Isotopic examination of links between diet, social differentiation, and DISH at the post-medieval Carmelite Friary of Aalst, Belgium.

Stable isotope ratios (δ(13) C and δ(15) N) were measured in human burials from the post-medieval (16th-18th c. AD) Carmelite friary burial grounds at Aalst, a town in Flanders, Belgium. Dietary patterns of 39 adult individuals were analyzed, from a mixed monastic and lay population buried in three different locations, reflecting groups with differing social status. The data show significant variation in the consumption of perhaps meat, but certainly also marine protein between females and males. This result represents a remarkable continuity with medieval dietary patterns, suggesting that the

The kinetics of intramolecular distribution of 15N in uric acid after administration of (15N) glycine. A reappraisal of the significance of preferential labeling of N-(3+9) of uric acid in primary gout.

THE CONCEPT OF AN ABNORMALITY OF GLUTAMINE METABOLISM IN PRIMARY GOUT WAS FIRST PROPOSED ON THE BASIS OF ISOTOPE DATA: when [(15)N]glycine was administered to gouty subjects, there was disproportionately great enrichment of N-(3 + 9) of uric acid, which derive from the amide-N of glutamine. An unduly high concentration of (15)N in glutamine was postulated, and attributed to a hypothetical defect in catabolism of glutamine. Excess glutamine was proposed as the driving force of uric acid overproduction. WE HAVE REEXAMINED THIS PROPOSITION IN FOUR GOUTY SUBJECTS: one mild overproducer of uric aci

Protein-energy status and 15N-glycine kinetic study of child a cirrhotic patients fed low- to high-protein energy diets.

In five male cirrhotic patients (Child A) and in four age- and sex-matched healthy control subjects, whole-body protein turnover was measured using a single oral dose of 15N-glycine as a tracer and urinary ammonia as end product. Subjects were studied in the fasting and feeding state, with different levels of protein and energy intake. The patients were underweight and presented lower plasma transthyretin and retinol-binding protein levels. When compared with controls, the kinetic studies showed patients to be hypometabolic in the fasting (D0) state and with the control diet [D1 = (0.85 g of p

Synthesis of ¹⁸F-labelled β-lactams by using the Kinugasa reaction.

Owing to their broad spectrum of biological activities and low toxicity, β-lactams are attractive lead structures for the design of novel molecular probes. However, the synthesis of positron emission tomography (PET)-isotope-labelled β-lactams has not yet been reported. Herein, we describe the simple preparation of radiofluorinated β-lactams by using the fast Kinugasa reaction between (18)F-labelled nitrone [(18)F]-1 and alkynes of different reactivity. Additionally, (18)F-labelled fused β-lactams were obtained through the reaction of a cyclic nitrone 7 with radiofluorinated alkynes [(18)F]-6

Decreased whole body endogenous nitric oxide production in patients with primary pulmonary hypertension.

Impaired pulmonary release of nitric oxide (NO) is one of the characteristic phenotypic changes of vascular cells in pulmonary hypertension. The aim of this study was to determine nitric oxide synthase (NOS)-dependent whole body NO production in patients with primary pulmonary hypertension. NOS-dependent whole body NO production was assessed by giving an intravenous infusion of L-[(15)N](2)-arginine (50 micromol/min for 30 min) and measuring isotopic urinary enrichment of (15)N-nitrite and (15)N-nitrate. Four female patients with no signs of infection were recruited and compared with 6 age-mat

Whole body and forearm substrate metabolism in hyperthyroidism: evidence of increased basal muscle protein breakdown.

Thyroid hormones have significant metabolic effects, and muscle wasting and weakness are prominent clinical features of chronic hyperthyroidism. To assess the underlying mechanisms, we examined seven hyperthyroid women with Graves' disease before (Ht) and after (Eut) medical treatment and seven control subjects (Ctr). All subjects underwent a 3-h study in the postabsorptive state. After regional catheterization, protein dynamics of the whole body and of the forearm muscles were measured by amino acid tracer dilution technique using [15N]phenylalanine and [2H4]tyrosine. Before treatment, triiod

The high-resolution structure of the triplex formed by the GAA/TTC triplet repeat associated with Friedreich's ataxia.

Expansions of the triplet repeat, GAA/TTC, inside the first intron of the frataxin gene causes Friedreich's ataxia (FRDA). It was of interest to us to examine whether the FRDA repeat forms an unusual DNA structure, since formation of such structure during replication may cause its expansion. Here, we show that the FRDA repeat forms a triplex in which the TTC strand folds on either side of the same GAA strand. We have determined the high-resolution NMR structures of two intramolecularly folded FRDA triplexes, (GAA)2T4(TTC)2T4(CTT)2 and (GAA)2T4(TTC)2T2CT2(CTT)2 with T.A.T and C+.G.C triads. T4

Acid-base regulation of hepatic glutamine metabolism and ureagenesis: study with 15N.

The metabolism of (5-15N)glutamine and (2-15N) glutamine has been studied by isolated hepatocytes obtained from either control, chronically acidotic, or alkalotic rats. The main goal was to elucidate the mechanism(s) by which altered acid-base state affects hepatic ureagenesis from glutamine. Isolated hepatocytes were incubated in Krebs buffer (pH 7.4) supplemented with 0.1 mM ornithine plus either 1 mM (5-15N)glutamine or (2-15N)glutamine. To elucidate the role of glutamine cycling in net ammonia metabolism, a separate series of experiments were performed with 1 mM unlabeled glutamine plus 1

Brain glutamate metabolism during metabolic alkalosis and acidosis.

Glutamate modifies ventilation by altering neural excitability centrally. Metabolic acid-base perturbations may also alter cerebral glutamate metabolism locally and thus affect ventilation. Therefore, the effect of metabolic acid-base perturbations on central nervous system glutamate metabolism was studied in pentobarbital-anesthetized dogs under normal acid-base conditions and during isocapnic metabolic alkalosis and acidosis. Cerebrospinal fluid transfer rates of radiotracer [13N]ammonia and of [13N]glutamine synthesized de novo via the reaction glutamate+NH3-->glutamine in brain glia were m

Challenges and Methodologies to Assess Protein Requirement and Quality Across Different Life Stages in Dogs: A Review.

Determining protein requirements (PRs) for dogs remains a longstanding challenge. During growth, the rapid rate of protein deposition increases the demand for amino acids. In adult dogs, differences in overall diet digestibility and lower energy requirements of domestic dogs have led to discrepancies between the minimum crude protein (CP) value proposed by the National Research Council (NRC; 80 g of CP/kg of diet) and the 180 g of CP/kg of diet proposed by the European Pet Food Industry Federation (FEDIAF) and the Association of American Feed Control Officials (AAFCO), although most commercial

Pharmaceutical pollution in a wastewater-impacted coastal ecosystem.

As global demand for pharmaceuticals continues to grow, pharmaceutical pollution is becoming increasingly pervasive. Research on the occurrence and distribution of pharmaceuticals in marine ecosystems remains limited, however, with coastal sewage outfalls considered to be a major source of pharmaceutical pollutants in marine environments. Here, we investigated the distribution of pharmaceuticals across a wastewater gradient in Port Phillip Bay, Melbourne, Australia, using stable nitrogen isotope ratios (δ15N) as a signal of wastewater influence. Marine invertebrates, macroalgae and marine/terr

Interfacial Confinement-Programmed Hydrogen Spillover on Ag/CoNiS Boosts Nitrate-to-Ammonia Electrosynthesis.

Electrochemical nitrate reduction (NO3RR) under ambient conditions offers a sustainable route for ammonia (NH3) synthesis; however, its efficiency is restricted by the kinetic mismatch between water dissociation and nitrate hydrogenation. Here, we design Ag/CoNiS heterostructures in which Ag loading density programs interfacial confinement to regulate hydrogen spillover from CoNiS water-activation domains to Ag-associated nitrate/nitrogen oxide (NOx) intermediates, thereby coupling *H generation, relay, and deep nitrate hydrogenation. The optimized AgM/CoNiS achieves an NH3 yield of 22.31 mg h

Biocrusts contribute to shrub nitrogen nutrition via soil transfer in resource-limited drylands.

Biocrusts and xeric shrubs commonly coexist in drylands, forming shrub-biocrust patches that shape soil processes and ecosystem functioning. While biocrusts are important sources of newly fixed nitrogen (N) and carbon (C), it remains unclear how C and N move between biocrusts and shrubs, whether transfer is bidirectional or asymmetric, and which pathways mediate exchange. Using whole-atmosphere isotope pulse labeling combined with experimental manipulation of hyphal access in a semiarid shrubland, we traced biocrust-fixed N (15N2), biocrust-fixed C (13CO2), and shrub-fixed C (13CO2). Biocrust-

Unraveling the Bimetallic Synergy: A Unified Framework for Synthesis, Dynamic Evolution, and Multi-Electron Electrocatalysis.

Bimetallic electrocatalysts provide additional geometric and electronic degrees of freedom for regulating multi-electron reactions and can relax the scaling constraints encountered at monometallic sites in favorable cases. However, rational development of such catalysts remains challenging due to the mismatch between static as-synthesized structures and dynamically evolving active sites under operating conditions. This Review presents an integrated framework connecting synthesis, structural evolution, characterization, and multi-electron electrocatalytic behaviors. Representative bimetallic sy

Pseudomonas vancouverensis as a platform for one-carbon (C1) assimilation and polyhydroxyalkanoate accumulation.

Metabolic engineering calls for new bacterial chassis that combine robust metabolic and physiological traits with genetic tractability. We developed Pseudomonas vancouverensis DhA-51, a native one-carbon (C1)-trophic strain, into a platform that broadens chassis options beyond the usual Pseudomonas hosts while retaining native capacities relevant to bioproduction. We closed and annotated the chromosome to provide stable coordinates for genome editing and omics, established antibiotic susceptibility profiles, and explored replication of common broad-host-range plasmid toolsets. An expression to

Sediment capping causes metabolic stress and hydrogen sulfide intrusion in Posidonia australis: Implications for seagrass restoration.

Anthropogenic pollution in developed coastal areas often causes widespread seagrass loss. In Cockburn Sound, Western Australia, industrial run-off drastically reduced cover of Posidonia spp. by 77% in the 1960s-1990s. Despite significant water quality improvements, natural recovery remains limited, potentially due to legacy pollution and phytotoxic hydrogen sulfide (H2S) production in sediments. Using a novel multidisciplinary approach combining metabolomics, nutrient (carbon, nitrogen, phosphorus), and δ34S isotope analysis, we assessed whether capping existing sediment with clean, dredged ma

Atmospheric CH4 plateau sustained by NOx emission rise and southward shift.

Methane exhibits decadal variability in atmospheric growth rates1. During the 1990s, atmospheric methane growth slowed relative to the 1980s, largely attributed to the stabilization or decline in anthropogenic emissions2-7. By contrast, economic expansion in the early 2000s (ref. 8) led to an increase in anthropogenic methane emissions9. The anticipated rise in atmospheric methane concentrations, however, was not detected by global monitoring networks1. Instead, almost no atmospheric methane growth was observed between 1999 and 2006 (ref. 1). Several hypotheses have been proposed to explain th

The Shapes of Sulfonamides: A Rotational Spectroscopy Study.

Benzenesulfonamides are a class of molecules of extreme interest in the biochemical field because many of them are active against a variety of diseases. In this work, the pharmacophoric group benzensulfonamide, its derivatives para-toluensulfonamide and ortho-toluensulfonamide, and the bioactive molecule sulfanilamide, were investigated using rotational spectroscopy to determine their conformations and the influence of different substituents on their structures. For all species, the hyperfine structure due to the 14N atom was analyzed, and this provided crucial information for the unambiguous

Anaerobic ammonium oxidation is a major N-sink in aquifer systems around the world.

Global-scale N-oxide contamination of groundwater within aquifers occurs due to the widespread use of N-bearing fertilizers and chemicals, threatening both human and environmental health. However, the conversion of these pollutants in active nitrogen (N) cycling processes in the subsurface biosphere still remains unclear. This study investigates the global occurrence of anaerobic ammonium oxidation (anammox) in aquifers, where anammox was found to be turned on and off between saturated and unsaturated soil horizons, and contributed 36.8-79.5% to N loss in saturated soil horizons, the remainder
