Morphogenetic alterations during endocardial cushion development in the trisomy 16 (Down syndrome) mouse.

Atrioventricular septal defect occurs with a high prevalence in both human Down syndrome (trisomy 21) and the animal model for this disorder, murine trisomy 16 (Ts-16). The embryologic basis of this defect is the failure of the endocardial cushions to fuse. Quantitatively, Ts-16 hearts, when compared to normal mouse embryos, were not significantly different in either the estimates of whole heart volume or endocardial cushion volume. However, both the raw number of cardiac mesenchyme cells and the cellular density were reduced significantly. Qualitatively, endocardial cushion shape was elongate

Immunophenotypic and structural signatures of severe granulation tissue in airway implant patients.

BACKGROUND: Granulation tissue formation is a frequent complication following airway implant procedures such as endobronchial valve (EBV) therapy in patients with chronic obstructive pulmonary disease (COPD). This hyperplastic response can impair implant function and clinical outcomes, yet the underlying immunological mechanisms remain poorly understood. METHODS: In a cohort of EBV patients (n = 147), we developed an optical scaling and scoring system to classify the severity of granulation tissue in relation to clinical outcomes. Based on this system, we prospectively collected endobronchial

Sequential vascular and neural changes underlying the progression of patellar tendinopathy.

PURPOSE: To investigate the temporal sequence of vascular, neural, and extracellular matrix (ECM) changes during the progression of collagenase-induced patellar tendinopathy and to test the hypothesis that early angiogenesis initiates a cascade leading to neural remodeling and tendon degeneration. METHODS: A collagenase-induced rat patellar tendinopathy model was established in 40 male Sprague-Dawley rats. Animals were divided into one control group (PBS injection, n = 8, sacrificed at 12 weeks) and four experimental groups (collagenase type I injection, n = 8 each, sacrificed at 2, 4, 8, and

All-Trans Retinoic Acid and Curcumin Exhibit Hormesis or Synergistic Anticancer Effects in U87 Glioblastoma Cells: Defining the Proteome Accompanying Synergism.

Persistently poor glioblastoma (GBM) survival necessitates better elucidation of tumor drug responses. After observing that low curcumin and all-trans retinoic acid (ATRA) doses stimulated cell proliferation and counteracted each other's high-dose antiproliferative effects in U87 GBM cells, drug influences on cell growth, migration, and death and the antiproliferative interaction proteome were further studied. Cell proliferation and migration were assessed by xCELLigence Real-Time Cell Analysis (RTCA). Cell death was defined using flow cytometry. Drug interactions were determined with CompuSyn

Temporal Gene Expression Changes in Rotator Cuff Tendon Injury and Repair.

Rotator cuff tendon injuries are common, particularly among the elderly. However, promising biomarkers to assess tendon degeneration and healing remain limited. This study aimed to identify dynamic biomarkers by analyzing temporal gene expression and histological changes in a chronic rotator cuff tear model. Using Sprague-Dawley rats, a chronic injury was induced by detaching the supraspinatus tendon and inserting a silastic drain to inhibit spontaneous healing. Tendon samples were collected at multiple time points after injury and after repair for gene expression profiling and histological an

Single cell RNA-seq characterization of non-fibrotic stromal wound repopulation in the rabbit.

Following injury or surgery, quiescent stromal keratocytes can transition into fibroblasts or myofibroblasts leading to either transient or protracted corneal haze. In this study, we investigate the transcriptional changes associated with non-fibrotic wound healing using a transcorneal freeze injury (FI) in the rabbit, which induces full-thickness stromal cell loss without inducing keratocyte-myofibroblast transformation. In control corneas, scRNA-seq revealed multiple clusters expressing markers associated with keratocyte identity (e.g. KERA, LUM, DCN, and ALDH1A1), suggesting heterogeneity i

A gene expression and a histostructural analysis of the palmar fascia of patients affected by Dupuytren's disease.

PURPOSE/AIM: Dupuytren's disease (DD) is a condition affecting the palmar fascia that may reduce the mobility of several fingers. Despite its clinical relevance, the genetic and structural mechanisms associated with this disease are still not well understood. In this work, we have carried out a genome-wide gene expression analysis to identify relevant genes associated with DD. MATERIALS AND METHODS: A genome-wide gene expression analysis was carried out using next generation sequencing (NGS) followed by a histological, histochemical and immunohistochemical analysis of some major components of

Cell-type specific effects of mineralocorticoid receptor gene expression suggest intercellular communication regulating fibrosis in skeletal muscle disease.

Introduction: Duchenne muscular dystrophy (DMD) is a fatal striated muscle degenerative disease. DMD is caused by loss of dystrophin protein, which results in sarcolemmal instability and cycles of myofiber degeneration and regeneration. Pathology is exacerbated by overactivation of infiltrating immune cells and fibroblasts, which leads to chronic inflammation and fibrosis. Mineralocorticoid receptors (MR), a type of nuclear steroid hormone receptors, are potential therapeutic targets for DMD. MR antagonists show clinical efficacy on DMD cardiomyopathy and preclinical efficacy on skeletal muscl

Expression of intramuscular extracellular matrix proteins in vastus lateralis muscle fibres between atrophic and non-atrophic COPD.

BACKGROUND: Extracellular matrix (ECM) proteins are the major constituents of the muscle cell micro-environment, imparting instructive signalling, steering cell behaviour and controlling muscle regeneration. ECM remodelling is among the most affected signalling pathways in COPD and aged muscle. As a fraction of COPD patients present muscle atrophy, we questioned whether ECM composition would be altered in patients with peripheral muscle wasting (atrophic COPD) compared to those without muscle wasting (non-atrophic COPD). METHODS: A set of ECM molecules with known impact on myogenesis were qua

Serum biomarkers and their relationship to axial spondyloarthritis associated with inflammatory bowel diseases.

Spondyloarthritis (SpA) constitute a group of chronic inflammatory immune-mediated rheumatic diseases characterized by genetic, clinical, and radiological features. Recent efforts have concentrated on identifying biomarkers linked to axial SpA associated with inflammatory bowel disease (IBD), offering predictive insights into disease onset, activity, and progression. Genetically, the significance of the HLA-B27 antigen is notably diminished in ankylosing spondylitis (AS) associated with IBD, but is heightened in concurrent sacroiliitis. Similarly, certain polymorphisms of endoplasmic reticulum

Comparative transcriptional profiling of regenerating damaged knee joints in two animal models of the newt Notophthalmus viridescens strengthens the role of candidate genes involved in osteoarthritis.

OBJECTIVES: To compare joint regeneration in adult newts (N. viridescens) upon both newly established surgical removal and previously reported enzymatic destruction of articular cartilage to identify molecular factors and functionally analyze potentially important regulators involved in osteoarthritis (OA). METHODS: Damage of knee cartilage was induced by either intra-articular injection of collagenase or by surgical removal of articular cartilage as a novel additional approach. Changes over time were clinically and histologically analyzed and studied by cDNA microarray analysis, real-time qu

Identification of the extracellular matrix protein Fibulin-2 as a regulator of spinal nerve organization.

During amniote peripheral nervous system development, segmentation ensures the correct patterning of the spinal nerves relative to the vertebral column. Along the antero-posterior (rostro-caudal) axis, each somite-derived posterior half-sclerotome expresses repellent molecules to restrict axon growth and neural crest migration to the permissive anterior half-segment. To identify novel regulators of spinal nerve patterning, we investigated the differential gene expression of anterior and posterior half-sclerotomes in the chick embryo by RNA-sequencing. Several genes encoding extracellular matri

Bone matrix, cellularity, and structural changes in a rat model with high-turnover osteoporosis induced by combined ovariectomy and a multiple-deficient diet.

In estrogen-deficient, postmenopausal women, vitamin D and calcium deficiency increase osteoporotic fracture risk. Therefore, a new rat model of combined ovariectomy and multiple-deficient diet was established to mimic human postmenopausal osteoporotic conditions under nutrient deficiency. Sprague-Dawley rats were untreated (control), laparatomized (sham), or ovariectomized and received a deficient diet (OVX-Diet). Multiple analyses involving structure (micro-computed tomography and biomechanics), cellularity (osteoblasts and osteoclasts), bone matrix (mRNA expression and IHC), and mineralizat

Periosteal Sharpey's fibers: a novel bone matrix regulatory system?

Sharpey's "perforating" fibers (SF) are well known skeletally in tooth anchorage. Elsewhere they provide anchorage for the periosteum and are less well documented. Immunohistochemistry has transformed their potential significance by identifying their collagen type III (CIII) content and enabling their mapping in domains as permeating arrays of fibers (5-25 μ thick), protected from osteoclastic resorption by their poor mineralization. As periosteal extensions they are crucial in early skeletal development and central to intramembranous bone healing, providing unique microanatomical avenues for

Proteomic identification of matrix metalloproteinase substrates in the human vasculature.

BACKGROUND: Matrix metalloproteinases (MMPs) play a key role in cardiovascular disease, in particular aneurysm formation and plaque rupture. Surprisingly, little is known about MMP substrates in the vasculature. METHODS AND RESULTS: We used a proteomics approach to identify vascular substrates for 3 MMPs, 1 of each of the 3 major classes of MMPs: Human arteries were incubated with MMP-3 (a member of stromelysins), MMP-9 (considered a gelatinase), and MMP-14 (considered a member of the collagenases and of the membrane-bound MMPs). Candidate substrates were identified by mass spectrometry based

MED and PSACH COMP mutations affect chondrogenesis in chicken limb bud micromass cultures.

Mutations in cartilage oligomeric matrix protein (COMP) cause pseudoachondroplasia (PSACH) and multiple epiphyseal dysplasia (MED). We studied the effects of over-expression of wild type and mutant COMP on early stages of chondrogenesis in chicken limb bud micromass cultures. Cells were transduced with RCAS virus harboring wild type or mutant (C328R, PSACH; T585R, MED) COMP cDNAs and cultured for 3, 4, and 5 days. The effect of COMP constructs on chondrogenesis was assessed by analyzing mRNA and protein expression of several COMP binding partners. Cell viability was assayed, and evaluation of

Expression and activation of TGF-beta isoforms in acute allergen-induced remodelling in asthma.

BACKGROUND: Airway wall remodelling and inflammation are features of chronic asthma. Transforming growth factor beta (TGF-beta) has been implicated in these processes. AIM: To determine the effect of allergen challenge on airway inflammation and remodelling and whether TGF-beta isoforms and the Smad signalling pathways are involved. METHODS: Thirteen patients with atopic asthma underwent inhalational challenge with 0.9% saline, followed by allergen 3-4 weeks later. After both challenges, fibreoptic bronchoscopy was undertaken to obtain bronchial biopsies and tissue samples were processed for
