Identification of host proteins involved in Japanese encephalitis virus infection by quantitative proteomics analysis.

Japanese encephalitis virus (JEV) enters host cells via receptor-mediated endocytosis and replicates in the cytoplasm of infected cells. To study virus-host cell interactions, we performed a SILAC-based quantitative proteomics study of JEV-infected HeLa cells using a subcellular fractionation strategy. We identified 158 host proteins as differentially regulated by JEV (defined as exhibiting a greater than 1.5-fold change in protein abundance upon JEV infection). The mass spectrometry quantitation data for selected proteins were validated by Western blot and immunofluorescence confocal microsco

Trisomy for synaptojanin1 in Down syndrome is functionally linked to the enlargement of early endosomes.

Enlarged early endosomes have been observed in neurons and fibroblasts in Down syndrome (DS). These endosome abnormalities have been implicated in the early development of Alzheimer's disease (AD) pathology in these subjects. Here, we show the presence of enlarged endosomes in blood mononuclear cells and lymphoblastoid cell lines (LCLs) from individuals with DS using immunofluorescence and confocal microscopy. Genotype-phenotype correlations in LCLs carrying partial trisomies 21 revealed that triplication of a 2.56 Mb locus in 21q22.11 is associated with the endosomal abnormalities. This locus

Current risk of iatrogenic Creutzfeld-Jakob disease in the UK: efficacy of available cleaning chemistries and reusability of neurosurgical instruments.

The initial cleaning of reusable surgical devices is critical to ensure the efficacy of the subsequent sterilisation process. Transmissible spongiform encephalopathies (TSEs) are incurable and fatal neurodegenerative diseases apparently transmitted simply by the absorption or ingestion of self-aggregating protease-resistant prions (PrP(Sc)), which are very resilient to most standard cleaning chemistries and heat-based decontamination techniques. Therefore there is a risk of iatrogenic transmission from reusable surgical devices if these are allowed to retain potentially infectious material aft

Spitting performance parameters and their biomechanical implications in the spitting spider, Scytodes thoracica.

Spitting spiders Scytodes spp. subdue prey by entangling them at a distance with a mixture of silk, glue, and venom. Using high-speed videography and differential interference contrast microscopy, the performance parameters involved in spit ejection by Scytodes thoracica (Araneae, Scytodidae) were measured. These will ultimately need to be explained in biomechanical and fluid dynamic terms. It was found that the ejection of "spit" from the opening of the venom duct (near the proximal end of the fang) was orderly. It resulted in a pattern that scanned along a lateral-medial axis (due to fang os

Macrocephaly in bull spermatozoa is associated with nuclear vacuoles, diploidy and alteration of chromatin condensation.

Spermatozoa from 2 dairy AI (artificial insemination) bulls (A and B), identified by their abnormal spermiogram with cells depicting frequent macrocephaly, double tails and nuclear vacuoles, were case-investigated and compared to normal spermatozoa from a control AI sire (C). Head sizes were measured and morphological abnormalities scored using brightfield and differential interference contrast microscopy. The degree of sperm maturation and of resistance to acid-induced DNA denaturation in situ were determined after uploading of acridine orange using flow cytometry of 5,000 cells/sample. Nucle

Effective inhibition of Japanese encephalitis virus replication by small interfering RNAs targeting the NS5 gene.

Japanese encephalitis virus (JEV), a mosquito-borne flavivirus, causes an acute infection of the central nervous system resulting in encephalitis of humans and many kinds of animals. NS5, the largest and most conserved flavivirus protein, is homologous to methyltransferase and RNA-dependent RNA polymerase. RNA interference is an effective anti-viral strategy to inhibit viral replication in vitro. In this study, four short hairpin RNA (shRNA) expression vectors (pS4.1-NS5-201, pS4.1-NS5-455, pS4.1-NS5-699, and pS4.1-NS5-804) targeting the NS5 gene of JEV were employed to target and destroy JEV

Proteome analysis of Legionella vacuoles purified by magnetic immunoseparation reveals secretory and endosomal GTPases.

Legionella pneumophila, the causative agent of Legionnaires' disease, replicates in macrophages and amoebae within 'Legionella-containing vacuoles' (LCVs), which communicate with the early secretory pathway and the endoplasmic reticulum. Formation of LCVs requires the bacterial Icm/Dot type IV secretion system. The Icm/Dot-translocated effector protein SidC selectively anchors to LCVs by binding the host lipid phosphatidylinositol-4-phosphate (PtdIns(4)P). Here, we describe a novel and simple approach to purify intact vacuoles formed by L. pneumophila within Dictyostelium discoideum by using m

Evolutionarily conserved nuclear migration genes required for early embryonic development in Caenorhabditis elegans.

The nudF and nudC genes of the fungus Aspergillus nidulans encode proteins that are members of two evolutionarily conserved families. In A. nidulans these proteins mediate nuclear migration along the hyphae. The human ortholog of nudF is Lis1, a gene essential for neuronal migration in the developing cerebral cortex. The mammalian ortholog of nudC encodes a protein that interacts with Lis1. We have identified orthologs of nudC and Lis1 from the nematode Caenorhabditis elegans. Heterologous expression of the C. elegans nudC ortholog, nud-1, complements the A. nidulans nudC3 mutant, demonstratin

Heparin inhibits leukocyte rolling in pial vessels and attenuates inflammatory changes in a rat model of experimental bacterial meningitis.

Heparin is a natural proteoglycan that was first described in 1916. In addition to its well characterized effect on blood coagulation, it is becoming clear that heparin also modulates inflammatory processes on several levels, including the interference with leukocyte-endothelium interaction. Anecdotal observations suggest a better clinical outcome of heparin-treated patients with bacterial meningitis. The authors demonstrate that heparin, a glycosaminoglycan, inhibits significantly in the early phase of experimental pneumococcal meningitis the increase of 1) regional cerebral blood flow (125 +

Microtubule-based filopodium-like protrusions form after axotomy.

The growth cone at the front of a growing neurite often has F-actin-rich structures--digitate filopodia and sheet-like veils and lamellipodia--whose protrusion advances the leading edge. Microtubules and other cytoplasmic constituents later fill the protruded area, transforming it into new neuritic length. Growth can be initiated from an axon by transecting it. We have used video-enhanced contrast-differential interference contrast microscopy to observe the early events following transection of Aplysia axons in culture. Many filopodium-like protrusions (FLPs) grew rapidly (average instantaneou

Subperineurial demyelination associated with reduced nerve blood flow and oxygen tension after epineurial vascular stripping.

Using laser Doppler measurements of nerve blood flow and electron microscopy, we determined that removal of the vasa nervorum from the surface of rat peripheral nerve results in an immediate 58.4% +/- (SD) 12.6% reduction in nerve blood flow (p less than 0.017) and subsequent subperineurial demyelination. To further assess the role of ischemia in demyelination, a second group of Sprague-Dawley rats (250 to 300 gm) was anesthetized and oxygen tensions were recorded with platinum microelectrodes in the tibial epineurial and endoneurial spaces before and 30 minutes after epineurial devascularizat

Absolute specificity for retrograde fast axonal transport displayed by lipid droplets originating in the axon of an identified Aplysia neuron in vitro.

Lipid droplets were found to form all along the axon of the giant cerebral neuron (GCN) of the sea hare Aplysia californica when the cell was placed in culture. The emission of yellow fluorescence by the droplets after exposure of the neuron to Nile red and their uniformly dark appearance in electron micrographs of axons fixed with glutaraldehyde and osmium tetroxide identified them as lipid droplets. In contrast to lipid droplets in fat cells and certain other cell types, these droplets were bounded by a membrane, indicating that the lipid droplet is a type of organelle that is membranated in

Neuropathology of "spinning syndrome" induced by prenatal intoxication with a PCB in mice.

A striking motor dysfunction, "spinning syndrome," developed with a high frequency in weaning mice whose dams received oral 3,4,3',4'-tetrachlorobiphenyl (4-CB) during gestation (day 10 through day 16). The syndrome is permanent and is characterized by swift circling movements sustained in one direction at a minimal rate of 40 turns/min (usually 50 to 150 turns/min), restlessness, and hyperkinesia. Twenty-four spinners and 4-CB nonspinners and age-matched controls were subjected to histopathologic, histofluorescent, histochemical, and electron microscopic studies. The most reliable histopathol

Human cytomegalovirus infection: recent developments in diagnosis and epidemiology.

Cytomegalic inclusion disease (CID) is caused by a horizontally or vertically transmitted human herpes virus infection and may persist for life without obvious clinical symptoms. A serious course of horizontal primary and recurrent infections, however, is often observed in immunocompromised persons such as recipients of organ transplants and patients receiving fresh blood transfusions. Vertical infection may cause fetopathies. The human cytomegalovirus (HCMV) is thought to inherit an oncogenic potential as lately discussed for AIDS and M. Kaposi. Laboratory diagnosis of HCMV infection is perfo

Composition-Dependent Microwave Shielding Performance of Kaolinite/Chopped-Strand Composites for Wide Frequency Range.

The increasing demand for high-frequency electronic and communication systems has intensified the need for lightweight and efficient electromagnetic interference (EMI) shielding materials. In this study, the conventional oxide mixing method was used to form kaolinite-chopped strand composites. Natural kaolinite powder was calcined at 450 °C for 4 h to remove volatile and organic components before composite fabrication. Kaolinite/chopped-strand epoxy composites were fabricated and evaluated for their microwave shielding effectiveness in the 0-4.4 GHz frequency range. Composites with different k

[Synchronous detection test strip of multiple exogenous mycotoxins in Chinese medicinal materials based on flower-like gold nanoparticles].

Mycotoxin contamination is one of the major causes of quality and safety risks in Chinese medicinal materials. To effectively ensure medication safety, this study established a multiplex and rapid detection method for exogenous fungal mycotoxins in Chinese medicinal materials. Flower-like gold nanoparticles(AuNPs) and gold-labeled probes were characterized using ultraviolet-visible spectroscopy and transmission electron microscopy. Key parameters, including the pH of the AuNP solution, antibody dosage, amount of gold-labeled probes, antigen-antibody jetting rate, nitrocellulose membrane select

Polarization-Interference Jones Matrix Sensors of Layer-by-Layer Scanning of Polycrystalline Dehydrated Blood Films. Fundamental and Applied Aspects.

To date, visual analysis is mainly used to evaluate images of dehydrated films (facies) of biological fluids-microscopy at various magnifications, illumination with white or polarized light, as well as using a dark field. At the same time, important information on the architectonics of optically anisotropic supramolecular networks of facies is unknown (inaccessible). In our work, a model of optical anisotropy of the architectonics of supramolecular networks of blood facies is proposed. Algorithms and a methodology for a new multifunctional method of polarization-interference visualization of t

Furan derivative affects the cell division and mitochondrial integrity of tachyzoites of Toxoplasma gondii.

Toxoplasma gondii is an obligate intracellular protozoan, the causative agent of toxoplasmosis. The current treatment against toxoplasmosis is based on the combination of sulfadiazine and pyrimethamine, which are toxic and unable to clear the infection. Due to the need for new active drugs against T. gondii, we have described here the effects of Furan derivatives on T. gondii in vitro. They were previously used with relevant activity against Leishmania amazonensis and Trypanosoma cruzi. The anti-Toxoplasma effects of the furan derivatives were evaluated using tachyzoites of T. gondii from RH s

Contact Dynamics of Cytoadhering Plasmodium falciparum-Infected Erythrocytes in Flow.

The virulence of the human malaria parasite Plasmodium falciparum is linked to the altered mechanical and adhesive properties of infected erythrocytes, which adhere to the microvascular endothelium to evade splenic clearance. The underlying biophysical mechanisms remain incompletely understood, particularly regarding the contact area and bond landscape, due in part to the rapid and transient nature of these interactions. In this study, we investigated the dynamic adhesion behavior of P. falciparum-infected erythrocytes on surfaces functionalized with intercellular adhesion molecule 1 (ICAM-1),

RNAi of 20S proteasome β subunit 3 inhibits midgut homeostasis and the survival of the migratory locust, Locusta migratoria.

BACKGROUND: Tissue homeostasis is essential for both morphogenesis and maintaining function. In insects, intestinal homeostasis is critical for growth and development. The midgut is key in food digestion, nutrient absorption, and immune response. Locusta migratoria is among the most damaging agricultural pests worldwide. Identifying a suitable target gene is important for RNA interference (RNAi)-based pest control. RESULTS: In this study, RNAi-mediated knockdown of the 20S proteasome β subunit 3 of L. migratoria (LmProsβ3) caused atrophy of the midgut and gastric caecum in fourth- and fifth-i
