Engineered surface topographies have proven to be an efficient method to replicate in vitro the architecture of highly organized tissues by controlling the spatial arrangement of cells. In this work, we studied the ability to guide muscle cell development using femtosecond-laser-induced topography on standard culture plates. This technique allows the creation of highly porous microstructured channels that exhibit strong hydrophobicity while offering mechanical cues for cell guidance. We characterized the resulting engineered surfaces by assessing their capacity to direct cellular organization.

Inflammatory bowel disease (IBD) patients disproportionately suffer from depression and anxiety, which exacerbate clinical outcomes yet remain inadequately addressed by current anti-tumor necrosis factor (TNF) biologics. To bridge this therapeutic gap, we engineered Escherichia coli Nissle 1917 (eEcN) as a clinically translatable living microbial therapeutics platform for dual-targeted intervention. eEcN is designed to autonomously secrete immunomodulatory outer membrane vesicles (OMVs) bearing TNFα-neutralizing nanobodies (TNFαnb). These bioengineered nanovesicles specifically target inflamed

Hepatocellular carcinoma, a malignant tumor with high incidence and mortality rates, urgently requires the development of novel therapeutic strategies that are highly efficient and minimally toxic. Sonodynamic therapy has garnered significant attention due to its non-invasiveness, deep tissue penetration capability, and low side effects, yet its efficacy is highly dependent on the development of high-performance sonocatalyst. This study successfully prepared Pd-integrated indium oxide with abundant structural defects as a novel sonocatalyst for the highly efficient sonocatalytic eradication of

Fabry disease is an X-linked lysosomal storage disorder caused by mutations in the GLA gene, leading to deficient α-galactosidase A (α-Gal A) activity and pathological accumulation of globotriaosylceramide (Gb3) and globotriaosylsphingosine (Lyso-Gb3) in various organs. To enhance α-Gal A expression and secretion for recombinant adeno-associated viral (rAAV) gene therapy, the signal peptide was engineered in HepG2 cells. Among 22 heterogeneous signal peptides, sp21 was identified and validated for this purpose. The enzyme activity of GLA with sp21 in conditioned media was 2.3 times higher than

BACKGROUND: Streptococcus agalactiae (GBS) causes severe tilapia streptococcosis with heavy aquaculture losses; existing vaccines have administration or efficacy limitations. This study used CRISPR-Cas9 to construct recombinant Escherichia coli DH5α-ORF4-GFP (targeting GBS scpB gene ORF4 fragment), optimized tilapia immersion immunization doses/frequencies, and evaluated the vaccine's protective efficacy, biosafety and regulatory effects via multi-dimensional assays.
RESULTS: The optimal regimen was single immersion at 1.5 × 104 CFU/mL, with a maximum RPS of 73.13% and stable 65.79% in valida

Inflammatory bowel disease (IBD) remains an incurable condition, often accompanied by high rates of anxiety and depression, further diminishing the quality of life of patients. Endogenous gasotransmitters, such as carbon monoxide (CO) and hydrogen sulfide (H₂S), exhibit potent anti-inflammatory and immunomodulatory effects. However, their therapeutic application is limited by challenges in targeted delivery to affected tissues. Here, we propose a novel strategy for targeted gut delivery of CO/H2S through engineering Escherichia coli Nissle 1917 (EcN) with CO/H2S-releasing copolymer (POSR) load

Vascularized composite allotransplantation (VCA) has emerged as a robust alternative for addressing anatomically complex defects but requires a toxic lifelong immunosuppressive regimen. Tissue engineering offers the promise of creating recipient-specific alternative grafts using a decellularization and recellularization approach. In this article, we establish a reliable protocol for human face decellularization by immersion as a new tool in the development of engineered graft alternatives for reconstructive surgery. Three cadaveric face grafts were immersed in 1% sodium dodecyl sulfate for 216…

mRNA delivery offers new opportunities for disease treatment by directing cells to produce therapeutic proteins. However, designing highly stable mRNAs with programmable cell type-specificity remains a challenge. To address this, we measured the regulatory activity of 60,000 5' and 3' untranslated regions (UTRs) across six cell types and developed PARADE (Prediction And RAtional DEsign of mRNA UTRs), a generative AI framework to engineer untranslated RNA regions with tailored cell type-specific activity. We validated PARADE by testing 15,800 de novo-designed sequences across these cell lines a

Adeno-associated virus (AAV) has emerged as a pivotal delivery tool in clinical gene therapy owing to its minimal pathogenicity and ability to establish long-term gene expression in different tissues. Recombinant AAV (rAAV) has been engineered for enhanced specificity and developed as a tool for treating various diseases. However, as rAAV is being more widely used as a therapy, the increased demand has created challenges for the existing manufacturing methods. Seven rAAV-based gene therapy products have received regulatory approval, but there continue to be concerns about safely using high-dos

Crosstalk between nerves and bone is essential for bone repair, for which Schwann cells (SCs) are crucial in the regulation of the microenvironment. Considering that exosomes are critical paracrine mediators for intercellular communication that exert important effects in tissue repair, the aim of this study is to confirm the function and molecular mechanisms of Schwann cell-derived exosomes (SC-exos) on bone regeneration and to propose engineered constructs that simulate SC-mediated nerve-bone crosstalk. SCs promoted the proliferation and differentiation of bone marrow mesenchymal stem cells (

Bladder cancer is an understudied area, particularly in genetically engineered mouse models (GEMMs). Inbred GEMMs with tissue-specific Cre and loxP sites have been the gold standards for conditional or inducible gene targeting. To provide faster and more efficient experimental models, an ex vivo organoid culture system is developed using adenovirus Cre and normal urothelial cells carrying multiple loxP alleles of the tumor suppressors Trp53, Pten, and Rb1. Normal urothelial cells are enzymatically disassociated from four bladders of triple floxed mice (Trp53f/f: Ptenf/f: Rb1f/f). The urothelia

The capacity to efficiently deliver the gene-editing enzyme complex to target cells is favored over other forms of gene delivery as it offers one-time hit-and-run gene editing, thus improving precision and safety and reducing potential immunogenicity against edited cells in clinical applications. Here we performed a proof-of-mechanism study and demonstrated that a simian adenoviral vector for DNA delivery can be repurposed as a robust intracellular delivery platform for a functional Cas9/guide RNA (gRNA) complex to recipient cells. In this system, the clinically relevant adenovirus was genetic

Bioavailable glutamine (Gln) is critical for metabolism, intestinal health, immune function, and cell signaling. Routine measurement of serum Gln concentrations could facilitate improved diagnosis and treatment of severe infections, anorexia nervosa, chronic kidney disease, diabetes, and cancer. Current methods for quantifying tissue Gln concentrations rely mainly on HPLC, which requires extensive sample preparation and expensive equipment. Consequently, patient Gln levels may be clinically underutilized. Cell-free protein synthesis (CFPS) is an emerging sensing platform with promising clinica

Immune checkpoint blockade exhibits considerable antitumor activity, but previous studies have reported instances of severe treatment-related adverse events. We sought to explore local immune checkpoint blockade, with an antibody (Ab) form that would be retained intra- or peritumorally, limiting systemic exposure. To accomplish this, we conjugated the checkpoint blockade Abs to an extracellular matrix (ECM)-super-affinity peptide derived from placenta growth factor-2 (PlGF-2123-144). We show enhanced tissue retention and lower Ab concentrations in blood plasma after PlGF-2123-144 conjugation,

Trinucleotide repeats can be highly unstable, mutating far more frequently than point mutations. Repeats typically mutate by addition or loss of units of the repeat. CAG repeat expansions in humans trigger neurological diseases that include myotonic dystrophy, Huntington disease, and several spinocerebellar ataxias. In human cells, diverse mechanisms promote CAG repeat instability, and in mice, the mechanisms of instability are varied and tissue-dependent. Dissection of mechanistic complexity and discovery of potential therapeutics necessitates quantitative and scalable screens for repeat muta

Despite advances in bone tissue engineering, repairing diabetic bone defects remains a formidable challenge. Here, we present an innovative strategy for diabetic bone regeneration, the BCPG-MF@Y scaffold, which features diamond lattices of 3D-printed biphasic calcium phosphate (BCP) integrating porous gelatine methacryloyl (pGelMA) hydrogels embedded with Yoda1-functionalized manganese dioxide-ferritin (MF@Y) biomimetic nanozymes. Our comprehensive analyses revealed that the MF@Y nanozyme orchestrates mitochondrial homeostasis in bone mesenchymal stem cells (BMSCs), effectively scavenges mtROS…

The rapid and nondestructive detachment of cells from culture surfaces is critical for obtaining high-quality cell populations in tissue engineering applications. Thermoresponsive coatings, such as poly-(N-isopropylacrylamide) (PNIPAM), enable enzyme-free cell release; however, their practical utility is often constrained by inherently slow detachment kinetics. Here, we report a hybrid strategy that synergistically integrates surface acoustic wave (SAW) stimulation with a PNIPAM-based coating to markedly accelerate cell detachment. Under optimized conditions, this system achieves a cell detach…

The differences in the phospholipid profile between bacterial and mammalian membranes present a promising direction for designing antimicrobial peptides (AMPs). A key challenge, however, is engineering AMPs with a high affinity for bacterial anionic phospholipid (phosphatidylglycerol, PG) while minimizing off-target interactions with mammalian phosphatidylserine (PS). Herein, we report a multiarmed radially amphiphilic AMP (maRAP) design that enhances PG recognition, exhibiting potent antibacterial activity and exceptional antibacterial selectivity. Compared to the single-arm RAP (1aRAP), maRA

The current management of end-stage liver diseases relies on organ transplantation but is constrained by donor scarcity and immune rejection. Induced pluripotent stem cell (iPSC)-derived cells represent a promising alternative, as they can be expanded indefinitely, genome-edited to reduce immunogenicity and employed to develop more mature three-dimensional cultures. Here, we combined tissue engineering with hypoimmunogenic iPSC technology to generate off-the-shelf hepatic organoids that integrate parenchymal and non-parenchymal cells: hepatocytes, cholangiocytes, hepatic stellate cells, Kupffe…
