Compound 17 Inhibits Lung Cancer Progression via Inducing Cellular Apoptosis and Blocking TNF Signaling Pathway Activation.

Lung cancer ranks among the most commonly diagnosed malignancies worldwide, with dismal prognosis largely due to its intrinsic drug resistance and high recurrence rate. Herein, we synthesized 30 glycyrrhetinic acid derivatives and evaluated their anti-lung cancer potential both in vitro and in vivo. The biological effects of compound 17 on A549 cells were determined using MTT, colony formation, and Transwell assays. Flow cytometry, transcriptomic profiling, and RT-qPCR were performed to identify differentially expressed genes, followed by GO and KEGG enrichment analyses and molecular docking v

N-acetyltransferase 10 participates in dopaminergic neuronal apoptosis in Parkinson's disease through ac4C-mediated transcriptomic regulation.

Parkinson's disease (PD) is characterized by the progressive loss of dopaminergic neurons in the substantia nigra. However, the underlying pathogenic mechanisms of PD remain incompletely defined, and effective treatments are lacking. N-acetyltransferase 10 (NAT10) catalyzes N4-acetylcytidine (ac4C) modification of mRNA, a process crucial for mRNA stability and translational efficiency. However, whether NAT10-mediated ac4C modification contributes to PD and the molecular mechanisms underlying its involvement remain unclear. This study aimed to investigate the role of NAT10 and its ac4C-mediated

Sigma-1 Receptor antagonism potentiates siramesine-induced apoptosis and altered autophagic signaling in human leukemia cells.

T-cell acute lymphoblastic leukemia (T-ALL) and chronic myelogenous leukemia (CML) are hematological malignancies arising from lymphoid and myeloid lineages, respectively. Although many patients achieve remission following chemotherapy, relapse and chemoresistance remain major clinical challenges. Sigma receptors, including sigma-1 (S1R) and sigma-2 (S2R), regulate pathways involved in cancer cell survival, stress responses, and cell death. While S2R is frequently overexpressed in tumors, S1R promotes protective mechanisms that support cancer cell survival, suggesting that dual targeting of th

MtROS-Drp1-mediated mitochondria fission contributes to microglia BV2 cell apoptosis induced by titanium dioxide nanoparticles.

The widespread use of titanium dioxide nanoparticles (TiO2 NPs) has raised serious concerns regarding their biosafety, particularly their potential effects on the central nervous system. As innate immune cells in the brain, microglia serve as key targets for neurotoxicity assessment. This study aims to systematically elucidate the toxic effects of TiO2 NPs on BV2 cells and their underlying molecular mechanisms. The results showed that TiO2 NPs were spherical, with an average particle size of approximately 10 nm, and primarily existed in the anatase phase. TiO2 NPs significantly inhibited BV2 c

IDO1 Silencing Enhances Cisplatin Sensitivity in Gastric Cancer Cells via Modulation of Apoptosis and Oxidative Stress.

Gastric cancer remains a leading cause of cancer-related mortality worldwide, largely due to resistance to platinum-based chemotherapy. Indoleamine 2,3-dioxygenase 1 (IDO1) has been implicated in tumor progression and immune evasion; however, its cell-intrinsic role in chemoresistance remains incompletely understood. This study demonstrates that IDO1 functions as a critical regulator of cisplatin sensitivity in gastric cancer cells through a ROS-mediated mechanism. IDO1 expression was suppressed using siRNA in two gastric cancer cell lines, AGS and MKN45, followed by cisplatin treatment. Cell

TRPM7-mediated calcium signaling contributes to Hyperglycemia-induced mitochondrial dysfunction and apoptosis in retinal Müller cells.

Calcium signaling dysregulation is a critical trigger of mitochondrial dysfunction in metabolic disorders, yet the upstream mechanisms linking hyperglycemic stress to organellar Ca2+ overload remain poorly defined. The transient receptor potential melastatin 7 (TRPM7) channel functions as a Ca2+-permeable signaling node with unique kinase activity, but its role in hyperglycemia-induced glial injury is unknown. Here, we investigated whether TRPM7 mediates mitochondrial dysfunction and apoptosis in retinal Müller cells under hyperglycemic stress. Using a streptozotocin/high-fat diet-induced diab

Multifunctional platinum(IV) complexes reverse cisplatin resistance in triple- negative breast cancer via ferroptosis and apoptosis.

Platinum(II) drugs are important chemotherapeutical strategies for the triple-negative breast cancer (TNBC) but are frequently hindered by resistance and systematic toxicity. To address these limitations, a series of novel platinum(IV) complexes were designed by integrating ferroptosis inducers (sulfasalazine derivatives) with platinum-based moieties. Among them, complex 11 emerged as the most effective agent against both cisplatin (CDDP)-sensitive and CDDP-resistant TNBC cell lines, but showed lower cytotoxicity toward the normal breast epithelial cell line MCF-10 A compared with CDDP or carb

Targeting G9a Exerts Pleiotropic Suppression in Triple-Negative Breast Cancer Cells: Cooperatively Inducing Pyroptosis and Apoptosis.

Background: Pyroptosis, a pro-inflammatory programmed cell death process, is a key player in tumor biology, including in triple-negative breast cancer (TNBC). Inhibiting G9a has been proven to exert anticancer effects; however, the molecular mechanism of the effects remains unclear. The study aimed to illustrate whether inhibiting G9a can suppress the process of TNBC cells by promoting pyroptosis and investigate the underlying mechanisms. Methods: MCF-10A, MDA-MB-231 and SUM159PT cell lines were used for in vitro study. CCK8 and EdU staining assay were used to examine the cell proliferation, a

Silencing OGFOD1 ameliorates hepatic ischemia-reperfusion injury through abrogating oxidative stress and apoptosis via downregulating SPARC.

Hepatic ischemia-reperfusion injury (HIRI) is a complicated pathological process impacting the prognosis of patients suffering from liver resection and transplantation surgery. 2-oxoglutarate and iron dependent oxygenase domain containing 1 (OGFOD1), a crucial factor involved in protein translation, was highly expressed during HIRI. However, its role as well as underlying mechanism is still unclear. In this study, two datasets from Gene Expression Omnibus database were analyzed and we focused on OGFOD1 which is associated with ischemia-reperfusion. Later, a HIRI rat model and a hypoxia/reoxyge

Structure-activity relationship and anticancer mechanism of febuxostat-1,2,3-triazole hybrids inducing DNA damage and apoptosis.

Leveraging febuxostat's liver-targeting properties and the pharmacological versatility of the 1,2,3-triazole scaffold, we designed and synthesized a series of novel febuxostat-linked 1,2,3-triazole derivatives and systematically evaluated their anticancer activity against HepG2 hepatocellular carcinoma cells. Structure-activity relationship analysis revealed that para-substitution of the triazole on the phenyl ring, particularly when combined with a fluorine atom on the benzyl group, markedly enhanced cytotoxicity, with compounds 10c, 10f, and 10 h reducing HepG2 cell viability below 50%. A catalytic redox-cycling nanoreactor enables robust oxidative stress amplification for synergistic tumor apoptosis and ferroptosis.

Intervening in the aberrant redox homeostasis of tumors, particularly toward reactive oxygen species (ROS) overload, holds considerable promise for cancer therapy, yet, is severely constrained by the robust compensatory antioxidant defense system (ADS) and the unavoidable disruption of redox homeostasis in normal tissues. Here, we present a catalytic redox-cycling nanoreactor, TEMPO radical-modified cross-linked lipoic acid nanoparticles (T@cLAN), designed to achieve robust oxidative stress amplification for cancer therapy. Lipoic acid (LA) characterized by a cyclic disulfide backbone enables

The antiviral effect of TRPV1 antagonist SB-705498 against GCRV infection via inflammation and apoptosis inhibition.

Grass carp reovirus (GCRV), the causative agent of grass carp hemorrhagic disease (GCHD), inflicts substantial economic losses in aquaculture, yet effective therapeutic interventions remain limited. Previous studies have demonstrated that the host factor CiTRPV1 facilitates GCRV infection by activating multiple pro-viral signaling pathways, underscoring its potential as an antiviral target. Here, we screened a library of TRPV1-targeting small molecules and identified three candidates with significant antiviral activity in Ctenopharyngodon idella kidney (CIK) cells. Based on its favorable selec

WWP2 underlies ROS-induced granulosa cell apoptosis by promoting ubiquitination of BAK in polycystic ovary syndrome.

Granulosa cell (GC) apoptosis is intrinsically linked to the ovarian dysfunction of polycystic ovary syndrome (PCOS). Although oxidative stress and apoptosis in GCs have been detected in PCOS patients, how reactive oxygen species (ROS) links to GC apoptosis in PCOS remains to be further elucidated. Here, by integrating public single-cell RNA-seq data with clinical GC sample validation, we found that the expression of the E3 ubiquitin ligase WWP2 was significantly reduced, whereas its role in PCOS has not been previously reported. Notably, we first demonstrated that WWP2 overexpression can effe
