Detection of protein homolougs with HIV-1 antigens in bacteria of positive patients - phase II.

OBJECTIVES: Human immunodeficiency virus type 1 is widely accepted as the cause of AIDS (Acquires Immunodeficiency Syndrome) but it is necessary to consider other factors, not only HIV, which may be involved in AIDS process. It is apparent that a viral reservoir persists in virtually all infected individuals receiving HAART. Reservoirs were detected in macrophages and other cells of the blood system, in which even very effective HAART was not able to eliminate the virus. Over the last period of time AIDS research has been focused on the gut and other mucosal tissue as the major site of HIV inf

The ectodomain sheddase ADAM10 restricts HIV-1 propagation and is counteracted by Nef.

HIV-1 Nef enhances virus propagation by down-regulating CD4 and SERINC5. However, recent evidence points to the existence of an additional Nef-sensitive restriction mechanism. We now show that Nef suppresses the aberrant cleavage of HIV-1 gp41 by ADAM10, a virion-associated cellular ectodomain sheddase, and thus increases the amount of HIV-1 envelope glycoprotein (Env) on virions. Additionally, Nef inhibits the shedding of at least some cellular ADAM10 substrates, resulting in their accumulation on HIV-1 virions. Whereas Nef+ HIV-1 replicated only marginally better in the absence of ADAM10, th

Mitophagy in Botulinum Toxin Type A-Induced Muscle Atrophy.

OBJECTIVE: Botulinum toxin type A (BTXA) is widely used in oral and maxillofacial surgery to treat masseter hypertrophy and bruxism, inducing transient masseter atrophy, but the underlying mechanisms remain unclear. Mitophagy, essential for muscle fiber homeostasis, plays a critical role in muscle atrophy. This study aims to investigate whether mitophagy mediates BTXA-induced masseter muscle atrophy. METHODS: Rats received BTXA injections into masseter for 2 and 8 weeks. Muscle fiber composition was assessed via histology and immunofluorescence. Mitophagy markers (LC3-II, p62, beclin-1, Tomm2

Biomarkers of intestinal permeability in major psychiatric disorders: Distinct biological roles call for a more nuanced application.

BACKGROUND: Zonulin, lipopolysaccharide-binding protein (LBP), and soluble CD14 (sCD14) are frequently used as proxy biomarkers of intestinal permeability in studies involving patients with psychiatric disorders. Although often used interchangeably, these biomarkers reflect distinct biological processes. OBJECTIVE: This review integrates evidence from basic research on the mechanisms of action of zonulin, LBP, and sCD14 with findings from studies on major depressive disorder, bipolar disorder, and schizophrenia spectrum disorders, aiming to refine their application in gut-brain axis research.

Synthesis and characterization of pseudocantharidins, novel phosphatase modulators that promote the inclusion of exon 7 into the SMN (survival of motoneuron) pre-mRNA.

Alternative pre-mRNA splicing is a central element of eukaryotic gene expression. Its deregulation can lead to disease, and methods to change splice site selection are developed as potential therapies. Spinal muscular atrophy is caused by the loss of the SMN1 (survival of motoneuron 1) gene. A therapeutic avenue for spinal muscular atrophy treatment is to promote exon 7 inclusion of the almost identical SMN2 (survival of motoneuron 2) gene. The splicing factor tra2-beta1 promotes inclusion of this exon and is antagonized by protein phosphatase (PP) 1. To identify new compounds that promote exo

The channel-forming protein proaerolysin remains a dimer at low concentrations in solution.

Proaerolysin, the proform of the channel-forming protein aerolysin, is secreted as a dimer by Aeromonas sp. The protein also exists as a dimer in the crystal, as well as in solution, at least at concentrations in the region of 500 microg/ml. Recently it has been argued that proaerolysin becomes monomeric at concentrations below 100 microg/ml and that only the monomeric form of the protoxin can bind to cell surface receptors (Fivaz, M., Velluz, M.-C., and van der Goot, F. G. (1999) J. Biol. Chem. 274, 37705-37708). Here we show, using non-denaturing polyacrylamide electrophoresis, chemical cros

FTY720 inhibited proinflammatory cytokine release and osteoclastogenesis induced by Aggregatibacter actinomycetemcomitans.

BACKGROUND: Periodontitis is a bacteria-driven inflammatory bone loss disease. Previous studies showed that the oral pathogen Aggregatibacter actinomycetemcomitans (A. actinomycetemcomitans) stimulated the generation of sphingosine 1 phosphate (S1P). In addition, S1P signaling regulated the migration of osteoclast precursors and affected osteoclastogenesis. Furthermore, treatment with FTY720 (also called fingolimod, a modulator of multiple S1P receptors) alleviated osteoporosis and suppressed arthritis in animals. This study determined the effect of FTY720 on proinflammatory cytokine productio

Observations on the energy and redox state and protein synthetic rate in animal and human placentas.

The energy and redox states of placentas in newly anesthetized or sacrificed guinea pigs and sheep were compared with that of human placentas immediately after delivery. Subsequently the effect of ischemia was observed in both human and animal placentas. The influence of perfusion, using a fluid containing oxygenated red cells (adult human) was studied in the human placenta. Adenine nucleotide precursors were added to the perfusion fluid in an attempt to improve the energy and redox states and the mixed protein synthetic rate of the tissue. The ATP and lactate concentrations in guinea pig plac

Adult Low-Hypodiploid Acute Lymphoblastic Leukemia Emerges from Preleukemic TP53-Mutant Clonal Hematopoiesis.

UNLABELLED: Low hypodiploidy defines a rare subtype of B-cell acute lymphoblastic leukemia (B-ALL) with a dismal outcome. To investigate the genomic basis of low-hypodiploid ALL (LH-ALL) in adults, we analyzed copy-number aberrations, loss of heterozygosity, mutations, and cytogenetics data in a prospective cohort of Philadelphia (Ph)-negative B-ALL patients (n = 591, ages 18-84 years), allowing us to identify 80 LH-ALL cases (14%). Genomic analysis was critical for evidencing low hypodiploidy in many cases missed by cytogenetics. The proportion of LH-ALL within Ph-negative B-ALL dramatically

High level of sialate-O-acetyltransferase activity in lymphoblasts of childhood acute lymphoblastic leukaemia (ALL): enzyme characterization and correlation with disease status.

Previous studies had established an over-expression of 9-O-acetylated sialoglycoproteins (Neu5,9Ac(2)-GPs) on lymphoblasts of childhood acute lymphoblastic leukaemia (ALL). Here, we report the discovery and characterization of sialate-O-acetyltransferase enzyme in ALL-cell lines and lymphoblasts from bone marrow of children diagnosed with B- and T-ALL. We observed a positive correlation between the enhanced sialate-O-acetyltransferase activity and the enhanced expression of Neu5,9Ac(2)-GPs in these lymphoblasts. Anti-PLA2R1 IgG-mediated podocyte PLA2R1 cleavage drives glomerular subepithelial immune complex deposition in membranous nephropathy.

INTRODUCTION: In membranous nephropathy (MN), subepithelial immune deposits and glomerular basement membrane (GBM) thickening are defining lesions, yet how circulating anti-phospholipase A2 receptor 1 (PLA2R1) IgG generates GBM-anchored complexes from podocyte antigen remains unclear. METHODS: To clarify this, human podocytes stably expressing PLA2R1 and HEK293 cells expressing full-length or antibody-binding-deficient PLA2R1 (ΔN1-9) were stimulated with PLA2R1-positive MN [MN(P+)] serum or purified IgG or the metalloproteinase activator phorbol 12-myristate 13-acetate (PMA). Predicting Sepsis in Heart Failure Patients Supported by Left Ventricular Assist Devices: The Role of VE-Cadherin and ADAM10.

Vascular endothelial cadherin (VE-Cadherin) is a major endothelial adhesion molecule and can be cleaved explicitly by metalloproteinase domain-containing protein 10 (ADAM10). Vascular hyperpermeability may contribute to a greater susceptibility to sepsis in left ventricular assist device (LVAD) support patients. We aim to evaluate the efficacy of VE-Cadherin and ADAM10 for predicting sepsis in LVAD patients. We prospectively recruited 50 patients with advanced heart failure receiving LVAD therapy. Baseline and weekly postoperative blood samples (weeks 1-4) were collected, and plasma VE-cadheri

ADAM10-dependent disruption of N-cadherin and ventricular zone in post-hemorrhagic hydrocephalus.

Intraventricular hemorrhage (IVH) and subsequent post-hemorrhagic hydrocephalus (PHH) are common, severe complications of preterm birth associated with substantial morbidity and mortality. The pathophysiology of IVH/PHH involves the disruption of N-cadherin-mediated cell junctions in the ventricular zone (VZ), impaired differentiation of radial glial cells into ependymal cells, and reactive astrogliosis. A disintegrin and metalloproteinase domain-containing protein 10 (ADAM10) is a widely distributed metalloproteinase known to cleave cell junction proteins under inflammatory conditions similar

Activated PIK3CD drives marginal zone B cell development from early transitional progenitors by enhancing ADAM10 expression.

BACKGROUND: Activated PI3K-delta syndrome (APDS) is a human monogenic primary immunodeficiency disorder caused by mutations in the gene encoding the p110δ catalytic subunit of phosphoinositide 3-kinase, PIK3CD. APDS is characterized by complex immune phenotypes, including increased serum IgM, susceptibility to encapsulated bacterial respiratory infections, poor vaccine response, and autoimmune disorders. Impaired B cell development and responses, involving an expansion at the transient transitional T1 stage and a strikingly selective expansion of autoreactive marginal zone B cells, may be larg

Higher soluble ADAM10 plasma levels are associated with decreased cognitive performance in older adults carrying APOEε4.

The APOE gene, particularly its ε4 allele, is a significant genetic risk factor for Alzheimer's disease (AD) and influences amyloid-β (Aβ) pathology and cognitive decline. This study explores the relationship between APOEε4 genotype, plasma levels of soluble ADAM10 (sADAM10), and cognitive performance in cognitively unimpaired (CU) older adults and those with AD dementia. It is a cross-sectional analysis that included 85 participants assessed for cognitive function, APOE genotype, and plasma sADAM10 levels. Jolkinolide B Activates Mitophagy to Exhibit Antipancreatic Cancer Activity and Alleviate Cognitive Deficits in Alzheimer's Disease.

Dysfunctional mitophagy leads to the pathological accumulation of damaged mitochondria, which is closely associated with the development of human diseases such as cancer and Alzheimer's disease. The identification of safer and more effective mitophagy regulators may provide a novel approach for treating mitochondrial diseases. Covalent-binding drugs have attracted substantial attention due to their high specificity, selectivity, and low resistance potential. In this study, we demonstrated that the natural epoxide compound jolkinolide B (JB) specifically induces mitophagy both in vitro and in v

Antisense oligonucleotides directed against App and Rab5 normalized endosomal Rab activity and reversed DS-AD-linked degenerative phenotypes in the Dp16 mouse model of Down syndrome.

INTRODUCTION: Down syndrome (DS) markedly raises the risk of Alzheimer's disease (DS-AD). Our findings identified widespread dysregulation of the endolysosomal network (ELN) in DS and DS-AD brains, driven by increased APP gene dose, hyperactivation of RAB5, and elevated levels of guanine nucleotide exchange factors (GEFs) for RABs 7 and 11. METHODS: We investigated whether increasing APP gene dose and RAB5 hyperactivation contributed to neuropathogenesis and whether a clinically feasible intervention could reverse ELN changes. The Dp16 DS-AD mouse model was treated with a mouse App-specific a

Truncated mini LRP1 transports cargo from luminal to basolateral side across the blood brain barrier.

BACKGROUND: The most crucial area to focus on when thinking of novel pathways for drug delivery into the CNS is the blood brain barrier (BBB). A number of nanoparticulate formulations have been shown in earlier research to target receptors at the BBB and transport therapeutics into the CNS. However, no mechanism for CNS entrance and movement throughout the CNS parenchyma has been proposed yet. Here, the truncated mini low-density lipoprotein receptor-related protein 1 mLRP1_DIV* was presented as blood to brain transport carrier, exemplified by antibodies and immunoliposomes using a systematic

Large-scale gene expression changes in APP/PSEN1 and GFAP mutation models exhibit high congruence with Alzheimer's disease.

Alzheimer's disease (AD) is a complex neurodegenerative disorder with both genetic and non-genetic causes. Animal research models are available for a multitude of diseases and conditions affecting the central nervous system (CNS), and large-scale CNS gene expression data exist for many of these. Although there are several models specifically for AD, each recapitulates different aspects of the human disease. In this study we evaluate over 500 animal models to identify those with CNS gene expression patterns matching human AD datasets. Approaches included a hypergeometric based scoring system th
