Implication of the thalamus in sleep alterations observed in Alzheimer's disease.

Alzheimer's disease (AD), the most common neurodegenerative disorder, is frequently accompanied by sleep disturbances. Alterations of sleep, in both quality and quantity, contribute to the progression of AD neuropathology. The thalamus, through its diverse nuclei, plays a central role in sleep regulation and sleep-dependent memory consolidation, making it a relevant structure to investigate sleep disturbances associated with AD. Structural studies reveal that AD-related changes emerge early within the thalamus: Higher-order nuclei show atrophy, degeneration, and tau and Aβ accumulation. AD is …

The effect of insomnia on development of Alzheimer's disease.

Alzheimer's disease (AD) is the most common type of dementia and a neurodegenerative disorder characterized by memory deficits especially forgetting recent information, recall ability impairment, and loss of time tracking, problem-solving, language, and recognition difficulties. AD is also a globally important health issue but despite all scientific efforts, the treatment of AD is still a challenge. Sleep has important roles in learning and memory consolidation. Studies have shown that sleep deprivation (SD) and insomnia are associated with the pathogenesis of Alzheimer's disease and may have …

Early detection of cryptic memory and glucose uptake deficits in pre-pathological APP mice.

Earlier diagnosis and treatment of Alzheimer's disease would greatly benefit from the identification of biomarkers at the prodromal stage. Using a prominent animal model of aspects of the disease, we here show using clinically relevant methodologies that very young, pre-pathological PDAPP mice, which overexpress mutant human amyloid precursor protein in the brain, exhibit two cryptic deficits that are normally undetected using standard methods of assessment. Despite learning a spatial memory task normally and displaying normal brain glucose uptake, they display faster forgetting after a long d

Long-term complete remission and peripheral biomarkers in Hodgkin lymphoma patients after decitabine-plus-camrelizumab epi-immunotherapy and treatment cessation.

Patients with relapsed/refractory classical Hodgkin lymphoma (cHL) achieve complete response (CR) after decitabine-plus-camrelizumab therapy, while long-term outcome especially after treatment discontinuation remains unclear. We present a retrospective analysis of 87 relapsed/refractory cHL patients who acquired CR after decitabine-plus-camrelizumab. Patients were divided into two groups and received consolidation treatment every 3-4 or 6-12 weeks, and 1-year of continuous CR was guaranteed for treatment cessation. At a median follow-up of 5.3 years, the median relapse-free survival (RFS) afte

Central activity of 2-amino-4-phosphonobutyric acid (AP4).

The influence of intracerebroventricular (icv) injections of 2-amino-4-phosphonobutyric acid (AP4) on the activity of central nervous system was examined in rats. The compound (100 and 200 micrograms icv) significantly enhanced apomorphine stereotypy, in doses of 50, 75, and 100 micrograms decreased haloperidol-induced catalepsy and did not change motility of rats estimated in the "open field" test. AP4 significantly improved learning and memory processes in passive and learning in active avoidance situation. The highest significant effect was observed after icv dose of 100 micrograms per rat,

Double dissociation in the involvement of noradrenergic and endocannabinoid systems in classical and higher-order conditioning in newborn rabbits.

Since Ivan Pavlov first demonstrated classical conditioning by pairing a neutral stimulus with a reinforcer, researchers have applied this approach to understand not only direct associations, but also more complex learning processes, such as sensory preconditioning and second-order conditioning. Despite their relative immaturity, very young mammals such as newborn rabbits exhibit robust classical and higher-order conditioning. However, the neuromodulatory systems involved in these different types of neonatal conditioning remain to be identified. Here, we compared the role played by the noradre

A neuromodulatory circuit-to-molecular pathway for reformatting aversive memories during recall.

Memories can be altered when they are recalled through the process of reconsolidation, requiring gene expression in brain cells that store these memories. How brain circuits reformat memories during recall by directing molecular signaling in specific neuronal populations is not known. Here, we show that brainstem noradrenaline projections to the amygdala, a brain region that stores aversive emotional memories, control memory reconsolidation in rats. During reconsolidation, this circuit regulates the nuclear translocation of CREB-regulated transcriptional coactivator-1 (CRTC1), a molecule impor

Pharmacological activation of the lateral orbitofrontal cortex on regulation of learned fear and extinction.

Obsessive-compulsive disorder (OCD) is usually accompanied with hyperactivity of the orbitofrontal cortex (OFC). OCD patients have anxiety issues, and there is high comorbidity of OCD and post-traumatic stress disorder (PTSD). One of the leading factors of PTSD is the failure of fear extinction. In this study, we examined whether hyperactivity of the OFC interfered with extinction processes. The lateral OFC (lOFC) was pharmacologically activated with N-methyl-d-aspartate (NMDA) in behaving rats during encoding, consolidation, and retrieval, of Pavlovian fear extinction. We found that when we b

Corticosterone injection into the dorsal and ventral hippocampus impairs fear memory reconsolidation in a time-dependent manner in rats.

Reconsolidation is an active process induced following the reactivation of previously consolidated memories. Recent studies suggest brain corticosteroid receptors may participate in the modulation of fear memory reconsolidation. Glucocorticoid receptors (GRs), with 10-fold lower affinity than mineralocorticoid receptors (MRs), are mainly occupied during the peak of the circadian rhythm, and after stress, so they probably have a more critical role than MRs in memory phases during stressful situations. This study investigated the role of dorsal and ventral hippocampal (DH and VH) GRs and MRs on

Studies on retrograde and anterograde amnesia of olfactory memory after denervation of the hippocampus by entorhinal cortex lesions.

The effect of hippocampal denervation on olfactory memory in rats was tested after interrupting the lateral olfactory tract projections at the level of the entorhinal cortex. When lesioned animals were trained to learn new odors, they showed no evidence of retention 3 h after acquisition. These results confirm earlier data on rapid forgetting in rats after hippocampal deafferentation and are in parallel to the anterograde amnesia typically found in humans with hippocampal damage. On the other hand, preoperatively learned information was minimally impaired after hippocampal deafferentation even

Odor-Cued Targeted Reactivation Was Unable to Selectively Benefit Declarative Memories During Sleep.

Declarative memories are reactivated-and thereby consolidated-during sleep. Real-life memories are typically nested hierarchically (e.g., memory for making coffee nested within memory for one's morning routine). We tested the specificity of memory reactivation during sleep in humans: is it limited to low-tier items or does it extend to wider contexts? To test this, we adapted a well-replicated design using targeted memory reactivation, which uses non-invasive sensory cues to preferentially reactivate memories during sleep. Thirty-two participants (18 women and 14 men) learned two sets of objec

Neural correlates of sleep-induced benefits on traumatic memory processing.

Recent findings indicate that sleep after trauma compared to sleep loss inhibits intrusive memory development, possibly by promoting adequate memory consolidation and integration. However, the underlying neural mechanisms are still unknown. Here, we examined the neural correlates underlying the effects of sleep on traumatic memory development in 110 healthy participants using a trauma film paradigm and an implicit memory task with fMRI recordings in a between-subjects design. To further facilitate memory integration, we used targeted memory reactivation (TMR) to reactivate traumatic memories d…

DNA methyltransferase activity in the basolateral amygdala is critical for reconsolidation of a heroin reward memory.

The persistence of drug memory contributes to relapse to drug seeking. The association between repeated drug exposure and drug-related cues leads to cravings triggered by drug-paired cues. The erasure of drug memories has been considered a promising way to inhibit cravings and prevent relapse. The re-exposure to drug-related cues destabilizes well-consolidated drug memories, during which a de novo protein synthesis-dependent process termed "reconsolidation" occurs to restabilize the reactivated drug memory. Disrupting reconsolidation of drug memories leads to the attenuation of drug-seeking be

Getting to know you: Gaze behaviours and biographical information in face-name associations.

BACKGROUND: A person's name serves as a unique identifier, yet retrieving it is considered the most challenging aspect of person recognition. Here, we examined the role of biographical information and gaze behaviours during the acquisition and short- and long-term retrieval of face-name associations. METHODS: During Familiarization, undergraduate participants (N = 47) were presented with new face-name associations, with a biographical fact (hobby) provided for half of the pairs. During Consolidation, participants repeatedly recognized the names of the newly learned faces until they reached a

Basolateral Amygdala Corticotrophin Releasing Factor Receptor 2 Interacts with Nonmuscle Myosin II to Destabilize Memory.

Inhibiting the actin motor ATPase nonmuscle myosin II (NMII) with blebbistatin (Blebb) in the basolateral amgydala (BLA) depolymerizes actin, resulting in an immediate, retrieval-independent disruption of methamphetamine (METH)-associated memory. The effect is highly selective, as NMII inhibition has no effect in other relevant brain regions (e.g. dorsal hippocampus [dPHC], nucleus accumbens [NAc]), nor does it interfere with associations for other aversive or appetitive stimuli, including cocaine (COC). To investigate a potential source of this specificity, pharmacokinetic differences in METH
