Laboratory infection of primates with Ascaris suum to provide a model of allergic bronchoconstriction.

Wild-caught non-human primates are naturally sensitive to Ascaris antigen and provide a useful model for studying atopic asthma. The present study was carried out to determine the effect of experimentally infecting home-bred macaques with the nematode Ascaris suum and hence provide an alternative for the naturally occurring model. Following oral infection with the parasite the animals developed a blood eosinophilia and specific antibodies to purified Ascaris antigen. These antibodies appeared to be of the IgE class as they could be detected by a radiometric assay using a radiolabelled antibody

Chemical phototoxicity in humans.

Phototoxic reactions may be induced by endogenous or exogenous chemicals. Endogenous photosensitizers made by the body include porphyrin molecules which are responsible for the photocutaneous syndromes noted in the porphyrias. Exogenous photosensitizers may arrive on the skin through topical applications or may be distributed through the vasculature. Topical photosensitizers are found in cosmetics, medications, plants, and industrial and air pollutant emissions. Systemic photosensitizers consist primarily of therapeutic agents. Acute and chronic phototoxic reactions may occur. The acute respon

Experimental infection of rhesus monkeys with Lassa virus and a closely related arenavirus, Mozambique virus.

As a model for the pathogenesis of Lassa fever in humans, nine rhesus monkeys were inoculated with Lassa virus. Three monkeys had had a previous asymptomatic experimental infection with Mozambique virus, a closely related arenavirus; these monkeys were protected from illness and viremia and manifested only mild pathologic lesions. The other animals developed severe disease and viremia. At necropsy, hepatocellular necrosis, interstitial pneumonia, a unique pulmonary arteritis, adrenal gland necrosis, encephalitis, and uveitis were prominent pathologic lesions which correlated with the organ tit

Skeletal muscle circulation during sufentanyl and morphine anesthesia in propranolol treated dogs.

Sufentanyl is a new, potent, short-acting, fentanyl-like morphinomimetic. In the present study we compared the effects of high doses of sufentanyl and morphine on the peripheral circulation in beta-blocked dogs. Skeletal muscle surface pH (m-pH) was recorded continuously as an index of the microcirculation. Sufentanyl (0.01 mg/kg) had no adverse effects on the peripheral perfusion. Morphine (4 mg/kg) caused a severe and rapid fall in m-pH from 7.34 to 7.14 during the 30-min experimental period. At the same time calculated blood volume decreased by 20%. This hypovolemic deterioration of the cir

Studies on human heart citrate synthase.

Citrate synthase from human heart was purified by affinity chromatography with Sepharose-ATP. The molecular weight (100,000) and the presence of two presumably identical subunits do not differ from other mammalian citrate synthases. However, the kinetics constants and immunologic characteristics of the enzyme differed from other mammalian citrate synthases. The Km values for acetyl-CoA (0.4 microM) and oxaloacetate (0.25 microM) were about an order of magnitude lower than those previously found for other mammalian (and eucaryotic) citrate synthases. The kinetics constants for the reverse react

Immunologically mediated toxicity.

A major consideration which precedes the release of a new chemical into the environment of human beings is the possibility of its producing an undesirable or toxic effect. Toxicity is the clinical end-point of a mechanism which results in tissue damage. One such mechanism is mediated via the immunological system. The effect of some chemicals on immunologically competent cells has been to induce perturbations of immunological competence. This is currently under debate as a toxic hazard, and the concept is explored here, particularly in relation to the action of alkyl tins. Hypersensitivity, whi

Human osteopetrosis: a histological, ultrastructural, and biochemical study.

UNLABELLED: Histological ultrastructural, and biochemical studies of the tissues of a patient with osteopetrosis were done. The bone contained an increased number of osteoclasts which were characterized ultrastructurally by the absence of ruffled borders and clear zones, hallmarks of actively resorbing osteoclasts. In contrast with normal human bone, tissue collagenase was not detected in osteopetrotic bone cultured in vitro, nor was tissue collagenase activity released when the osteopetrotic bone was incubated with parathyroid hormone. No striking abnormalities of parathyroid hormone or calci

Experimental quantification of balance using whole-body stability regions from postural sway exercises.

BACKGROUND: Comprehensive balance assessment is essential for evaluating balance deficiencies, particularly in individuals with limited mobility and increased fall risk. However, clinical balance assessments often rely on subjective scoring and simplified models that may not capture the full dynamics of human postural control. Additionally, center of pressure (COP)-based metrics and inverted pendulum models offer limited insight into whole-body balance ability. Balance regions (BRs) offer a more holistic approach by quantifying balanced center of mass (COM) states. Previously, BRs were used to

DHCR7 inhibition ameliorates MetALD and HCC in mice and human 3D liver spheroids.

BACKGROUND & AIMS: Metabolic dysfunction and alcohol-associated liver disease (MetALD) results in the development of liver steatosis, inflammation, fibrosis, and hepatocellular carcinoma (HCC). De novo lipogenesis and cholesterol synthesis play an important role in the pathogenesis of MetALD. DHCR7 (7-dehydrocholesterol reductase) regulates the last stages of cholesterol production. METHODS: We investigated whether targeting DHCR7 can ameliorate the development of MetALD and HCC using experimental models and 3D human liver spheroids. RESULTS: Here, we demonstrate that partial genetic ablatio

Thermodynamic limitations on brain oxygen metabolism: physiological implications.

Recent thermodynamic modelling indicates that maintaining the brain tissue ratio of O2 to CO2 (abbreviated tissue O2 /CO2 ) is critical for preserving the entropy increase available from oxidative metabolism of glucose, with a fall of that available entropy leading to a reduction of the phosphorylation potential and impairment of brain energy metabolism. This provides a novel perspective for understanding physiological responses under different conditions in terms of preserving tissue O2 /CO2 . To enable estimation of tissue O2 /CO2 in the human brain, a detailed mathematical model of O2 and C

Studies in a Murine Granuloma Model of Instilled Carbon Nanotubes: Relevance to Sarcoidosis.

Poorly soluble environmental antigens, including carbon pollutants, are thought to play a role in the incidence of human sarcoidosis, a chronic inflammatory granulomatous disease of unknown causation. Currently, engineered carbon products such as multiwall carbon nanotubes (MWCNT) are manufactured commercially and have been shown to elicit acute and chronic inflammatory responses in experimental animals, including the production of granulomas or fibrosis. Several years ago, we hypothesized that constructing an experimental model of chronic granulomatosis resembling that associated with sarcoid

Proteomic analysis of skin invasion by blood fluke larvae.

BACKGROUND: During invasion of human skin by schistosome blood fluke larvae (cercariae), a multicellular organism breaches the epidermis, basement membrane, and dermal barriers of skin. To better understand the pathobiology of this initial event in schistosome infection, a proteome analysis of human skin was carried out following invasion by cercariae of Schistosoma mansoni. METHODOLOGY AND RESULTS: Human skin samples were exposed to cercariae for one-half hour to two hours. Controls were exposed to water used to collect cercariae in an identical manner, and punctured to simulate cercarial tu

Cyclopentenylcytosine. A carbocyclic nucleoside with antitumor and antiviral properties.

Cyclopentenylcytosine (CPE-C, 2), a pyrimidine analogue of the fermentation derived carbocyclic nucleoside neplanocin A, has been synthesized from the optically active cyclopentenylamine 3b by two synthetic routes. CPE-C demonstrates significant antitumor activity against both the sensitive and ara-C resistant lines of L1210 leukemia in vivo. Multiple long term survivors are produced in both tumor models. The compound also gives 100% growth inhibition of the solid human A549 lung and MX-1 mammary tumor xenografts grown in athymic mice. Good activity is also observed against a third human tumor

Dengue viruses serotypes 2 and 4 exhibit distinct infection kinetics and modulation of anti-viral immune responses in human tonsil histocultures.

Dengue virus (DENV) is the most prevalent mosquito-borne viral disease with over ten million cases worldwide. There are four antigenically distinct, co-circulating DENV serotypes (DENV 1-4) capable of infecting humans. Given the lack of immunocompetent animal models and the limitations of known cell culture models, more physiologically relevant experimental models are needed to recapitulate DENV infection and host immune responses. Building on previous observations that DENV-2 and DENV-4 serotypes elicit distinct innate immune responses in monocyte-derived dendritic cells (moDC) in vitro, we u

Ulcerative colitis reporting across medications: FAERS analysis and cross-species transcriptomic triangulation.

BACKGROUND: Ulcerative colitis (UC) is reported with medications from different therapeutic classes. We examined these reporting associations together with recorded clinical context and tested whether experimental cytokine-blockade signatures align with human UC mucosal expression. METHODS: FAERS reports from 2004Q1-2024Q4 were restricted to health professionals and primary-suspect medications. Ten previously identified candidates were evaluated using unique report-generic counts, reporting odds ratios (RORs), proportional reporting ratios, and a supportive Bayesian information component. Continuous mechanical compression-induced intervertebral disc degeneration and pain in the caudal spine in rats.

BACKGROUND: Chronic low back pain imposes substantial social and economic burdens due to its rising prevalence and associated disability. Intervertebral disc degeneration is the primary driver of discogenic back pain, though the mechanistic links between degenerative changes and persistent pain remain incompletely defined. Aberrant mechanical loading is recognised as a key trigger of disc degeneration. This work built a caudal spine compression rat model to explore how the P2Y2 receptor mediates load-induced disc degeneration and associated pain. METHODS: Fifty-six eight-week-old male laborat

Tri-dimensional pattern analysis of foodstuff bitemarks: A quantitative method of comparison.

Bitemark analysis has long been an important field within of forensic odontology, yet it remains controversial due to its inherent subjectivity and the limitations of currently adopted 2D methodologies. The National Institute of Standards and Technology has highlighted the lack of sufficient scientific foundation for conventional bitemark analysis, particularly with respect to bitemarks impressed on human skin, concerning uniqueness, reproducibility, and evidentiary reliability. Rather than addressing the limitations identified for human skin bitemark analysis, this study proposes a three-dime

What is the influence of knee joint movement on the maximal average mechanical power output of human quadriceps femoris muscle?

Performance during motor activities, such as wheelchair riding, cycling, rowing, and speed skating, depends critically on the average mechanical power output (AMPO) produced by the muscles. To maximize short-duration performance, limb movements should allow muscles to deliver maximal AMPO. However, it is unclear which movement maximizes AMPO of human muscle. In this study, we employed a Hill-type muscle-tendon-complex (MTC) model to predict the maximally attainable AMPO of human m. quadriceps femoris for various imposed periodic knee joint movements. Based on these predictions, we selected one
