Sex- and site-specific effects of GPER-1 activation on saccharin vs cocaine preference in male and female rats.

Estradiol (E2) receptor signaling has a sex-specific impact on the brain's reward pathway, enhancing cocaine reinforcement in females but not in males. Selective activation of G-Protein Coupled Estradiol Receptor 1 (GPER-1) in the dorsolateral striatum (DLS) attenuates the reinforcing effects of 0.1 % saccharin (SACC) and cocaine in males but not females. This study investigated GPER-1 activation in the DLS and systemically using the GPER-1 agonist G1 to assess its effect on SACC and cocaine preference in male and female rats. Five experiments were conducted using gonad-intact and gonadectomiz

The biosynthesis of steroid hormones is a fascinating process . In this reaction cascade, cholesterol, a normal constituent of the lipid membrane bylayers, is coverted through a number of hydroylation, oxidation and reduction steps into a variety of biologically active substances, such as mineralocorticoids, glucocorticoids and sex hormones. Most of these modifications take place in the adrenal glands, testes and ovaries, although other tissues such as liver, kidney, placenta, brain, and skin are also quite active. The effect of steroid hormones is a complex, not yet fully understood process i

Sexual differentiation of the adolescent rodent brain: hormonal influences and developmental mechanisms.

This article is part of a Special Issue "Puberty and Adolescence". Sexual differentiation is the process by which the nervous system becomes structurally and functionally dissimilar in females and males. In mammals, this process has been thought to occur during prenatal and early postnatal development, when a transient increase in testosterone secretion masculinizes and defeminizes the developing male nervous system. Decades of research have led to the views that structural sexual dimorphisms created during perinatal development are passively maintained throughout life, and that ovarian hormon

Ontogeny of gonadotropin-inhibitory hormone cells and fibers in the brain and pituitary gland of the pejerrey fish, Odontesthes bonariensis.

The gonadotropin-inhibitory hormone is a neuropeptide belonging to the RF-amide family of peptides, first characterized in birds. This peptide can inhibit the synthesis and release of gonadotropins in both avians and mammals. Although gonadotropin-inhibitory hormone (Gnih) variants have also been detected in different fish species, knowledge of their physiological action is still inconclusive and controversial. In addition, there is very little information on the neuronal development of Gnih cells in early stages. In this context, the objective of the present study is to characterize Gnih-cont

ERα-Dependent Regulation of Adropin Predicts Sex Differences in Liver Homeostasis during High-Fat Diet.

Non-alcoholic fatty liver disease (NAFLD) represents a public health issue, due to its prevalence and association with other cardiometabolic diseases. Growing evidence suggests that NAFLD alters the production of hepatokines, which, in turn, influence several metabolic processes. Despite accumulating evidence on the major role of estrogen signaling in the sexually dimorphic nature of NAFLD, dependency of hepatokine expression on sex and estrogens has been poorly investigated. Through in vitro and in vivo analysis, we determined the extent to which hepatokines, known to be altered in NAFLD, can

The etiology of anomalous sexual preferences in men.

People discover rather than choose their sexual interests. The process of discovery typically begins before the onset of puberty and is associated with an increase in the secretion of sex hormones from the adrenal glands. However, the determinants of the direction of sexual interest, in the sense of preferences for the same or opposite sex, are earlier. These preferences, although not manifest until much later in development, appear to be caused by the neural organizational effects of intrauterine hormonal events. Variations in these hormonal events likely have several causes and two of these

Neuronal stress-coping mechanisms in postpartum females.

Animals exhibit a wide range of stress responses aimed at restoring homeostasis and promoting adaptation. In response to stress, they employ coping mechanisms to maintain physiological balance. Dysregulated stress-coping strategies have been associated with mental disorders, including depression, anxiety, and post-traumatic stress disorder. Understanding the neuronal mechanisms that regulate stress-coping is critical for elucidating normal physiological responses and addressing the pathological processes underlying these disorders. Stress responses are influenced by sex and life stage, with no

Sex-dependent serotonergic signaling across development: molecular mechanisms shaping vulnerability to neurodevelopmental and mental disorders.

Psychiatric disorders and several neurodevelopmental conditions, including autism spectrum disorder, display marked sex biases in prevalence, symptom profiles, and treatment response. Converging evidence has positioned the serotonergic system as a key organizer of the brain during development and across the lifespan; however, the principles determining when serotonin acts permissively or instructively remains unresolved. In this Review, we critically examine interventional studies across sensitive developmental and adult windows to assess whether serotonergic signaling acts in a sex-dependent

Sex and Gender-related Disparities in Clinical Characteristics and Outcomes in Heart Transplantation.

PURPOSE OF REVIEW: Limited research has been conducted on sex disparities in heart transplant (HT). The aim of this review is to analyse the available evidence on the influence of sex and gender-related determinants in the entire HT process, as well as to identify areas for further investigation. RECENT FINDINGS: Although women make up half of the population affected by heart failure and related mortality, they account for less than a third of HT recipients. Reasons for this inequality include differences in disease course, psychosocial factors, concerns about allosensitisation, and selection

Sex differences in gene regulatory networks during mid-gestational brain development.

Neurodevelopmental disorders differ considerably between males and females, and fetal brain development is one of the most critical periods to determine risk for these disorders. Transcriptomic studies comparing male and female fetal brain have demonstrated that the highest difference in gene expression occurs in sex chromosomes, but several autossomal genes also demonstrate a slight difference that has not been yet explored. In order to investigate biological pathways underlying fetal brain sex differences, we applied medicine network principles using integrative methods such as co-expression

Sex-dependent differential transcript expression in the placenta of growth restricted infants.

INTRODUCTION: The pathological decrease of fetal growth during gestation can lead to subsequent poor health outcomes for the fetus. This process is commonly controlled by the placenta, the interface between mother and baby during gestation. Sex-specific gene expression has been implicated in placental function, therefore, there is a need to determine if it is important during reduced fetal growth. We therefore aimed to characterise placental gene expression at term to evaluate sex-specific genetic changes that occur in small for gestational age (SGA) infants. METHODS: RNA-sequencing of twelve

Global and Sex-Stratified Genome-Wide Association Study of Long COVID Based on Patient-Driven Symptom Recall.

We aimed to explore the global and sex-specific genetic variants associated with long COVID, as defined by patient-driven symptom recall. A 1-year cohort study of 2411 COVID-19 patients collected long COVID symptoms with an open-ended, non-directed questionnaire, and long COVID incidence was determined according to the World Health Organization definition. Global and sex-stratified genome-wide association analyses were conducted by logistic regression models adjusted for age, sex (in the global analysis), and the first 10 principal components. We assessed sex-variant interactions and performed

Randomised controlled trial on whether advance knowledge of prostate-specific antigen testing improves participant reporting of unprotected sex.

OBJECTIVES: To determine whether the process of informing research participants that they would be tested for the presence of a biological marker of semen exposure would reduce bias in their reports of unprotected sex. METHODS: A randomised trial of 210 female sex workers from Mombasa, Kenya, was conducted, where half the group had advance knowledge (via the request for informed consent) that they would be tested for prostate-specific antigen (PSA) in their vaginal fluid before they reported on sex and condom use for the past 48 h. The other half were invited to participate (via additional in

Sex Disparities in ICH: Presentation, Care, and Outcomes in the TICH-2 Trial.

BACKGROUND: Sex-based disparities in ischaemic stroke outcomes are more widely reported than those for intracerebral haemorrhage. This study aims to examine sex differences in baseline characteristics, imaging findings, care processes, and functional outcomes following ICH, and to determine whether the treatment effect of tranexamic acid (TXA) differed by sex. METHODS: We performed a post hoc analysis of the TICH-2 trial, a multicentre, double-blind, randomised, placebo-controlled study of TXA in spontaneous ICH. Baseline characteristics and care processes were compared between men and women.

Sex-dimorphic oxytocin regulation of CA1-dependent spatial memory and synaptic plasticity in juvenile rats.

Research into memory mechanisms has predominantly centered on adult male rodents, often overlooking the influences of sex and developmental stage. Memory processes vary significantly between juveniles and adults, with sex acting as a critical determinant. Oxytocin (OXT) has emerged as a key modulator of fear responses and extinction in a sex-dependent manner, with prepubertal females displaying OXT-dependent contextual extinction patterns akin to adult males. These differences likely stem from diverse trajectories of hippocampal and prefrontal cortex maturation. This study examines the CA1 reg

Disorders of sex development: a genetic study of patients in a multidisciplinary clinic.

Sex development is a process under genetic control directing both the bi-potential gonads to become either a testis or an ovary, and the consequent differentiation of internal ducts and external genitalia. This complex series of events can be altered by a large number of genetic and non-genetic factors. Disorders of sex development (DSD) are all the medical conditions characterized by an atypical chromosomal, gonadal, or phenotypical sex. Incomplete knowledge of the genetic mechanisms involved in sex development results in a low probability of determining the molecular definition of the geneti

Cerebellar Neuromodulation Impacts Reading Fluency in Young Adults.

The cerebellum is traditionally associated with the control of coordinated movement, but ample evidence suggests that the cerebellum also supports cognitive processing. Consistent with this, right-lateralized posterolateral cerebellar regions are engaged during a range of reading and reading-related tasks, but the specific role of the cerebellum during reading tasks is not clear. Based on the cerebellar contribution to automatizing movement, it has been hypothesized that the cerebellum is specifically involved in rapid, fluent reading. We aimed to determine whether the right posterolateral cer

Sex Differences in the Serum Proteomic Profile During Acute Low Back Pain-A Preliminary Study of the Relationship to Future Low Back Pain.

The molecular processes driving the transition from acute to chronic low back pain (LBP) remain poorly understood and are likely to be sexually dimorphic. This study aimed to explore sex differences in the serum proteomic profile of people experiencing an acute LBP episode and determine if serum protein concentrations were associated with three-month outcome. Serum samples were collected through venepuncture from 30 female and 29 male participants experiencing an acute LBP episode. Serum samples underwent trypsin digestion and fractionation using hydrophobic interaction chromatography and were

The Effect of Short-Term Physical Activity on the Oxidative Stress in Rats with Different Stress Resistance Profiles in Cerebral Hypoperfusion.

Oxidative stress associated with chronic cerebral hypoperfusion is one of the fundamental factors leading to neurodegenerative diseases. To prevent oxidative stress, physical activity is effective. Physical exercise enables development of rehabilitation techniques that can progressively increase patients' stress resistance. We determined the oxidative stress dynamics in experimental hypoperfusion and modeled rehabilitation measures, comparing sex and stress resistance levels. The experiment was performed on 240 Wistar rats of both sexes over a period of 90 days. Based on behavioral test result
