GPRC5B protects osteoarthritis by regulation of autophagy signaling.

Osteoarthritis (OA) is one of the most common chronic diseases in the world. However, current treatment modalities mainly relieve pain and inhibit cartilage degradation, but do not promote cartilage regeneration. In this study, we show that G protein-coupled receptor class C group 5 member B (GPRC5B), an orphan G-protein-couple receptor, not only inhibits cartilage degradation, but also increases cartilage regeneration and thereby is protective against OA. We observed that Gprc5b deficient chondrocytes had an upregulation of cartilage catabolic gene expression, along with downregulation of ana…

Viability of loose body fragments in osteochondritis dissecans of the knee. A series of cases.

The purpose of this study was to determine if the cartilage from loose osteochondral fragments remains viable. Five patients with OCD of the knee who had undergone surgical treatment (arthroscopic reduction and internal fixation of the loose body) were included. The average age of patients was 13 years (range 10-14 years). Cartilage samples were obtained from the loose body fragments before reattachment was performed (study group) and from the healthy native cartilage (intercondyle area, control group) from each of the five patients. Tissue viability was assessed using live-dead assay in both …

Morphological analysis of the femoral trochlea in patients with recurrent dislocation of the patella using three-dimensional computer models.

We investigated the three-dimensional morphological differences of the articular surface of the femoral trochlea in patients with recurrent dislocation of the patella and a normal control group using three-dimensional computer models. There were 12 patients (12 knees) and ten control subjects (ten knees). Three-dimensional computer models of the femur, including the articular cartilage, were created. Evaluation was performed on the shape of the articular surface, focused on its convexity, and the proximal and mediolateral distribution of the articular cartilage of the femoral trochlea. The ext…

Growth and development of the acetabulum in the normal child. Anatomical, histological, and roentgenographic studies.

Postmortem studies of ten normal full-term infants and of three children, seven, nine, and fourteen years old, showed that the acetabular cartilage complex is a unit that is triradiate medially and cup-shaped laterally and is interposed between the ilium, ischium, and pubis. This complex is composed of epiphyseal growth-plate cartilage adjacent to these bones, of articular cartilage adjacent to these bones, of articular cartilage around the acetabular cavity, and, for the most part, of hyaline carilage. Interstitial growth within the triradiate part of the cartilage complex causes the hip sock…

Macromolecular markers in joint disease.

Cartilage contains a number of matrix macromolecules not present in other connective tissues. In tissue processes in disease and in normal turnover of the matrix these molecules are fragmented and released into surrounding fluids, in the case of joints into the synovial fluid. Immunoassay techniques have been developed to quantify these fragments. It appears that increased levels in synovial fluid (SF) correlate to a process in the joint cartilage. With successful therapy the level returns to normal. Interestingly, in early rheumatoid arthritis, i.e., with no discernible cartilage damage by ra…

Depletion of aquaporin 1 decreased ADAMTS‑4 expression in human chondrocytes.

Inflammation serves an important role in the progression of osteoarthritis (OA), and IL‑1β may act as a catabolic factor on cartilage, reducing the synthesis of primary cartilage components type II collagen and aggrecan. Aquaporin 1 (AQP1) is a 28‑kDa water channel formed of six transmembrane domains on the cell membrane. AQP1 is highly expressed in the anus, gallbladder and liver, and is moderately expressed in the hippocampus, ependymal cells of the central nervous system and articular cartilage. It was hypothesized that AQP1 may be highly expressed in OA cartilage and that it may increase t…

NGF-BMSC-SF/CS composites for repairing knee joint osteochondral defects in rabbits: evaluation of the repair effect and potential underlying mechanisms.

BACKGROUND: With the rapid growth of the ageing population, chronic diseases such as osteoarthritis have become one of the major diseases affecting the quality of life of elderly people. The main pathological manifestation of osteoarthritis is articular cartilage damage. Alleviating and repairing damaged cartilage has always been a challenge. The application of cartilage tissue engineering methods has shown promise for articular cartilage repair. Many studies have used cartilage tissue engineering methods to repair damaged cartilage and obtained good results, but these methods still cannot be …

Tissue-engineered trachea regeneration using decellularized trachea matrix treated with laser micropore technique.

UNLABELLED: Tissue-engineered trachea provides a promising approach for reconstruction of long segmental tracheal defects. However, a lack of ideal biodegradable scaffolds greatly restricts its clinical translation. Decellularized trachea matrix (DTM) is considered a proper scaffold for trachea cartilage regeneration owing to natural tubular structure, cartilage matrix components, and biodegradability. However, cell residual and low porosity of DTM easily result in immunogenicity and incomplete cartilage regeneration. To address these problems, a laser micropore technique (LMT) was applied in …

Effects of mechanical loading on the tissues of the rabbit knee.

We studied changes in subchondral bone and articular cartilage in an animal model of osteoarthrosis. In this model we applied repetitive impulsive loads to rabbits' knees. Their legs were held in short leg splints so the rabbits were unable to dampen the peak applied load with ankle flexion. After sacrifice, at 1 day to 6 weeks, we studied proximal tibial load-bearing cartilage histologically, biochemically, and with radioactive sulfate uptake. We also studied the subchondral bone under that cartilage histologically, histomorphometrically, with bone scan (99mTc pyrophosphate), and by tetracycl…

Mice lacking link protein develop dwarfism and craniofacial abnormalities.

Link protein (LP), an extracellular matrix protein in cartilage, stabilizes aggregates of aggrecan and hyaluronan, giving cartilage its tensile strength and elasticity. Cartilage provides the template for endochondral ossification and is crucial for determining the length and width of the skeleton. During endochondral bone formation, hypertrophic chondrocytes die and the cartilage is replaced with bone matrix. Here, we have generated targeted mutations in mice in the gene encoding LP (Crtl1). Homozygotes showed defects in cartilage development and delayed bone formation with short limbs and cr…

Visualisation of the intralaryngeal joints: a low voltage radiological study.

Low-voltage radiography of the neck was adjusted for visualisation of the crico-arytenoid joints. Sixty-four patients (48 males, 16 females) with laryngeal tumours were examined. The degree of ossification in the laryngeal cartilage around the crico-arytenoid joints was different in the males and the females. All males had ossification in the plate of the cricoid, and 94% in the arytenoids. The articular facets on the cricoid were visible in 94%, and on the arytenoid in 85%. All females had ossification in the arytenoids, and 94% in the plate of the cricoid. The articular facets on the aryteno…

The Effect of Intraarticular Insulin on Chondral Defect Repair.

OBJECTIVE: The aim of this study is to evaluate the effects of intraarticular insulin on the treatment of chondral defects. DESIGN: Twenty-four mature New Zealand rabbits were randomly divided into 3 groups as control (Group 1), microfracture (Group 2), and microfracture and insulin (Group 3). Four-millimeter full-thickness cartilage defects were created to the weight-bearing surface on the medial femoral condyles of each rabbit. In the first group, any additional interventions were not performed. Microfracture was performed on defects in groups 2 and 3. Additionally, 10 IU of insulin glargin…

The Posterior Part Influences the Anterior Part of the Mouse Cranial Base Development.

The cranial base is a critical structure in the head, which is composed of endoskeletal and dermal skeletal. The braincase floor, part of the cranial base, is a midline structure of the head. Because it is a midline structure connecting the posterior skull with the facial region, braincase floor is critical for the orientation of the facial structure. Shortened braincase floor leads to mid-facial hypoplasia and malocclusions. During embryonic development, elongation of the braincase floor occurs through endochondral ossification in the parachordal cartilage, hypophyseal cartilage, and trabecul…

Fabrication of three-dimensional cartilaginous framework in auricular reconstruction.

OBJECTIVE: One of the greatest challenges in facial plastic surgery is total auricular reconstruction, as it demands precise technique as well as artistic creativity. The crucial step to reconstruct satisfactory auricles is sculpting a three-dimensional cartilaginous framework, which determines the success of surgery. In this paper we describe our new techniques in fabricating the three-dimensional auricular framework. METHODS: To assemble satisfactory auricular frameworks, we designed four different methods according to the number of rib cartilages used. (1) Method I, the framework was made …

Significance of synovitis in Legg-Calvé-Perthes disease.

Synovitis is an important feature in Legg-Calvé-Perthes disease (LCPD) with a significant prognostically negative impact on clinical symptoms, cartilage biochemistry, mechanical properties of the cartilage, joint biomechanics, and prognosis toward healing with a congruent, spherical head of femur. Synovitis causes cartilage edema, deterioration of the cartilage's mechanical properties, cartilage hypermetabolism, and, subsequently, cartilage hypertrophy. This sequence of events could explain the clinical course, which consists of cartilage hypertrophy, lateral subluxation, anterolateral deforma…

Histochemical study of normal and collapsed tracheas in dogs.

Tracheas from 15 toy breed dogs with normal tracheas and 6 dogs with collapsed tracheas were examined histologically and histochemically. Tracheal cartilage was analyzed for chondroitin sulfate by means of alcian blue (CEC method) and for calcium with alizarin red S. Cartilage arcs from dogs with collapsed tracheas had areas that were apparently hypocellular, and some had other areas that appeared like fibrocartilage or fibrous tissue. Histochemically, collapsed tracheal cartilage had less chondroitin sulfate and calcium than did normal tracheal cartilage. Cartilage arcs from the collapsed tra…

Sternocostal instability after Ravitch repair in adolescents; 3 case-reports and a review of surgical techniques in the literature.

UNLABELLED: Ravitch repair is a common surgical procedure to correct chest wall deformities. In this procedure, a subperichondreal cartilage resection of the deformed parasternal cartilage, and if necessary a repositioning of the sternum, is performed. Insufficient regeneration of the resected cartilage may result in sternocostal instability or even floating sternum. This rare complication presents with symptoms of pain and exercise intolerance. METHODS: We describe sternocostal instability in 3 adolescent patients after the Ravitch procedure for pectus carinatum and reviewed the literature o…
