NLA proteins: Master regulators that integrate nutrient homeostasis, stress adaptation, and plant development.

As sessile organisms, plants rely on intricate molecular regulatory networks to navigate complex, fluctuating environments and diverse stress conditions. Nitrogen Limitation Adaptation (NLA), a RING-type ubiquitin ligase harboring an SPX domain, has emerged as a central hub integrating plant responses to nutrient availability, pathogen attack, and cold stress. This review provides a systematic overview of the structural features of NLA, with a particular focus on the regulatory role of inositol pyrophosphates (PP-InsPs) in plant phosphorus signaling. We highlight the core functions of NLA in m

PcXTH22 as a core regulator enhancing aluminum toxicity tolerance in Populus cathayana.

Aluminum (Al) toxicity in acidic soils restricts plant growth. Xyloglucan endotransglucosylase/hydrolases (XTHs) modulate Al responses, but their mechanisms in poplar remain unknown. Here, we characterized PcXTH22-mediated Al tolerance in Populus cathayana. The PcXTH22OE lines showed negligible injury, whereas the pcxth22 mutants suffered severe damage, confirming the role of PcXTH22 as a positive Al regulator. The pcxth22 lines showed increased xyloglucan endotransglucosylase/hydrolase (XET/XTH) activities and xyloglucan content under Al toxicity. This offered more Al³ ⁺ binding sites and res

Overcoming serious incompatibilities resulting from intergeneric hybridisation between Diplotaxis tenuifolia and Brassica oleracea to develop CMS wall-rocket.

Alloplasmic male-sterile breeding lines of Diplotaxis tenuifolia were developed by intergeneric hybridisation between cytoplasmic male-sterile cauliflower (Brassica oleracea L.) and perennial wall-rocket. Amphidiploid filial generation (F1) plants (2n = 2x = 20, CD) were obtained by manual crosses and ovary culture. The phenotype of the hybrids was mostly similar to that of the wall-rocket parent. The plants were male-sterile, but absolutely no seed set was achieved after pollination with D. tenuifolia. Amphitetraploid F1 hybrid plants (4n = 4x = 40, CCDD) obtained by colchicine treatment were

Tomato DOR encodes a signal peptide peptidase-like protein required for adventitious organogenesis and rooting.

The identification of the tomato DOR gene that is essential for adventitious organogenesis provides a genetic target for improving regeneration efficiency and root system performance in tissue culture and plant breeding. Plant rooting and adventitious organogenesis represent major bottlenecks in the application of plant tissue culture techniques. In this study, we characterize the dor mutant (defective in organogenesis and rooting) identified in our collection of tomato T-DNA lines. This mutant exhibits normal callus proliferation but fails to differentiate adventitious buds. Additionally, it …

The Stress Detection and Segmentation Strategy in Tea Plant at Canopy Level.

As compared with the traditional visual discrimination methods, deep learning and image processing methods have the ability to detect plants efficiently and non-invasively. This is of great significance in the diagnosis and breeding of plant disease resistance phenotypes. Currently, the studies on plant diseases and pest stresses mainly focus on a leaf scale. There are only a few works regarding the stress detection at a complex canopy scale. In this work, three tea plant stresses with similar symptoms that cause a severe threat to the yield and quality of tea gardens, including the tea green

Virus-induced gene silencing suggests (1,3;1,4)-β-glucanase is a susceptibility factor in the compatible russian wheat aphid-wheat interaction.

The Russian wheat aphid (RWA), Diuraphis noxia (Kurdjumov), is a significant insect pest of wheat (Triticum aestivum L.) and has a major economic impact worldwide, especially on winter wheat in the western United States. The continuing emergence of new RWA biotypes virulent to existing resistance genes reinforces the need for more durable resistance. Studies have indicated that resistance in previously susceptible plants can be produced by knock-down of susceptibility genes or other genes involved in host plant susceptibility. Therefore, investigation into genes involved in compatible RWA-whea

Genetic variation in jasmonic acid- and spider mite-induced plant volatile emission of cucumber accessions and attraction of the predator Phytoseiulus persimilis.

Cucumber plants (Cucumis sativus L.) respond to spider-mite (Tetranychus urticae) damage with the release of specific volatiles that are exploited by predatory mites, the natural enemies of the spider mites, to locate their prey. The production of volatiles also can be induced by exposing plants to the plant hormone jasmonic acid. We analyzed volatile emissions from 15 cucumber accessions upon herbivory by spider mites and upon exposure to jasmonic acid using gas chromatography-mass spectrometry. Upon induction, cucumber plants emitted over 24 different compounds, and the blend of induced vola

Genetic diversity and yield trait associations in soybean germplasm using SSR markers.

Soybean (Glycine max (L.) Merrill), a vital food and oilseed crop native to China, has the development of high-yield and high-quality new varieties as a core objective in soybean breeding. However, studies on genetic diversity and marker-trait associations for yield-related traits across multiple environments using simple sequence repeat (SSR) markers remain limited. This study utilized 20 pairs of SSR primers to investigate genetic diversity and marker-trait associations (MTAs) between SSR markers and yield-related traits in 150 soybean accessions of diverse origins, which were grown under tw

Integrated Transcriptomics and Metabolomics Analysis of the Formation Mechanism of Low Bitterness and Astringency in "Huangjinya" Tea.

The flavor variation among different tea plant cultivars significantly influences their economic value. 'Huangjinya' (HJY), a typical light-sensitive etiolated tea cultivar, is prized for its fresh and mellow taste with low bitterness and astringency, and commands a high market value, serving as excellent germplasm for breeding superior-flavor tea varieties. Previous studies have largely focused on the mechanisms underlying fresh and umami tastes, whereas the regulatory mechanisms underlying bitterness and astringency remain poorly understood. In this study, two tea plant (Camellia sinensis (L

Plant histone variants: deposition mechanisms, functional diversification, and translational prospects.

Histone variants are non-allelic isoforms of canonical histones that assemble into nucleosomes via replication-independent pathways, reshaping chromatin biophysical properties and transcriptional control at specific genomic loci. Constrained by their sessile lifestyle, plants have evolved a rich set of histone variants, including lineage-specific innovations such as heterochromatin-stabilizing H2A.W, stress-inducible H1.3, gametophyte-restricted H2B.8, Arabidopsis H3.14, and Oryza-specific H4.V. This review systematically summarizes recent advances in plant histone variants from three dimensio

Mutations of the brassinosteroid biosynthesis gene HvDWARF5 enable balance between semi-dwarfism and maintenance of grain size in barley.

Brassinosteroids (BRs) are phytohormones which regulate various developmental processes in plants. They are exceptional phytohormones, as they do not undergo long-distance transport between plant organs. However, knowledge about the function of the enzymes that catalyse BR biosynthesis (particularly its early stages) in cereal crops remains limited. Therefore, this study identifies and analyses the function of the HvDWARF5 (HvDWF5) gene, involved in the early stage of BR biosynthesis in barley (Hordeum vulgare), an important cereal crop, using the TILLING (Targeting Induced Local Lesions IN Ge

The concepts of "domestication" and "artificial selection" and their significance in the work of Charles Darwin.

This analysis examines the terms "domestication" and "selection" presented in Darwin's The variation of animals and plants under domestication, and the significance he gave to these concepts. This approach focuses on how Darwin drew upon knowledge generated centuries earlier by experts in breeding and cultivation to modify prevailing definitions and use this semantic shift as a primary instrument to defend his theory. The construction of the "analogy" between natural and artificial selection was not merely a heuristic resource, but rather the result of an inductive generalization grounded in i

Chlamydia infection in ducks: preliminary communication.

Chlamydia psittaci was isolated from cloacal swab pools from ducks in processing plants, and from cloacal and conjunctival swab pools from ducks on farms, in a small proportion of the samples examined. These isolates were difficult to passage. This difficulty was partly overcome by the use of emetine as the cell growth inhibitor. The direct complement fixation (CF) test was modified for testing duck sera by using complement at the los dose of 1.5 units. By this method, positive titres were demonstrated in experimentally prepared duck antisera, and in some of the samples collected from breeding

Genome editing beyond transgenes: DNA-free technologies in crop improvement.

Conventional and transgenic approaches to crop improvement are increasingly constrained by biosafety concerns, stringent regulatory frameworks, and limited public acceptance, necessitating new precision breeding methods. DNA-free genome editing using the CRISPR/Cas9 approach has emerged as a transformative approach that enables precise, targeted genetic modifications without the stable integration of a foreign gene (transgene). This strategy transiently introduces CRISPR/Cas9 ribonucleoprotein (RNP) complexes, or in vitro transcribed mRNA, into plant cells. This review focuses primarily on CRI

Looking for non-hermaphrodite cacti: multidisciplinary studies in Gymnocalycium bruchii endemic to central Argentina.

Through a multidisciplinary study we found that Gymnocalycium bruchii, an endemic cactus from central Argentina, acts as a dioecious species, which is the first record in this genus. Cactaceae species are typically hermaphroditic; however, about 2% have other different reproductive systems. These non-hermaphroditic species may develop sexual dimorphism in flowers or other reproductive, vegetative or ecological traits, besides a specific breeding system and floral ontogeny. Therefore, multidisciplinary research is necessary to fully understand reproduction in those species. For this purpose, we

The transcription factor CsPAT1 from tea plant (Camellia sinensis) is involved in drought tolerance by modulating phenylpropanoid biosynthesis.

Tea plants, in particular, leafy cash crops, prefer warm and humid climates. Our previous work identified CsPAT1 as a facilitator of lignin biosynthesis in tea plants. The specific role of CsPAT1 in tea plants' abiotic stress response remains unclear. In this study, we found that the expression of CsPAT1 in tea plants was induced under drought, cold, heat, and ABA treatments. CsPAT1 transgenic Arabidopsis lines displayed enhanced drought tolerance compared with wild-type (WT) controls. The SOD and POD activities, proline content, and expression levels of drought-responsive genes were significa

Enhanced folate content in bioengineered lettuce stably expressed under field conditions.

Folate (vitamin B9) is an essential nutrient that plays a crucial role in various bodily functions. Its deficiency can lead to health issues, such as megaloblastic anemia, neural tube defects in the developing fetus and increased risk of cardiovascular diseases. Consequently, there is interest in increasing folate content in food crops by classical and molecular breeding. Since folate is a water-soluble vitamin that is sensitive to heat, we chose to manipulate its synthesis pathway in lettuce, which is often used for salads and sandwiches, without heat treatment, which makes it a good vehicle
