Bovine In Vitro Fertilization Embryo Development Predicted by Capacitation-Induced Zinc Ion Flux and Acrosome Remodeling.

Sire-dependent variation in bovine in vitro fertilization (IVF) success is poorly explained by conventional semen parameters once samples meet standard quality thresholds. Sperm capacitation is required for acquisition of fertilization competence, yet its contribution to sire-dependent variation in bovine IVF outcomes remains unclear. Fifteen bulls (Bos taurus) were evaluated across five IVF replicates using 30 abattoir-derived oocytes per bull per replicate. Sperm were analyzed before (0 h) and after 3 h of in vitro capacitation using image-based flow cytometry to quantify zinc ion localizati

Male Reproductive and Cellular Damage After Prenatal 3.5 GHz Radiation Exposure: One-Year Postnatal Effects.

This study investigates the long-term effects of prenatal exposure to 3.5 GHz radiofrequency radiation (RFR) on male reproductive health. Pregnant Wistar Hannover rats were divided into sham control, full-gestation exposure (3T RFR), and late-gestation exposure (2T RFR) groups (2 h/day). Male offspring were euthanized at 12 months for testicular analysis. In the 3T RFR group, seminiferous tubule diameter and epithelial height were significantly reduced compared to controls (adjusted p = 0.03 and 9.71 × 10-8), along with lower Johnsen scores (adjusted p = 0.022). Abnormal sperm morphology incre

Lipidomics and transcriptomics analyses reveal high sperm storage ability-related lipids/genes in the liver-blood-utero-vaginal junction mucosa axis of female ducks.

The sperm storage trait of female poultry refers to their ability to store sperm in the oviduct, thereby enabling them to continuously produce fertilized eggs. The analysis of the underlying principles of this ability not only helps to improve reproductive efficiency, but also contributes to driving innovation in semen preservation technologies. However, the underlying mechanisms of this ability remain unclear. This study successfully quantified the sperm storage ability in 98 female ducks. Histological comparisons between female ducks with higher sperm storage ability (HFSS) and lower sperm s

Dynamic architecture of mammalian paternal chromatin: histone-to-protamine exchange and post-fertilization reprogramming.

Chromatin remodeling in male germ cells and after fertilization plays a pivotal role in genetic transmission and early embryonic development. During spermatogenesis, histone-based chromatin undergoes progressive reorganization: canonical histones are gradually replaced by testis-specific variants, then by transition proteins, and ultimately by highly basic protamines (PRM1 and PRM2). This hierarchical replacement, modulated by histone post-translational modifications-including hyperacetylation, ubiquitination, and dynamic methylation-and supported by molecular chaperones and chromatin remodele

Protective effect of exosomes derived from platelet-rich plasma during human sperm cryopreservation.

Fertility preservation is a critical aspect of improving human life quality. Sperm cryopreservation is highly recommended for men at risk of infertility; however, it can have detrimental effects on sperm quality. In this study, we evaluate the effects of platelet-rich plasma (PRP) and PRP-derived exosomes on cryopreservation outcomes in men with asthenoteratozoospermia referred to a public infertility center. The semen samples from 75 men with asthenoteratozoospermia (based on WHO 2020 criteria) were randomly divided into five groups: Control (conventional freezing), Exo/before, Exo/After, PRP

Phosphoproteomic profiling of Siberian sturgeon (Acipenser baerii, Brandt, 1869) sperm: insights into cryopreservation-induced changes with DMSO and methanol†.

Semen cryopreservation is a crucial technique in aquaculture, enabling the long-term preservation of genetic material from endangered species such as the Siberian sturgeon (Acipenser baerii, Brandt, 1869). While methanol (MeOH) and dimethyl sulfoxide (DMSO) similarly affect sturgeon semen quality post-thaw, DMSO significantly reduces hatching rates. Given our prior findings on cryoprotectant-specific differences in the sturgeon sperm proteome, we investigated the molecular basis of DMSO-induced dysfunction using label-free quantitative phosphoproteomics. In fresh sperm, 1649 phosphorylation si

In vitro fertilization induces reproductive changes in male mouse offspring and has multigenerational effects.

In vitro fertilization (IVF) is a noncoital method of conception used to treat human infertility. Although IVF is viewed as largely safe, it is associated with adverse outcomes in the fetus, placenta, and adult offspring. Because studies focusing on the effect of IVF on the male reproductive system are limited, we used a mouse model to assess the morphological and molecular effects of IVF on male offspring. We evaluated 3 developmental stages: 18.5-day fetuses and 12- and 39-week-old adults. Regardless of age, we observed changes in testicular-to-body weight ratios, serum testosterone levels,

Chemosensory signalling in human sperm is controlled by Ca2+ influx via CatSper and Ca2+ clearance via plasma membrane Ca2+ ATPases.

BACKGROUND AND PURPOSE: Loss of function of the sperm-specific Ca2+ channel CatSper is a common channelopathy that causes male infertility. CatSper controls the intracellular Ca2+ concentration and, thereby, the motility of human sperm. Activation of CatSper by oviductal ligands evokes a transient Ca2+ increase, which entails changes in the flagellar beat that are required for fertilisation. The CatSper-mediated Ca2+ influx has been studied extensively, whereas the mechanisms underlying Ca2+ clearance and recovery from Ca2+ influx have remained ill-defined. EXPERIMENTAL APPROACH: We examined

The effects of radiofrequency radiation on male reproductive health and potential mechanisms.

Recent studies have demonstrated that radiofrequency (RF) radiation emanating from devices such as mobile phones and Wi-Fi may have adverse effects on male reproductive health. This radiation can elevate testicular temperature, potentially compromising sperm quality and DNA integrity, and influence the specific absorption rate (SAR) across different body regions, leading to detrimental reproductive outcomes. Furthermore, exposure to RF radiation has been linked to conditions that could affect male reproductive function, such as oxidative stress, alterations in ion transitions across cell membr

Inexpensive thermal containers and insulation materials prevent deterioration of semen parameters for less than 90 minutes.

Male infertility contributes substantially to overall infertility, with temperature changes adversely affecting the sperm quality. During infertility treatments, the exposure of home-collected semen to extreme temperatures during transport deteriorates the semen parameters. This study investigates the effectiveness of thermal containers and insulation materials for preserving semen quality at low temperatures. Semen samples from 35 healthy male partners undergoing fertility treatments were analyzed. The samples were segregated in three groups and assessed: standard collection containers (Group

Deciphering the microbiome, lipopolysaccharides, and metabolome interplay: Unveiling putrescine's mechanism for enhancing sperm quality in heat-stressed boars.

Global warming has added to concerns regarding declining male fertility due to high temperatures. As a metabolite of arginine, putrescine improves gut health and promotes testicular development in boar; however, its action in improving semen quality under heat stress is unknown. Therefore, we assessed the effect of putrescine on the semen quality of boars in a heat stress model. Results showed that putrescine ameliorated the heat stress-induced decline in semen quality and testosterone levels in boars, confirmed by sperm viability, immobility rate, and apoptosis levels. A novel sperm FcR binding assay that assesses sperm fertilizing potential.

Freshly ejaculated mammalian sperm have poor fertilizing ability, with fertility only gained after sperm undergo capacitation and the acrosome reaction. To visualize exposed Fc receptors (FcRs), which occur during the acrosome reaction and whose absence has been related to infertility, a novel sperm FcR binding assay (FcR assay) was developed to assess fertilizing potential of sperm in proof-of-concept studies. A competition binding assay between sperm FcR and exogenously added FcR was used to assess whether the FcR was a functioning ligand in bull sperm. Once FcR was confirmed as a functional

Potential effects of environmental toxicants on sperm quality and potential risk for fertility in humans.

Semen quality is a key factor in male fertility, but defining normal reference values for semen parameters remain challenging. Over the past four decades, several authors have reported a noticeable decline in sperm parameters, raising concerns about male reproductive health. While the exact causes remain unclear, potential contributors include environmental pollution, endocrine disruptor chemicals (EDCs) and oxidative stress, with the latter becoming a growing concern. Environmental changes and increased exposure to EDCs, such as pesticides, herbicides, bisphenol A (BPA), phthalates, polychlor
