Human monocarboxylate transporter 2 (MCT2) is a high affinity pyruvate transporter.

The transport of pyruvate and lactate across cellular membranes is an essential process in mammalian cells and is mediated by the H+/monocarboxylate transporters (MCTs). We have molecularly cloned and characterized a novel human monocarboxylate transporter, MCT2. The cDNA is 1,907 base pairs long and encodes a polypeptide of 478 amino acids with 12 predicted transmembrane domains. Human MCT2 is the product of a single gene that mapped to chromosome 12q13 by fluorescence in situ hybridization. The kinetic properties of human MCT2 fulfill the criteria to establish it as a H+/monocarboxylate tran

Discrete thalamic lesions attenuate winter adaptations and increase body weight.

The midline thalamus (e.g., the paraventricular thalamic nuclei and the reuniens nucleus) of Siberian hamsters and other mammals has been reported to contain specific binding sites for melatonin, a hormone that is essential for photoperiodically induced winter adaptations such as reproductive quiescence, loss of body weight, daily torpor, and the winter molt. The first experiment investigated whether the midline thalamus is necessary for these winter adaptations. Adult Siberian hamsters received discrete neurotoxic lesions of the paraventricular thalamic nuclei or the reuniens nucleus while un

Human immunodeficiency virus type 1 infection of choriocarcinoma-derived trophoblasts.

An in vitro model of placental infection by human immunodeficiency virus type 1 (HIV-1) was established using human choriocarcinoma-derived trophoblast lines exposed to free HIV-1 or HIV-1-infected lymphocytic and monocytic cells. Virus infectivity was evaluated by measuring both the levels of p24 HIV-1 antigen and reverse transcriptase activity either from indicator MT-4 lymphocytes after co-cultivation with infected trophoblasts or directly from trophoblast cultures. None of the tested trophoblast lines were permissive, in a detectable manner, to infection by cell-free virus. Furthermore, th

The effects of ICRF-154 in combination with other anticancer agents in vitro.

We studied the effects of ICRF-154 in combination with 11 anticancer agents on four human leukaemia cell lines. Cells were incubated for 3 days in the presence of two drugs (ICRF-154 and one other), and cell growth inhibition was determined by MTT assay. Effects of drug combinations at the ID50 level were analysed using the isobologram method (Steel). In the lymphoblastic leukaemia cell lines, MOLT-3, HSB, and B-ALL, supra-additive effects were observed for ICRF-154 in combination with amsacrine, bleomycin, doxorubicin, and etoposide. Additive effects were observed for its combinations with ci

Avian reproductive endocrinology.

Hypothalamic-releasing factors regulate the secretion of anterior pituitary hormones. The anterior pituitary gland secretes the same six hormones as found in mammals: FSH, LH, prolactin, GH (somatotropic hormone), ACTH, and TSH, plus the melanotropic hormone. The endocrine hormones of the avian posterior pituitary gland concerned with reproduction are mesotocin and AVT. The pineal gland, through the secretion of the hormone melatonin, modulates the periodic autonomic functions of the central nervous system. The ovary produces estrogens, progestogens, and androgenic compounds. The testes produc

Autumnal territorial aggression is independent of plasma testosterone in mockingbirds.

Mockingbirds (Mimus polyglottos) show intense territorial activity in the autumn as newcomers attempt to establish space within resident populations. Examination of autumnal territorial behavior showed that unmated males sing more and engage in more territorial fights than mated males. Newcomers that have just acquired space also sing more and show more territorial fights than birds resident to the population for at least one prior season. Among established residents, the average number of territorial fights was greater in birds that shared more territory boundaries with new residents. Radioim

The Hodgkin-associated Ki-1 antigen exists in an intracellular and a membrane-bound form.

The Hodgkin-associated Ki-1 antigen occurs in two different molecular forms. The 120-kDa membrane-associated form is a phosphorylated glycoprotein, which is derived from a non-phosphorylated intracellular 84-kDa apoprotein that is co-translationally N-glycosylated with a carbohydrate portion of 6 kDa. The other form of the Ki-1 antigen is a non-glycosylated phosphoprotein of 57 kDa which only occurs intracellularly. Both forms of the antigen are phosphorylated at serine residues. Enzymatic cleavage with sialidase reduced the 120-kDa membrane antigen by about 15 kDa, while its 90-kDa precursor

Expression of Thy-1 and effect of phosphatidylinositol-specific phospholipase C on primate and murine cell lines.

BALC/c mice were immunized with isolated human brain Thy-1. The antisera at an appropriate dilution only reacted with a doublet of an apparent molecular weight (MW) around 25,000 among all the glycoproteins of brain tissue isolated by lentil lectin affinity chromatography when tested by enzyme-linked immunosorbent assay and immunoblotting. When the antisera were used to test a number of human cell lines and a marmoset T-cell line (70N2) by flow cytometry, an astrocytoma cell line (U-373), a T-lymphoblastoid cell line (MOLT-3), and two cutaneous T-lymphoma cell lines (HUT-78 and HUT-102) as wel

A two-color flow cytometry assay for detection of hairy cells using monoclonal antibodies.

We have developed a simple two-color immunofluorescence assay equally suited for microscopy and flow cytometry detecting hairy cells (HCs) in single cell suspensions, based on the concomitant reactivities with the B cell-specific monoclonal antibody B1 (CD20) and the monocyte/HC-associated antibody SHCL-3 (CD11c). Thus, HCs can be demonstrated in peripheral blood, bone marrow, and spleen specimens from hairy cell leukemia (HCL) patients even when they constitute less than 1% of the cell suspension. Likewise, admixture experiments with normal mononuclear cells and the MOLT-4 T-acute lymphocytic

Budding by oozooids in the polystyelid ascidian Metandrocarpa tayori Huntsman.

Larvae of the stolidobranch ascidian Metandrocarpa taylori molt a thin sheath upon settling, then metamorphose and radiate a larval complement of vascular ampullae upon the substrate. These ampullae thereafter regress, "rest" in a reduced condition for several weeks, and then regrow into the oozooid's definitive array of vascular ampullae in accompaniment to the development of the oozooidal vascular nest of test-vessels. Pallial buds emerge some four months after the larva settles; the oozooid has by then grown to a length of at least 2 mm and its vascular nest is surrounded by at least 16 vas

The Emerging Role of Prolactin as a Biomarker for Reproduction, Health, and Welfare in Wildlife Species: Elephants as a Model.

Prolactin is a key hormone that plays significant roles in reproduction, extending far beyond its well-known function in mammalian lactation. It is vital for reproductive success across diverse taxa, including birds, fish, reptiles, and amphibians, where it regulates critical reproductive behaviors and physiological processes like parental care, pregnancy maintenance, embryonic diapause, immunity, osmoregulation, metamorphosis, and molting. Environmental cues often fine-tune its actions to ensure reproduction occurs under optimal conditions. Conversely, dysregulation of prolactin can severely …

Scaffold hopping approach to the novel hexacyclic pyrazol-3-amide derivatives as potential multi-target insect growth regulators candidates.

Ecdysone receptor (EcR) and three insect chitinases (OfChtI, OfChtII, and OfChi-h) are considered as attractive targets for the development of novel insect growth regulators (IGRs) since they are closely related to the insect molting. In this study, to develop potent multi-target IGRs, a series of hexacyclic pyrazol-3-amide derivatives were rationally designed by utilizing the scaffold hopping strategy with the previously reported compound 6j (N-(4-bromobenzyl)-2-phenyl-4,5,6,7-tetrahydro-2H-indazole-5-carboxamide) as a lead compound. The bioassay results indicated that most of the target comp…
