A serological test for leprosy based on competitive inhibition of monoclonal antibody binding to the MY2a determinant of Mycobacterium leprae.

A novel serological assay for leprosy has been devised on the basis of serum inhibition of binding of 125I-labelled ML04 monoclonal antibody to Mycobacterium leprae sonicate-coated microtitre plates. Antibodies were detected in 93% of lepromatous leprosy patients, whereas controls from the endemic area, including leprosy contacts and patients with tuberculosis, were serologically negative. The specificity and efficacy of the test may offer an advantage over previously used techniques.

Serology of Legionnaires disease: comparison of indirect fluorescent antibody, immune adherence hemagglutination, and indirect hemagglutination tests.

An immune adherence hemagglutination (IAHA) test for the measurement of antibodies to Legionella pneumophila was developed and evaluated for the diagnosis of Legionnaires disease. Its sensitivity was compared to that of the indirect fluorescent antibody (IFA) test and a recently developed indirect hemagglutination (IHA) test. The sensitivity of the three tests appeared to be similar, with the IFA test giving slightly higher titers. Both the IHA and IAHA tests appear useful for the serodiagnosis of Legionnaires disease; the IAHA test has the advantage that it can be used with many other serolog

Completion of Paired Serological Testing Algorithms for Spotted Fever Rickettsiosis and Ehrlichiosis, North Carolina: 2017-2020.

BACKGROUND: Ehrlichia and Rickettsia are tickborne pathogens capable of causing severe disease. Paired serological testing, involving both acute and convalescent samples, remains the primary method of diagnostic confirmation and source of surveillance data. Yet, few patients complete recommended testing algorithms. METHODS: We examined the demographic, clinical, and geographic factors associated with obtainment of convalescent samples for patients with suspected ehrlichiosis and spotted fever rickettsiosis using results from a large academic center in North Carolina between 2017 and 2020. An evaluation of the clinical impact of serological testing for herpes simplex virus in a regional UK sexual health service.

UK guidelines recommend herpes simplex virus (HSV) serology in limited situations, including distinguishing recent from established infections, excluding HSV in recurrent PCR negative lesions, identifying serodiscordant partners, or determining if a pregnant person with known HSV has both types. Challenges with testing include cost, delayed results, and interpretation. We aimed to determine whether HSV serology impacted on patient management. All HSV serology results between 04/10/2021 and 12/12/2025 at our large sexual health service in northwest England were reviewed and matched with HSV PCR

Developing a multi-antigen fusion protein (Tp15-17-47) serological test to improve neurosyphilis diagnosis.

OBJECTIVES: Our previous research identified a luciferase immunosorbent assay (LISA) for detecting serum antibodies against Tp17 and Tp47 as a potential tool for diagnosing neurosyphilis (NS). In this study, we developed a serological test based on a multi-antigen fusion protein (Tp15-17-47) to simplify the detection process and improve the diagnostic accuracy for NS. METHODS: The Tp15, Tp17, and Tp47 genes were amplified separately by polymerase chain reaction (PCR) and assembled into a fusion gene coding for a tripartite fusion protein antigen using overlap PCR. This recombinant Tp15-17-47

Asymptomatic Human Infections With Avian Influenza A(H5N1) Virus Confirmed by Molecular and Serologic Testing: A Scoping Review.

IMPORTANCE: Since 1997, more than 1000 infections with highly pathogenic avian influenza A(H5N1) virus among humans have been reported globally. Given ongoing A(H5N1) outbreaks in animals, understanding the frequency of A(H5N1) virus infections among asymptomatic persons can inform public health risk assessments and infection prevention guidance. OBJECTIVE: To identify and characterize reported cases of asymptomatic A(H5N1) virus infection among humans with confirmation by both molecular testing of 1 or more respiratory specimens and 1 or more serum specimens meeting World Health Organization

Evaluating the technical feasibility of serological testing and treatment for Plasmodium vivax in mobile at-risk of malaria Cambodian populations.

BACKGROUND: Cambodia is targeting malaria elimination by 2025, aligning with the WHO's Mekong Malaria Elimination program. While elimination of Plasmodium falciparum is nearly achieved, Plasmodium vivax elimination presents challenges inherent to this species due to the occurrence of dormant parasite stages, known as hypnozoites. A new approach has been proposed to serologically identify individuals likely carrying hypnozoites that should receive appropriate antimalarial treatment: P. vivax serological testing and treatment (PvSeroTAT). This study aims to determine the technical feasibility of

COVID-19, infection fatality rate (IFR) implied by the serology, antibody, testing in New York City.

This paper estimates COVID-19 infection fatality rate (IFR) in early 2020 before pharmaceutical interventions were available on a large population in the United States. The better estimates of COVID-19 deaths in New York City and its high COVID-19 infection rate made it ideal to accurately estimate the IFR. Further, we analyze the deaths and infections in New York City to estimate an overall IFR for the United States of 0.86 percent.

A serological test based on mutated recombinant Fasciola hepatica cathepsin L protease for the diagnosis of equine fasciolosis.

Fasciola hepatica is a common trematode parasite of livestock in many regions, causing significant economic losses and affecting animal welfare. Horses rarely develop patent liver fluke infection. However, liver damage can affect animal health and welfare. Therefore, F. hepatica infection in horses may be underreported. Recently, a serological test for the antibody detection has been reported based on recombinant parasite cathepsin L1 (FhCL1) protease. Here, we optimized this enzyme-linked immunosorbent assay (ELISA) for antibody-detection in horses with equine fasciolosis based on recombinant

Serological and histolomorphological investigation of camel bulls testes (Camelus dromedaries) during the rutting and non-rutting seasons.

BACKGROUND: Camels are bred for their milk, meat, wool and hair, transportation, and their excrement as fuel. The seasonal reproduction of camel bull is accompanied by changes in sexual activity, the morphology, and function of the testes. This study aimed to evaluate the seasonal fluctuations in serum testosterone (T) levels as well as total antioxidant capacity (TAC) and malondialdehyde (MDA) levels in the testes of dromedary bulls (Camelus dromedarius) during the rutting and non-rutting seasons. Moreover, the impact of rutting season on the testicular size and histomorphology was also obser

The Epidemiology, Transmission, Genotypes, Replication, Serologic and Nucleic Acid Testing, Immunotolerance, and Reactivation of Hepatitis B Virus.

The epidemiology of Hepatitis B virus (HBV) has drastically changed in recent decades due to public health initiatives, including universal infant vaccination programs, urbanization driving global travel, and migration patterns. Despite screening of pregnant women and newborns significantly reducing the rate of perinatal transmission in certain parts of the world, other, perhaps more uncommon, routes (e.g., parenteral) have led to outbreaks in specific areas affected by the opioid epidemic and injection drug use. Although our current understanding of the effect of genetic variants of HBV is lac

Capillary driven microfluidic sequential flow device for point-of-need ELISA: COVID-19 serology testing.

A capillary-driven microfluidic sequential flow device, designed for eventual at-home or doctor's office use, was developed to perform an enzyme-linked immunosorbent assay (ELISA) for serology assays. Serology assays that detect SARS-CoV-2 antibodies can be used to determine prior infection, immunity status, and/or individual vaccination status and are typically run using well-plate ELISAs in centralized laboratories, but in this format SARs-CoV-2 serology tests are too expensive and/or slow for most situations. Instead, a point-of-need device that can be used at home or in doctor's offices fo

Evaluation of the Usefulness of a Serological Test for Diagnosis of Celiac Disease Simultaneously Detecting Specific Antibodies and Total IgA.

The diagnosis of celiac disease (CD) at the first diagnostic step requires the detection of specific class A antibodies to tissue transglutaminase type-2 (TG2 IgA) and the measurement of total immunoglobulin A (tIgA) to exclude IgA deficiency. The aim of the study was to evaluate the new quantitative immunoassay panel allowing for the detection of celiac-specific antibodies with the simultaneous determination of tIgA from the same sample of blood at one time. This retrospective study included 104 pediatric patients divided into groups with recognized CD and IgA deficiency (n = 20; 19%), immuno
