Bone mineral apposition rates at early implantation times around differently prepared titanium surfaces: a study in beagle dogs.

PURPOSE: This study evaluated the bone mineral apposition rate (MAR) at the bone-implant interface region of alumina-blasted/acid-etched (AB/AE), plasma-sprayed hydroxyapatite (PSHA), and nanometric-scale bioceramic-coated surfaces at early implantation times in a dog tibia model. MATERIALS AND METHODS: Implants (n = 12 per group) with three different surfaces-AB/AE, PSHA, and a bioceramic coating in the 300- to 500-nm thickness range-were placed bilaterally along the proximal tibiae of six male beagles. Implants remained for 3 and 5 weeks in vivo. Ten and 2 days prior to euthanization, calce

Sustained local drug delivery from a novel polymeric ring to inhibit intimal hyperplasia.

The long-term clinical success of autologous vein and synthetic vascular grafts are limited because of the development of anastomotic intimal hyperplasia (IH). We have previously published data suggesting that cyclosporine (CyA) may reduce the development of IH in a canine model (Hirko et al., J Vasc Surg 1993;17:877-887). However, systemic administration of CyA could create serious adverse effects. Therefore, it is our long-term goal to test the hypothesis that the controlled local release of CyA from a polymeric vascular wrap would prevent the development of IH. To test this hypothesis, we d

Guided bone regeneration by poly(lactic-co-glycolic acid) grafted hyaluronic acid bi-layer films for periodontal barrier applications.

A novel protocol for the synthesis of biocompatible and degradation controlled poly(lactic-co-glycolic acid) grafted hyaluronic acid (HA-PLGA) was successfully developed for periodontal barrier applications. HA was chemically modified with adipic acid dihydrazide (ADH) in the mixed solvent of water and ethanol, which resulted in a high degree of HA modification up to 85 mol.%. The stability of HA-ADH to enzymatic degradation by hyaluronidase increased with ADH content in HA-ADH. When the ADH content in HA-ADH was higher than 80 mol.%, HA-ADH became soluble in dimethyl sulfoxide and could be gr

In vitro leakage susceptibility of tracheoesophageal shunt prostheses in the absence and presence of a biofilm.

Although leakage through a tracheoesophageal shunt prosthesis is the main cause of prosthesis failure in a laryngectomy patient, this has never been the subject of in vitro evaluation. The aim of this study was to compare three commercially available voice prostheses by comparison of their in vitro leakage patterns, in absence or presence of a biofilm. To compare in vitro leakage patterns, a model comprised of an artificial throat equipped with a single prosthesis coupled to a water reservoir was developed. By varying the height of the water reservoir, different pressures on the voice prosthes

Morphological characteristics of total joint arthroplasty-derived ultra-high molecular weight polyethylene (UHMWPE) wear debris that provoke inflammation in a murine model of inflammation.

It is recognized that the chronic inflammation in peri-prosthetic tissue that contributes to implant failure frequently is provoked by the presence of wear debris. Some wear debris is inevitable because of the nature of the prosthesis, but not all patients develop severe inflammatory responses. The precise factors that mediate the severity of tissue inflammation to wear debris has yet to be fully defined. Because wear debris retrieved from peri-prosthetic tissue consists of a heterogeneous mixture of materials with various sizes and shapes, this study evaluated the influence of two major physi

Radiotheranostics With Nanoparticles: Precision Targeting in Receptor-Overexpressing Tumors.

Cancer remains one of the most challenging medical conditions. Nanotechnology is revolutionizing the approach to cancer management as an efficient tool for early diagnosis, therapy and monitoring. Nanoparticles (NPs) have shown promising results in advancing precision medicine by integrating diagnostic imaging and targeted therapy within a single platform, a concept of theranostics. Nanocarriers are synthesized via the use of biocompatible materials that are functionalized with specific ligands that bind to receptors that are overexpressed on the surface of cancer cells. For theranostic purpos…

Biomedical applications of lignin nanoparticles and their nanoconjugates: Advancements in synthesis, therapy, imaging, and drug delivery.

Multidrug resistance (MDR) in cancer and infectious diseases is a major global challenge. It reduces the effectiveness of antibiotics and chemotherapeutics, leading to higher treatment costs and mortality. Conventional agents suffer from poor selectivity, systemic toxicity, and resistance linked to efflux pump overexpression. This review addresses how lignin nanoparticles (LNPs) and their nanoconjugates can provide relatively sustainable, biocompatible alternatives to overcome MDR through tunable surface chemistry, stimuli responsiveness, and multifunctionality. Developments in LNP synthesis u

Mesoporous engineering of metal-phenolic artificial enzymes for enhanced oxidative stress mitigation.

Catalytic antioxidant materials are crucial in medical research, yet current research efforts focus mainly on coordination environments while overlooking physical structures, such as porosity and surface area, a limitation that hinders free radical interaction and reduces antioxidant efficacy. Herein, a conceptual framework is proposed for the development of mesoporous metal-polyphenolic artificial enzymes (MPAEs) (mPDA-Cu), effectively addressing these limitations by integrating catalytically activate centers and maximizing the accessible surface area. The pore confinement achieved by interme

Core-shell commensal biocapsules for in situ gut microbiome engineering.

Despite the profound influence that gut microbiome composition has on human health, the development of live microbe treatments is constrained by a lack of knowledge on single-species functions within the gastrointestinal (GI) tract. A key barrier is the absence of broadly accessible tools capable of overcoming colonization resistance and enabling spatiotemporal control of microbiome ecology. To address this gap, we have developed a core-shell capsular material, termed a biocapsule, designed to promote the engraftment of a defined bacterial payload to a modified GI niche, thereby enabling preci

Modular Control of PEG-Poly(2-Oxazoline) Co-Assembly Enables Tunable Aggregate Properties for Intraocular Pressure Management.

Poly(2-oxazoline) (POx) polymers are hydrophilic and biocompatible materials with tunable physicochemical properties for biomedical use. Postoperative hypotony, which currently lacks approved pharmacological or device-based treatment, requires minimally invasive and controllable methods for temporary intraocular pressure restoration. Herein, we present a modular strategy to control POx aggregation via thermally triggered co-assembly with thermoswitchable poly(2-n-propyl-2-oxazoline) (PnPrOx) and hydrophilic PEG-modified PnPrOx (PnPrOx-PEG). These mixtures remain dissolved at low temperatures b…

Toward Bright Polymetallic Biopolymer-Protected Luminescent Nanoclusters.

Polymetallic nanoclusters (NCs) offer a powerful route to brighter and more tunable luminescent materials, while biopolymer ligands provide additional advantages for sensing and bioimaging. However, the mechanisms responsible for luminescence enhancement remain poorly understood, particularly for DNA- and protein-protected NCs. Here, we critically review the relationship between metal composition, cluster structure, ligand environment, and excited-state dynamics in luminescent polymetallic NCs. Rather than treating alloying as a single "synergistic" effect, we distinguish three recurrent contr

Nanoconfined Photopolymerization in Self-Assembled Glycerol Monooleate, Hydroxyethyl Acrylate, and Water Suprastructures.

The nanoconfinement of lipid self-assembled structures offers a dynamic environment for chemical reactions. It enables the integration of regents of various polarities, their alignment at the lipid/water interface for regiospecific reactions, and tailored reaction dimensions from restricted diffusion in the nanoconfinement. However, the effects of monomer integration and polymerization on the self-assembly structure and reaction products remain to be fully understood. Here, we report the photoinitiated polymerization of hydroxyethyl acrylate (HEA) within the inverse bicontinuous cubic structur

Evaluation of Sericin/Polyvinyl Alcohol Mixtures for Developing Porous and Stable Structures.

Fibrous by-products, including defective or double cocoons, are obtained during silk processing. These cocoons primarily contain fibroin and sericin (SS) proteins along with minor amounts of wax and mineral salts. In conventional textile processes, SS is removed in the production of smooth, lustrous silk threads, and is typically discarded. However, SS has garnered attention for its antioxidant, antibacterial, biocompatible, and anticancer properties as well as its excellent moisture absorption, making it a promising polymer for biomedical applications. Owing to its functional groups (carboxyl

Transdermal Delivery of Valproate-Choline Ionic Liquid Induces Hair Regrowth.

Alopecia treatment research has been increasingly focused on innovative materials systems and drug delivery methods. Valproic acid (VPA) has been shown to promote hair regrowth by upregulating the Wnt/β-catenin signaling pathway, a key regulator of hair papilla cell proliferation. However, topical administration of VPA typically requires organic solvents or complex transdermal devices to enhance skin penetration. Biocompatible ionic liquids have gained attention as customizable solvents for transdermal drug delivery (TDD). In this study, we introduce a novel choline-based valproic acid ionic l

3D printed Aloe barbadensis loaded alginate-gelatin hydrogel for wound healing and scar reduction: In vitro and in vivo study.

Wounds are one of the most critical clinical issues in plastic surgery repair and restoration. Conventional wound dressing materials cannot absorb enough wound exudates and shield the site from microbial infection. Also, despite their healing prowess, bioactive molecules from medicinal plants are less bioavailable at the wound sites. This study developed a 3D-printed hydrogel of sodium alginate and gelatin loaded with freeze-dried Aloe barbadensis extract for enhanced wound healing. The hydrogel was hydrophilic and showed an average pore size of 163.66 ± 14.45 μm, moderate swellability, and id
