Targeted ablation of cholinergic interneurons in the dorsolateral striatum produces behavioral manifestations of Tourette syndrome.

Gilles de la Tourette syndrome (TS) is characterized by tics, which are transiently worsened by stress, acute administration of dopaminergic drugs, and by subtle deficits in motor coordination and sensorimotor gating. It represents the most severe end of a spectrum of tic disorders that, in aggregate, affect ∼ 5% of the population. Available treatments are frequently inadequate, and the pathophysiology is poorly understood. Postmortem studies have revealed a reduction in specific striatal interneurons, including the large cholinergic interneurons, in severe disease. We tested the hypothesis th

Long-term effects of neonatal exposure to MK-801 on recognition memory and excitatory-inhibitory balance in rat hippocampus.

Blockade of the N-methyl-d-aspartate receptors (NMDARs) during the neonatal period has been reported to induce long-term behavioral and neurochemical alterations that are relevant to schizophrenia. In this study, we examined the effects of such treatment on recognition memory and hippocampal excitatory and inhibitory (E/I) balance in both adolescence and adulthood. After exposure to the NMDAR antagonist, MK-801, at postnatal days (PND) 5-14, male Sprague-Dawley rats were tested for object and object-in-context recognition memory during adolescence (PND 35) and adulthood (PND 63). The parvalbum

Developmental abnormalities of cortical interneurons precede symptoms onset in a mouse model of Rett syndrome.

Rett syndrome (RTT, MIM312750), a neurodevelopmental disorder predominantly occurring in females, is caused in the majority of cases by sporadic mutations in the gene encoding the transcriptional modulator methyl-CpG-binding protein 2 (MECP2). In mice, impaired MeCP2 function results in severe motor, cognitive, and emotional defects. The lack of Mecp2 in γ-aminobutyric acid-(GABA) releasing forebrain interneurons (INs) recapitulate many RTT features, however, the role of this gene in the development of the cortical inhibitory system is still unknown. Here, we found that MeCP2 expression varies

Layer 6 corticothalamic neurons activate a cortical output layer, layer 5a.

Layer 6 corticothalamic neurons are thought to modulate incoming sensory information via their intracortical axons targeting the major thalamorecipient layer of the neocortex, layer 4, and via their long-range feedback projections to primary sensory thalamic nuclei. However, anatomical reconstructions of individual layer 6 corticothalamic (L6 CT) neurons include examples with axonal processes ramifying within layer 5, and the relative input of the overall population of L6 CT neurons to layers 4 and 5 is not well understood. We compared the synaptic impact of L6 CT cells on neurons in layers 4

Immunohistochemical features of dystrophic axons in Papillon dogs with neuroaxonal dystrophy.

The immunohistochemical features of dystrophic axons in brain tissues of Papillon dogs with neuroaxonal dystrophy (NAD) were examined in comparison with 1 dog with cerebellar cortical abiotrophy (CCA) and a dog without neurologic signs. Histologically, many dystrophic axons were observed throughout the central nervous system of all dogs with NAD. These axonal changes were absent in the dog with CCA and in the control dog. Severe Purkinje cell loss was found in the dog with CCA, whereas the lesions were milder in all dogs with NAD. Immunohistochemically, the many dystrophic axons were positive

Gene expression of glutamate receptors GluR1 and NR1 is differentially modulated in striatal neurons in rats after 6-hydroxydopamine lesion.

In the present study, we attempted to address the modulation of the gene expression of N-methyl-D-aspartate (NMDA) and alpha-amino-3-hydroxy-5-methyl-4-isoxazole-propionate (AMPA) glutamate receptors in the neostriatum of the 6-hydroxydopamine (6-OHDA)-lesioned rat, an animal model of Parkinson's disease. After 2 weeks of lesion, reverse transcriptase-polymerase chain reactions (RT-PCRs) revealed significant reduction in GluR1 mRNA expression but a significant enhancement of NR1 mRNA expression in the striatal tissues of the lesioned side. No modulation in the mRNA expression of GluR2, GluR3,

Selective neuronal vulnerability following mild focal brain ischemia in the mouse.

The evolution of cellular damage over time and the selective vulnerability of different neuronal subtypes was characterized in the striatum following 30-minute middle cerebral artery occlusion and reperfusion in the mouse. Using autoradiography we found an increase in the density of [3H]PK11195 binding sites--likely reflecting microglial activation--in the lesion border at 3 days and in the whole striatum from 10 days to 6 weeks. This was accompanied by a distinct loss of [3H]flumazenil and [3H]CGP39653 binding sites from 10 days up to 6 weeks reflecting neuronal loss. Brain ischemia resulted

Expression of connexin36 in the adult and developing rat brain.

The distribution of connexin36 (Cx36) in the adult rat brain and retina has been analysed at the protein (immunofluorescence) and mRNA (in situ hybridization) level. Cx36 immunoreactivity, consisting primarily of round or elongated puncta, is highly enriched in specific brain regions (inferior olive and the olfactory bulb), in the retina, in the anterior pituitary and in the pineal gland, in agreement with the high levels of Cx36 mRNA in the same regions. A lower density of immunoreactive puncta can be observed in several brain regions, where only scattered subpopulations of cells express Cx36

Selective neuronal vulnerability in HIV encephalitis.

Recent studies of human immunodeficiency virus type 1 (HIV-1) encephalitis have shown that in addition to well established white matter damage, the neocortex shows thinning, loss of large neurons and dendritic damage. In order to identify neuronal populations affected in HIV encephalitis and to determine how neuronal damage relates to the severity of HIV infection within the nervous system, we quantified parvalbumin (PV+) and neurofilament (NF+) immunoreactive neurons in the frontal cortex and hippocampus. We found that in the neocortex, the density of NF+ and PV+ neurons was independent of se

A comparison of behavioural effects and morphological features of grafts rich in cholinergic neurons placed in two sites of the denervated rat hippocampus.

This study compared the morphological characteristics and the behavioural effects of intrahippocampal septal cell suspension grafts injected either just above the pyramidal cell layer of the hippocampal region CA1 or within the dorsal leaf of the dentate gyrus (DG) in rats subjected to electrolytic fimbria-fornix lesions. The behavioural tests determined home-cage and open-field activity, as well as radial-maze performance. DNMT1 function in cortical PV interneurons modulates cortical network activity, mood disorder‑related behavior, and perineuronal nets.

Parvalbuminergic (PV) interneurons are central to cortical network stability and psychiatric vulnerability. Here, we identify DNA methyltransferase 1 (DNMT1) as a key epigenetic regulator linking PV interneuron function to glial and extracellular matrix remodeling. Conditional PV-specific Dnmt1 deletion, combined with single-nucleus RNA-seq (snRNA-seq), snATAC-seq, in vivo electrophysiology, histology, and behavioral analyses, revealed that DNMT1 loss increases PV spiking but reduces inhibitory efficacy, driving network desynchronization and depression- and anxiety-like behavior in mice. These

The dark side of light exposure: impacts on object recognition memory and spatial associative memory in Wistar rats.

Permanent exposure to constant light (LL) induces arrhythmicity in locomotor activity and impairs spatial memory, an effect that has been associated with reduced hippocampal neurogenesis in rodents. However, it remains unclear whether these memory deficits result from circadian disruption or from a sustained (tonic) effect of light exposure. In this study, rats were exposed to LL during lactation (21 days) to prevent the development of arrhythmicity in adulthood. Subsequently, animals were maintained under a 12:12 light and dark (LD) cycle for 39 days and then re-exposed to LL. Behavior assess

Differential impacts of germline and adult aggrecan knockout in PV+ neurons on perineuronal nets and PV+ neuronal function.

Perineuronal nets (PNNs) are a condensed form of extracellular matrix primarily found around parvalbumin-expressing (PV+) interneurons. The postnatal maturation of PV+ neurons is accompanied with the formation of PNNs and reduced plasticity. Alterations in PNN and PV+ neuron function have been described for mental disorders such as schizophrenia and autism. The formation of PNNs is highly dependent on aggrecan, a proteoglycan encoded by the ACAN gene, but it remains unknown if it is produced by the PV+ neurons themselves. Thus, we established a knockout (KO) mouse model (ACANflx/PVcre) and an

Microglia drive synaptic and functional connectivity deficits in the Ts65Dn mouse model of Down syndrome by affecting inhibition.

Microglia, the resident immune cells of the brain, play a crucial role in sculpting neuronal circuits during development, and their dysfunction is increasingly implicated in neurodevelopmental disorders such as Down syndrome (DS). Here, we reveal a previously unrecognized pathological mechanism whereby microglia contribute to synaptic and neuronal activity deficits in DS: a selective disruption of microglia–interneuron interactions. Using primary neuron–microglia co-cultures from Ts65Dn mice, we show that while trisomy in neurons drives excitatory synaptic deficits and major microglial morphol

A vagal reflex evoked by airway closure.

Airway integrity must be continuously maintained throughout life. Sensory neurons guard against airway obstruction and, on a moment-by-moment basis, enact vital reflexes to maintain respiratory function1,2. Decreased lung capacity is common and life-threatening across many respiratory diseases, and lung collapse can be acutely evoked by chest wall trauma, pneumothorax or airway compression. Here we characterize a neuronal reflex of the vagus nerve evoked by airway closure that leads to gasping. In vivo vagal ganglion imaging revealed dedicated sensory neurons that detect airway compression but

GABAergic subiculum-medial entorhinal projection in mice and rats.

The subiculum (SUB) is a major hippocampal output hub that routes information to cortical and subcortical targets, and its long-range projections are considered excitatory. Using enhancer-driven adeno-associated viral vectors to selectively label γ-aminobutyric acid (GABA)-releasing neurons across species, here we show that the dorsal SUB also sends an inhibitory projection to the dorsal part of the medial entorhinal cortex (MEC) in mice and rats. Anterograde tracing in mice revealed that the dorsal SUB contains GABAergic neurons that project sparsely to all layers of the dorsal MEC with enric

AUT00206, a novel and selective Kv3.1/3.2 channel modulator, restores cognitive dysfunction and negative symptoms in an animal model for schizophrenia symptomatology.

BACKGROUND: Voltage-gated Kv3.1 and KV3.2 potassium channels are mainly located on parvalbumin (PV) containing interneurons, where they play a key role in synchronising the coordinated firing of pyramidal neurons and regulating cognitive function. AIMS: We aim to explore the efficacy of acute treatment with AUT00206 (5,5-dimethyl-3-[2-(7-methylspiro[2H-benzofuran-3,1'-me]-4-yl)oxypyrimidin-5-yl]imidazolidine-2,4-dione), a novel and selective positive modulator of Kv3.1/3.2 channels, to improve cognitive and social behaviour deficits in our validated animal model relevant to schizophrenia. Extracellular matrix and perineuronal nets in neural plasticity, memory, and epileptogenesis.

In the mature brain, the extracellular matrix (ECM) molecules derived from neurons and glia accumulate in spatially distinct extracellular compartments, including a diffuse interstitial matrix and condensed perineuronal nets (PNNs), where they regulate receptor mobility, ion homeostasis, cell-surface signaling, structural stabilization, and experience-dependent synaptic remodeling. PNNs, in particular, assemble around selected neuronal populations - most prominently parvalbumin-positive fast-spiking interneurons - and thereby couple extracellular molecular architecture to inhibitory control, c

Nonlinear dynamics of a corticothalamic coupled neural mass model reproduce the mechanism of 40 Hz ASSR generation.

Auditory steady-state responses (ASSR) provide a powerful tool for investigating oscillatory dynamics and network synchronization along the auditory pathway. However, existing neural mass models of 40 Hz ASSR have largely focused on cortical microcircuits or macroscopic phenomenology, while neglecting the closed-loop regulation formed by thalamocortical feedforward transmission and thalamic reticular nucleus (TRN)-mediated feedback inhibition. To address this, we constructed an Auditory Corticothalamic Neural Mass Model (ACT-NMM) that investigates cortical pyramidal neurons (Pyr), parvalbumin-

Toll-like Receptor 8 (TLR8) Is Expressed in Mouse Hippocampal Neurons and Modulates Neuronal Excitability.

Toll-like receptors (TLRs) are essential in the innate immune response. Furthermore, neuronal TLRs have been involved in regulating neuronal dendritic outgrowth and excitability. However, a deeper understanding of neuronal-TLR function is essential. We investigated the expression and role of TLR8 using mouse postnatal hippocampal neuronal cultures. We assessed TLR8 (and TLR7) expression by immunocytochemistry and TLR8 functionality via electrophysiological recordings in a high-density multi-electrode array system and calcium flux imaging. Pathway activation following fast TLR8 stimulation was
