A Pilot Study: Automatically Monitoring the Physical Dysfunction of Idiopathic REM Sleep Behaviour Disorder with Wearables and Deep Learning.

Idiopathic Rapid eye movement (REM) sleep behavior disorder (iRBD) is considered a marker of Parkinson's disease (PD). The diagnosis and follow-up approach is video polysomnography (vPSG), which is too expensive to be accessible to the general population, and its precision could be affected by the first-night effect. Therefore, this study explores an economic approach to continuously monitoring iRBD at home. In accordance with the findings of previous studies, which suggest that iRBD exhibits symptoms of motor dysfunction similar to PD, this study proposes automatically recognizing the physica

Dissociation of Arousal Index Between REM and NREM Sleep in Elderly Adults with Cognitive Impairment, No Dementia: A Pilot Study.

BACKGROUND: Sleep disruption in elderly has been associated with an increased risk of cognitive impairment and its transition into Alzheimer's disease (AD). High arousal indices (AIs) during sleep may serve as an early-stage biomarker of cognitive impairment non-dementia (CIND). OBJECTIVE: Using full-night polysomnography (PSG), we investigated whether CIND is related to different AIs between NREM and REM sleep stages. METHODS: Fourteen older adults voluntarily participated in this population-based study that included Mini-Mental State Examination, Neuropsi battery, Katz Index of Independenc

Couple Relationships Are Associated With Increased REM Sleep-A Proof-of-Concept Analysis of a Large Dataset Using Ambulatory Polysomnography.

Background/Objectives: Rapid Eye Movement (REM) sleep is associated with memory consolidation and several health effects including stress response, mental health, and longevity. Recently, it has been shown that regularly co-sleeping couples have increased and stabilized REM sleep when co-sleeping as compared to sleeping individually. However, it remained unclear whether this is due to a specific effect of altering the usual sleeping environment by partner deprivation or due to a generalizable REM-sleep promoting effect of couple relationships. The present study aims to clarify this ambiguity.

A Discrete Glycinergic Neuronal Population in the Ventromedial Medulla That Induces Muscle Atonia during REM Sleep and Cataplexy in Mice.

During rapid eye movement (REM) sleep, anti-gravity muscle tone and bodily movements are mostly absent, because somatic motoneurons are inhibited by descending inhibitory pathways. Recent studies showed that glycine/GABA neurons in the ventromedial medulla (VMM; GlyVMM neurons) play an important role in generating muscle atonia during REM sleep (REM-atonia). However, how these REM-atonia-inducing neurons interconnect with other neuronal populations has been unknown. In the present study, we first identified a specific subpopulation of GlyVMM neurons that play an important role in induction of

Association of probable REM sleep behavior disorder with pathology and years of contact sports play in chronic traumatic encephalopathy.

Probable rapid eye movement (REM) sleep behavior disorder (pRBD) is a synucleinopathy-associated parasomnia in which loss of REM sleep muscle atonia results in motor behavior during REM sleep, including dream enactment. Traumatic brain injury is independently associated with increased risk of pRBD and Lewy body disease, and both pRBD and Lewy body disease are often observed in chronic traumatic encephalopathy (CTE). However, the frequency and pathological substrate of pRBD in CTE have not been formally studied and remain unknown. Of the total sample of 247 men, age at death of 63.1 ± 18.8 year

A rare case of REM sleep-related bradyarrhythmia syndrome with concomitant severe hypertension: a case report and a review of literature.

INTRODUCTION: Rapid eye movement (REM) sleep-related bradyarrhythmia syndrome is characterized by pathological asystoles during the REM sleep phase. It is a rare rhythm disorder, being reported only few times in the literature. Due to non-specific symptoms, REM sleep-related bradyarrhythmia might be often underdiagnosed. Other cardiac diseases associated with pathological sinus arrests must be excluded to establish the correct diagnosis of and appropriate therapy for REM sleep-related bradyarrhythmia. We report a case of this syndrome followed by hypertension and diastolic heart failure. THE

Enhanced connectivity between visual cortex and other regions of the brain in autism: a REM sleep EEG coherence study.

Functional interregional neural coupling was measured as EEG coherence during REM sleep, a state of endogenous cortical activation, in 9 adult autistic individuals (21.1±4.0 years) and 13 typically developed controls (21.5±4.3 years) monitored for two consecutive nights in a sleep laboratory. Spectral analysis was performed on 60 s of artefact-free EEG samples distributed equally throughout the first four REM sleep periods of the second night. EEG coherence was calculated for six frequency bands (delta, theta, alpha, sigma, beta, and total spectrum) using a 22-electrode montage. The magnitude

The H1 histamine receptor blocker, chlorpheniramine, completely prevents the increase in REM sleep induced by immobilization stress in rats.

Chlorpheniramine is a selective antagonist of the H1 histaminergic receptor subtype and its effects in humans include somnolence. Chlorpheniramine affects sleep in rats, mainly by decreasing REM sleep. On the other hand, stress by immobilization induces an important increase in the percentage of REM sleep. In this study we analyzed the effects of blocking histaminergic receptors on REM sleep induced by immobilization stress. Adult male Wistar rats were chronically implanted for sleep recording. Immobilization stress was induced by placing the rat in a small cylinder for 2 h. Experimental condi

Home-Based REM Sleep Without Atonia in Patients with Parkinson's Disease: A Post Hoc Analysis of the ZEAL Study.

REM sleep behavioral disorder (RBD) is of increasing interest in Parkinson's disease (PD). Previous studies exploring the association between REM sleep without atonia (RWA) and clinical PD features or other objective sleep metrics are scarce and have used PSG findings. A mobile electroencephalography (EEG)/electrooculography (EOG) recording system with two channels can objectively measure sleep parameters, including RWA, during natural sleep at home. We investigated whether RWA measured on a portable recording device at home could be associated with clinical PD features or other sleep metrics

The effect of citalopram treatment on memory performance, mood state, catalase, BDNF, and MDA in REM sleep-deprived rats.

Rapid-eye movement (REM) sleep is a critical stage of sleep that plays a pivotal role in memory formation, consolidation, and mood regulation. While REM sleep deprivation (REM SD) has been shown to impair cognitive and affective functions, findings across studies remain inconsistent. REM SD may also influence the expression of brain-derived neurotrophic factor (BDNF), a key protein involved in memory and mood-related processes. In this study, we investigated the effects of REM SD (6 h/day for 7 days) on behavioral and cognitive performance, BDNF levels in the prefrontal cortex, and serum marke

The Interplay Between Emotion Dysregulation and Repetitive Thoughts in Insomnia Disorder: The Impact of Worry, Rumination and REM Sleep Instability.

While previous research has identified emotional dysregulation and repetitive thinking as contributors to insomnia, the interplay between these factors remains unclear. Building upon data previously collected in our laboratory, this exploratory study extends prior findings by examining the mediating role of rumination, worry and rapid eye movement (REM) sleep instability in the relationship between emotional dysregulation and the functional impact of insomnia and depressive symptoms, aiming to generate hypotheses about the psychological and neurophysiological mechanisms linking these construct

A FAM8A1 frameshift variant is associated with REM sleep behavior disorder, urinary retention, and mydriasis in Russian Blue cats.

REM sleep behavior disorder (RBD) is a disease characterized by the loss of lower motor neuron inhibition responsible for skeletal muscle atonia during REM sleep. It has been reported in humans, dogs and cats, and can be idiopathic or secondary to a neurodegenerative disease. Five young adult Russian Blue cats from two related families were presented for progressively worsening RBD episodes frequently associated with urinary loss. Three of these cats also suffered urinary retention with overflow incontinence between RBD episodes. Neurological examination revealed a large bladder in three cats

The protective effect of bromelain on testicular dysfunction and oxidative stress induced by REM sleep deprivation in adult male rats.

Male infertility is a significant global health concern, with sleep deprivation emerging as a potential contributing factor due to its impact on oxidative stress and reproductive function. This study investigates the protective effects of bromelain, a natural anti-inflammatory enzyme, on testicular dysfunction induced by Rapid eye movement (REM) sleep deprivation in adult male rats. Thirty-six rats were divided into control, REM sleep-deprived (REM-dep), and REM-deprived with bromelain treatment (REM-dep+Br) groups. REM sleep deprivation was induced for a 21-day period using the flower pot tec

Free-Water Imaging of the Nucleus Basalis of Meynert in Patients With Idiopathic REM Sleep Behavior Disorder and Parkinson Disease.

BACKGROUND AND OBJECTIVES: Cognitive impairments are common in idiopathic REM sleep behavior disorder (iRBD), in which the cholinergic degeneration of nucleus basalis of Meynert (NBM) may play an important role. However, the progressive changes of NBM, the relationship between progressive NBM degeneration and progression of cognitive impairments, and whether degeneration of the NBM can predict cognitive decline in patients with iRBD remain unclear. This study aimed to investigate the cross-sectional and longitudinal microstructural alterations in the NBM of patients with iRBD using free-water

Heart rate variability-based Model for estimating the severity of orthostatic hypotension in patients with REM sleep behavior disorder.

BACKGROUND: Orthostatic hypotension (OH) is a typical autonomic dysfunction in patients with idiopathic REM sleep behavior disorder (iRBD). The Schellong test is a well-known method to evaluate the presence of OH; however, it is burdensome for both patients and the medical staff from the viewpoint of ensuring patient safety. In this study, we developed a machine learning (ML) model to discriminate the presence or absence of OH based on heart rate variability (HRV) in the supine position. METHODS: We recruited elderly healthy participants (HC) and iRBD patients and measured participants' R-R i

The Kir channel in the nucleus tractus solitarius integrates the chemosensory system with REM sleep executive machinery for homeostatic balance.

The locus coeruleus (LC), nucleus tractus solitarius (NTS), and retrotrapezoid nucleus (RTN) are critical chemosensory regions in the brainstem. In the LC, acid-sensing ion channels and proton pumps serve as H+ sensors and facilitate the transition from non-rapid eye movement (NREM) to rapid eye movement (REM) sleep. Interestingly, the potassium inward rectifier (KIR) channels in the LC, NTS, and RTN also act as H+-sensors and are a primary target for improving sleep in obstructive sleep apnea and Rett syndrome patients. However, the role of Kir channels in NREM to REM sleep transition for H+

Targeting retrieval of methamphetamine reward memory in the context of REM sleep deprivation: Age-dependent role of GABAB receptors.

GABAB receptors play a modulatory role in the mechanisms underlying drug addiction, sleep problems, and aging; however, there are few studies addressing their relationships to each other. Therefore, this study aimed to examine whether blockade of these receptors affects methamphetamine (METH) reward memory in adult and adolescent rapid-eye movement sleep-deprived (RSD) rats. Adolescent and adult male Wistar rats were subjected to RSD for seven days. They were then conditioned to receive methamphetamine (METH; 2 mg/kg, ip) during an eight-day conditioning period. METH reward memory was then rea

NPC1 variants are not associated with Parkinson's disease, REM-sleep behavior disorder or dementia with Lewy bodies in European cohorts.

NPC1 encodes a lysosomal protein involved in cholesterol transport. Biallelic mutations in this gene may lead to Niemann-Pick disease type C (NPC), a lysosomal storage disorder. The role of NPC1 in alpha synucleinopathies is still unclear, as different genetic, clinical, and pathological studies have reported contradictory results. This study aimed to evaluate the association of NPC1 variants with the synucleinopathies Parkinson's disease (PD), dementia with Lewy bodies (DLB), and rapid eye movement-sleep behavior disorder (RBD). We analyzed common and rare variants from 3 cohorts of European
