildae (Anura: Hylidae) by Notochaeta bufonivora (Diptera: Sarcophagidae) in the Neotropical area.

Myiasis in anurans is usually caused by diptera from the Calliphoridae, Sarcophagidae, Chloropidae, and Muscidae families. In this study, one case of cloacal myiasis and one of oral myiasis were registered in Aplastodiscus arildae, caused by Notochaeta bufonivora in Teresópolis, RJ, Brazil. With this report, a new host is listed for N. bufonivora, the first case of tree-frog myiasis caused by sarcophagid flies and the first occurrence of hylid myiasis in the Neotropical area.

Structural insights into mechanisms of zinc scavenging by the Candida albicans zincophore Pra1.

Candida albicans causes over 400,000 life-threatening, and an additional half a billion of mucosal infections annually. In response to infection, the host limits essential micronutrient availability, including zinc, to restrict growth of the invading pathogen. As assimilation of zinc is essential for C. albicans pathogenicity, limitation induces secretion of the zincophore protein Pra1 to scavenge zinc from the host. Pra1 also plays a number of important roles in host-pathogen interactions and is conserved in most fungi. However, the structure of fungal zincophores is unknown. Here, we present…

Double trouble: how co- and superinfections shape viral dynamics and host responses.

Concurrent infection of humans by multiple distinct viruses is a common biological phenomenon with important consequences for virus-host interactions. While coinfection refers to the simultaneous infection of an individual or cell with two or more viruses, superinfection denotes the establishment of a secondary infection following a primary one, in which the initial infection influences the susceptibility, replication, or outcome of the secondary infection. Virus-virus interactions can be antagonistic, facilitative, dependent/assisted, or neutral. Antagonistic interactions include superinfecti

Plant viruses and the microbiome: a complex network shaping plant health and disease resistance.

Plant viruses and microbiomes are pivotal to plant health, growth, and their ability to withstand environmental challenges. However, the extent to which viruses engage in direct or indirect crosstalk with plant-associated microbial communities remains largely unexplored. Traditionally, plant viruses are considered as harmful agents that disrupt normal plant functions, but new research indicates they may also engage in complex interactions with the plant's microbiome. On the other hand, the plant microbiome, comprising a diverse assembly of bacteria, fungi, and other microbes, can influence the

Identification of Virus-Host Interactions.

No protein acts on its own to carry out its designated function. Proteins interact with other biomolecules to form complexes that perform a coordinated function. While proteins most often form complexes involving other proteins, they also interact with nucleic acids, lipids, carbohydrates, ions, and other small molecules and metabolites (Wang et al., Proteomics 22(15-16):e2100190, 2022; Veenstra, Proteomics 21:e2000235, 2021). While the early days of proteomics were focused on identifying and quantifying as many proteins as possible, scientists quickly recognized the power of proteomics to rap

Genomic Structure Predicts Granulovirus Resistance Without a Detectable Life-History Trade-Off in Plodia interpunctella.

Understanding the genetic architecture of pathogen resistance and its relationship to host life-history traits is a fundamental problem in evolutionary biology, yet the conditions under which resistance costs manifest as detectable trade-offs remain poorly understood. Here, we examine resistance in Plodia interpunctella (Indian meal moth) to its granulovirus (PiGV) across 12 inbred lines maintained without deliberate pathogen exposure, alongside an independently assembled outbred stock population as a genomic reference. We quantify resistance using LD50 values from dose-response assays, summar

A trade-off between fungal biofilm formation and persistence in the host.

Fungal biofilm formation has long been considered an important virulence trait. A new study on Candida parapsilosis outbreak isolates in PLOS Biology challenges this, finding that low biofilm-producing strains are better adapted for immune evasion and survival.

Host genetic ancestry and immunity to pathogens.

Recent studies have reported increased susceptibility to or reduced severity of viral and bacterial infections between distinct ancestral populations. In this review, we will outline the current state of knowledge regarding the interplay between genetic ancestry and the immune response to pathogens. We will concentrate on the ancestry-associated drivers of these immunological variations through the lens of different viral and bacterial infections in humans. Understanding how different ancestral backgrounds influence the immune response to pathogens can reveal previously unrecognized mechanisms

The study of the Drosophila innubila nudivirus in cells and flies.

Species of the fruit fly, Drosophila, are essential models for investigating host/virus interactions. Research aimed at understanding how hosts mount immune defenses against viruses and how viruses evade those immune defenses relies on host/virus pairs that have evolved together over time, thus necessitating a need for a DNA virus model system for Drosophila with a virus that naturally infects the host. The Drosophila innubila nudivirus (DiNV) is an emerging model system poised to take on the role as the DNA virus model for Drosophila. In this paper we describe the development of the DiNV mode

The molecular arms race: mechanisms of fungal antiviral defense and viral counterstrategies.

Advances in high-throughput sequencing have accelerated the discovery of novel mycoviruses, shedding light on the dynamic coevolution between fungi and their viral partners. Recent studies indicate that mycoviruses act as key ecological drivers, modulating fungal growth, pathogenicity, and environmental adaptability. While most infections remain asymptomatic, some display a broad spectrum of interactions ranging from hypovirulence and hypervirulence to mutualism. Fungal hosts possess multilayered antiviral systems - such as vegetative incompatibility and RNA interference - that restrict viral
