Sporothrix schenckii Cell Wall Peptidorhamnomannans.

This mini-review article is dedicated to clarifying certain important biochemical aspects of Sporothrixschenckii cell wall peptidorhamnomannans. Cell wall components involved in the host interaction such as antigens as well as a gp70 adhesin are important molecules present on the surface of the yeast parasitic phase. Other structural glycoconjugates present on the fungus cell surface are also described here. Knowledge of the fine structure of carbohydrate epitopes expressed on the surface in both morphological phases of S. schenckii permitted the development of non-invasive immunochemical meth

Molluscum-like lesions in a patient with sporotrichosis.

Sporotrichosis is a subcutaneous fungal infection caused by Sporothrix schenckii and acquired by direct inoculation. Although the majority of cases consist of the classic lymphocutaneous presentation, the frequency of atypical and severe clinical forms of the disease has increased progressively. Systemic and disseminated cutaneous sporotrichosis constitute rare variants and such cases are generally associated with cellular immunodeficiency or debilitated states. The present paper describes the first published case of molluscum-like lesions in disseminated mucocutaneous sporotrichosis. Direct m

Sporotrichosis in north-east India.

Reports on the occurrence of sporotrichosis in some parts of north-east India are scarce. We report here five cases of lymphocutaneous sporotrichosis from north-east India, including one from Sikkim State and four from distant places in the north-eastern part of West Bengal. All patients were full or part-time farmers or gardeners. Diagnosis of sporotrichosis was confirmed by isolation of Sporothrix schenckii in culture and in vitro conversion of the isolates to yeast form. The cases were treated with 50% solution of potassium iodide.

Blastomyces dermatitidis of the perianal skin: report of a case.

Isolated fungal infections of the perianal skin are rare and their diagnosis is frequently overlooked. We report a case of a 78-year-old male patient who presented with a friable, violaceous, papulopustular lesion, with heaped-up edges along the anal verge. Biopsy revealed unicellular yeast consistent with blastomycosis. The patient was treated with itraconazole with resolution of this lesion. An extensive MEDLINE literature review from 1958 to the present indicates that this is an uncommon manifestation of cutaneous blastomycosis. A summary of the medical literature is presented with a review

Paradoxical inflammatory reaction during treatment of Cryptococcus neoformans var. gattii meningitis in an HIV-seronegative woman.

A human immunodeficiency virus (HIV)-seronegative woman was admitted to the hospital with postpartum onset Cryptococcus neoformans var. gattii meningitis and markedly increased intracranial pressure. A poor initial response to antifungal therapy was followed, 2 months after hospital admission, by severe acute meningeal and cerebral inflammation and clearance of yeast cells from cerebrospinal fluid. This first reported case of paradoxical inflammatory reaction to C. neoformans illustrates important aspects of the host-pathogen interaction and highlights possible effects of immunomodulatory ther

Hairy tongue: a case report.

Hairy tongue or black hairy tongue is a benign condition characterized by hypertrophy of the filiform papillae that give the dorsum of the tongue a furry appearance. The color of the elongated papillae varies from yellowish white to brown or black. The etiology is unclear, but the disorder has been associated with numerous predisposing conditions such as heavy smoking, poor oral hygiene, use of topical or systemic antibiotics, systemic corticosteroid therapy, yeast infections, and radiotherapy for head and neck malignancies. A case is presented in which a hairy tongue appeared in a 67-year-old

Molecular mycology: a genetic toolbox for Histoplasma capsulatum.

Research in medical mycology has traditionally been a mix of exciting biology and frustrating genetics, although the excitement has been steadily increasing as genetic obstacles have been successfully overcome. Now, a variety of fungal pathogens can be studied using molecular techniques derived from classical bacterial and yeast genetics, but with selective and strategic adaptations. Histoplasma capsulatum is the best-studied of the primary pathogens known as 'dimorphic' fungi, and tailored molecular genetic strategies are beginning to reveal a repertoire of genes and gene products intimately

Intraspecies diversity of Cryptococcus laurentii as revealed by sequences of internal transcribed spacer regions and 28S rRNA gene and taxonomic position of C. laurentii clinical isolates.

The intraspecies diversity of an opportunistic yeast pathogen, Cryptococcus laurentii, was revealed by analysis of the sequences of the internal transcribed spacer regions and the 28S rRNA gene. Ten strains of C. laurentii were grouped into two major phylogenetic groups and were further divided into at least seven species. Four of the strains isolated from patients did not represent a single species but showed heterogeneity. These results suggest that C. laurentii is a genetically heterogeneous species, and this must be taken into consideration when identifying C. laurentii clinical isolates.

Cryptococcosis in captive cheetah (Acinonyx jubatus): two cases.

Cryptococcus neoformans is a yeast-like organism associated with pulmonary, meningoencephalitic, or systemic disease. This case report documents 2 cases of cryptococcosis with central nervous system involvement in captive cheetah (Acinonyx jubatus). In both cases the predominant post mortal lesions were pulmonary cryptococcomas and extensive meningoencephalomyelitis. Both cheetahs tested negative for feline immunodeficiency virus and feline leukaemia virus. The organism isolated in Case 2 was classified as Cryptococcus neoformans var. gattii, which is mainly associated with disease in immunoco

Cryptococcus neoformans var neoformans isolated from droppings of captive birds in Nigeria.

The yeast, Cryptococcus neoformans, was found in apparently healthy birds at the Jos Wildlife Park and Zoo in Jos, Nigeria. Cryptococcus neoformans var neoformans was isolated from feces of four captive bird species. Five isolates belonged to serotype A while two were serotype D. Serotype A of C. neoformans was isolated from a white face duck (Dendrocygna viduata), eagle owl (Bubo africanus cinerascene) and peacock (Pavo cristatus). The other two (serotype D), were isolated from a spotted eagle owl.

Human XPMC2H: cDNA cloning, mapping to 9q34, genomic structure, and evaluation as TSC1.

XPMC2 is a Xenopus gene identified on the basis of its ability to correct a mitotic defect in fission yeast. Here we report the identification of cDNA clones for human XPMC2H, its mapping to the tuberous sclerosis gene TSC1 region on 9q34, determination of genomic structure, and identification of several coding region polymorphisms. The predicted protein has strong sequence similarity to the Xenopus gene. Through SSCP and heteroduplex analysis of genomic DNA, we found two intragenic polymorphisms but no evidence for significant mutations in patients with tuberous sclerosis in this gene.

Sporothrix schenckii isolated from a cat in Japan.

A yeast-like fungus isolated from a granulomatous and ulcerative skin lesion in a mongrel cat was mycologically examined. The isolate was identified as Sporothrix schenckii from the morphological findings and its pathogenicity in a mouse, confirming the first case of feline sporotrichosis in Japan. Fortunately, no transmission to humans occurred in this case, however the risk of humans contracting Sporothrix schenckii infection increases with the increase in the number of animals with sporotrichosis. Consequently when handling such animals protective clothing such as gloves should be worn to r

Protective and nonprotective monoclonal antibodies to Cryptococcus neoformans originating from one B cell.

Two immunoglobulin M monoclonal antibodies (mAbs) derived from the same B cell recognize different epitopes on the capsular polysaccharide of the pathogenic yeast, Cryptococcus neoformans. Their respective epitopes are located in spatially distinct regions of the capsule. Passive administration of one mAb prolonged survival whereas the other mAb did not. The results indicate that specificity is an important determinant of antibody efficacy against C. neoformans and that somatic mutations occurring during the antibody response can affect the protective efficacy of antibodies to C. neoformans.

[The action of bacillary inhibitory substances on human-pathogenic, phytopathogenic and saprotrophic fungi].

The study has shown that the growth of many soil fungi is inhibited by the effect of substances which are produced into the environment by some bacilli-saprotrophs. Besides, these substances can bring about a dimorphism of the Trichoderma, Blastomyces, Emmonsia, Alternaria and Verticillium fungi from the mycelial form into the yeast form, which results in the death of the fungi. The existence of bacilli capable of producing the inhibitory substances, in the soil in the territory of the USSR may explain, to a certain extent, the absence of the coccidioidomycosis and North American blastomycosis

[Secondary candidal infection in patients with chronic dermatoses].

Clinical and laboratory examinations of 250 patients with chronic dermatoses (eczema, neurodermatitis, psoriasis, pemphigus, etc.) have revealed secondary candidal involvement of the skin and mucosa in 121 (46.6%). C. albicans, often associated with bacillary (33.3%) or bacterial (7.3%) microflora, predominated among the yeast-like fungi isolated from the patients. These data point to the necessity of prescribing, besides the antimycotic agents, a complex of pathogenetic therapy including immunomodulators, in order to cut down the terms of treatment and prevent secondary candidal infection in

Controlling fermentation of lignocellulose hydrolysates in a continuous hollow-fiber reactor using biosensors.

Fermentation of lignocellulose hydrolysates as spent sulfite liquor or as hydrolysate from sulfur-dioxide-treated wood to ethanol has been controlled by using biosensors for glucose and ethanol. Yield and productivity were studied with respect to concentration level of the metabolites in a continuous hollow-fiber reactor. High constant yield was achieved by controlling the glucose to low concentration levels. Reduced productivity were obtained when fermenting at high ethanol concentrations as an effect of inhibition of the yeast cells. The observations emphasize the advantage of controlling th

Host-parasite relationships in paracoccidioidomycosis.

A viewpoint of host-parasite relationships in paracoccidioidomycosis is presented. The characteristics of the fungus which are important to the host-parasite interaction are discussed. Aspects of inhibition of mycelium-to-yeast transformation by estrogens acting at receptors on the fungal wall and in the cytoplasm, and the role of polysaccharide components of the cell wall in virulence are reviewed. The natural mechanisms of host defense are also examined, including phagocytosis, complement system, natural-killer cells and genetic control of resistance and susceptibility. Finally, a discussion

Systemic salmonellosis in patients with disseminated histoplasmosis. Case for 'macrophage blockade' caused by Histoplasma capsulatum.

Five patients are described with disseminated histoplasmosis and systemic salmonellosis. Four of these patients were also immunocompromised because of the acquired immunodeficiency syndrome in two patients and renal transplantation in another two patients. Histologic studies in two patients showed histiocytes that were heavily laden with Histoplasma capsulatum yeast-phase organisms. We postulate that diffuse parasitization of the reticuloendothelial system (RES) by Histoplasma organisms may cause "RES blockade," which then predisposes to systemic salmonellosis, as reported in certain other inf

A modified method for rapid mycological diagnosis of Tinea unguium in Taiwan, Republic of China.

This is a study to modify and simplify the classical laboratory procedures of mycological diagnosis of Tinea unguium. By means of a tube KOH digestion technique for microscopic examination of nail clippings and a paper disc transfer technique using a more uniform wool-fiber-yeast extract culture fluid as inoculum for all tests necessary for fungal identification, dermatophytes can be identified to a species level. To authenticate the advantage of the etiologic diagnosis of Tinea unguium by mycological studies, the promising result of our previous clinical therapeutic trial with Griseofulvin wa
