This chapter focuses on Epstein-Barr virus (EBV)-mediated tumorigenesis in nasopharyngeal carcinoma (NPC). EBV-mediated epithelial cell transformation is a crucial early event in the development of NPC. NPC and other EBV-associated lymphoepithelioma-like carcinoma have latency II program and a genomic landscape characterized by NF-κB pathway activating mutations, implicating the presence of viral latency program-specific oncogenesis. After the establishment of persistent EBV infection in nasopharyngeal epithelial cells, latency II viral genes (such as EBNA1, LMP1) are expressed to drive malign

Nasopharyngeal carcinoma (NPC) is a kind of malignant tumour originating from the mucosal epithelium of the nasopharynx, which has apparent regional distribution characteristics, and its incidence is increasing yearly. At present, the primary treatment method for nasopharyngeal carcinoma is radiotherapy, but radioresistance has become a complex problem to overcome to improve the therapeutic effect. Recently, ferroptosis has been proposed as a new type of iron-dependent programmed cell death, distinct from apoptosis, cell necrosis and autophagy. Many studies have shown that ferroptosis is invol

BACKGROUND: Cutaneous metastasis (CM) is an uncommon manifestation of internal malignancies, with nasopharyngeal carcinoma (NPC) as an exceptionally rare primary source. NPC, a subtype of head and neck carcinoma, demonstrates geographic variability and aggressive behavior, including potential for distant metastasis. Despite the rarity of cutaneous dissemination from NPC, accurate diagnosis requires comprehensive clinical evaluation supported by histopathological and immunohistochemical assessment. CASE PRESENTATION: A 59-year-old male with previously treated non-keratinizing squamous cell carc

BACKGROUND: Nasopharyngeal cancer (NPC) patients experience significant anatomical changes during radiation treatment (RT). Adaptive radiotherapy (ART) can be initiated in response to specific events or at scheduled intervals during treatment, prompting questions about patient selection, timing, and reliable triggers for re-CT and replanning. This study aimed to develop a method for selecting NPC patients for ART before treatment and identify the optimal timing for its implementation.
MATERIALS AND METHODS: We retrospectively evaluated NPC patients treated at University Hospitals Leuven betwe

BACKGROUND: The therapeutic benefit of concurrent chemoradiotherapy (CCRT) in elderly nasopharyngeal carcinoma (NPC) patients remains controversial. This study aimed to investigate the efficacy and toxicity of lobaplatin-based CCRT in elderly patients with NPC.
METHODS: We included stage II-IVA NPC patients aged ≥65 years who received lobaplatin concomitant with intensity-modulated radiation therapy (IMRT) between March 2019 and January 2023. Objective response rates and treatment-related toxicity were assessed. Kaplan-Meier's analysis was performed to calculate survival rates.
RESULTS: A to

Nasopharyngeal carcinoma (NPC) is a malignant epithelial tumor that develops in the nasopharynx. It has a distinct ethnic and geographical distribution, and emerging evidence suggests that it is an ecological disease. Most patients respond well to radiation combined with chemotherapy as the primary treatment for NPC. However, some patients will eventually develop radio resistance and chemoresistance, resulting in recurrence and metastasis, which is a primary cause of poor prognosis. The processes underlying radio resistance and chemoresistance in NPC are complex and unknown. MicroRNAs (miRNAs)

The nasopharyngeal tract contains a complex microbial community essential to maintaining host homeostasis. Recent studies have shown that SARS-CoV-2 infection changes the microbial composition of the nasopharynx. Still, little is known about how it affects the fungal microbiome, which could provide valuable insights into disease pathogenesis. Nasopharyngeal swabs were collected from 55 patients, during three distinct COVID-19 waves that occurred in the Campania Region (southern Italy). An RNA-seq-based analysis was performed to evaluate changes in mycobiota diversity, showing variations depend

Nasopharyngeal carcinoma (NPC) is characterised by its remarkable geographical and ethnic distribution. The interplay between genetic susceptibility, environmental exposures, and Epstein-Barr virus (EBV) infections is indicated in the development of NPC. Exposure to tobacco smoking, dietary factors, and inhalants has been associated with the risk of NPC. Genetic association studies have revealed NPC-associated susceptibility loci, including genes involved in immune responses, xenobiotic metabolism, genome maintenance, and cell cycle regulation. EBV exposure timing and strain variation might pl

Juvenile nasopharyngeal angiofibroma (JNA) is a rare, locally aggressive, highly vascularized benign tumor classically found in males from 13 to 20 years old. The most common clinical presentations are recurrent epistaxis and nasal congestion. Imaging plays a significant role in differentiating and determining the extension of nasopharyngeal lesions. The case below highlights a 15-year-old male that presented with nasal congestion and recurrent epistaxis, who underwent nasal endoscopy and ablation. Persisting symptoms led to further evaluation by computed tomography and magnetic resonance imag

Nasopharyngeal carcinoma (NPC) originates from the nasopharynx epithelium, and luteolin is recognized as an important anti-cancer agent. This study investigated the effects of luteolin on ferroptosis in NPC cells. NPC cells were cultured and exposed to varying concentrations of luteolin. Cell viability, malondialdehyde (MDA) levels, superoxide dismutase (SOD) activity, glutathione (GSH) levels, Fe2+ concentration, and glutathione peroxidase 4 (GPX4) protein level were assessed. Additionally, SRY-related high-mobility-group box 4 (SOX4) expression was measured. Subsequently, the binding of SOX4

Nasopharyngeal middle turbinate is an extremely rare anatomical variant which has been suggested to correlate with chronic rhinosinusitis. However, due the rarity of this variant, its clinical implications remain inadequately defined. Here, we present a case of a 37-year-old male patient with bilateral nasopharyngeal middle turbinates, one of which had a large cystic lesion causing nasal obstruction. The patient's presentation, radiological and endoscopic findings, as well as the management approach are discussed in the context of the limited literature on this topic.

Nasopharyngeal carcinoma (NPC) is a prevalent malignant tumor of the head and neck, with approximately 70 % of patients being diagnosed at a locally advanced stage. Despite the responsiveness to radiotherapy and chemotherapy, the 5-year survival rate of locally advanced NPC (LANPC) remains at approximately 80 %. Hence, there is an urgent need for novel treatment strategies to improve the prognosis of patients with LANPC. Numerous studies have illustrated the efficacy of immune checkpoint inhibitors (ICIs) in recurrent/metastatic NPC. Hence, the potential of immunotherapy for LANPC is under inv

Nasopharyngeal cancer is a relatively rare cancer type, it is most common in southeast Asia and north Africa, it can be presented with nonspecific symptoms, making it challenging to diagnose. However, it remains a significant cancer to diagnose and treat early, as it can be aggressive and difficult to manage in its advanced stages. We report the case of a 48-year-old man who presented with isolated neck swelling, which was later found to be caused by multiple lymphadenopathies and suspected nasopharyngeal neoplasm. Imaging confirmed a large mass in the nasopharynx and bilateral cervical adenop

CASE SUMMARY: An 11-month-old female domestic shorthair cat presented with a 24 h history of inspiratory dyspnoea, abnormal upper respiratory tract sounds, gagging, retching and making exaggerated swallowing motions. Retroflexed nasopharyngoscopy revealed a large, right-sided nasopharyngeal mass that was seen to exude purulent material and a possible small foreign body when pressure was applied with forceps. Thorough expression of the mass alleviated clinical signs. Cytology revealed septic neutrophilic inflammation, and a Pasteurella species with no noted antimicrobial resistance was cultured
