Sex-specific differences in transcriptomic profiles and cellular characteristics of oligodendrocyte precursor cells.

The susceptibility to neurological and psychiatric disorders reveals sexual dimorphism in the structure and function of human brains. Recent evidence has also demonstrated the sex-related differences in cellular components of the brain, including neurons, microglia, astrocytes, and endothelial cells. Oligodendrocyte precursor cells (OPCs) regulate the neuronal system in various ways and play crucial roles in brain homeostasis beyond their well-known role as a reservoir for mature oligodendrocytes. Although recent studies have shown regional diversities and heterogeneities of OPCs, sex-related …

Noninflammatory Changes of Microglia Are Sufficient to Cause Epilepsy.

Microglia are well known to play a critical role in maintaining brain homeostasis. However, their role in epileptogenesis has yet to be dete … ion into hippocampal pyramidal layers, moderate neuronal degeneration, and massive proliferation of astrocytes. Moreover, the mice thus affected develop severe early-onset spontaneous recurrent seizures (SRSs). Therefore, we have revealed an epileptogenic mechanism that is independent of the microglial inflammatory response. Our data suggest that microglia c …

Discovery and implications of transcellular mitophagy.

The mitochondrial quality control system regulating mitochondria biogenesis, dynamics, and degradation has been extensively studied because of its roles in normal cell homeostasis and dysfunction due to aging or disease. Mitochondria degradation is generally thought to occur by autophagy and has therefore been viewed as a cell-autonomous process. In a recent study, we demonstrated that a large fraction of retinal ganglion cell mitochondria undergo lysosomal degradation within the astrocytes of the optic nerve head. It will be important to determine whether other neurons with long axons also us…

Intracerebral transplantation of mesenchymal stem cells derived from human umbilical cord blood alleviates hypoxic ischemic brain injury in rat neonates.

AIMS: Fetal hypoxic-ischemic brain injury (HIBI) is a severe condition for which no effective therapy exists. In this study mesenchymal stem … al study showed alleviation of brain tissue injury after MSCs treatment. Differentiation of MSCs to astrocytes, but not neurons, was observed. CONCLUSIONS: The results indicate a beneficial effect of intracerebral transplantation of MSCs on the functional recovery of rat neonates with HIBI.

Erythropoietin enhances long-term neuroprotection and neurogenesis in neonatal stroke.

Neonatal stroke leads to mortality and severe morbidity, but there is no effective treatment currently available. Erythropoietin (EPO) has b … rthermore, EPO increased the percentage of newly generated neurons while decreasing newly generated astrocytes following brain injury, without demonstrating long-term differences in the subventricular zone. These results suggest that EPO may neuroprotect and direct cell fate toward neurogenesis and away from gliogenesis in neonatal stroke.

CASE REPORTS: Four cases of desmoplastic infantile ganglioglioma (DIG) seen in India are described. These patients presented with large, sup … tumour, probably arising from neural progenitor cells in subcortical zone along with mature subpial astrocytes. CONCLUSIONS: In view of its favourable prognosis, this tumour has to be diagnosed accurately by immunohistochemical techniques using glial and neuronal markers. Expression of alphaB-crystallin in a mouse model of inherited retinal degeneration.

AlphaB-crystallin, which is abundantly expressed in the lens but also in a diversity of other tissues, functions as a stress-inducible molec … s in the degenerative state. Immunocytochemistry confirmed increased expression at Müller cells and astrocytes, together with transiently increased localization to the degenerating photoreceptors. These findings suggest that increased alphaB-crystallin expression is associated with glial cell reaction to neuronal damage in the retina, and may comprise part o …

In vitro-generated neural precursors participate in mammalian brain development.

During embryogenesis, pluripotent stem cells segregate into daughter lineages of progressively restricted developmental potential. In vitro, this process has been mimicked by the controlled differentiation of embryonic stem cells into neural precursors. To explore the developmental potential of these cell-culture-derived precursors in vivo, we have implanted them into the ventricles of embryonic rats. The transplanted cells formed intraventricular neuroepithelial structures and migrated in large numbers into the brain tissue. Embryonic-stem-cell-derived neurons, astrocytes, and oligodendrocyte…

Evolutionary perspectives on amyloid and inflammatory features of Alzheimer disease.

We propose that the amyloid deposits in senile plaques of Alzheimer's Disease (AD) result from ancient mechanisms in wound-healing and infla … ammatory proteins of ancient origin that appear to be made by local brain cells, including neurons, astrocytes, and microglia. We describe a highly conserved 16-mer found in pentrakins from mammals and from the horseshoe crab. The senile plaque thus provides a novel opportunity to study primitive features of complement-mediated inflammatory responses in the …

Phocine distemper in a harp seal (Phoca groenlandica) from the Gulf of St. Lawrence, Canada.

The first case of phocine distemper in a seal from Canadian waters and the first case of clinical phocine distemper in a harp seal, Phoca groenlandica, is reported. A two-month-old female harp seal stranded on Prince Edward Island in May 1991. Significant clinical findings were lethargy and severe conjunctivitis. Pulmonary congestion was the main necropsy finding, and histological lesions included diffuse demyelinating nonsuppurative encephalitis and mild multifocal interstitial pneumonia. Acidophilic intracytoplasmic and intranuclear inclusions were present in cerebral neurons and astrocytes.…

Basal ganglia mass lesions in juvenile rheumatoid arthritis.

A 2-year-old boy suffered aphasia, hypotonia, dystonia, and loss of activity and spontaneous speech during an active stage of juvenile rheumatoid arthritis with pericarditis, fever, anemia, and a high antinuclear antibody titer. These neurologic signs slowly improved with corticosteroid treatment but fluctuated over 1 year. The neuroimaging studies revealed irregular mass lesions in the basal ganglia bilaterally mainly involving the globus pallidus. They gradually decreased in size and almost disappeared after 1 year. A stereotactic brain biopsy revealed a slight proliferation of astrocytes. C…

Morphogenesis of amyloid plaques in mice with Creutzfeldt-Jakob disease.

Amyloid plaques were experimentally produced in the brains of mice inoculated with human Creutzfeldt-Jakob disease (CJD) brain homogenate. Light-microscopically, these plaques were mostly round and oval and were surrounded by macrophages and astrocytes. Ultramicroscopically, amyloid plaques were present in the cytoplasm of macrophages or were surrounded by these cells. The macrophages had numerous Golgi apparatuses, endoplasmic reticula (ER), ribosomes, polysomes, and lysosomes with inoculated materials or degenerating products. The bundles of amyloid fibrils were intermingled with the cytopla…

S-100 protein in amniotic fluid of anencephalic fetuses.

S-100 protein, which is found essentially in the astrocytes of the nervous system, was assayed in amniotic fluids by Particle Counting ImmunoAssay. It was present in 19 cases of anencephaly out of 26, in 1 case of open spina bifida out of 5 and in each of the 4 cases of fetal death, whereas it was not detected in the 48 control amniotic fluids collected between the 16th and the 35th week of gestation. Thirty-one amniotic fluids from fetuses with other congenital malformations were devoid of detectable S-100. The presence of S-100 in amniotic fluid of anencephalic fetuses can presumably be cons…

Central pontine myelinolysis following saline treatment of a diabetic rat for dehydration.

Central pontine myelinolysis (CPM) was observed at necropsy in an emaciated 333-day-old BB Wistar insulin-dependent diabetic rat that had been treated for dehydration by subcutaneous injection of normal saline. There was a large area of nearly complete demyelination involving most of the basal portion of the pons, part of the tegmentum, and the base of the middle cerebellar peduncles. There was a relative sparing of neurons and axons, but some unidentifiable large cells with degenerative changes in the cytoplasm were present. No oligodendrocytes or reactive astrocytes were seen and there was n…

[A 65-year-old man with rigid-bradykinetic parkinsonism, vertical gaze palsy, difficulty of eye-lid opening, and marked pseudo-bulbar palsy].

We report a 65-year-old man with rigid-bradykinetic parkinsonism, vertical gaze palsy, difficulty in eye-lid opening, and marked pseudo-bulbar palsy. He felt difficulty of it, hand movement at 59 years old. When he was 60 years old, monotonous speech and slowness of movement appeared. He visited a neurologist who noted vertical gaze palsy, neck rigidity, and bradykinesia. He was diagnosed as progressive supranuclear palsy (PSP) and given 300 mg L-Dopa/Benserazide by the neurologist. This medication improved his rigidity and bradykinesia. At 62 years of the age, his eye-lids closed involuntary
