Drug interactions in controlled clinical trials.

As much information as possible should be obtained in clinical trials to assess possible interactions between test drugs and concomitant medications prescribed for other medical indications. Side effect profiles were compared in patients taking buspirone, mean = 20 mg/day; diazepam, 20 mg/day; clorazepate, 23 mg/day; and placebo, with or without concomitant medications. Approximately 1,000 anxious patients were included in the analysis; 700 received buspirone. The use of a variety of common medications did not affect the side effect profile in the buspirone, clorazepate, and placebo groups, bu

Effect of nolinium bromide, cimetidine, and oxyphencyclimine on gastric hypersecretion induced by some secretagogues in the rat.

Gastric hypersecretion was induced in the pylorus-ligated rat by histamine, pentagastrin, carbachol, or insulin following the peroral administration of either nolinium bromide, a new non-anticholinergic antisecretory drug, cimetidine, an H2-histamine receptor antagonists, or oxyphencyclimine hydrochloride, a specific anticholinergic drug. Nolinium bromide cimetidine or oxyphencycline. Nolinium bromide and cimetidine weakly antagonized hypersecretions in response to carbachol and to insulin while oxyphencyclimine caused profound antagonism. It was concluded that there was incomplete parallelism

Evaluation of trimethoprim-sulphamethoxazole compound in treatment of salmonella infections.

Fifty patients suffering from infections caused by various salmonella species were treated with trimethoprim-sulphamethoxazole compound. Twenty-three had enteric fever and two were biliary carriers of Salmonella typhi. The other 25 suffered from infections caused by salmonella species other than S. typhi or S. paratyphi B. Twenty-one of the patients with enteric fever responded clinically to the drug, one failed treatment, and one died. Two patients suffering from typhoid fever relapsed and three temporarily excreted S. typhi in stools following treatment. One of the typhoid carriers was succe

Frespaciguat (MK-5475): A Novel Inhaled Soluble Guanylate Cyclase Stimulator for Pulmonary Hypertension Associated with COPD.

Soluble guanylate cyclase (sGC) stimulators are a validated therapeutic approach for pulmonary vasodilation; however, oral agents can cause systemic hypotension and may worsen ventilation-perfusion mismatch in pulmonary hypertension associated with chronic obstructive pulmonary disease (PH-COPD). To address these limitations, we applied a property-driven medicinal chemistry strategy to develop an inhaled sGC stimulator that maximizes pulmonary exposure while minimizing systemic exposure. Structure-activity relationship and pharmacokinetic studies identified high systemic clearance and low perm

Janus kinase inhibitors in the management of lichen planus: a systematic review.

BACKGROUND AND OBJECTIVES: Lichen planus (LP) is a chronic, lymphocytic, inflammatory skin disorder that can affect the skin, hair follicles, nails, and mucous membranes. It has limited treatment options. The pathogenesis of LP includes inflammation driven by interferons and the Janus kinase-signal transducer and activator of transcription (JAK-STAT) signaling pathway. Thus, Janus kinase inhibitors (JAKi) have been proposed as a viable therapeutic approach for LP. JAKi have recently been approved to treat some lymphocytic skin disorders such as alopecia areata and atopic dermatitis. The purpos

Ruxolitinib treatment for myelofibrosis with anemia: a retrospective assessment of real-world patient characteristics, treatment patterns, and survival.

Characterization of real-world outcomes in ruxolitinib-treated patients with myelofibrosis and anemia is limited. This retrospective analysis of the US Flatiron Health electronic health record-derived deidentified database focused on patients with myelofibrosis who did or did not have anemia and received ruxolitinib. More baseline anemic vs nonanemic patients received <20 mg/day of ruxolitinib throughout the first 3 months post baseline (25% vs 16%), and baseline anemic patients had numerically shorter times to first dose reduction (median, 5.6 vs 12.8 months) and discontinuation (16.7 vs 24.8

Characterization of a rare EGFR resistance mutation in a lung cancer patient with a long response to osimertinib: case report.

INTRODUCTION: Osimertinib is the standard of care in first-line therapy for advanced EGFR-mutant NSCLC, but resistance invariably develops through EGFR-dependent or independent mechanisms. CASE DESCRIPTION: Here, we report the case of a stage IVA adenocarcinoma patient with several metastases harboring an EGFR exon 19 deletion, which progressed locally after five years on osimertinib. Sequencing of the re-biopsy revealed the emergence of an acquired exon 20 mutation, p.(Cys797Ala), not previously characterized and absent from variant databases. Immunohistochemical analysis (IHC) showed phosph

Differential Responses of Botrytis Isolates to Pristine, Luna Tranquility, Switch, and Their Active Ingredients.

Botrytis cinerea is an adaptable, high-risk pathogen capable of developing resistance to several site-specific fungicides. To manage gray mold, blueberry growers in Washington State and Oregon follow spray regimes that include rotation of site-specific fungicides, tank-mixing site-specific fungicides with a multisite fungicide such as captan, and using premix fungicides to slow down selection of B. cinerea populations with resistance to site-specific fungicides. In this study, we conducted conidial germ tube growth assays on media to assess responses of Botrytis isolates to discriminatory dose

Characterising the urinary excretion of thymidine dimer photolesions in mice.

DNA damage in the form of thymidine dimers (T <  > Ts) is formed in the skin in response to ultraviolet radiation (UVR). T <  > Ts are excised from the skin and excreted into the urine at a quantifiable concentration. Recently, detection of T <  > Ts were described in murine urine, but little is known about the excretion pattern or kinetics of T <  > Ts in murine models following UVR. This study presents a characterisation of the urinary excretion of T <  > Ts in hairless mice exposed to UVR. Using ultrahigh-performance liquid chromatography coupled to mass spectrometry, T <  > T excretion sho

Clinical Outcomes of Ruxolitinib Treatment in Patients With IPSS Intermediate-1-Risk Myelofibrosis: Interim Analysis From an Italian, Prospective Study (ROMEI).

Ruxolitinib (RUX), a JAK1/2 inhibitor, demonstrated treatment benefits for myelofibrosis (MF) in intermediate-1 (Int-1)-risk patients with a significant disease burden; however, the evidence is scarce. This interim analysis investigated the efficacy and safety of ruxolitinib in patients with Int-1-risk MF. ROMEI, a multicenter, observational, prospective study, enrolled 508 adult patients with MF receiving ruxolitinib according to approved indications. The present interim analysis was focused on 107 eligible patients in the Int-1-risk group. Primary endpoints included changes in symptoms respo

A meta-analysis of the efficacy and safety of Janus kinase inhibitors in patients with ulcerative colitis.

BACKGROUND: Ulcerative colitis (UC) is a chronic inflammatory bowel disease characterized by relapsing intestinal inflammation and substantial impairment of quality of life. Janus kinase (JAK) inhibitors are orally administered small-molecule agents that interfere with intracellular cytokine signaling pathways central to UC pathogenesis. This systematic review and meta-analysis aimed to assess the therapeutic benefits and safety profile of JAK inhibitors in adults with moderate-to-severe UC. METHODS: A systematic literature search was conducted in PubMed (from inception to November 2025), Emb

Optimized UPLC-MS/MS Method for the Simultaneous Quantification of Pazopanib and GSK-1268997 and Its Application to Drug-Drug Interaction Studies.

INTRODUCTION: Pazopanib, an oral vascular endothelial growth factor receptor tyrosine kinase inhibitor (VEGFR-TKI), is approved for treating advanced renal cell carcinoma (RCC). However, pazopanib can cause hypertension, often necessitating co-administration with antihypertensives like nicardipine. Given their shared metabolic pathway via cytochrome P450 3A4 (CYP3A4), this study investigated their potential drug-drug interaction (DDI). METHODS: An ultra performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS) method was developed for the simultaneous quantification of pazopanib
