Bidirectional imprinting of a single gene: GNAS1 encodes maternally, paternally, and biallelically derived proteins.

The GNAS1 gene encodes the alpha subunit of the guanine nucleotide-binding protein Gs, which couples signaling through peptide hormone receptors to cAMP generation. GNAS1 mutations underlie the hormone resistance syndrome pseudohypoparathyroidism type Ia (PHP-Ia), so the maternal inheritance displayed by PHP-Ia has raised suspicions that GNAS1 is imprinted. Despite this suggestion, in most tissues Gsalpha is biallelically encoded. In contrast, the large G protein XLalphas, also encoded by GNAS1, is paternally derived. Because the inheritance of PHP-Ia predicts the existence of maternally, rath

Does paternal occupational lead exposure increase the risks of low birth weight or prematurity?

The relation between paternal occupational lead exposure and low birth weight/prematurity was examined in a retrospective cohort study. Birth weight and gestational age (1981-1992), obtained from New York State birth certificates, were compared between lead-exposed and nonexposed workers. The exposed group (n = 4,256) consisted of births to male workers of reproductive age reported to the New York State Heavy Metals Registry. The control group (n = 2,318) consisted of the offspring of a random sample of male bus drivers, frequency matched by age and residence. There were no statistically signi

A case of full triploidy (69,XXX) of paternal origin with unusually long survival time.

Triploidy is a rare disorder in live-born children and these infants generally die within the first hours after birth. We report here on a girl with full triploidy and multiple malformations, who survived for 10 1/2 weeks. The extra set of haploid chromosomes was of paternal origin, as shown by chromosomal banding techniques.

Paternal mononuclear cell immunization therapy for repeated miscarriage: predictive variables for pregnancy success.

Pregnancy outcomes for 125 women with unexplained recurrent abortion conceiving after immunologic testing for possible paternal leukocyte immunization were analyzed. Pregnancy success was related to the number of previous miscarriages (relative risk 0.36 for each additional miscarriage after 3), a history of a late pregnancy loss (relative risk 0.18), any other relevant treated or untreatable diagnosis (relative risk 0.27), immunization with paternal mononuclear cells (relative risk 5.6), and time in weeks from test date to LMP of the next pregnancy (relative risk 0.93 for each additional week

"Partial trisomy 22 and 11" due to a paternal 11;22 translocation associated with Hirschsprung disease.

The 11;22 translocation seems to be the most frequent, non-Robertsonian, translocation in man. Approximately 50 cases with an unbalanced karyotype 47,XX (or XY),+der(22), t(11q;22q), due to a 3:1 meiotic disjunction in the parental translocation carrier, have been reported in the literature. We present an additional patient with that chromosome aberration, whose father was shown to be the translocation carrier. He presented with many of the more or less typical signs of the syndrome, but had an extraordinary additional finding, namely Hirschsprung disease. Although anal stenosis is a rather fr

A pupal lethal mutation with a paternally influenced maternal effect on embryonic development in Drosophila melanogaster.

The maternal effect and zygotic phenotype of l(1)pole hole (l(1)ph) is described. l(1)ph is a zygotic lethal mutation which affects cell division of adult precursor cells in Drosophila larvae. The locus is located in 2F6 on the salivary gland chromosome map and four alleles have been characterized. Germ-line clonal analysis of amorphic alleles indicates that l(1)ph has a maternal effect lethal phenotype. Two lethal phenotypes are observed among embryos derived from female germ-line clones homozygous for amorphic alleles dependent upon the zygotic activity of l(1)ph+ introduced via the sperm. C

Relationship of the estrogen surge and multiple mates to cub paternity in the giant panda (Ailuropoda melanoleuca): implications for optimal timing of copulation or artificial insemination.

The effectiveness of ex situ breeding programs for endangered species can be limited by challenges in mimicking mating competitions that naturally occur among multiple mates in the wild. The objective of this study was to evaluate the impact of timed natural matings and/or artificial inseminations in the context of the urinary estrogen surge on cub production in the giant panda (Ailuropoda melanoleuca). We used a large cohort of giant pandas, including 12 females and 17 males. DNA paternity exclusion was used to pinpoint accurately the interval during the estrogen surge that coincided with the…

Sperm TLR2/1 activation modulates the paternal RNA cargo and enhances bovine embryonic development.

In vitro fertilization (IVF) and in vitro embryo production (IVP) are crucial technologies for accelerating genetic gain in cattle. However, IVP efficiency remains suboptimal, as in vitro-derived embryos frequently exhibit reduced developmental competence compared to their in vivo counterparts. Previous studies indicate that activation of the sperm Toll-like receptor 2/1 (TLR2/1) complex via Pam3CSK4 (PAM3) enhances sperm-oocyte interactions and improves cleavage and blastocyst rates; however, the underlying molecular mechanisms remain unexplored. In this study, frozen-thawed active sperm from

Paternal genetic background orchestrates fetal development through tissue-specific cis-regulatory divergence.

Paternal influences on offspring development extend beyond Mendelian inheritance, but how sire genetics shape fetal transcription and cis-regulatory landscapes across tissues is unclear. Using a reciprocal backcross in pigs (F1×German Landrace vs. F1×Piétrain) and a haplotype-agnostic RNA-seq pipeline, we profiled six fetal tissues to test sire-breed effects. We identified paternal breed-driven transcriptional divergence, with 1,176 genes differentially expressed. Transcriptional divergence was most pronounced in metabolic tissues, revealing a tissue-specific trade-off: F1 × GL conceptuses pri

Paternal Metabolic Reversal Remodels Sperm RNA Profiles and Ameliorates Intergenerational Metabolic Disorder in Mice.

UNLABELLED: Paternal obesity increases metabolic risk in offspring, but whether this risk can be reduced by restoring paternal health before conception remains unresolved. We developed a within-sire induction-and-reversal model in outbred CD1 mice in which high-fat diet (HFD)-exposed males generated offspring before and after transition to an ingredient-matched control diet with voluntary exercise. HFD caused obesity, glucose intolerance, insulin resistance, and extensive remodeling of sperm mRNA, lncRNA, and sncRNA profiles, together with transcriptomic changes in metabolic tissues. Diet and

Paternal SIRT1 Inhibition During Capacitation Imprints Histone 4 Hyperacetylation in Mouse Zygotes and Blastocysts.

Sirtuin 1 (SIRT1) is a NAD+-dependent deacetylase involved in chromatin remodelling. In this study, we evaluated whether sperm SIRT1 inhibition affects early embryonic chromatin dynamics. Sperm were treated with sirtinol during capacitation or during both capacitation and fertilization. SIRT1 inhibition increased acH4K16 in the male pronucleus without affecting SIRT1 levels, whereas the female pronucleus remained largely unchanged, except for reduced SIRT1 when inhibition was extended to fertilization. Notably, embryos derived from treated sperm maintained elevated acH4K16 at the blastocyst st

Fathers at risk: Effects of threatened and actual preterm birth on the course of paternal peripartum depression and anxiety.

BACKGROUND: While it is established that preterm birth (PB) increases the risk of psychological distress in fathers postpartum, the impact of a threatened PB on peripartum depressive and anxiety symptoms, as well as their course over time, remains insufficiently studied. METHODS: We assessed 116 men during their partner's pregnancy (T1) and 6 weeks postpartum (T2), using the Edinburgh Postnatal Depression Scale and the State-Trait Anxiety-Depression Inventory. Participants were categorized into a control group (CG) with term birth and two risk groups based on PB occurrence: 33 men with partne

Welfare paternalism and the paradox of protection: towards a rights-based framework to mediate the deprivation of liberty of children.

This article examines the acute ambiguity in the law governing the deprivation of liberty of children in England and Wales, where protective interventions often rely on judicial direction and on unclear statutory safeguards. It is argued that the current framework is reactive, welfare-driven, and lacking in procedural coherence. Drawing upon Fineman's theory of vulnerability, Fraser's parity of participation, and Lundy's analysis of children's agency in relation to the participatory guarantees of Article 12 of the United Nations Convention on the Rights of the Child, the article reconceptualiz

The paternal brain: longitudinal insights into structural and functional plasticity and attachment over 24 weeks postpartum.

Over the last two decades, interest in the neurobiological basis of parenthood has grown, with the primary focus being on maternal neuroplasticity. However, research on paternal brain adaptations remains limited and inconclusive. This longitudinal study investigated gray matter volume (GMV) and resting-state functional connectivity (rsFC) changes in 25 fathers immediately after childbirth and at 3, 6, 9, 12, and 24 weeks postpartum. Morphological changes were most evident as reductions in GMV within the bilateral occipital, frontal, temporal and parietal cortices, as well as the temporo-pariet

Maternal and paternal perceptions of child vulnerability in children with familial mediterranean fever: a cross-sectional study.

OBJECTIVES: Familial Mediterranean fever (FMF) is a chronic autoinflammatory disease that may affect not only children but also parental psychological well-being. This study aimed to evaluate parental perceptions of child vulnerability in children with FMF and to investigate factors associated with maternal and paternal vulnerability scores. METHODS: This cross-sectional study included 64 children aged 3-17 years with FMF who were followed in a pediatric rheumatology outpatient clinic and their parents. All patients were evaluated during attack-free periods. Parents completed the Child Vulner

Experiences of discrimination, paternal support, and hypertensive disorders of pregnancy (HDP) in the MIEHR study, Houston, Texas.

BACKGROUND: Discrimination is a psychosocial stressor that, through activation of stress-response pathways, may influence risk of adverse perinatal health outcomes. However, few studies have evaluated the association between discrimination and hypertensive disorders of pregnancy (HDP), and none have explored effect modification by race or paternal support, which may buffer the adverse effects of discrimination. METHODS: We included 1,847 non-Hispanic Black and White participants from the Maternal and Infant Environmental Health Riskscape (MIEHR) study. Krieger's validated Experiences of Discr

A systematic review on the role of paternal factors in human placental development, function, and pregnancy-related disorders.

BACKGROUND: The human placenta plays an important role in pregnancy and offspring health. The paternal genome contributes significantly to placental growth and development. While the maternal factors affecting gestational health are thoroughly investigated, the paternal factors are often overlooked. Thus, it is important to understand various paternal factors affecting placental development and function. OBJECTIVE: To assess the effect of various paternal factors on placental development, function, and pregnancy-related disorders. MATERIALS AND METHODS: This review was registered in PROSPERO
