Genes affecting tooth morphogenesis.

The development of dentition is a fascinating process that encompasses a complex series of epithelial-mesenchymal interactions involving growth factors, transcription factors, signal receptors and other soluble morphogens. It is not surprising that such a complex process is prone to disturbances and may result in tooth agenesis. Initial discoveries indicating that the homeo-domain protein MSX1 and the paired-domain transcription factor PAX9 are causative genes in tooth morphogenesis were made in mice. Both genes are co-expressed in dental mesenchyme and either one, when homozygously deleted, r

Cellular expression of eve1 suggests its requirement for the differentiation of the ameloblasts and for the initiation and morphogenesis of the first tooth in the zebrafish (Danio rerio).

even-skipped-related (evx) genes encode homeodomain-containing transcription factors that are involved in a series of developmental processes such as posterior body patterning and neurodifferentiation. Although evx1 and evx2 were not reported to be expressed during mammalian tooth development, we present here evidence that eve1, the closest paralog of evx1 in the actinopterygian lineage, is expressed during pharyngeal tooth formation in the zebrafish, Danio rerio. We have performed whole-mount in situ hybridization on zebrafish embryos and larvae ranging from 24 to 192 hours postfertilization

Is tooth agenesis related to brainstem anomalies in myelomeningocele patients with Chiari II malformations?

Defects in the prenatal development of the brainstem can result in cranial nerve deficiencies. As the development of tooth germ is dependent on n. trigeminus, which originates from the brainstem, the hypothesis underlying this study was that anomalies of the brainstem would lead to an increased prevalence of tooth agenesis. Twenty-three patients (13 F and 10 M, age range 6-37 years) were studied, all with myclomeningocele and brainstem anomalies (Chiari II). They were examined retrospectively from the data in journals and dental radiographs and compared to available data on the prevalence of t

Relationship of third molar development and root angulation.

Angled roots are seen in mandibular third molars, which have a high frequency of incomplete impaction. We examined the relationship between incomplete impaction and angled roots. We enrolled orthopantomographs to determine the prevalence of angled roots in 239 men and 222 women aged 21--35 years with bilateral mandibular third molars. Angled roots were more frequent in subjects in whom the third molars had a different status on each side than in those with the same status on both sides (men: P<0.05; women: P<0.01). The incidence of women with angled roots in those with bilateral incomplete imp

The effects of local trauma to the enamel-related periodontal tissues in the eruption of the rat incisor.

The periodontal tissues related to enamel (PTE) of the rat incisor comprise a connective tissue derived from the dental follicle and the enamel organ with its successive stages of development. Localized damage to these tissues in rat lower incisors was done surgically in three ways: with an endodontic file introduced into the labial periodontal space through either (i) its basal or (ii) its incisal extremities, or (iii) by the partial removal of the mandibular lower border, at the level of the molar teeth, together with the introduction of an endodontic file into the incisal part of that space

Prenatal traces of aberrant neurofacial growth.

The interrelation between the development of the brain/peripheral nerves and that of the surrounding bone tissue is termed neuro-osteology. In orthodontic and pediatric practice the development of the hard tissues is evaluated radiographically, but the development of the neural tissue within the bone tissue is not evaluated. In this review the emphasis is placed on two neuro-osteologic interrelations that can be observed on profile radiographs and orthopantomograms, respectively. One is the connection between the pituitary gland of the central nervous system and the sella turcica (profile radi

Meningococcal septicemia and disseminated intravascular coagulation affecting the premaxillary permanent tooth germs.

The following case describes the dental effects resulting from a case of meningococcal septicemia which caused a disseminated intravascular coagulation and premaxillary osteomyelitis at age two years. The effects went unnoticed for eight years when delayed development of the maxillary incisors was noted. Treatment involved surgical removal of the dental remnants and provision of a removable partial denture. Implants and ridge augmentation will be considered in early adulthood.

Dental development and molar root length in children with cleidocranial dysplasia.

Cleidocranial dysplasia (CD) is an autosomal dominant skeletal disorder with characteristic dental findings of numerous supernumerary teeth and noneruption of permanent teeth. This investigation compared the dental development and root lengths of the mandibular first permanent molar in 11 CD patients with those of 22 healthy, normal children matched for race, age and sex. The results showed that children with CD experienced a delay in dental development of approximately 3 years compared with normal children (P < 0.05). In addition, the root lengths of the mandibular first permanent molar were

Assessing the eruption of lower third molars on the basis of radiographic features.

To determine radiographic features by which one could estimate whether lower third molars are likely to erupt after the age of 20 years, university students with a total of 84 unerupted or partially erupted lower third molars were followed up. Five radiographic features of each third molar were measured from the panoramic tomogram taken at age 20 years, and the status of third molars was examined for a second time at age 26 years. The results showed that the lower third molars that did erupt after the age of 20 years were initially: 1) root formation complete; 2) impacted in soft tissue; 3) ve

Radiographic and histological analysis of tooth eruption through calcium phosphate ceramics in the cat.

The effect of implanting calcium phosphate ceramics (CPC) into metabolically active sites within kitten mandibles during permanent premolar tooth eruption was examined. Forty kittens, 3-4 months of age were used: the deciduous second and third mandibular premolars were extracted and their sockets implanted with autologous blood clot, autogenous cancellous marrow, and the calcium phosphate ceramics, non-porous beta-tricalcium phosphate or porous hydroxylapatite. Animals were killed at 1, 2, 3, 4 and 5 months after implantation and undermineralized sagittal sections were evaluated by light micro

Interrelationships among measures of somatic, skeletal, dental, and sexual maturity.

The interrelationships among five measures of physiologic maturity for 50 French-Canadian girls are evaluated--menarche, peak height velocity (PHV), 75% skeletal maturity, appearance of the ulnar sesamoid, and 90% dental development. The mean ages of occurrence of the events differed significantly (P less than 0.01). Menarche and 90% dental maturity showed the least variability. Ages of PHV, menarche, and 75% skeletal maturity were significantly correlated (P less than 0.05). Age of menarche was most closely associated with PHV. The appearance of the ulnar sesamoid was highly correlated with 7

Nutrition in growth and development of oral tissues.

The development of teeth, salivary glands, and other oral tissues can be seriously compromised by a deficiency of a number of essential nutrients. Because nutritional stress during critical development periods may decrease the ultimate resistance of the oral tissues to challenge, the prevention of disease and maintenance of oral health are intimately dependent on the nutrient supply during development. Appropriate dietary counseling, particularly with reference to critical developmental times and intake of essential nutrients, can insure optimal oral tissue development.

Injectable laponite nanocomposite hydrogel with synergistic antibacterial and odontogenic activity for endodontic regeneration.

Persistent pulpitis often leads to irreversible dentin defects and pulp necrosis, posing significant challenges for functional dental restoration. To address the critical limitation of insufficient odontogenic differentiation in dental pulp stem cells (DPSCs), we develop an injectable photo-crosslinking methacrylated hyaluronic acid hydrogel (PLSr2+@HAMA) containing laponite-based ternary nanocomposites which self-assemble by laponite, strontium ions (Sr2+), and antibacterial peptide P-113. We demonstrate that PLSr2+@HAMA features three distinctive advantages: (1) Unique injectability enabling

Miniaturization: How many cells are needed to build a tooth?

BACKGROUND: Organs that develop early in life, and are replaced by a larger version as the animal grows, often represent a miniature version of the adult organ. Teeth constituting the first functional dentition in small-sized teleost fish, such as medaka (Oryzias latipes), are examples of such miniature organs. With a dentin cone as small as the size of one human cell, or even smaller, these teeth raise the question how many dentin-producing cells (odontoblasts) are required to build such a tooth, and whether this number can be as little as one. RESULTS: Based on detailed observations with tr

DPP promotes odontogenic differentiation of DPSCs through NF-κB signaling.

Dentin phosphophoryn synthesized and processed predominantly by the odontoblasts, functions as both structural and signaling protein. Mechanistic studies revealed that DPP stimulation of DPSCs positively impacted the differentiation of DPSCs into functional odontoblasts. Results show that NF-κB signaling and transcriptional activation of genes involved in odontoblast differentiation were influenced by DPP signaling. Specifically, RelA/p65 subunit of NF-κB was identified as being responsible for the initiation of the differentiation cascade. Confocal imaging demonstrated the nuclear translocati

Role of the Inferior Alveolar Nerve in Rodent Lower Incisor Stem Cells.

In developing teeth, the sequential and reciprocal interactions between epithelial and mesenchymal tissues promote stem/progenitor cell differentiation. However, the origin of the stem/progenitor cells has been the subject of considerable debate. According to recent studies, mesenchymal stem cells originate from periarterial cells and are regulated by neurons in various organs. The present study examined the role of innervation in tooth development and rodent incisor stem/progenitor cell homeostasis. Rodent incisors continuously grow throughout their lives, and the lower incisors are innervate

Mesenchymal Wnt/β-catenin signaling limits tooth number.

Tooth agenesis is one of the predominant developmental anomalies in humans, usually affecting the permanent dentition generated by sequential tooth formation and, in most cases, caused by mutations perturbing epithelial Wnt/β-catenin signaling. In addition, loss-of-function mutations in the Wnt feedback inhibitor AXIN2 lead to human tooth agenesis. We have investigated the functions of Wnt/β-catenin signaling during sequential formation of molar teeth using mouse models. Continuous initiation of new teeth, which is observed after genetic activation of Wnt/β-catenin signaling in the oral epithe
