Development of resistance to antibiotics is a major problem worldwide. The normal oropharyngeal flora, the intestinal flora and the skin flora play important roles in this development. Within a few days after the onset of antibiotic therapy, resistant Escherichia coli, Haemophilus influenzae and Staphylococcus epidermidis can be detected in the normal flora of volunteers or patients. Horizontal spread of the resistance genes to other species, e.g. Salmonella spp., Staphylococcus aureus and Streptococcus pneumoniae, occurs by conjugation or transformation. An ecologically sound antibiotic polic

Nocardioform actinomycetes are Gram-positive bacteria with high G+C content. Several species of Mycobacterium, Corynebacterium, Nocardia and Rhodococcus are important human or animal pathogens. Transposon mutagenesis and homologous recombination are powerful genetic tools used for the identification of pathogenicity determinants. Transposable elements with potential application to mutagenize pathogenic nocardioform actinomycetes are described. Homologous recombination experiments which have been recently achieved in mycobacteria and related actinomycetes are commented on.

Borrelia spp. are transmitted to humans by the bite of an infected tick. In Europe, Borrelia afzelii and Borrelia garinii are the main causative agents of Lyme borreliosis, one of the most prevalent tick-borne diseases in the northern hemisphere. In bacteria such as Borrelia spp., a restriction-modification system (RMS) protects against the harmful introduction of foreign DNA. The RMS comprises two activities: methyltransferase and endonuclease. This study is aimed to characterize the RMS of B. afzelii and B. garinii. First, we identified potential RMS genes. The predicted genes were cloned in

Research on biofuel production by cyanobacteria has largely focused on unicellular species. To date, there are no reports on engineered filamentous cyanobacteria for bioethanol or biohydrogen production. However, biofilm formation in filamentous cyanobacteria could provide an advantage for biotechnological applications. We used natural transformation and homologous recombination to establish recombinant expression at the psaD locus of the filamentous cyanobacterium Phormidium lacuna. A similar approach has already been successful in the unicellular cyanobacterium Synechocystis sp. PCC 6803 (Ap

Streptococcus pneumoniae (pneumococcus) remains a significant cause of pneumonia, bacteremia, sepsis, meningitis, and otitis media. The ability of pneumococcus to enter a state of genetic competence is central to pneumococcal adaptability and virulence. Activation of the competence state enables pneumococcus to uptake exogenous DNA for genetic transformation, acquire genome plasticity and antimicrobial resistance, while also regulating stress tolerance, biofilm formation, and coordinated expression of virulence factors. This review summarizes current evidence on competence-regulated virulence

Mycoplasma synoviae, an important poultry pathogen, belonging to the class Mollicutes, causes airsacculitis, synovitis, decreased egg production and produces significant economic losses. Efforts to determine M. synoviae virulence factors and their role in pathogenicity require suitable tools for genetic manipulation of this pathogen. This study describes, for the first time, the identification and cloning of the origin of replication (oriC) of M. synoviae to develop a replicable oriC vector for this mycoplasma. Shuttle vectors containing different putative oriC regions along with tetracycline

Valsa mali is a causal agent of apple and pear trees canker disease, which is a destructive disease that causes serious economic losses in eastern Asia, especially in China. The lack of an efficient transformation system for Valsa mali retards its investigation, which poses difficulties to control the disease. In this research, a transformation system for this pathogen was established for the first time using A. tumefaciens-mediated transformation (ATMT), with the optimal transformation conditions as follows: 10(6)/mL conidia suspension, cocultivation temperature 22°C, cocultivation time 72 ho

Bacterial exopolysaccharides have always been suggested to play crucial roles in the bacterial initial adhesion and the development of complex architecture in the later stages of bacterial biofilm formation. However, Escherichia coli group II capsular polysaccharide was characterized to exert broad-spectrum biofilm inhibition activity. In this study, we firstly reported that a bacterial exopolysaccharide (A101) not only inhibits biofilm formation of many bacteria but also disrupts established biofilm of some strains. A101 with an average molecular weight of up to 546 KDa, was isolated and puri

Attenuated total reflection Fourier transform infrared spectroscopy has been used to study the secondary structure of anthrax lethal toxin proteins: protective antigen (PA) and lethal factor (LF), as a function of pH in the absence and in the presence of phospholipid vesicles. We first characterized the binding of LF and PA to the lipid membrane and demonstrated the strong pH dependence of the association of PA and LF to the lipid bilayer as well as the effect of pH neutralization on this binding. Binding of LF to the lipid membrane can be, at least partially, reversed when the pH is brought t

To more effectively study the genetic basis of Legionnaires' disease, we characterized a system for mini-Tn10 mutagenesis in Legionella pneumophila. The mini-transposons were first electroporated into Legionella on counterselectable vectors expressing altered target site transposases. Then, by simultaneously selecting for the kanamycin-resistance gene within the transposon and counterselecting against the maintenance of the plasmid, we directly and readily isolated strains bearing single chromosomal insertions. Southern hybridization analysis further demonstrated that the insertions were rando

Of 46 viridans streptococci isolated from cases of probably true bacteremia, 26 strains or 56% could be grouped serologically in the Lancefield system. Eight strains (17%) were identified as serogroup H (characteristic of Streptococcus sanguis), ten strains (22%) as serogroup W (a new group antigen found in S. sanguis) and eight strains (17%) as serogroup K (related to S. mitis (mitior)). The high number of serogroupable viridans streptococci was specially due to the introduction of serogroup W and indicates the importance of this serogroup in cases of bacteremia. A set of six diagnostic tests

The genetic locus of staphylococcal enterotoxin B (SEB) was investigated in the Staphylococcus aureus food-poisoning isolates, strains S6 and 277. Direct neutral sucrose gradient centrifugation analysis of sodium dodecyl sulfate-sodium chloride-mediated cleared lysates demonstrated that strain S6 contained a single 37S plasmid. Transductional analysis revealed that the 37S plasmid in S6 encoded for cadmium resistance (Cad) but not SEB. Additionally, elimination of cadmium resistance in S6 provided a plasmid-negative derivative that produced SEB at the same level as the parent. Examination of s

Bacterial species demonstrate remarkable adaptability in hostile environments and develop resistance to multiple antibacterial molecules, creating significant challenges for infection treatment in clinical settings. At the core of this challenge lies peptidoglycan, a key component in cell wall synthesis that provides both structural integrity and environmental protection for bacterial cells. The peptidoglycan biosynthesis pathway operates across three distinct cellular compartments: the cytoplasm, membrane, and periplasmic space. The process initiates in the cytoplasm with nucleotide precursor

A common vaginal dysbiosis, bacterial vaginosis (BV), has serious consequences for reproductive health, including an elevated risk of infertility and pelvic inflammatory disease. Its distinctive feature is a robust polymicrobial biofilm, mainly composed of Gardnerella species (spp.), which protects infections against antibiotic therapy and promotes high rates of recurrence. Concurrently, Chlamydia trachomatis (CT) is the most common bacterial sexually transmitted infection globally, with over 130 million new cases annually. A primary cause of tubal factor infertility, CT infection promotes adn

The causal agent of tomato leaf mold, Cladosporium fulvum, is prevalent in greenhouses worldwide, especially under high humidity conditions. Despite its economic impact, studies on antifungal agents targeting C. fulvum remain limited. This study evaluates biocontrol agents (BCAs) as alternatives to chemical controls for managing this disease, alongside the strobilurin fungicide azoxystrobin. From a Moroccan collection of potential BCAs, five bacterial isolates (Alcaligenes faecalis ACBC1, Pantoea agglomerans ACBC2, ACBP1, ACBP2, and Bacillus amyloliquefaciens SF14) and three fungal isolates (T

The simultaneous development of antibiotic resistance in bacteria due to metal exposure poses a significant threat to the environment and human health. This study explored how exposure to both arsenic and antibiotics affects the ability of an arsenite oxidizer, Achromobacter xylosoxidans CAW4, to transform arsenite and its antibiotic resistance patterns. The bacterium was isolated from arsenic-contaminated groundwater in the Chandpur district of Bangladesh. We determined the minimum inhibitory concentration (MIC) of arsenite, cefotaxime, and tetracycline for A. xylosoxidans CAW4, demonstrating

INTRODUCTION: Changes in the gut microbiota of patients with partial small intestinal obstruction (PSIO) have not been widely clarified. We aimed to explore bacterial diversity in a PSIO mouse model.
METHODS: A PSIO mouse model was established using male C57BL/6 mice, and feces samples from the distal ileum and ileum epithelium tissues were collected. MiSeq sequencing of the 16S rRNA gene was conducted to characterize microbiota diversity and composition. RNA sequencing for differences in transcriptomic programming of the ileum tissue was performed between the PSIO and (Control) Ctrl groups.
