The liquorice root derivative glycyrrhetinic acid can ameliorate ionoregulatory disturbance in rainbow trout (Oncorhynchus mykiss) abruptly exposed to ion-poor water.

To consider the idea that a dietary botanical supplement could act as an adaptogen in a teleost fish, the effect of a liquorice root derivative (18β-glycyrrhetinic acid, 18βGA) on rainbow trout following an acute ionoregulatory stressor was examined. Freshwater (FW) trout were fed a control or 18βGA supplemented diet (0, 5, or 50μg 18βGA/g diet) for 2weeks, then abruptly exposed to ion-poor water (IPW) for 24h. Following IPW exposure, muscle moisture content and serum cortisol levels elevated and serum [Na(+)] and/or [Cl(-)] reduced in control and 50μg/g 18βGA-fed fish. However, these endpoint…

Immune responses to oral pcDNA-VP2 vaccine in relation to infectious pancreatic necrosis virus carrier state in rainbow trout Oncorhynchus mykiss.

The VP2 gene of infectious pancreatic necrosis virus, encoded in an expression plasmid and encapsulated in alginate microspheres, was used for oral DNA vaccination of fish to better understand the carrier state and the action of the vaccine. The efficacy of the vaccine was evaluated by measuring the prevention of virus persistence in the vaccinated fish that survived after waterborne virus challenge. A real-time RT-qPCR analysis revealed lower levels of IPNV-VP4 transcripts in rainbow trout survivors among vaccinated and challenged fish compared with the control virus group at 45 days post-inf…

Cardiorespiratory adjustments during hypercarbia in rainbow trout Oncorhynchus mykiss are initiated by external CO(2) receptors on the first gill arch.

Experiments were performed to test the hypothesis that the marked ventilatory and cardiovascular responses to hypercarbia in rainbow trout Oncorhynchus mykiss arise from specific stimulation of chemoreceptors localised to the first gill arch. This was accomplished by measuring cardiorespiratory variables during acute hypercarbia (20 min at P(CO(2))=8 mmHg; 1 mmHg=0.133 kPa) in fish subjected to selective bilateral extirpation of the first gill arch. The cardiovascular responses to hypercarbia in the intact fish included a significant bradycardia (from 75.0+/-1.6 to 69.0+/-2.0 beats min(-1); me…

Detection of estrogen receptor mRNA in trout pineal and retina: estradiol-17 beta modulates melatonin production by cultured pineal photoreceptor cells.

Pineal photoreceptor cells produce the neurohormone melatonin, a major "Zeitgeber" of the organism. This compound is involved in the control of development, growth, sexual maturation, and seasonal reproductive cycles. The present study reports that the photosensitive pineal organ and the retina of the trout express a 3.5-kb mRNA corresponding to the estradiol-17 beta receptor. The effects of estradiol-17 beta on melatonin production by cultured pineal photoreceptor cells were also studied. Under a light/dark (LD:12/12) cycle, these cells maintained a rhythmic secretion of melatonin, with highe…

Pineal gland hydroxyindole-O-methyl transferase (HIOMT) from rainbow trout (Salmo gairdneri) subject to temperature-dependent substrate inhibition by N-acetylserotonin.

The effect of different incubation temperatures on the activity of trout hydroxyindole-O-methyltransferase (HIOMT) was determined in order to study the potential effects of changes in environmental temperature on production of melatonin. In the study, N-acetylserotonin (NAS) was found to inhibit trout pineal HIOMT, inhibitory concentration decreasing with a decrease in incubation temperature. An increase in incubation temperature was accompanied by an increase in maximal velocity (Vm) and a decrease in affinity of the substrate NAS for the enzyme, while the affinity of the inhibitor NAS remain…

Combined amino acid mutations in the glycoprotein of infectious hematopoietic necrosis virus attenuate virulence and confer strong immunogenicity in rainbow trout.

Infectious hematopoietic necrosis virus (IHNV) is a major threat to the salmonid aquaculture industry, causing mortality rates exceeding 90% in juvenile salmonids. Our previous study showed that single-site mutations at amino acid residues 67, 107, 221 or 256 of the IHNV G protein reduced IHNV virulence, and immunization of rainbow trout with attenuated mutants improved survival after challenge with wild-type (wt) IHNV-HLJ09. To investigate the effects of combining these G-protein substitutions on IHNV virulence and protective efficacy, we employed a reverse genetics system to construct four r…

Rainbow Trout (Oncorhynchus mykiss) as a Model Organism for Assessing Long-Term Impacts of Municipal Wastewater Pollution on Early Fish Development.

The increasing release of micropollutants from wastewater treatment plant (WWTP) effluents poses growing risks to aquatic ecosystems; however, linking environmental exposure to biological responses under environmentally realistic conditions remains challenging. This study aimed to evaluate biological responses associated with WWTP effluent exposure during the early life stages of rainbow trout (Oncorhynchus mykiss) using an integrated field-based approach. Five floating incubations containing 1000 eggs each were deployed at each of an upstream reference site and a downstream WWTP-affected site…

Hematological and biochemical alterations in rainbow trout Oncorhynchus mykiss during a natural Aeromonas sp. outbreak in high-altitude Ladakh, trans-Himalayas.

An outbreak of bacterial septicemia associated with Aeromonas sp. was investigated in intensive high-altitude aquaculture of rainbow trout, Oncorhynchus mykiss, at the Drass fish farm, Ladakh, during the seasonal transition of May-June 2025. This observational study documents physiological associations rather than experimental causality. A total of 72 fish were sampled: 36 severely diseased and 36 asymptomatic but exposed specimens from adjacent raceways of the same facility. Phenotypic, biochemical, and 16S rRNA gene sequencing of isolate SFB099 identified the agent as Aeromonas sp. with sequ…

Metal-induced inhibition of Na+/K+-ATPase and SERCA in the rainbow trout (Oncorhynchus mykiss) heart: Implications for cardiac ion regulation.

Metal ions are known to inhibit sarcolemmal ion currents and induce cardiac arrhythmias in fish, but their actions are not fully explained by ion channel blockade. To further clarify the effects of di- and trivalent cations on fish heart function, we examined the impact of eight metals on the activities of Na+/K+-ATPase and Sarco/Endoplasmic Reticulum Calcium ATPase (SERCA) in vitro in ventricular homogenates of rainbow trout (Oncorhynchus mykiss). Unfractionated ventricular homogenates were exposed in vitro to eight metals (Al3+, Mn2+, Fe2+, Co2+, Ni2+, Cu2+, Zn2+ or Hg2+) at concentrations o…

Systematic reassessment of Ambiphrya and Apiosoma (Ciliophora, Peritrichia), with morpho-molecular characterization of two epibiotic ciliates from rainbow trout in Korea.

Many peritrich ciliates are parasites of, or epibionts on, aquatic animals. Among them, the genera Ambiphrya and Apiosoma are common fish ectocommensals in aquacultures. Here, we report infections by two poorly known forms, Ambiphrya cf. ameiuri and Apiosoma pseudopiscicola, on rainbow trout, a commonly farmed and economically important food fish in South Korea, and provide detailed descriptions and diagnostic illustrations of both species. Their SSU rDNA sequences were obtained, filling the molecular data gap for Apiosoma, a genus for which no sequences had previously been available. Our phyl…

Replacing fish oil with Tetraselmis chui microalgae biomass does not compromise rainbow trout health: Biochemical, histologic, antioxidant and immune gene expression.

Microalgae offer a nutritionally robust alternative to fishmeal and fish oil, helping reduce pressure on wild stocks and supporting more sustainable aquafeed production. This study explored the potential of replacing fish oil with Tetraselmis (Tetraselmis chui) microalgae biomass in the diet of juvenile rainbow trout (89.0 ± 1.10 g) (Oncorhynchus mykiss), assessing its effects on the fish's health. A control diet containing 53% crude protein and fish oil (FO) was modified by replacing FO with Tetraselmis at three graded inclusion levels: 33% (Tetra33), 66% (Tetra66), and 100% (Tetra100). The 8…

Transcriptomics, antioxidant enzyme activities, and immune-associated parameter analysis reveal the molecular responses of rainbow trout (Oncorhynchus mykiss) to transportation stress.

To explore how transportation stress affects the physiology of rainbow trout, we measure antioxidant and immunity-related indicators in spleen tissues of adult fish during transportation (at 0, 2, and 6 h) and 24 h following it (r24 h). We report a significant reduction in spleen catalase activity among the 2, 6, and r24 h groups compared with the 0 h group (P < 0.05). Levels of glutathione peroxidase activity and total antioxidant capacity, and lipid peroxide, complement C4, and immunoglobulin M contents first decreased, then increased; complement C3 content and lysozyme activity first increa…

Divergent miRNA-mRNA networks contribute to tissue-specific lipid homeostasis in triploid rainbow trout skeletal muscle during high-lipid diet challenge.

Rainbow trout (Oncorhynchus mykiss) is one of the most widely farmed salmonid species globally. To investigate the mechanisms regulating muscle fat content and enhancing tolerance to high-lipid diets, transcriptomic changes in muscle tissues (myotomes and myosepta) were analyzed after treatment with an elevated dietary lipid level (30 %). The results revealed that myotomes and myosepta employed distinct strategies to maintain lipid homeostasis under elevated dietary lipids. Myotomes suppressed lipid deposition by upregulating lipoic acid synthesis to enhance lipolysis and reducing lipoprotein …

Characterization of volatile compounds in the water samples from rainbow trout aquaculture ponds eliciting off-odors: understanding locational and seasonal effects.

The quality of water used in aquaculture ponds is one of the crucial factors influencing the smell and sensory properties of fish. The water samples were taken from the rainbow trout fish ponds from three different fish farms in three provinces in Türkiye in four different seasons. The samples were analyzed for the volatile components by employing HS-SPME/GC-MS. Seven different volatile groups including aldehydes, ketones, esters, alcohols, volatile phenols, terpenes and other aromatic substances were identified in the samples. Among these, aldehydes were found to be the most dominant. (E)-2-H…

Parasite abundance distribution as a model of host-parasite relationships between monogeneans Gyrodactylus spp. and cage-reared rainbow trout Oncorhynchus mykiss.

Aggregation is a fundamental feature of parasite distribution in the host population, but the biological implications of the aggregation indices used to describe the relationships between the populations of parasites and hosts are not evident. It is speculated that the form of distribution in each case is predicated on the host's varying resistance to the infection, which is hard to control, making it difficult to adequately interpret the index values. This paper examines several cases from trout farms in Russian Karelia to explore the monogenean Gyrodactylus spp. infection in rainbow trout of…

Beyond the surface: Consequences of methyl tert-butyl ether (MTBE) exposure on oxidative stress, haematology, genotoxicity, and histopathology in rainbow trout.

Concerns have been raised about the possible environmental effects of methyl tert-butyl ether (MTBE), which is widely used as a gasoline additive. This research aimed to look at the consequences of MTBE contamination on rainbow trout (Oncorhynchus mykiss), emphasizing oxidative stress, genotoxicity, and histopathological damage. After determining the LC50-96 h value, the effects of sub-lethal doses of MTBE (0 (control), 90, 180, and 450 ppm) on rainbow trout were investigated. In fish tissues, the levels of oxidative stress indicators such as malondialdehyde (MDA) and superoxide dismutase (SOD…
