Subunit recombinant vaccine protects against monkeypox.

The smallpox vaccine Dryvax, a live vaccinia virus (VACV), protects against smallpox and monkeypox, but is contraindicated in immunocompromised individuals. Because Abs to VACV mediate protection, a live virus vaccine could be substituted by a safe subunit protein-based vaccine able to induce a protective Ab response. We immunized rhesus macaques with plasmid DNA encoding the monkeypox orthologs of the VACV L1R, A27L, A33R, and B5R proteins by the intradermal and i.m. routes, either alone or in combination with the equivalent recombinant proteins produced in Escherichia coli. Animals that rece

Altered Glycosylation Patterns Increase Immunogenicity of a Subunit Hepatitis C Virus Vaccine, Inducing Neutralizing Antibodies Which Confer Protection in Mice.

UNLABELLED: Hepatitis C virus (HCV) infection is a global health problem for which no vaccine is available. HCV has a highly heterogeneous RNA genome and can be classified into seven genotypes. Due to the high genetic and resultant antigenic variation among the genotypes, inducing antibodies capable of neutralizing most of the HCV genotypes by experimental vaccination has been challenging. Previous efforts focused on priming humoral immune responses with recombinant HCV envelope E2 protein produced in mammalian cells. Here, we report that a soluble form of HCV E2 (sE2) produced in insect cells

Efficient production of functional cholera toxin B subunit using geminiviral replicons in Nicotiana benthamiana.

The cholera toxin B subunit (CTB) has the potential to be a carrier molecule and an effective adjuvant for mucosal vaccines because of its ability to enhance immune responses to antigens. CTB proteins have been expressed in plant-based expression systems. In this study, we used geminiviral replicon systems to transiently express CTB in Nicotiana benthamiana. We developed a high-level expression system that uses combinations of the replication machinery of geminivirus, including tomato yellow leaf curl virus (TYLCV), honeysuckle yellow vein virus (HYVV), and beet mild curly top virus (BMCTV). T

Brucella antigens (BhuA, 7α-HSDH, FliC) in poly I:C adjuvant as potential vaccine candidates against brucellosis.

A promising strategy for controlling animal brucellosis is vaccination with commercial vaccine strains (Brucella melitensis Rev.1 and Brucella abortus RB51). Owing to safety concerns associated with these vaccines, developing a more effective and safe vaccine is essential. In this study, we examined the capacity of BhuA, 7α-HSDH or FliC antigens in the presence or absence of adjuvant in eliciting immune responses against brucellosis. After cloning, expression and purification, these proteins were used to examine immunologic responses. All immunized mice induced a vigorous IgG, with a predomina

In-silico design of a multi-epitope vaccine candidate against onchocerciasis and related filarial diseases.

Onchocerciasis is a parasitic disease with high socio-economic burden particularly in sub-Saharan Africa. The elimination plan for this disease has faced numerous challenges. A multi-epitope prophylactic/therapeutic vaccine targeting the infective L3 and microfilaria stages of the parasite's life cycle would be invaluable to achieve the current elimination goal. There are several observations that make the possibility of developing a vaccine against this disease likely. For example, despite being exposed to high transmission rates of infection, 1 to 5% of people have no clinical manifestations

A Lyme disease mRNA vaccine targeting Borrelia burgdorferi OspA induces strong immune responses and prevents transmission in mice.

Lyme borreliosis (LB), caused by Borrelia burgdorferi sensu lato, is one of the most common tick-borne diseases in the northern hemisphere. Given its increasing global incidence, LB remains a major public health concern and the development of an effective vaccine is recognized as a key component of the overall disease prevention strategy. Here, we present results obtained with newly developed lipid nanoparticle-encapsulated mRNA vaccine candidates encoding the outer surface protein A (OspA) of B. burgdorferi sensu stricto (Bbss) serotype 1 (mRNA-OspA) with or without a secretion signal (SS) or

In silico identification of novel vaccine candidates against multidrug-resistant pseudomonas aeruginosa using subtractive proteomics and immunoinformatics.

Pseudomonas aeruginosa, a notorious pathogen, frequently presents in hospitals and poses numerous health issues due to its drug resistance, making treatment challenging. The rise of multidrug-resistant microorganisms emphasizes the crucial need for novel therapeutic approaches, such as vaccines. This work employed immunoinformatics and subtractive proteomics to find possible universal vaccine candidates. After analyzing the proteome of P. aeruginosa (PAO1 strain), three pathogenic or antigenic proteins were identified as potential targets for vaccines. T and B cell epitopes were predicted from

An optimized secretory expression system and immunogenicity evaluation for glycosylated gp90 of avian reticuloendotheliosis virus.

Reticuloendotheliosis is an important immunosuppressive disease, associated with avian reticuloendotheliosis virus (REV) infection, and causes notable economic losses worldwide. Glycoprotein gp90 is an important structural protein of REV, and considered to be the most important immunogenic antigen, which can induce neutralizing antibodies against REV. In this study, an optimized suspension culture system was developed and applied to secretory express the immunogenic surface antigen gp90. To achieve an optimal glycosylation, the gp90 was designed to secretory expressed into the supernatant of t

Multi-Modal Characterization of the Interactions between a Recombinant Protein Antigen and a Liposomal Adjuvant.

Subunit vaccines rely on adjuvants to enhance immunogenicity, yet antigen-adjuvant interactions in liposomal systems remain underexplored, with unknown implications that could impact the function and stability of coformulated system designs. We report a multimodal study aimed at elucidating interactions between a recombinant protein antigen, gE, and a liposomal adjuvant system, AS01B, using a commercially available vaccine as a model. Quantitative binding assays revealed no significant binding for the antigen-adjuvant pair, with only weak, dose-dependent biolayer interferometry (BLI) signals s

Burkholderia pseudomallei Complex Subunit and Glycoconjugate Vaccines and Their Potential to Elicit Cross-Protection to Burkholderia cepacia Complex.

Burkholderia are a group of Gram-negative bacteria that can cause a variety of diseases in at-risk populations. B. pseudomallei and B. mallei, the etiological agents of melioidosis and glanders, respectively, are the two clinically relevant members of the B. pseudomallei complex (Bpc). The development of vaccines against Bpc species has been accelerated in recent years, resulting in numerous promising subunits and glycoconjugate vaccines incorporating a variety of antigens. However, a second group of pathogenic Burkholderia species exists known as the Burkholderia cepacia complex (Bcc), a grou

Mucosal delivery of antigen-coated nanoparticles to lungs confers protective immunity against tuberculosis infection in mice.

Mucosal boosting of BCG-immunised individuals with a subunit tuberculosis (TB) vaccine would be highly desirable, considering that the lungs are the principal port of entry for Mycobacterium tuberculosis (MTB) and the site of the primary infection and reactivation. However, the main roadblock for subunit TB vaccine development is the lack of suitable adjuvants that could induce robust local and systemic immune responses. Here, we describe a novel vaccine delivery system that was designed to mimic, in part, the MTB pathogen itself. The surface of yellow carnauba wax nanoparticles was coated wit

Construction of cholera toxin B subunit-producing Vibrio cholerae strains using the Mariner-FRT transposon delivery system.

The most widely used oral whole-cell-recombinant B subunit cholera vaccine contains the nontoxic cholera toxin B subunit (CTXB) and either heat- or formalin-killed Vibrio cholerae O1 strains. Vibrio cholerae O1 strains in the vaccine provide antibacterial immunity, and CTXB contributes to the vaccine's efficacy by stimulating production of anti-CTXB antibody. Various attempts have been made to increase CTXB production. In this study, the mariner-FRT transposon delivery system developed by Chiang and Mekalanos was used to place the ctxB gene under the control of a strong chromosomal promoter in

Genome-wide screening of vaccine targets prioritization and reverse vaccinology aided design of peptides vaccine to enforce humoral immune response against Campylobacter jejuni.

Campylobacter jejuni, gram-negative bacteria, is an infectious agent of foodborne disease-causing bloody diarrhea, abdominal pain, fever, Guillain-Barré syndrome (GBS) and Miller Fisher syndrome in humans. Campylobacter spp. with multidrug resistance to fluoroquinolones, tetracycline, and erythromycin are reported. Hence, an effective vaccine candidate would provide long-term immunity against C. jejuni infections. Thus, we used a subtractive proteomics pipeline to prioritize essential proteins, which impart a critical role in virulence, replication and survival. Five proteins, i.e. Single-stra

Tetrahymena thermophila granule lattice protein 3 improves solubility of sexual stage malaria antigens expressed in Escherichia coli.

The requirement for low cost manufacturing makes bacterial cells a logical platform for the production of recombinant subunit vaccines for malaria. However, protein solubility has been a major stumbling block with prokaryotic expression systems. Notable examples include the transmission blocking vaccine candidates, Pfs25 and Pfs48/45, which are almost entirely insoluble when expressed as recombinant proteins in Escherichia coli. Various solubility tags have been used with limited success in improving solubility, although recent studies with granule lattice protein 1 (Grl1p) from the ciliated p

A novel albumin-based nanoparticle vaccine including the globular knob domain of the egg drop syndrome virus fiber protein for egg drop syndrome: Immunogenicity and preliminary structural analyses.

Egg drop syndrome (EDS), caused by the avian adenovirus known as EDS virus (EDSV), is a viral disease characterized by reduced egg production and abnormal egg quality in laying hens, resulting in substantial economic losses. While conventional vaccines can have some safety issues, the subunit vaccines circumvent these concerns. The globular knob domain of the EDSV fiber protein (EDSVFP) harbors critical neutralizing epitopes, making it an ideal vaccine target. In this study, chicken serum albumin nanoparticles (CSA-NPs) were employed as a delivery vehicle. The coding sequence for the knob doma

Development of a cross-protective subunit cocktail vaccine against diverse serotypes of Glaesserella parasuis infection in pigs.

Glässer's disease, caused by Glaesserella parasuis (GPS), causes substantial economic losses in the swine industry worldwide. This situation is largely attributable to the poor cross-protective efficacy of currently available vaccines. In this study, a multi-antigen cocktail subunit vaccine against GPS infection was developed. In silico antigenicity analysis, the immunoprotective efficacy, and antibody titers of 14 GPS antigens were assessed in a murine model. Finally, VacJ, PtsG, and GAPDH were identified as the component of the cocktail vaccine, with Gel-01 as the optimal adjuvant. Immunizat

Design of a novel epitope-based tetravalent subunit vaccine against dengue virus: An immunoinformatic approach.

Dengue imposes a profound global impact, with millions affected annually. Its transmission by Aedes mosquitoes poses significant challenges to combat, aggravated by urbanization and climate change. Despite efforts, no impeccable antiviral treatment exists to date, highlighting the urgency for a vaccine. Developing one encounters hurdles like the four distinctive serotypes of the virus and complex immune responses. In this study, we employed an immunoinformatics approach to design an epitope-based tetravalent subunit vaccine aimed at confronting all DENV serotypes. The study contemplates epitop

A novel multi-epitope peptide vaccine targeting immunogenic antigens of Ebola and monkeypox viruses with potential of immune responses provocation in silico.

The emergence or reemergence of monkeypox (Mpox) and Ebola virus (EBOV) agents causing zoonotic diseases remains a huge threat to human health. Our study aimed at designing a multi-epitope vaccine (MEV) candidate to target both the Mpox and EBOV agents using immunoinformatics tools. Viral protein sequences were retrieved, and potential nonallergenic, nontoxic, and antigenic epitopes were obtained. Next, cytotoxic and helper T-cell (CTL and HTL, respectively) and B-cell (BCL) epitopes were predicted, and those potential epitopes were fused utilizing proper linkers. Design of an epitope-based peptide vaccine against Cryptococcus neoformans.

Cryptococcus neoformans is the highest-ranked fungal pathogen in the Fungal Priority Pathogens List (FPPL) released by the World Health Organization (WHO). In this study, through in silico simulations, a multi-epitope vaccine against Cryptococcus neoformans was developed using the mannoprotein antigen (MP88) as a vaccine candidate. Following the retrieval of the MP88 protein sequences, these were used to predict antigenic B-cell and T-cell epitopes via the bepipred tool and the artificial neural network, respectively. Conserved B-cell epitopes AYSTPA, AYSTPAS, PASSNCK, and DSAYPP were identifi
