Combined effects of ambient particulate matter exposure and a high-fat diet on oxidative stress and steatohepatitis in mice.

BACKGROUND: Chronic exposure to ambient particulate matter with aerodynamic diameters < 2.5 (PM2.5) induces oxidative injury and liver pathogenesis. The present study assessed the effect and mechanism of long-term, real-world airborne particulate matter (PM) exposure on oxidative stress and hepatic steatosis in the context of a standard chow diet (STD) and a high-fat diet (HFD); the study further explored whether a combination of PM exposure and HFD treatment exacerbates the adverse effects in mice. METHODS: C57BL/6J mice fed with STD or HFD (41.26% kcal fat) were exposed to PM or filtered ai…

Contribution of soluble and particulate-matter fractions to the total glyphosate and AMPA load in water bodies associated with horticulture.

Glyphosate-based herbicides are used in horticulture to prepare the soil for planting and to remove weeds surrounding greenhouses. Superfici … lated partition coefficients Kd confirmed that these compounds had tended to be associated with the particulate matter, the soluble glyphosate and AMPA contributed, on the average, to more than 90% of the total water volume. Although the analysis of suspended particulates is considered a more sensitive strategy for glyphosate and AMPA detection in surface water, the …

Oxidation of Reduced Peat Particulate Organic Matter by Dissolved Oxygen: Quantification of Apparent Rate Constants in the Field.

Peat particulate organic matter (POM) is an important terminal electron acceptor for anaerobic respiration in northern peatlands provided that the electron-accepting capacity of POM is periodically restored by oxidation with O2 during peat oxygenation events. We employed push-pull tests with dissolved O2 as reactant to determine pseudo-first-order rate constants of O2 consumption ( kobs) in anoxic peat soil of an unperturbed Swedish ombrotrophic bog. Dissolved O2 was rapidly consumed in anoxic peat with a mean kobs of 2.91 ± 0.60 h-1, corresponding to an O2 half-life of ∼14 min. POM dominated

Metabolic insults-initialised nonalcoholic steatohepatitis promoted by fine particulate matter challenge: A mechanism involving RNF15-driven ASK1 degradation.

Epidemiological studies have found that long-term fine particulate matter (PM2.5) exposure is closely linked to the incidence of nonalcoholic steatohepatitis (NASH). However, no approved clinical medication or effective treatment strategy is available because its complex pathological processes are poorly understood. RING-finger-protein 15 (RNF15) is an E3 ubiquitin-protein ligase that abounds in the liver. We investigated whether and by what mechanisms RNF15 influences hepatic metabolic homeostasis to mitigate steatohepatitis. RNF15 expression was confirmed in human patients with NASH and rode…

Ultrafine particulate matter exposure induces gut microbiota dysbiosis together with ER stress in the liver and worsened atherosclerosis.

Air pollution exposure is associated with increased cardiovascular morbidity and mortality worldwide. Previous studies provide a causal relationship between exposure to particulate matter (PM) and atherosclerosis development. We have previously demonstrated increased aortic atherosclerosis and adverse metabolic effects in hyperlipidemic mice exposed to ambient ultrafine PM. However, the underlying mechanisms by which ambient PM promotes systemic effects leading to worsened atherosclerosis remain unknown. We have recently shown that the gut microbiota composition was altered in mice exposed to …
