This report describes the development of an experimental head injury model capable of producing diffuse brain injury in the rodent. A total of 161 anesthetized adult rats were injured utilizing a simple weight-drop device consisting of a segmented brass weight free-falling through a Plexiglas guide tube. Skull fracture was prevented by cementing a small stainless-steel disc on the calvaria. Two groups of rats were tested: Group 1, consisting of 54 rats, to establish fracture threshold; and Group 2, consisting of 107 animals, to determine the primary cause of death at severe injury levels. Data

Preclinical research in neuroscience has traditionally relied on animal models to investigate disease mechanisms and develop new therapeutic strategies. While these models are valuable to gain mechanistic insights, their translational power remains limited due to interspecific differences and, hence, frequent failures in clinical translation. The uniqueness of the human brain calls for alternative approaches in neuroscientific research that more faithfully capture human physiology and pathology. In recent years, a variety of human-based model systems have emerged, ranging from dissociated neur

Malignant glioma remains one of the most aggressive and therapy-resistant brain tumors, with limited treatment options and a poor prognosis. Overcoming the blood-brain barrier (BBB) is a major challenge for effective drug delivery. Here, we developed a triple-targeted nanodelivery system, denoted BCNU-FeSeH@EVLF, which integrates magnetic guidance, chemotactic homing, and receptor-mediated transport for enhanced brain penetration and glioma treatment. The system consists of carmustine (BCNU)-loaded, hyaluronic acid-modified superparamagnetic Fe3O4 nanoparticles with a mesoporous selenium shell

Wiskott-aldrich syndrome (WAS) is a rare X-linked primary immunodeficiency characterized by thrombocytopenia, eczema, and recurrent infections. Live vaccines, such as Bacillus Calmette-Guérin (BCG), routinely administered in tuberculosis-endemic regions, may cause severe complications in undiagnosed patients. We report a 2-year-old boy with genetically confirmed WAS who developed a rare BCG-related brain abscess. The patient initially presented with neonatal thrombocytopenia and eczema, followed by recurrent respiratory and gastrointestinal infections. Whole exome sequencing identified a patho

Microelectrode arrays (MEAs) are widely used platforms for monitoring neuronal activity in biological systems. The advent of brain organoid technology-three-dimensional (3D) neural tissue models derived from human stem cells-has opened unprecedented opportunities to study network connectivity and developmental processes of brain-like tissues in vitro. However, most commercial MEAs are restricted to planar, two-dimensional configurations, limiting recordings to a single surface and failing to capture the multilayered architecture of organoids. To overcome this limitation, we develop a multilaye

CASE PRESENTATION: Mitochondrial complex III deficiency nuclear type 2 (MC3DN2) is a rare inherited neurometabolic disease. A 34-year-old male had neuropsychiatric episodes, progressive cerebellar degeneration, myopathy, polyneuropathy, and brain stem and basal ganglion lesions since childhood. Muscle biopsy revealed mitochondrial abnormalities. Two novel TTC19 pathogenic variants were detected.
LITERATURE REVIEW: To analyze phenotypic characteristics of MC3DN2 related to clinical onset age, neurological presentation and brain MRI regarding infantile and childhood-onset (ICO) and adolescent a…

Neural development is a highly intricate process that relies on the precise regulation of gene expression. While a significant focus has been placed on understanding the transcriptional control of brain development, the regulation of mRNA translation plays a fundamental role in controlling gene expression. mRNA translation in subcellular compartments distant from the cell body, such as neuronal growth cones and astrocytic processes, allows for a rapid response to the local environment. Thus, the regulation of mRNA translation influences neurodevelopmental mechanisms such as cell fate decisions

Hypoxic-ischemic brain injury (HIBD) is a major cause of neonatal mortality and long-term neurological deficits, with limited treatment options. Extracellular vesicles (EVs) from human umbilical cord mesenchymal stem cells (hUC-MSC-EVs) have shown promise in neuroprotection, but the mechanisms remain unclear. This study explores how hUC-MSC-EVs protect neonatal rats from HIBD. hUC-MSC-EVs were isolated, characterized, and administered to neonatal rats subjected to HIBD. Behavioral reflexes and brain infarction were assessed, along with cellular and molecular analyses of hippocampal tissue. An

Mammalian sleep consists of non-rapid eye movement sleep (NREMS) and rapid eye movement sleep (REMS), accounting for approximately 75% and 2 … rain anatomy and neural circuits, which constitute highly distributed networks spanning the cortex, brain stem, hypothalamus, and other regions. We also summarize the role of REMS in negative emotion processing. Finally, we propose key open questions that need to be addressed in future studies.

The WWOX gene encodes a 414-amino-acid protein composed of two N-terminal WW domains and a C-terminal short-chain dehydrogenase/reductase (SDR) domain. WWOX protein is highly conserved among species and mainly expressed in the cerebellum, cerebral cortex, brain stem, thyroid, hypophysis, and reproductive organs. It plays a crucial role in the biology of the central nervous system, and it is involved in neuronal development, migration, and proliferation. Biallelic pathogenic variants in WWOX have been associated with an early infantile epileptic encephalopathy known as WOREE syndrome. Both miss…

Tau inclusions are a shared feature of many neurodegenerative diseases, among them frontotemporal dementia caused by tau mutations. Treatment approaches for these conditions include targeting posttranslational modifications of tau proteins, maintaining a steady-state amount of tau, and preventing its tendency to aggregate. We discovered a new regulatory pathway for tau degradation that operates through the farnesylated protein, Rhes, a GTPase in the Ras family. Here, we show that treatment with the farnesyltransferase inhibitor lonafarnib reduced Rhes and decreased brain atrophy, tau inclusion

Cerebral X-linked adrenoleukodystrophy is a devastating neurodegenerative disorder caused by mutations in the ABCD1 gene, which lead to a rapidly progressive cerebral inflammatory demyelination in up to 60% of affected males. Selective brain endothelial dysfunction and increased permeability of the blood-brain barrier suggest that white matter microvascular dysfunction contributes to the conversion to cerebral disease. Applying a vascular model to conventional dynamic susceptibility contrast magnetic resonance perfusion imaging, we demonstrate that lack of ABCD1 function causes increased capil
