BACKGROUND AND PURPOSE: Although subcortical vascular dementia, the major subtype of vascular dementia, is caused by a disruption in white matter integrity after cerebrovascular insufficiency, no therapy has been discovered that will restore cerebral perfusion or functional cerebral vessels. Because adrenomedullin (AM) has been shown to be angiogenic and vasoprotective, the purpose of the study was to investigate whether AM may be used as a putative treatment for subcortical vascular dementia.
METHODS: A model of subcortical vascular dementia was reproduced in mice by placing microcoils bilat

Platelet factor-4 (PF4), a platelet-derived chemokine, has two important functions in the vasculature. It has a pro-atherogenic role while also having anti-angiogenic effects. The activity of platelet factor-4 (PF4), unlike other chemokines that bind to specific receptors, depends on its unusually high affinity for proteoglycans and other negatively charged molecules. High affinity for heparan sulfates was thought to be central to all of PF4's biological functions. However, other mechanisms have been described such as direct growth factor binding, activation of the CXCR3B chemokine receptor is

Cell-based therapy has emerged as a promising therapeutic tool for treatment of ischemic cardiovascular disease. Both unselected bone marrow-derived mononuclear cells (BMNCs), which include stem/progenitor cells and several other cell types, and endothelial progenitor cells (EPCs), a subpopulation of BMNCs, display regenerative potential in ischemic tissue. Abundant evidence supports the involvement of EPCs in capillary growth, and EPCs also appear to participate in the formation of collateral vessels. Collectively, these effects have led to improved perfusion and functional recovery in animal

Vasculopathy in patients with connective tissue diseases (CTDs), including systemic sclerosis (SSc) and systemic lupus erythematosus (SLE), is a serious complication that mainly affects small arteries and capillaries, reduces the blood flow and causes progressive tissue ischemia. Recently, CTD patients have been reported to have abnormalities in circulating endothelial progenitor cells (EPCs); these abnormalities are believed to contribute to the pathophysiology of vasculopathy and to the premature and accelerated development of atherosclerosis in CTD patients. Furthermore, we are currently co

INTRODUCTION: Rheumatoid Disease (RD) is associated with increased rates of cardiovascular disease (CVD). Angiogenesis is central to RD, and well-recognized in CVD. We hypothesised that plasma levels of two indices associated with angiogenesis, vascular endothelial growth factor (VEGF) and angiogenin, would be higher among RD patients compared to healthy controls (HC), would relate to CVD risk factors, calculated 10-year coronary heart disease (CHD) and stroke risk scores.
METHODS: 144 clinic patients with established RD and 63 HC were recruited in a cross-sectional study. RD patients were gr

Systemic sclerosis (SSc, scleroderma) is a connective tissue disease of unknown etiology. Perivascular inflammatory infiltrates and endothelial apoptosis with an impaired angiogenesis are observed in early stages of the disease, whereas later stages are characterized by an excessive accumulation of extracellular matrix proteins in the skin and various internal organs. Consistent with the ongoing endothelial cell damage, various markers of endothelial cells such as endothelin-1, sICAM-1, s-VCAM-1 and thrombomodulin are found in high levels in the serum of SSc patients. Surprisingly, the vascula

Angiogenesis or the formation of new vessels out of pre-existing capillaries is a sequence of events that is fundamental to physiology of the female reproductive tract and also pathologic processes such as ovarian hyperstimulation syndrome. Many factors include vascular endothelial growth factor (VEGF), angiopoietins and others involved in the regulation of angiogenesis have been identified. There are some endocrine control mechanisms, which stimulate or inhibit the angiogenesis. The studies indicate that the normal processes of folliculogenesis, ovulation and corpus luteum function in the ova

OBJECTIVE: Our previous work demonstrated that angiogenesis is inhibited in nonhealing venous ulcers. The object of this study was to determine whether local expression of vascular endothelial growth factor (VEGF) and other major regulators of vessel growth are related to healing of venous ulcers.
SUBJECTS AND METHODS: The study included 35 patients with venous ulcers (CEAP 6) and 9 patients whose ulcers had healed (CEAP 5). Control subjects were 18 patients undergoing routine operations (8 with closed suction drains, 10 standard skin biopsies). Healing ulcers were defined as having healed in

Within the past few years, transmyocardial laser revascularization (TMR) has attracted the attention of cardiologists and cardiac surgeons as a therapy for patients suffering from otherwise treatable coronary artery disease. Clinical studies have consistently shown symptomatic improvement that lasts at least 1 year in a majority of patients. The original hypothesis that prompted development of the technique was that direct myocardial perfusion from the chamber could be achieved through chronically patent channels, as is the case in reptilian hearts. Results of our early studies failed to suppo

Transforming growth factor-beta (TGF-beta) has profound effects on all cell types making up the vasculature, including endothelial cells, smooth muscle cells, and adventitial connective tissue. As such, it plays a prominent role not only in the physiologic vasculogenesis and angiogenesis characteristic of embryogenesis and inflammation and repair but also in vascular disorders such as the arterial thickening associated with pulmonary hypertension. The actions of TGF-beta on these vascular cells in vitro and in vivo are extremely complex. In vitro, TGF-beta inhibits both the proliferation and m

Choroidal neovascularization (CNV) is the pathological growth of new blood vessels in the sub-retinal pigment epithelial (RPE) space from the choroid through a break in the Bruch's membrane (BM). Despite its importance in studying biological processes and drug discovery, the development of an in vitro CNV model that achieves the physiological structures of native RPE-BM-choroidal capillaries (CC) is still challenging. Here, we develop a novel 3D RPE-BM-CC complex biomimetic system on an ultra-thin, free-standing nanofiber membrane. The thickness of the pristine nanofiber membrane is 2.17 ± 0.8

Apelin is an endogenous ligand of seven-transmembrane G-protein-coupled receptor APJ. Apelin and APJ are distributed in various tissues, including the heart, lung, liver, kidney, and gastrointestinal tract and even in tumor tissues. Studies show that apelin messenger RNA is widely expressed in gastrointestinal (GI) tissues, including stomach and small intestine, which is closely correlated with GI function. Thus, the apelin/APJ system may exert a broad range of activities in the digestive system. In this paper, we review the role of the apelin/APJ system in the digestive system in physiologica

Connective tissue growth factor (CTGF) is a member of the recently described CCN gene family which contains CTGF itself, cyr61, nov, elm1, Cop1, and WISP-3. CTGF is transcriptionally activated by several factors although its stimulation by transforming growth factor beta (TGF-beta) has attracted considerable attention. CTGF acts to promote fibroblast proliferation, migration, adhesion, and extracellular matrix formation, and its overproduction is proposed to play a major role in pathways that lead to fibrosis, especially those that are TGF-beta-dependent. This includes fibrosis of major organs

In the retinal pigment epithelium (RPE) several factors within the macular compared to peripheral regions cause differences in physiological aging. The molecular mechanisms during aging in the context of topography are not well known. The proteome of RPE of different aged macular-bearing primates Callithrix jacchus was thus analysed with ion mobility mass spectrometry. Macular and periphery of neonate RPE were well differentiated from aged tissues as demonstrated by principal component analysis. This finding was mainly due to proteins involved in major developmental processes and the visual cy

Disulfiram (DSF), which has been traditionally used to treat alcoholism, has been shown to inhibit tumor growth, indicating its potential as an anticancer agent. However, its development and application are hindered by its poor water solubility, instability in physiological environments, and low bioavailability. In this study, phenylboronic acid-chitosan (PBA-CS) grafts were synthesized using the carbodiimide method. PBA-CS-modified DSF PLGA nanoparticles (DSF@PBA-CS-PLGA NPs) were constructed by coating the nanoparticle surfaces with PBA-CS to improve the stability of DSF in physiological env

Porous biomaterials that integrate tunable biophysical and biochemical cues have been extensively studied for guiding cell behavior and harnessing the body's intrinsic regenerative potential. Aerogels, characterized by their ultralight structure, high porosity, and large surface area, have emerged as promising porous scaffolds for tissue engineering; however, their limited pore tunability may hinder efficient cell infiltration and functional tissue integration. To address this persistent limitation, we develop a new class of porous biomaterials called granular aerogel scaffolds (GAS), assemble

Mesenchymal stromal cell-derived artificial microvesicles (MSC-MVs) hold significant promise as a cell-free alternative to traditional stem cell therapy for the treatment of lower limb ischemia. However, their fragile plasma membrane is highly susceptible to oxidative damage, environmental fluctuations, and long-term storage, often leading to membrane rupture, vesicle disintegration, and leakage of bioactive cargoes. Additionally, MSC-MVs can be contaminated by nuclear genes, limiting their safety and therapeutic applicability. In this study, we developed gelated microvesicles (gel-MVs) derive

Angiogenesis is essential not only during embryogenesis but also in adult physiology and disease, relying on vascular endothelial growth factor (VEGF) gradients to guide tip cell sprouting. Cellular prion protein (PrPC), expressed in several neurovascular unit (NVU) cell types, regulates neuronal and astrocytic differentiation through laminin binding, yet its role in angiogenesis remains understudied. Here, we investigated its function in retinal vascular development. In PrPC knockout mice, the superficial retinal plexus exhibited increased tip cell density and branching, accompanied by microg

Angiogenesis is a multistep biological process involving endothelial cell activation, migration, elongation, lumen formation, branching, and anastomosis. Conventional two-dimensional culture systems are useful for the preliminary evaluation of drugs, genes, or culture conditions, but they do not adequately recapitulate the spatial organization of endothelial morphogenesis within a three-dimensional extracellular matrix. The present protocol describes a microcarrier bead-based three-dimensional angiogenesis assay using human umbilical vein endothelial cells (HUVECs). The HUVECs are first seeded
