Resveratrol engages SIRT1/AMPK networks to restore BDNF signaling, autophagy, and mitochondrial function in age-related cognitive decline: A geroscience perspective.

Age-related cognitive decline and neurodegenerative disorders present significant challenges in neuropsychiatry, as current therapeutic strategies largely fail to adequately address the complexities of cerebral aging. Resveratrol, a natural polyphenol, has become a promising multi-target neuroprotective agent that can affect multiple important pathways that are involved in aging and neurodegeneration at the same time. This review consolidates evidence indicating that resveratrol, particularly through bioavailability-enhanced formulations, activates SIRT1, stimulates AMPK, inhibits mTORC1, rest

Lichen secondary metabolite atraric acid alleviates chronic intermittent hypoxia-induced liver injury by activating autophagy to inhibit the NF-κB pathway.

Chronic intermittent hypoxia (CIH) is an alternating state of recurrent hypoxia and reoxygenation triggered by disorders such as sleep apnea, leading to systemic oxidative stress and organ damage. In this study, we investigate the protective effect and mechanism of atraric acid (AA), a secondary metabolite produced by lichens, against CIH-induced liver injury. By establishing a CIH mouse model (10 h/d, oxygen cycling 5%-21% for 4 weeks) and an in vitro model of AML-12 cell hypoxia/reoxygenation (H/R), we perform H&E staining, biochemical assays (ALT/AST/MDA/CAT/T-AOC/GSH-Px), qPCR, western blo

Protein kinase C θ deletion ameliorates bleomycin-induced pulmonary fibrosis via the improvement of SIRT1/AMPK signaling on autophagy and senescence.

OBJECTIVES: Protein kinase C theta (PKC θ) is involved in the regulation of various inflammatory diseases, such as allergic asthma and ConA-induced hepatitis. However, its role in pulmonary fibrosis has not been reported. This study aimed to explore the role of PKC θ in bleomycin-induced pulmonary fibrosis and the potential mechanisms involved. MATERIALS AND METHODS: WT C57BL/6 and PKC-θ deletion mice were administered a single intratracheal injection of bleomycin at a dosage of 3 mg/kg to prepare an animal model of pulmonary fibrosis. Histopathological changes of lung fibrosis were observed.

Targeting Long Noncoding RNA LUCAT1 Alleviates Insulin Resistance of Ovarian Granulosa Cells in Polycystic Ovary Syndrome by Blocking HMGB1-Mediated Autophagy.

PROBLEM: Dysfunctional autophagy in ovarian granulosa cells (GCs) represents a key pathological feature of insulin resistance (IR) in polycystic ovary syndrome (PCOS), though regulatory roles of autophagy-associated lncRNAs remain poorly characterized. METHOD OF STUDY: Comparing GCs from IR-PCOS patients, non-IR PCOS patients, and non-PCOS controls revealed significantly elevated LUCAT1 expression in IR-PCOS. Functional validation via LUCAT1 knockdown in primary IR-PCOS GCs assessed cell viability, apoptosis, autophagy markers, and insulin sensitivity. Mechanistic studies employed Ago2-RIP, d

Discovering host-viral protein interactions in autophagy: A LIR discovery pipeline for identifying LC3-interacting region motifs in highly virulent viruses.

Hemorrhagic fever virus (HFV) infections are highly fatal, posing a significant global pandemic threat as they continue to emerge in new locations. HFVs and other highly virulent viruses (HVVs), such as Nipah virus, exploit host cell pathways, including the conserved host autophagy pathway, to promote viral replication. The Atg8/microtubule-associated protein 1 light chain 3 (LC3) proteins are necessary for autophagosome formation and maturation. Proteins interact with Atg8-family proteins through LC3-interacting region (LIR) motifs, which is a short linear motif (SLiM) found in intrinsically

iNOS/NO-Induced autophagy primes PANoptosis as the central driver of streptococcal toxic shock-like syndrome in zoonotic Streptococcus suis infection.

Streptococcus suis is a major zoonotic pathogen that causes human fulminant meningitis and streptococcal toxic shock-like syndrome (STSLS), the latter being driven by dysregulated inflammatory responses and explosive cytokine storms. It remains unclear whether PANoptosis acts as a coordinated host-protective response or instead serves as a detrimental driver of cytokine storm during bacterial infection. Here, we identify autophagy as an upstream orchestrator of PANoptosis during zoonotic Streptococcus suis infection, redefining STSLS as a disease driven by an integrated PANoptosis cell-death p

Liposomal honokiol attenuates dimethylhydrazine-induced colon carcinogenesis in rats through modulation of oxidative stress, inflammation, apoptosis, autophagy, and lipid metabolism pathways.

Colorectal cancer is one of the leading causes of cancer-related mortality worldwide, and its progression is strongly associated with oxidative stress, chronic inflammation, dysregulated apoptosis, and impaired autophagy. Although honokiol (HNK) possesses promising anticancer properties, its therapeutic application is limited by poor bioavailability and low aqueous solubility. Therefore, this study investigated the potential of liposomal honokiol nanoparticles (HNK-LNPs) as an advanced nanotherapeutic strategy to enhance the chemoprotective efficacy of HNK against dimethylhydrazine (DMH)-induc

H4K12 lactylation-regulated NLRP3 is involved in cigarette smoke-accelerated Alzheimer-like pathology through mTOR-regulated autophagy and activation of microglia.

Cigarette smoke (CS), an indoor environmental pollution, is an environmental risk factor for diverse neurological disorders. However, the neurotoxicological effects and mechanisms of CS on Alzheimer's disease (AD) progression remain unclear. We found that CS accelerated the progression of AD, including increasing β-amyloid (Aβ) plaque deposition and exacerbating cognitive decline. Mechanistically, CS exposure increased the levels of NOD-like receptor protein 3 (NLRP3), which impaired autophagic flux in microglia by activating the mammalian target of rapamycin (mTOR) signal. Metabolomics analys

Neurotrophic factor-α1/carboxypeptidase E regulates critical protein networks to rescue neurodegeneration, defective synaptogenesis and impaired autophagy in Alzheimer's disease mice.

BACKGROUND: The global aging population is increasingly inflicted with Alzheimer's disease (AD), but a cure is still unavailable. Neurotrophic factor-α1/carboxypeptidase E (NF-α1/CPE) gene therapy has been shown to prevent and reverse memory loss and pathology in AD mouse models. However, the mechanisms of action of NF-α1/CPE are not fully understood. We investigated if a non-enzymatic form of NF-α1/CPE-E342Q is efficient in reversing AD pathology and carried out a proteomic study to uncover the mechanisms of action of NF-α1/CPE in AD mice. METHODS: AAV-human NF-α1/CPE or a non-enzymatic form

HBCOC attenuates cerebral ischemia-reperfusion injury in mice by inhibiting the inflammatory response and autophagy via TREM-1/ERK/NF-κB.

OBJECTIVE: Hemoglobin-based carbon monoxide carrier (HBCOC) can dissociate carbon monoxide and ameliorate organ damage by inhibiting inflammation and oxidative stress. In this study, we evaluated its effect on cerebral ischemia-reperfusion injury in mice and explored its potential mechanism. METHODS: A middle cerebral artery occlusion/reperfusion (MCAO/R) mouse model was established using the wire embolization method, and HBCOC or equivalent normal saline was administered via the tail vein during reperfusion. HE staining and TEM were used to observe the injury in the tissue. The levels of IL-

Multi-omics analysis of ketogenic diet-mediated neural repair in spinal cord injury: Targeting of lysosomal autophagy through CTSB/LAMP2 regulation.

Spinal cord injury (SCI) initiates secondary pathologies characterized by dysregulated autophagy and neuroinflammation Although the ketogenic diet (KD) has shown potential in promoting functional recovery after SCI, the mechanisms underlying KD-mediated neural repair remain unclear. We employed an integrated multi-omics approach combining 4D proteomics, transcriptomics, and single-cell RNA sequencing in a C5 hemi-contusion mouse model. This was combined with in vitro validation using β-hydroxybutyrate (β-OHB)-treated BV2 microglia cells to investigate KD's effects on lysosome-mediated autophag

Changes to the Autophagy-Related Muscle Proteome Following Short-Term Treatment with Ectoine in the Duchenne Muscular Dystrophy Mouse Model mdx.

The most severe form of muscular dystrophy (MD), known as Duchenne MD (DMD), remains an incurable disease, hence the ongoing efforts to develop supportive therapies. The dysregulation of autophagy, a degradative yet protective mechanism activated when tissues are under severe and prolonged stress, is critically involved in DMD. Treatments that harness autophagic capacities therefore represent a promising therapeutic approach. Osmolytes are protective organic molecules that regulate osmotic pressure and cellular homeostasis and may support tissue-repairing autophagy. We therefore explored the e

The Olive Polyphenol Hydroxytyrosol Enhances Autophagy and Heme Oxygenase-1 Expression in Aortic Endothelial Cells and Reduces Arterial Stiffness ex vivo.

Age-related arterial stiffening is a hallmark of vascular ageing and a key driver of cardiovascular disease. Oxidative stress, impaired autophagy, and extracellular matrix remodelling play an important role in the progression of aortic stiffening. Hydroxytyrosol (HT), a phenolic compound in olives, has demonstrated antioxidant properties and the ability to modulate autophagy, positioning it as a potential therapeutic for vascular ageing. In this study, we investigated the effects of HT on autophagy flux and antioxidant protein expression in human aortic endothelial cells (HAoECs). In parallel,

CEBPB, C19MC, and Defective Autophagy Drive Novel Podosomal Belt to Macropinocytosis Transition, Lipid Accumulation, and HBV A-to-I RNA-editing.

Obesity and neurodegeneration are clinically associated diseases with defective autophagy. However, the genetic, biological, and metabolic underpinnings connecting these diseases are not well-understood. Here we identified a Mitochondriaobesity/neurodegeneration (Mon) gene-signature that is shared between obesity, and neurodegenerative diseases. We demonstrate that, CEBPB elevates Mon-gene-signature, to form podosomal belts, and enhance ROS production. Inhibiting autophagy collapses podosomal-belts through macropinocytosis to accumulate vacuoles, lipid-droplets, nuclear Notch-1 (nNICD), DEPTOR

Neuroglobin-promoted autophagy inhibits the proliferation, migration and invasion of human glioblastoma cells through the EGFR/PI3K/AKT/mTOR signaling pathway.

BACKGROUND: Neuroglobin (Ngb) is a novel oxygen-binding or sensor protein that is present primarily in neurons and the brain. It plays a critical role in sensing oxygen and hypoxia signals and in transducing intracellular signaling pathways. However, the specific functions and mechanism of Ngb in the glioblastoma remain unclear. METHODS: The expression of Ngb in glioma brain tissues and cells was measured by Western blot, immunohistochemical staining and retrieved from TCGA databases. Loss or gain of function assays, CCK-8 assay, scratch and transwell assay, transmission electron microscopy (

Bipolar and schizophrenia risk gene AKAP11 encodes an autophagy receptor coupling the regulation of PKA kinase network homeostasis to synaptic transmission.

Human genomic studies have identified protein-truncating variants in AKAP11 associated with both bipolar disorder (BD) and schizophrenia (SCZ), implicating a shared disease mechanism driven by loss-of-function. AKAP11, a protein kinase A (PKA) adapter, plays a key role in degrading the PKA-RI complex through selective autophagy. However, the neuronal functions of AKAP11 and the impact of its loss-of-function remains largely uncharacterized. Through multi-omics approaches, cell biology, and electrophysiology analysis in mouse models and human induced neurons, we delineate a central role of AKAP

The lncRNA MIR503HG/miR-16-5p/FOSL1 pathway mediates autophagy to promote esophageal epithelial cells proliferation and EMT in esophageal restenosis.

Esophageal stents have been used increasingly to relieve malignant dysphagia and are widely employed in the treatment of esophageal stenosis. However, the clinical application of esophageal stents has been limited by esophageal restenosis. Our preliminary studies have indicated that long non-coding RNAs (lncRNAs) may be involved in the development of esophageal restenosis. The aim of this study was to investigate the role of the lncRNA MIR503HG (MIR503HG)/miR-16-5p/Fos-like antigen-1 (FOSL1) pathway in esophageal restenosis. We demonstrate that MIR503HG and FOSL1 expression levels were signifi
