Sox17 regulates liver lipid metabolism and adaptation to fasting.

Liver is a major regulator of lipid metabolism and adaptation to fasting, a process involving PPARalpha activation. We recently showed that the Vnn1 gene is a PPARalpha target gene in liver and that release of the Vanin-1 pantetheinase in serum is a biomarker of PPARalpha activation. Here we set up a screen to identify new regulators of adaptation to fasting using the serum Vanin-1 as a marker of PPARalpha activation. Mutagenized mice were screened for low serum Vanin-1 expression. Functional interactions with PPARalpha were investigated by combining transcriptomic, biochemical and metabolic a

Impact of eight weeks of repeated ischaemic preconditioning on brachial artery and cutaneous microcirculatory function in healthy males.

BACKGROUND: Ischaemic preconditioning has well-established cardiac and vascular protective effects. Short interventions (one week) of daily ischaemic preconditioning episodes improve conduit and microcirculatory function. This study examined whether a longer (eight weeks) and less frequent (three per week) protocol of repeated ischaemic preconditioning improves vascular function. METHODS: Eighteen males were randomly allocated to either ischaemic preconditioning (22.4 ± 2.3 years, 23.7 ± 3.1 kg/m(2)) or a control intervention (26.0 ± 4.8 years, 26.4 ± 1.9 kg/m(2)). Age-related dedifferentiation and compensatory changes in the functional network underlying face processing.

Recent evidence has shown that older adults fail to show adaptation in the right fusiform gyrus (FG) to the same face presented repeatedly, despite accurate detection of the previously presented face. We used functional magnetic resonance imaging to investigate whether this phenomenon is associated with age-related reductions in face specificity in brain activity and whether older adults compensate for these face-processing deficiencies by increasing activity in other areas within the face-processing network, or outside this network. A comparison of brain activity across multiple stimulus cate

The olfactory system as a puzzle: playing with its pieces.

The mammalian olfactory bulb (OB) has all the features of a whole mammalian brain but in a more reduced space: neuronal lamination, sensory inputs, afferences, or efferences to other centers of the central nervous system, or a contribution of new neural elements. Therefore, it is widely considered as "a brain inside the brain." Although this rostral region has the same origin and general layering as the other cerebral cortices, some distinctive features make it very profitable in experimentation in neurobiology: the sensory inputs are driven directly on its surface, the main output can be acce

Changes in electromyographic activity of trunk muscles within the sub-acute phase for individuals deemed recovered from a low back injury.

Evidence indicates that previous low back injury (LBI) is a strong predictor for re-injury. The purpose of this study was to examine whether neuromuscular patterns remain altered in a LBI group who were deemed recovered. Surface electromyograms from 12-abdominal and 12-back extensors sites and motion variables were recorded from 33 LBI individuals (sub-acute phase) and 54 asymptomatic controls. Pain-related variables were recorded and a clinical assessment performed for LBI participants. Subjects performed a symmetrical lift and replace task in two reaches. Pattern recognition techniques were

An ultrasonographic investigation of cleft-type compensatory articulations of voiceless velar stops.

Ultrasound imaging was used to investigate the articulation of the voiceless velar stop [k] in five speakers with compensatory articulation related to cleft palate. The perceptual evaluation of the acoustic realization and the visual assessment of the tongue movement for the target sound were made by three examiners. The analysis revealed a variety of different compensatory strategies that included glottal stops, pharyngeal stops, midpalatal stops and glottal and velar co-productions. One patient produced palatal click sounds together with a midpalatal stop. The ultrasound imaging also reveale

Signaling aggression.

From psychological and sociological standpoints, aggression is regarded as intentional behavior aimed at inflicting pain and manifested by hostility and attacking behaviors. In contrast, biologists define aggression as behavior associated with attack or escalation toward attack, omitting any stipulation about intentions and goals. Certain animal signals are strongly associated with escalation toward attack and have the same function as physical attack in intimidating opponents and winning contests, and ethologists therefore consider them an integral part of aggressive behavior. Aggressive sign

Adenosine A₂a receptors and O₂ sensing in development.

Reduced mitochondrial oxidative phosphorylation, via activation of adenylate kinase and the resulting exponential rise in the cellular AMP/ATP ratio, appears to be a critical factor underlying O₂ sensing in many chemoreceptive tissues in mammals. The elevated AMP/ATP ratio, in turn, activates key enzymes that are involved in physiologic adjustments that tend to balance ATP supply and demand. An example is the conversion of AMP to adenosine via 5'-nucleotidase and the resulting activation of adenosine A(₂A) receptors, which are involved in acute oxygen sensing by both carotid bodies and the bra

Foot placement variability as a walking balance mechanism post-spinal cord injury.

BACKGROUND: Spinal cord injury affects walking balance control, which necessitates methods to quantify balance ability. The purposes of this study were to 1) examine walking balance through foot placement variability post-injury; 2) assess the relationship between measures of variability and clinical balance assessments; and 3) determine if spatial parameter variability might be used as a clinical correlate for more complex balance measurements. METHODS: Ten persons with spinal cord injury walked without devices on a split-belt treadmill at self-selected speeds. Ten healthy controls walked at

Contact pressure on ACL hamstring grafts in the bone tunnel with interference screw fixation--dynamic adaptation under load.

INTRODUCTION: Interference screws used in fixation of anterior cruciate ligament (ACL) hamstring grafts create mechanical hold by forcing the graft into frictional contact with the bone tunnel. We analyzed the resultant graft-tunnel contact pressure using an in vitro model of human cadaver 8mm hamstring grafts. METHODS: Contact characteristics were assessed using both pressure sensitive films and a force sensor. Two screw sizes were investigated (8 and 9 mm in an 8mm Sawbone tunnel), both with and without a bone wedge between graft and screw. Separately, time dependent relaxation of contact f

Muscle activation changes during body weight support treadmill training after focal cortical ischemia: A rat hindlimb model.

The study used a focal ischemia rat hindlimb model to investigate muscle activity changes during a 10-day body weight support (BWS) treadmill training program. The changes being studied included fatigue effects, EMG burst duration in the gait cycle, and the symmetry of muscle activation between affected and unaffected sides. Intramuscular EMG of medial gastrocnemius (MG) and tibialis anterior (TA) muscles in male Sprague Dawley rats at affected side (n=10) and unaffected side (n=10) were recorded during the treadmill running before a middle cerebral artery occlusion/reperfusion (MCAo/r) surger

Adaptively controlling deep brain stimulation in essential tremor patient via surface electromyography.

OBJECTIVES: We present patient test outcomes to show that on-off control of deep brain stimulation sequences in essential tremor patients is achievable in a self-adaptive manner via non-invasive surface-electromyography, to prevent tremors in these patients. METHOD: In our study, an essential tremor patient, who underwent bilateral deep brain stimulation implantation 8 years earlier, was subjected to deep brain stimulation at 130 pulses/second, with a 90-microsecond pulse-width, in packets of durations from 20 to 73 seconds and was monitored with surface-electromyography. RESULTS: At the end

First aid kit for hypoxic survival: sensors and strategies.

Survival success under conditions of acute oxygen deprivation depends on efficiency of the central and peripheral chemoreception, optimization of oxygen extraction from the hypoxic environment and its delivery to the periphery, and adjustments of energy production and consumption. This article uses a comparative approach to assess the efficiency of adaptive strategies used by anoxia-tolerant and hypoxia-sensitive species to support survival during the first minutes to 1 h of oxygen deprivation. An aquatic environment is much more demanding in terms of diurnal and seasonal variations of the amb

Survival of clinical and food isolates of Listeria monocytogenes through simulated gastrointestinal tract conditions.

Twenty-seven strains of Listeria monocytogenes previously isolated from food (n = 16) and human patients of listeriosis (n = 11) were characterized and compared based on their ability to survive through the simulated gastrointestinal tract conditions. Cells were exposed (60 or 120 min) to low pH in the presence of pepsin, to simulate the digestion in the stomach, and subsequently to bile salts to simulate the digestion in the small intestine (60 or 120 min). Their survival was shown to be origin- (food and clinical) and strain dependent (p < 0.001) and also significantly dependent on the impos

Active stiffening of mitral valve leaflets in the beating heart.

The anterior leaflet of the mitral valve (MV), viewed traditionally as a passive membrane, is shown to be a highly active structure in the beating heart. Two types of leaflet contractile activity are demonstrated: 1) a brief twitch at the beginning of each beat (reflecting contraction of myocytes in the leaflet in communication with and excited by left atrial muscle) that is relaxed by midsystole and whose contractile activity is eliminated with beta-receptor blockade and 2) sustained tone during isovolumic relaxation, insensitive to beta-blockade, but doubled by stimulation of the neurally ri

Comparison of two lower-body modes of endurance training on lower-body strength development while concurrently training.

The most recent American College of Sports Medicine (1998) recommendations for quantity and quality of exercise includes both resistance and endurance exercise components. Skeletal muscle adaptations to resistance-only and endurance-only programs may be different and possibly antagonistic when both types of training are imposed concurrently. The present study examined the effect of two different modes of lower-body endurance exercise (i.e., cycle ergometry and incline treadmill walking) on lower-body strength development with concurrent resistance training designed to improve lower-body streng

Invariant template matching in systems with spatiotemporal coding: A matter of instability.

We consider the design principles of algorithms that match templates to images subject to spatiotemporal encoding. Both templates and images are encoded as temporal sequences of samplings from spatial patterns. Matching is required to be tolerant to various combinations of image perturbations. These include ones that can be modeled as parameterized uncertainties such as image blur, luminance, and, as special cases, invariant transformation groups such as translation and rotations, as well as unmodeled uncertainties (noise). For a system to deal with such perturbations in an efficient way, they

Hypertension and heart failure: a dysfunction of systole, diastole or both?

The pathological myocardial hypertrophy associated with hypertension contains the seed for further maladaptive development. Increased myocardial oxygen consumption, impaired epicardial coronary perfusion, ventricular fibrosis and remodelling, abnormalities in long-axis function and torsion, cause, to a varying degree, a mixture of systolic and diastolic abnormalities. In addition, chronotropic incompetence and peripheral factors such as lack of vasodilator reserve and reduced arterial compliance further affect cardiac output particularly on exercise. Many of these factors are common to hyperte
