Carcinogen adducts as an indicator for the public health risks of consuming carcinogen-exposed fish and shellfish.

A large variety of environmental carcinogens are metabolically activated to electrophilic metabolites that can bind to nucleic acids and protein, forming covalent adducts. The formation of DNA-carcinogen adducts is thought to be a necessary step in the action of most carcinogens. Recently, a variety of new fluorescence, immunochemical, and radioactive-postlabeling procedures have been developed that allow the sensitive measurement of DNA-carcinogen adducts in organisms exposed to environmental carcinogens. In some cases, similar procedures have been developed for protein-carcinogen adducts. In

[Recent concepts of the organization and structure of human trabecular bone--results of combined 2- and 3-dimensional analysis].

Every loss of bone mass caused by dysfunction of the endocrine glands and mechanically, respectively, is accompanied by changes of the bone structure. For the pathogenetic mechanisms which in man lead to the abolition of the bone structure in osteoporosis the knowledge of the physiological principles of construction is a basic prerequisite. Hitherto performed studies have shown that with the help of two-dimensional section-cuttings a tridimensional reconstruction is either not possible or can be carried out only by means of serial sections and extensive computer procedures in circumscribed par

Preoperative embolization of the head and neck: indications, patient selection, goals, and precautions.

Preoperative embolization was performed in 39 patients with 44 paragangliomas of the head and neck. Because of their complex vascular supply and their relation to vital structures such as the internal carotid artery and the lower cranial nerves, paragangliomas of the temporal bone represent challenging lesions to both the neuroradiologist and the otoneurosurgeon. Detailed classification by high-resolution CT and recognition of the multi- or monocompartmental vascular composition and of dangerous situations by selective angiography are essential prerequisites for safe and effective devasculariz

Long-term behavior of 239Pu, 241Am and 233U in different bones of one-year-old rats: macrodistribution and macrodosimetry.

Female and male rats of the Sprague-Dawley strain, aged about 13 months, were injected intravenously with monomeric 239Pu-(30.7 kBq/kg), 241Am-(54.8 kBq/kg) or 233U-citrate (56.6 kBq/kg) and killed between 7 and 540 days after injection. In both sexes the wet skeletal weight was proportional to body weight; however, the skeletal weight of female rats remained constant, whereas the skeletal weight, and body weight, of male rats increased as a function of age. The initial skeletal deposition decreased in the order 239Pu greater than 241Am greater than 233U and for americium and uranium was great

Prognostic value of cachexia index in HR +/HER2 - metastatic breast cancer treated with CDK4/6 inhibitors.

PURPOSE: The prognostic significance of body composition parameters in patients with HR + /HER2 − metastatic breast cancer treated with CDK4/6 inhibitors remains poorly defined. The cachexia index (CXI), a composite marker integrating skeletal muscle mass, nutritional status, and systemic inflammation, may provide additional prognostic information in this setting. This study aimed to evaluate the prognostic value of body composition parameters, with a particular focus on CXI, in this patient population. METHODS: This retrospective single-center study included 84 patients with HR + /HER2 − meta

Clinical Implication of Mid-Range Dynamic Instability in Lumbar Degenerative Spondylolisthesis.

STUDY DESIGN: Retrospective evaluation. PURPOSE: To determine the prevalence of mid-range dynamic instability in patients with degenerative spondylolisthesis (DS) and to evaluate the clinical implication of mid-range instability (MI). OVERVIEW OF LITERATURE: Instability is identified by measuring vertebral body anterior-posterior translation on static end-range flexion and extension lateral radiographs. Mid-range kinematics could evince occult dynamic instability in which motion is not appreciated at the terminal-range of motion. METHODS: In this study, 30 patients with DS with checked stan…

Ectopic bone formation and systemic bone loss in a transmembrane TNF-driven model of human spondyloarthritis.

BACKGROUND: The transmembrane-TNF transgenic mouse, TgA86, has been shown to develop spontaneously peripheral arthritis with signs of axial involvement. To assess similarity to human spondyloarthritis, we performed detailed characterization of the axial, peripheral, and comorbid pathologies of this model. METHODS: TgA86 bone pathologies were assessed at different ages using CT imaging of the spine, tail vertebrae, and hind limbs and characterized in detail by histopathological and immunohistochemical analysis. Cardiac function was examined by echocardiography and electrocardiography and bone

Impact of sarcopenia in borderline resectable and locally advanced pancreatic cancer patients receiving stereotactic body radiation therapy.

BACKGROUND: Total psoas area (TPA), a marker of sarcopenia, has been used as an independent predictor of clinical outcomes in gastrointestinal (GI) cancers as a proxy for frailty and nutritional status. Our study aimed to evaluate whether TPA, in contrast to traditional measurements of nutrition like body mass index (BMI) and body surface area (BSA), was predictive of outcomes in borderline resectable pancreatic cancer (BRPC) and locally advanced pancreatic cancer (LAPC) patients receiving stereotactic body radiation therapy (SBRT). METHODS: Retrospective analysis of an institutional review b

High doses of caffeine reduce in vivo osteogenic activity in prepubertal rats.

Caffeine adversely affects endochondral ossification during fetal skeletal growth, and results in increased incidence of delayed and abnormal fetal skeletal development. Chronic caffeine intake also decreases growth hormone secretion. Thus, it is conceivable that caffeine may disrupt bone growth during the peripubertal period. This study aimed to investigate the impact of high-caffeine consumption on bone growth throughout puberty. A total of 51 male rats (21 days old) were divided randomly into three groups: a control group and two groups fed caffeine via gavage with 120 and 180 mg kg(-1)  da

Metolachlor exposure induces immune impairment and activates TLR4/NF-κB signaling in zebrafish embryos.

As a herbicide, Metolachlor (MET) has emerged as an 'invisible pollutant' due to its prolonged half-life, high water solubility, and extensive application scope, which pose potential threats to aquatic ecosystems and human health. Nonetheless, systematic evaluations of MET's effects on early growth development and immune mechanisms in vertebrates have been limited. This study is the first to systematically assess the immunotoxic effects of MET on non-target organisms, utilizing zebrafish as a model. The results indicate that exposure to MET reduces embryo hatching rates, slows heart rates, inc

A plant DNMT3 induces non-CG methylation and affects gene expression in human cells.

CG methylation is the predominant form of DNA methylation across eukaryotes, whereas non-CG methylation in vertebrates is largely restricted to neurons and embryonic stem cells and is mediated by DNMT3 enzymes. In contrast, plant DNMT3 homologs catalyze extensive non-CG methylation across diverse tissues. To test whether a plant DNMT3 can establish non-CG methylation and influence transcription in human cells, we expressed Physcomitrium patens DNMT3b (PpDNMT3) in HEK293 cells, which lack this modification. PpDNMT3 induced robust, genome-wide non-CG methylation in transiently and stably transfe

Resolving thyroid lineage cell trajectories merging into a dual endocrine gland in mammals.

The thyroid has a remarkable evolution, transforming from an exocrine constituent of the chordate endostyle to an endocrine gland in basal vertebrates. In mammals, a second endocrine cell type of presumed neural crest origin appears, although recent lineage tracing has firmly established thyroid C-cells are also endoderm-derived. Here, we characterize the global gene expression in both embryonic thyroid lineages at single-cell level and identify lineage-specific transcription factors and their network regulation of target genes implicated in thyroid development into a dual endocrine organ. Maternal thyroid hormones affect zebrafish embryo development.

Thyroid hormones (THs), including thyroxine (T4) and triiodothyronine (T3), regulate vertebrate development, influencing critical processes during embryogenesis and larval stages. However, their maternal transfer and developmental roles in fish remain poorly understood. This study investigates maternal THs' impact on zebrafish development, focusing on the somatotropic axis, a key regulator of growth and metabolism. Female zebrafish were fed diets with 12.5 or 25 μg T4/g body weight/day for five days, while controls received vehicle-only food. Plasma TH levels were measured, and females were br

The Contribution of the Double Rib Contour Sign and the Rib Index to the Study of Scoliogeny, Thoracic Deformity, Progression, Outcome of Treatments and Costoplasty for Idiopathic Scoliosis.

This opinion article refers to the "double rib contour sign" and to the rib index (DRCS and RI), to their reliability study results in the chest radiographs of a control group and to their validity study results. These two parameters were introduced by the first author in this report. The introduction of the Segmental Rib Index (SRI) and its relation to spinal deformity is also discussed. The RI has been confirmed to be a strong surrogate for scoliometric readings in idiopathic scoliosis (IS). The clinical applications of the RI are analyzed for the following: (a) the documentation of deformit

Neural crest cells: bridging embryology and regenerative medicine.

Neural crest cells are an embryonic stem cell population unique to vertebrates. Neural crest cells, which arise from the embryonic ectoderm germ layer, are thought to give rise to neurons and glia of the peripheral nervous system, as well as melanocytes throughout the body. In addition, neural crest cells give rise to many ectomesenchymal derivatives in the head, including cells typically derived from the mesoderm (cartilage, bone, and adipose tissue). Neural crest cells are a group of multipotent and migratory cells found throughout the developing embryo. These cells migrate to many different

Re-examination of the Subalyuk Neanderthal remains uncovers signs of probable TB infection (Subalyuk Cave, Hungary).

In 1932, skeletal remains of two Neanderthal individuals, a young adult female and a 3-4-year-old child, were discovered in Subalyuk Cave in Northern Hungary [1,2]. Results of the anthropological examination were published some years after this important discovery. Methodological progress encouraged re-examination of the material during the last few years. Radiocarbon dating revealed a chronological age of 39,732-39,076 cal. BP for the adult female and 36,117-35,387 cal. BP for the child [3]. Morphological paleopathological studies of these Neanderthal remains uncovered distinct evidence of sk

Pathogenicity and virulence of Borrelia burgdorferi.

Infection with Borrelia burgdorferi often triggers pathophysiologic perturbations that are further augmented by the inflammatory responses of the host, resulting in the severe clinical conditions of Lyme disease. While our apprehension of the spatial and temporal integration of the virulence determinants during the enzootic cycle of B. burgdorferi is constantly being improved, there is still much to be discovered. Many of the novel virulence strategies discussed in this review are undetermined. Lyme disease spirochaetes must surmount numerous molecular and mechanical obstacles in order to esta
