Congenital Anomalies of the Nose: From Embryologic to Surgical Treatment.

The nose, a facial organ, not only plays a crucial role in olfaction and respiration but also has a major impact on the overall anatomy of the face. However, congenital anomalies affecting the nose can be challenging as they require both functional impairments and cosmetic concerns. Facial maldevelopments during the early embryonic period induce a wide-ranging nasal deformity. Understanding the etiology of these anomalies is essential for healthcare professionals involved in diagnosis, treatment, and management. Based on current literature, this review aims to present the spectrum of congenita

Establishment and characterization of induced pluripotent stem cells from placental mesenchymal stromal cells.

Human term placenta is a bulky organ which harbours abundant mesenchymal stromal cells. This study reports isolation and characterization of placental mesenchymal stromal cells (PMSCs) followed by reprogramming of PMSCs to induced pluripotent stem cells (iPSCs). The placental human iPSC (PhiPSC) line is pluripotent with high telomerase activity, genetically identical to parental PMSCs, transgene free, karyotypically normal and differentiates into ectoderm, mesoderm and endoderm both in vitro and in vivo. Thus PMSCs serve as a unique cell type for human perinatal iPSC derivation from extra embr

Primary umbilical melanoma.

We report a 72-year-old woman who presented with a primary melanoma of the umbilicus and periumbilical skin of 4 years' duration. Pathological examination of a biopsy specimen showed Clark's level IV, and tumour thickness 2.3 mm. The patient underwent preoperative lymphatic mapping followed by sentinel lymph node biopsy, and wide local excision of the primary tumour, including its attachment to the peritoneum. Because of the variations in vascularity and the residual embryonal connections of the umbilicus with the peritoneum and other intra-abdominal organs, an aggressive surgical approach is

Amniotic band sequence: Streeter's hypothesis reexamined.

Recently published reports of 54 subjects with the amniotic band syndrome (ABS) were reviewed, paying particular attention to internal anomalies. Evidence from the internal anomalies suggests that in most cases reviewed, damage occurred in a definable time period, probably prior to 26 days postconception and before the establishment of effective embryonic circulation. Most defects are explicable in terms of interference with neuropore closure, malmigration of cephalic neural crest tissue, and damage to the mesonephros consistent with local interference of the graded expression of organizationa

Biological consequences of interactions between hepatitis B virus and human nonhepatic cellular genomes.

The incidence of free and integrated hepatitis B virus (HBV) in various human tissues is discussed. Although hepadnavirus replication is restricted to a more or less stringent host cell range, it is clear that viral integration is not restricted to any particular organ but occurs in many tissues, including the placenta, embryo, and spermatozoa. The evidence for mother-to-infant transmission of HBV is presented. Direct transfer of HBV through the germ line cells and its implication in the development of embryonal tumours are discussed. HBV may thus possess potential oncogenicity not only for th

Trapping of large numbers of larvae in the livers of Toxocara canis-reinfected mice.

Congenitally athymic nude mice (BALB/c-nu/nu) and BALB/c-nu/+ were infected with 500 embryonated Toxocara canis eggs. Six weeks later they were reinfected with the same number of eggs. The liver and other organs were examined for numbers of 2nd-stage larvae at 2, 4, and 6 weeks after reinfection. Far more larvae were trapped in the liver after reinfection than after the primary infection but fewer were found in the livers of BALB/c-nu/nu than in BALB/c-nu/+ mice.

Differentiation of pinopsin-immunoreactive cells in the developing quail pineal organ: an in-vivo and in-vitro immunohistochemical study.

The avian pineal organ contains several types of photoreceptors with different photopigments: rhodopsin, iodopsin, and pinopsin. We have previously examined the differentiation of both rhodopsin-like and iodopsin-like immunoreactive cells during pineal development in quail embryos to determine the onset of synthesis of specific proteins and their cellular localization. In the present study, we have performed pinopsin immunohistochemistry on in-vivo developing and in-vitro cultured pineal organs of quail embryos. The results were compared with those obtained with rhodopsin and iodopsin immunohi

Allele-specific CRISPR perturbation of the imprinted Dlk1-Dio3 domain reveals regulation of BMP-NOTCH-VEGF signaling in embryonic organogenesis.

The Dlk1-Dio3 imprinted domain is essential for mammalian development, yet its role in coordinating multi-organ embryogenesis is not fully understood. Here we generated four distinct mouse models via CRISPR/Cas9-mediated insertion of a transcriptional termination cassette within the Dlk1-Dio3 domain, encompassing homozygous (HOMO), maternally inherited (MK), paternally inherited (PK), and wild-type alleles (WT), to elucidate parent-of-origin-specific effects. Integrative transcriptomic analyses across multiple scales revealed allele-specific dysregulation of imprinted genes and altered epigene

Guanine nucleoside alleviates mycophenolic acid-induced toxicity in mouse embryonic stem cells.

INTRODUCTION: Mycophenolic acid (MPA), an immunosuppressant widely used in organ transplantation and the treatment of autoimmune diseases, poses a teratogenic risk to mammalian fetuses. Guanine nucleoside (GUO), an important intermediate in purine metabolism, has the potential to modulate immune responses. METHODS: In this study, we systematically evaluated the effects of MPA at various concentrations (0.05, 0.075, 0.1 μM) on the proliferation, apoptosis, and differentiation of mouse embryonic stem cells (PGK12.1), and investigated whether GUO at different concentrations (2, 4, 8 mM) could mit

Androgen-regulated gene expression of developing external genitalia in mice.

Androgen signaling during a critical embryonic window is essential for male embryonic external genitalia (eExG) development, including urethral tubularization, and defects in the androgen signaling pathway result in common congenital anomalies, such as hypospadias. To identify key regulators for eExG differentiation, we manipulated the hormonal environment of developing mouse embryos by administering testosterone propionate (TP) to pregnant dams to induce masculinization in female embryos or the androgen receptor (AR) antagonist flutamide (FLU) to induce feminization in male embryos. We perfor

Medicine designed to combat diseases of affluence affects the early development of fish. How do plastic microparticles contribute?

The incidence of diseases of affluence, such as diabetes mellitus, cardiovascular diseases, high blood pressure, and high cholesterol has been reported to rise. Consequently, the concentrations of residues of drugs designed to treat these diseases have been rising in water bodies. Moreover, the toxicity of these pharmaceuticals towards fish and other non-target organisms can be even enhanced by microplastic particles that are reportedly present in surface water. Therefore, the aim of this study was to describe the effects of three highly prescribed drugs, in particular metoprolol, enalapril, a

Stage-dependent function of Wnt5a during male external genitalia development.

External genitalia development in mice involves multiple developmental processes under the regulation of various signaling pathways. Wnt5a, one of the major Wnt ligands, is a crucial developmental regulator of outgrowing organs such as the limb, the mandible, and the external genitalia. Defects in Wnt5a signaling have been linked to Robinow syndrome, a genetic disorder in which male patients manifest a micropenis and defective urethral tube formation. Whereas Wnt5a is required for cell proliferation during embryonic external genitalia outgrowth, its role for urethral tube formation has yet to

Inhibition of cell-cell adhesion impairs directional epithelial migration on micropatterned surfaces.

The development of the vertebrate body plan with left-right (LR) asymmetry (also known as handedness and chirality) requires the emerging chiral morphogenesis of epithelial cells at specific embryonic stages. In this process, cell-cell adhesions coordinate cellular organization and collective cell migration, and are critical for the directional looping of developing embryonic organs. However, the underlying biophysical mechanism is not yet well understood. Here we modeled normal and delayed epithelial LR symmetry breaking with patterned epithelial chiral morphogenesis on microscale lines with

Dual effect of CTCF loss on neuroprogenitor differentiation and survival.

An increasing number of proteins involved in genome organization have been implicated in neurodevelopmental disorders, highlighting the importance of chromatin architecture in the developing CNS. The CCCTC-binding factor (CTCF) is a zinc finger DNA binding protein involved in higher-order chromatin organization, and mutations in the human CTCF gene cause an intellectual disability syndrome associated with microcephaly. However, information on CTCF function in vivo in the developing brain is lacking. To address this gap, we conditionally inactivated the Ctcf gene at early stages of mouse brain

[Stem cell therapy: an update].

Medicine will be faced with a major challenge in coming years, namely how to treat for tissue dysfunction due to disease and aging There are two basic options: drug therapy and cell therapy. Stem cells have been the subject of intense speculation and controversy for several years, as they open up radically new therapeutic possibilities. Classical drugs can only smoothen consequences of tissue dysfunction, whereas cell therapy has the potential to restore tissue function by providing fresh cells. Cell therapy is totally different from organ transplantation, which can only benefit a limited numb

CCN5 Expression in mammals. II. Adult rodent tissues.

CCN5 is a secreted heparin- and estrogen-regulated matricellular protein that inhibits vertebrate smooth muscle cell proliferation and motility. CCN5 is expressed throughout murine embryonic development in most organs and tissues. However, after embryonic development is complete, we hypothesized that CCN5 distribution would be largely restricted to small set of tissues, including smooth muscle cells of the arteries, uterus, airway, and digestive tract. Because CCN5 inhibits proliferation of smooth muscle cells in vitro, it might function to prevent excessive growth in vivo. In contrast, anothe

Doubly mutant mice, deficient in connexin32 and -43, show normal prenatal development of organs where the two gap junction proteins are expressed in the same cells.

The connexins are a family of proteins that form the intercellular membrane channels of gap junctions. Genes encoding 13 different rodent connexins have been cloned and characterized to date. Connexins vary both in their distribution among adult cell types and in the properties of the channels that they form. In order to explore the functional significance of connexin diversity, several mouse connexin-encoding genes have been disrupted by homologous recombination in embryonic stem cells. Although those experiments have illuminated specific physiological roles for individual connexins, the resu

Maternal Huluwa regulates postfertilization microtubule array organization for asymmetrical transport of dorsal determinants in zebrafish.

The dorsal organizer, essential for vertebrate embryonic axis formation, is induced by microtubule-mediated transport of maternal determinants. Maternal Huluwa (Hwa) has been identified as an essential organizer inducer in zebrafish and frogs, functioning at midblastula stages to activate β-catenin signaling in the preorganizer. It remains unknown if maternal Hwa functions at or before fertilization. Here, we report that maternal Hwa protein is critical for organizing the vegetal parallel microtubule array immediately after fertilization in zebrafish. Hwa protein and mRNA are enriched at the v

Renal epithelial development in organotypic culture.

An in vitro model system for the experimental study of renal epithelial differentiation is described. Fetal murine metanephric tissue consisting of nephrogenic blastema and branched ureteric bud is isolated following 24-36 h of natural embryonic inductive interaction (13 +/- 0.4 days gestation) and cultured as an intact organ in a Trowell-type assembly. During 120 h of organ culture incubation in completely defined serum-free medium, advanced organotypic proximal tubular and glomerular epithelial differentiation proceed in the absence of vascularization, perfusion, and urine production. The sy

Understanding laterality disorders and the left-right organizer: Insights from zebrafish.

Vital internal organs display a left-right (LR) asymmetric arrangement that is established during embryonic development. Disruption of this LR asymmetry-or laterality-can result in congenital organ malformations. Situs inversus totalis (SIT) is a complete concordant reversal of internal organs that results in a low occurrence of clinical consequences. Situs ambiguous, which gives rise to Heterotaxy syndrome (HTX), is characterized by discordant development and arrangement of organs that is associated with a wide range of birth defects. The leading cause of health problems in HTX patients is a
