FLVCR1-related diseases: from clinical heterogeneity to mechanistic insights.

Feline Leukemia Virus subgroup-C Receptor 1 (FLVCR1) is an ubiquitously expressed choline and ethanolamine importer that is involved in the control of multiple aspects of cell biology including the regulation of phospholipids metabolism, heme homeostasis, mitochondria-ER contact sites and cellular bioenergetics. Mutations in the FLVCR1 gene cause a spectrum of autosomal-recessive disorders mainly affecting the nervous system. Research conducted in the last decade highlighted the complexity of the clinical features associated with FLVCR1 mutations, ranging from dysfunction of specific sensory m

Illuminating epilepsy: A deep NIR-I fluorogenic sensor reports peroxynitrite dynamics in vivo.

This work presents BDP-NIR-S-Fa, a deep NIR-I fluorogenic probe designed to report peroxynitrite (ONOO-) dynamics in epilepsy. The probe features a specific emission at 805 nm and responds rapidly and sensitively to ONOO-. It successfully visualized elevated endogenous ONOO- flux in both kainic acid-induced epileptic cells and a corresponding mouse model, demonstrating its potential as a diagnostic tool. Beyond detection, BDP-NIR-S-Fa was further applied to evaluate the therapeutic efficacy of antiepileptic drugs by monitoring the reduction of ONOO- levels in treated disease models. The probe

Nanopore-Based Profiling of PEGylation in Nucleic Acid Therapeutics.

Nucleic acid therapeutics (NATs), including aptamers, offer effective strategies for programmable and targeted disease treatment. To improve their stability and circulation time, oligonucleotides are often conjugated to hydrophilic polymers, such as polyethylene glycol (PEG). However, current bulk techniques fail to resolve PEG heterogeneity, especially in complex biological environments. Here, we use nanopore sensing to quantify the PEG conjugation efficiency of the FDA-approved RNA aptamer pegaptanib. We assembled DNA nanostructures that bind pegaptanib, and then we used solid-state nanopore

Recent advances in electrochemical synthesis of sulfur-containing compounds using elemental sulfur, carbon disulfide, and sulfur dioxide.

Sulfur-containing compounds occupy a central position in pharmaceutical chemistry, agrochemical development, and functional materials. Traditional synthetic methods often rely on hazardous reagents, transition-metal catalysts, or stoichiometric oxidants, which are incompatible with the principles of green chemistry. In recent years, electrochemical organic synthesis has emerged as a novel platform for the construction of sulfur-containing compounds, owing to its mild, green, and sustainable characteristics. Elemental sulfur (S8), carbon disulfide (CS2), and sulfur dioxide (SO2) are inexpensive

The EXTra-uterine Environment for Neonatal Development (EXTEND) - future treatment for prematurity and potentially other congenital disease.

The EXtra-uterine Environment for Neonatal Development (EXTEND) was developed to support extremely premature infants (EPIs) with the goal to mimic in-utero conditions as closely as possible and therefore reduce morbidity and mortality associated with organ immaturity and iatrogenic injury caused by current neonatal management. In an animal model using fetal lambs at a gestational age equivalent to EPIs of 22-24 weeks of gestation, EXTEND was able to support fetuses for up to four weeks while enabling normal organ growth, particularly normal lung development. Recent advances in EXTEND have allo

[Pathophysiological process of vocal fold injury repair and new advances in regenerative therapy].

The repair of vocal fold injury is a complex process involving inflammatory response, cell proliferation, and extracellular matrix(ECM) remodeling. Aberrant repair can lead to scar formation and voice dysfunction. Recent research has revealed that this process is tightly regulated by a sophisticated network of cytokines, including TGF-β and HGF, and signaling pathways such as JAK/STAT and Wnt/β-catenin. Building on this deepened understanding of molecular mechanisms, vocal fold repair strategies are entering a new era of regenerative medicine. This article systematically elaborates on the path

A milestone in liver xenotransplantation: The first 10-gene-edited pig-to-living-human auxiliary transplantation and the road ahead.

The global shortage of human donor livers poses a formidable barrier to treating end-stage liver diseases and unresectable hepatic malignancies, leaving millions of patients without life-saving options each year. While genetically engineered pig-to-human xenotransplantation has achieved significant breakthroughs in cardiac and renal fields, liver xenotransplantation has long lagged due to the liver's intricate metabolic, synthetic, and immunological functions. The recent report by Zhang et al. in the Journal of Hepatology-documenting the world's first successful auxiliary liver xenotransplanta

Challenges and perspectives in xenoliver research: lessons from decedent and primate models.

Liver xenotransplantation is a potential strategy to address the shortage of donor livers, either as a temporary bridge to allotransplantation, bridge to native liver recovery, or destination therapy. Advances in genetic engineering and immunosuppression have enabled liver xenografts to evade hyperacute rejection, yet durable graft survival remains elusive. Profound thrombocytopenia, consumptive coagulopathy, and xenotransplantation-associated thrombotic microangiopathy limit long-term graft survival. Evidence from nonhuman primates, decedents, and recent clinical models suggests that hematolo

Immune-stromal interactions at the crossroads of tissue injury, repair, and tumor progression.

Macrophage-fibroblast crosstalk plays a critical yet incompletely understood role in cancer, fibrosis, and tissue repair. A central challenge is capturing the diversity of these cells, whose phenotypes are shaped by microenvironmental cues and developmental origins. Macrophage heterogeneity reflects the tissue-resident and bone marrow-derived lineages, while fibroblasts adopt distinct subsets defined by tissue context. Myofibroblasts can emerge from fibroblasts, monocytes, and other stromal cells. The lack of markers to distinguish these subpopulations remains a major barrier to defining their

Robust and interpretable unit level causal inference in neural networks for pediatric myopia.

Understanding causal mechanisms in deep learning is essential for clinical adoption, where interpretability and reliability are critical. Most existing AI systems act as black boxes, limiting transparency in medicine. We propose a causal inference framework integrated into neural networks to assess the influence of individual features on predictions. Using a prospective pediatric ophthalmology cohort of over 3000 children with longitudinal follow-up, our method estimates direct and indirect causal effects through intervention. Applied to myopia progression in children, our approach not only ac

Patient-derived organoids in hematologic malignancies: Fidelity and translation beyond animals.

Hematologic patient-derived organoids (PDOs) reconstruct human bone marrow and lymph node microenvironments in 3D, preserving malignant cell states, clonal diversity, and clinically relevant drug responses. Leukemia PDOs artificially mimic the complex bone marrow niche and cytokine dependencies. Lymphoma PDOs recapture immune-stromal interactions and enable testing of targeted and immunotherapies, including bispecific antibodies and chimeric antigen receptor (CAR) T-cells. Multiple myeloma PDOs model the bone marrow microenvironment and maintain plasma tumor cell survival cues that drive drug

Intraoperative diagnosis of Amyand's hernia: inguinal herniorrhaphy, appendicectomy and orchidectomy.

Amyand's hernia, a rare condition where the appendix is contained within an inguinal hernia sac, occurs in ~1% of inguinal hernias, with <0.1% involving acute appendicitis. Though often diagnosed intraoperatively, imaging advancements like CT and ultrasound have improved preoperative detection. We present a case of a male in his 70s with a right-sided direct inguinal hernia, diagnosed via ultrasound. Intraoperatively, a non-inflamed appendix was found in the hernial sac. Despite the absence of appendicitis, an appendectomy was performed due to age, comorbidities and potential future risk. Hern

A practical toolbox for modelling fibrosis in vitro.

Fibrosis is a widespread disease implicated in millions of deaths worldwide and impacting diverse organs, including the heart, lung, kidney, liver and synovium. Characterized by excessive extracellular matrix deposition, fibrosis leads to tissue scarring and dysfunction, ultimately resulting in organ failure. The development of faithful experimental models of fibrosis is challenged by its complexity, limiting the discovery of effective therapeutics. Human in vitro tissue models are emerging as powerful systems that capture the pathophysiology of fibrosis to uncover disease mechanisms, identify

Cholinergic reflex control of inflammation: mechanistic and translational advances in transcutaneous auricular vagus nerve stimulation across rheumatic, metabolic, and postoperative disorders.

Transcutaneous auricular vagus nerve stimulation (taVNS) has recently emerged as a focal noninvasive neuromodulatory approach in anti-inflammatory therapeutics. The vagus nerve functions as a critical neuroimmune interface, tonically suppressing proinflammatory cytokine release via the cholinergic anti-inflammatory pathway (CAP). This mechanism provides substantial therapeutic potential across a spectrum of inflammatory disorders, including postoperative systemic inflammation. Clinical trials have demonstrated the anti-inflammatory efficacy of taVNS, supporting its expanded use in rheumatoid a
