PURPOSE OF REVIEW: The goal of this review is to summarize recent advances into the pathogenesis and treatment of systemic sclerosis (SSc) from genomic and proteomic studies.
RECENT FINDINGS: Intrinsic gene expression-driven molecular subtypes of SSc are reproducible across three independent datasets. These subsets are a consistent feature of SSc and are found in multiple end-target tissues, such as skin and esophagus. Intrinsic subsets as well as baseline levels of molecular target pathways are potentially predictive of clinical response to specific therapeutics, based on three recent clinic

A nitric oxide (NO)-boosting molecular agent (CyHU) was developed for enhanced mild-temperature photothermal therapy (MPTT). This photothermal agent can specifically target mitochondria and efficiently produce NO, inhibiting adenosine triphosphate-dependent heat shock proteins, reversing tumor heat tolerance and ultimately achieving MPTT of cancer cells.

Approximately 25% of newly diagnosed AML patients display an internal tandem duplication (ITD) in the fms-like tyrosine kinase 3 (FLT3) gene. Although both multi-targeted and FLT3 specific tyrosine kinase inhibitors (TKIs) are being utilized for clinical therapy, drug resistance, short remission periods, and high relapse rates are challenges that still need to be tackled. RNA interference (RNAi), mediated by short interfering RNA (siRNA), presents a mechanistically distinct therapeutic platform with the potential of personalization due to its gene sequence-driven mechanism of action. This stud

Sepsis-associated acute kidney injury (SA-AKI) is a common and severe complication in critically ill patients, independently associated with significantly increased mortality and a high risk of progression to chronic kidney disease. The pathogenesis of SA-AKI is complex and involves not only traditional concepts such as renal hypoperfusion, microcirculatory dysfunction, and direct tubular injury, but also, more importantly, the disordered activation of the host immune response. In recent years, regulated cell death (RCD) has emerged as a central mechanism linking immune dysregulation to tissue

The epidemic of heart failure continues apace, and development of novel therapies with clinical efficacy has lagged. Now, important insights into the molecular circuitry of cardiovascular autophagy have raised the prospect that this cellular pathway of protein quality control may be a target of clinical relevance. Whereas basal levels of autophagy are required for cell survival, excessive levels - or perhaps distinct forms of autophagic flux - contribute to disease pathogenesis. Our challenge will be to distinguish mechanisms that drive adaptive versus maladaptive autophagy and to manipulate t

Cardiovascular imaging has rapidly advanced over the past decades. Traditional imaging techniques such as echocardiography, computed tomography, and cardiovascular magnetic resonance are essential for assessing the structural and functional aspects of the cardiovascular system but often fall short in providing direct insights into disease activity. This gap is increasingly being bridged by molecular nuclear imaging techniques, including positron emission tomography and single-photon emission computed tomography, which enable the visualization of disease processes at the molecular and cellular

Treatment with molecular targeted agents together with immune checkpoint inhibitors will most likely improve the efficacy of current cancer immunotherapy. Because molecular targeted agents not only directly affect cancer cells, but also influence immune cells and modulate the tumor microenvironment, a better understanding of the overall immunological effects of these drugs will contribute to the rational design of combination therapies. Therefore, this study performed extensive immune monitoring of patients' peripheral blood mononuclear cells (PBMCs) to investigate the immunological effects of

Rhizomes of Drynaria quercifolia have long been traditionally used to manage rheumatic pain. However, there is limited research supporting this traditional practice and insufficient evidence demonstrating the molecular mechanisms of action of plant-derived bioactives in rheumatoid arthritis (RA). The current study aims to identify the effective components in Drynaria quercifolia methanol rhizome extract (DME) and their probable pharmacological mechanisms in alleviating Rheumatoid Arthritis (RA) using network-pharmacology, molecular docking, molecular-dynamics simulations, and gene expression-b

Alport syndrome (AS) is a hereditary glomerulopathy caused by mutations in the COL4A3, COL4A4, or COL4A5 genes, leading to progressive kidney decline and extrarenal manifestations. Advances in genetic testing have enabled the reclassification of AS into X-linked, autosomal recessive, and autosomal dominant forms, facilitating more accurate diagnosis and risk stratification. While renin-angiotensin-aldosterone system (RAAS) blockade remains the foundation of treatment to delay kidney failure, it does not directly target the underlying molecular pathology. Notwithstanding their key roles in therapy and as biological probes, 7% of approved drugs are purported to have no known primary target, and up to 18% lack a well-defined mechanism of action. Using a chemoinformatics approach, we sought to "de-orphanize" drugs that lack primary targets. Surprisingly, targets could be easily predicted for many: Whereas these targets were not known to us nor to the common databases, most could be confirmed by literature search, leaving only 13 Food and Drug Administration-approved drugs with unknown targets; the number of drugs without molecular targets likely i

Molecular characterisation of odontogenic cysts and tumours has advanced recently and molecular pathogenesis for several tumour types is included in the 5th edition World Health Organisation (WHO) classification of head and neck tumours. This narrative review examines critically the genetic and signalling alterations reported in odontogenic cysts and tumours and groups them by the signalling pathways affected: MAPK/ERK (mostly BRAF and KRAS mutations), Wnt/beta-catenin (mostly CTNNB1 mutations) and HH (PTCH1 mutations). Odontogenic tumours characterized by gene rearrangements are also discusse

Tongue squamous cell carcinoma (TSCC) is the most common and highly malignant oral squamous cell carcinoma (OSCC) with lymph node metastasis or distant organ metastasis and, thus, low overall survival rates. Studies on the molecular mechanism of the biologic characteristics of TSCC and exploration of targeted therapies have become the frontiers in the research of this disease. Long noncoding RNA (lncRNA), a type of noncoding RNA, plays a key role in the occurrence and development of tumors. In recent years, more and more studies on lncRNA have been focusing on tongue cancer. lncRNA affects the

Acute myeloid leukemia (AML) is a heterogeneous disease with outcomes dependent upon several factors, including patient age, karyotype, mutational status, and comorbid conditions. For younger patients, approximately 60% to 80% achieve complete remission with standard therapy involving cytarabine and an anthracycline. However, only 20% to 30% have long-term disease-free survival. For adults older than 60 years of age, only 40% to 55% achieve a complete remission, with dismal long-term survival rates. Unfortunately, the median age at diagnosis for AML is 70 years. Significant advances in our und

Therapeutic resistance remains a major cause of treatment failure and disease recurrence across cancer types, considerably limiting the long-term efficacy of chemotherapies, targeted therapies, and immunotherapies. Growing evidence indicates that resistance cannot be fully explained by static genetic alterations but rather arises from dynamic and reversible adaptive processes. Epigenetic regulation governs transcriptional plasticity, cellular state transitions, and tumor heterogeneity under therapeutic stress. Alterations in DNA methylation, histone modifications, chromatin accessibility, and

Estrogen receptor (ER)-positive breast cancer (BrCa) accounts for two-thirds of all BrCa cases worldwide. Therefore, ER-targeted endocrine therapy is the standard treatment for this disease. There has been a recent trend towards developing combination therapies using molecularly targeted drugs to improve outcomes. This study aimed to identify therapeutic targets demonstrating efficacy when combined with fulvestrant (a selective ER downregulator/degrader). We generated microRNA (miRNA) signatures from fulvestrant-treated MCF-7 cells by RNA sequencing. From the signature, we evaluated miR-374b-5

The oncogenic activation of the PI3K/AKT/mTOR (PAM) pathway, which is often associated with loss of PTEN, is an important adaptive mechanism to androgen-targeted therapy in castration-resistant prostate cancer (CRPC). The concomitant targeting of the PAM pathway and the androgen receptor (AR) pathway is a promising therapeutic strategy for CRPC. Many PAM pathway inhibitors only target one component of the PAM pathway, which can limit efficacy due to the activation of the uninhibited components. We previously showed that the multi-target pan-PI3K-mTORC1/2 inhibitor, gedatolisib, exerts greater

Adenoid cystic carcinoma (ACC) is a lethal salivary gland malignant neoplasm. Lung metastasis is the primary cause of mortality in ACC patients while there is no effective treatment available at present. In this study, a precise and biomimetic nanoplatform, CG/MC/U-M, is designed to combine cuproptosis, gas therapy and immunotherapy against metastatic adenoid cystic carcinoma. CG/MC/U-M was derived from ACC cell membranes, encapsulating carbonyl manganese (Mn2(CO)10), efferocytosis inhibitor (UNC2025) and metal-phenolic network (CuGA). After targeted accumulation in pulmonary metastases and ce
