Novel ACE-inhibitory peptides from egg yolk protein by stepwise enzymatic hydrolysis: Optimization, structural-activity characterization and molecular dynamics simulation.

To overcome the limited functionality of egg yolk protein from single-protease hydrolysis, this study developed a stepwise enzymatic strategy using alkaline (AH) and flavor (FH) proteases to concurrently enhance physicochemical properties and generate antihypertensive peptides. The optimal AH + FH treatment significantly improved powder solubility (78.88%, P < 0.05) and protein solubility (67.31%, P < 0.05). These improvements were associated with a reduction in β-sheet content and enhanced molecular flexibility. The hydrolysis also modified surface properties by increasing net negative charge…

Screening of ACE inhibitory peptides in digestive products of TGase-induced GCSG and evaluation of their absorption and transport properties.

The digestion characteristics of grass carp skin gelatin (GCSG) with varying degrees of cross-linking induced by transglutaminase (TGase) were investigated. Specific peptide segments with potential angiotensin-converting enzyme (ACE) inhibitory activity were screened from the digestive products of GCSG with a medium crosslinking degree (45.89 ± 0.99%) which had the most abundant digestive peptide segments. Then they were synthesized to confirm their ACE inhibitory efficacy. Additionally, the absorption and transport mechanisms of the active peptides were examined. The findings revealed that th…

Negatively Charged Peptides-Assisted Self-Assembled Mitochondria-Targeting Nanoprobe for Detecting Enterokinase and Image-Guided Navigation in Human Breast Cancer.

Surgical resection remains the primary treatment in human breast cancer. However, the accurate and rapid delineation of tumor margins intraoperatively is still a significant challenge. Herein, we designed a near-infrared fluorescent probe DCIP-ENTK by incorporating dicyanoisophorone dye with a negatively charged peptide, which can be specifically cleaved by enterokinase (ENTK, a tumor biomarker for breast cancer). In vitro experiments confirmed that the probe could quickly (<10 min), specifically, and sensitively (LOD = 0.031 U/mL) respond to ENTK. Notably, this probe was engineered as a pepti

Computational Discovery of Angiotensin-(1-7)-like Peptides Targeting the MAS Receptor from the Genomic Dark Matter of Saccharomyces cerevisiae.

Non-coding regions of eukaryotic genomes, once considered transcriptionally inert, are increasingly recognized as potential sources of bioactive peptides. Here, we mined the intergenic "genomic dark matter" of Saccharomyces cerevisiae to identify peptide ligands targeting the angiotensin-(1-7)/Mas receptor axis. Intergenic sequences were computationally translated in all reading frames and screened for short peptides with sequence and structural similarity to angiotensin-(1-7), a key modulator of the renin-angiotensin system. Two candidates showing 72-86% similarity were identified and structu…

Optimized LL-37-Derived Peptides Exhibit Antitubercular Activity, Induce Membrane Disruption, and P-Type ATPase Transcriptional Responses in Mycobacterium tuberculosis.

Tuberculosis (TB), caused by Mycobacterium tuberculosis (Mtb), remains a major cause of morbidity and mortality worldwide, particularly due to the emergence of drug-resistant strains. Membrane-active antimicrobial peptides (AMPs) represent attractive therapeutic candidates because they target bacterial envelope integrity and disrupt essential cellular processes. We evaluated two rationally designed LL-37-derived peptides: a truncated C-terminally amidated analog (LL37-1) and a modified variant incorporating N-terminal acetylation and a single D-amino acid substitution (D-LL37). Dose-response a…

CD4+ T-cell responses directed towards A/H5N1-derived haemagglutinin peptides increase in patients with seasonal influenza A virus infection.

OBJECTIVES: Recent outbreak of influenza A/H5N1 viral infections in birds and mammals worldwide prompted renewed concerns over an emerging H5N1-related influenza pandemic in the generally immunologically-naive global population. Cross-reactive CD4+ T cells between A/H1N1 and A/H5N1 subtypes targeting internal and surface influenza proteins have been identified in humans, indicating some protection against severe and fatal A/H5N1 infection. METHODS: To detect T-cell responses towards H1 and H5 viruses, we stimulated PBMCs from patients hospitalised with seasonal influenza A viruses, alongside

Impact of Structural Flexibility on Tunneling Contribution to the Reaction Mechanism of Spontaneous Succinimide Formation in Asn-Gly-Containing Peptides.

Spontaneous deamidation and isomerization of asparagine (Asn) residues is a major pathway of nonenzymatic protein aging, where Asn-Gly-containing peptides (NG motifs) represent the most reactive sequence. In this context, the first step of isomerization is succinimide formation, which is initiated by a concerted proton-transfer and nucleophilic activation step. Although the overall mechanism is established, the detailed nature of the rate-determining succinimide formation step and the possible role of nuclear quantum effects remain unclear. Here, we combine density functional theory (DFT), int…
