[Alternative sources of hepatocytes for cell therapy].

There is an urgent need to search for alternatives to whole organ transplantation. Several methods have been proposed. Among these strategies, cell transplantation is currently one of the most promising. To achieve this aim, in addition to highly differentiated adult hepatocytes, the use of stem cells is considered a highly attractive therapeutic method for the treatment of liver disease and for temporary support of hepatic function until a liver becomes available for organ transplantation. This strategy is based on the ability of stem cells to differentiate into different cellular types accor

Present and future cell therapies for pancreatic beta cell replenishment.

If only at a small scale, islet transplantation has successfully addressed what ought to be the primary endpoint of any cell therapy: the functional replenishment of damaged tissue in patients. After years of less-than-optimal approaches to immunosuppression, recent advances consistently yield long-term graft survival rates comparable to those of whole pancreas transplantation. Limited organ availability is the main hurdle that stands in the way of the widespread clinical utilization of this pioneering intervention. Progress in stem cell research over the past decade, coupled with our decades-…

[Advances in stem cell and exosome-based therapies for dry eye disease].

Conventional treatments for dry eye disease, such as artificial tears and anti-inflammatory drugs, can only alleviate symptoms, and it is difficult to achieve etiological repair with these treatments. In recent years, stem cell and exosome-based therapies have emerged as promising research hotspots for dry eye disease treatment, owing to their immunomodulatory, tissue-regenerative, and anti-inflammatory properties. Multiple basic and clinical studies have preliminarily confirmed the therapeutic potential of stem cells and exosomes in treating dry eye disease. This review summarizes recent adva

[Research progress on corneal organoids].

Corneal organoids are cornea-like tissue structures that are developed from induced pluripotent stem cells or embryonic stem cells in vitro. They have similar anatomical characteristics and gene expression profiles to the human cornea. Although the culture of corneal organoids is difficult, expensive, and time-consuming, they can be tailored to reconstruct the physiological environment and the interaction between corneal cells in each layer, which suggests broad application prospects. In this article, we review the latest research progress and summarize the application of corneal organoids in

The impact of stem cell secretome, exosomes, and photobiomodulation on enhancing diabetic wound recovery.

Chronic wounds, such as diabetic ulcers, represent a substantial clinical difficulty because of impaired healing, often resulting from vascular and nerve damage associated with hyperglycemia. Traditional treatments primarily offer supportive care but frequently fail to fully restore tissue function, highlighting the urgent need for advanced regenerative strategies. Stem cell-based therapies, especially those leveraging secretome and exosomes, have gained attention as effective alternatives to direct stem cell transplantation. These cell-free approaches offer several advantages, including low i

High-dose chemotherapy with hematopoietic stem cell transplantation for the treatment of primary central nervous system lymphoma.

Primary central nervous system lymphoma (PCNSL) is a rare B-cell lymphoid neoplasm for which current regimens utilizing standard-dose chemotherapy and/or radiation therapy lead to high relapse rates and/or unacceptable neurologic sequelae. High-dose chemotherapy followed by hematopoietic stem cell transplantation may overcome limitations of current treatment schemas. A search was performed of all English-language literature (1968 to June 2009) within the MEDLINE, EMBASE and Cochrane Library databases to identify relevant clinical trials using the terms stem cell transplantation, bone marrow tr

Stem cell model of spinal muscular atrophy.

Human embryonic stem cells provide a useful source of material for studying basic human development and various disease states. However, ethical issues concerning their procurement limit their acceptance and possible clinical applicability. Recent advances in stem cell technology have provided an alternative source of pluripotent stem cells that does not require the use of an embryo. This review addresses the generation of induced pluripotent stem cells from skin fibroblasts taken from various patient populations, with a specific focus on the pediatric disorder spinal muscular atrophy. These p

The application of regenerative medicine in colorectal surgery.

Tissue reconstruction and regeneration represent one of the greatest challenges in any surgical field. Regenerative medicine combined with stem cell-based therapy is a novel and promising field of medicine. Stem cells possess the ability to differentiate into specialized cells and to decrease inflammation and therefore can play a role in repair or regeneration of damaged tissues. Colorectal surgery often deals with infected, poorly vascularized, radiated, and inflamed tissue, as well as instances where imperfect healing might have grave implications. This problem has led researchers to study u

Comprehensive analysis of the value of angiogenesis and stemness-related genes in the prognosis and immunotherapy of ovarian cancer.

Tumor angiogenesis and the presence of cancer stem cells (CSCs) are critical characteristics of tumors. Previous research has demonstrated that cancer stem cells promote tumor angiogenesis, while increased vascularity, in turn, fosters the growth of cancer stem cells. This creates a detrimental cycle that contributes to tumor progression. However, studies investigating the angiogenesis and stemness characteristics in ovarian cancer (OV) are limited. In this study, we employed cluster analysis and LASSO methods to assess the significance of angiogenesis- and stemness-related genes in the effica

Human dental pulp stem cells for spinal cord injury.

Spinal cord injury (SCI) is a serious neurological disorder that causes loss of mobility, pain, and autonomic dysfunction, resulting in altered sensation and devastating loss of function. Current treatments for SCI mainly focus on surgery and drug therapy to promote neurological recovery. However, there are virtually no effective remedies for irreversible nerve damage that result in a victim's loss of motor function and sensory changes that occur after an injury. With the continuous development of medical technology, stem-cell-based regenerative medicine provides researchers with new treatment

Stem Cell Niches for Olfactory Regeneration and Their Therapeutic Applications.

Olfactory dysfunction affects many individuals and may result from infections, trauma, neurodegenerative diseases, or genetic disorders. Unlike most neurons in the mammalian nervous system, olfactory sensory neurons (OSNs) continuously regenerate throughout life, a process facilitated by specialized stem cell niches in the olfactory epithelium. Horizontal basal cells are typically dormant under normal conditions but become activated following injury, differentiating into globose basal cells (GBCs) and other cell types. GBCs serve as the primary source for OSN regeneration and are regulated by

Neuroinflammation and Graft Survival in Pluripotent Stem Cell-Derived Therapies for the Central Nervous System.

Recently, Japan conditionally approved the first cell transplantation therapy for a central nervous system disorder using induced pluripotent stem cells as the source of dopaminergic progenitors to treat Parkinson's disease. This milestone was achieved almost 40 years after the first cell therapy trial for Parkinson's disease in Lund, Sweden, and 18 years after the first clinical trial using oligodendrocyte progenitors derived from pluripotent stem cells for spinal cord injury. In this review, we update the current state of development of cell therapies based on pluripotent stem cells for dise

Properties of skin stem cells and their potential clinical applications in modern dermatology.

Stem cells play an important role in medical science, and scientists are investing large sums in order to perform sophisticated studies designed to establish potential clinical applications of stem cells. Growing experience has enabled researchers to determine the precise nature of stem cell division. Although the properties of this particular population of cells have been known and used for some time, mainly with regards to bone marrow-derived mesenchymal stem cell transplantation, we now face a significant challenge in implementing the practical use of skin-derived precursors, making it poss

[Stem cells--potential therapeutic use in neurological diseases].

There is a growing interest in medical potential of stem cells which may be used someday to create new tissue. Suggested applications involve a board spectrum ranging from the replacement for cells destroyed by diseases to even organ transplants. As ethical and legal controversy makes uncertain the future of embryonic stem cells research, attention has been recently turned to adult stem cells to discover whether they also can serve to transplantation. In this article we present data indicating that adult stem cells found in areas of the body like blood, skin, lymph and nervous systems, may be
