Reconstitution of human thymic implants is limited by human immunodeficiency virus breakthrough during antiretroviral therapy.

Human immunodeficiency virus type 1 (HIV-1)-infected SCID-hu thymic implants depleted of CD4(+) cells can support renewed thymopoiesis derived from both endogenous and exogenous T-cell progenitors after combination antiretroviral therapy. However, successful production of new thymocytes occurs transiently. Possible explanations for the temporary nature of this thymic reconstitution include cessation of the thymic stromal support function, exhaustion of T-cell progenitors, and viral resurgence. Distinguishing between these processes is important for the development of therapeutic strategies aim

Chronic HIV infection affects the expression of the 2 transcription factors required for CD8 T-cell differentiation into cytolytic effectors.

CD8 T cells lose the capacity to control HIV infection, but the extent of the impairment of CD8 T-cell functions and the mechanisms that underlie it remain controversial. Here we report an extensive ex vivo analysis of HIV-specific CD8 T cells, covering the expression of 16 different molecules involved in CD8 function or differentiation. This approach gave remarkably homogeneous readouts in different donors and showed that CD8 dysfunction in chronic HIV infection was much more severe than described previously: some Ifng transcription was observed, but most cells lost the expression of all cyto

Quantitation of immunosuppression by tacrolimus using flow cytometric analysis of interleukin-2 and interferon-gamma inhibition in CD8(-) and CD8(+) peripheral blood T cells.

The authors have determined the frequency of intracellular interleukin-2 (IL-2) and interferon-gamma (IFN-gamma) synthesis by T-cell subsets in whole blood (WB) and isolated lymphocytes in 16 transplant recipients treated with tacrolimus and 10 control patients who were not transplant recipients. The authors also determined the impact of varying amounts of red blood cells (RBC) on immunosuppression by tacrolimus. Samples were analyzed by two-color flow cytometry, and the results were expressed as a ratio of whole blood to isolated lymphocytes. In healthy subjects who were not transplant recipi

Bronchoalveolar lavage cell findings in three types of eosinophilic pneumonia: acute, chronic and drug-induced eosinophilic pneumonia.

There are clinically different types of eosinophilic pneumonia (EP) but no study to date has compared pulmonary inflammatory cells between different types of EP, such as acute eosinophilic pneumonia (AEP), chronic eosinophilic pneumonia (CEP) and drug-induced eosinophilic pneumonia (drug-EP). The present study compared bronchoalveolar lavage fluid (BALF) cell findings to elucidate whether the profiles of the pulmonary inflammatory cells were different among the three types of EP. Clinical records of 28 patients with EP, consisting of eight AEP patients, 10 CEP patients and 10 drug-EP patients,

The Th17 pathway in cystic fibrosis lung disease.

RATIONALE: Cystic fibrosis (CF) is characterized by bronchoalveolar neutrophilia and submucosal lymphocytosis. We hypothesized that Th17 lymphocytes are part of this submucosal infiltrate. OBJECTIVES: Quantification and phenotyping of the lymphocytic infiltrate in the bronchial submucosa of patients with CF (n = 53, of which 20 were newly diagnosed), non-CF bronchiectasis (n = 17), and healthy control subjects (n = 13). METHODS: We measured IL-17 levels in bronchoalveolar lavage and CD4(+), CD8(+), and IL-17(+) cell counts in endobronchial biopsies. Correlations were made with infection stat

Upregulation of an Epithelial miRNA Is Associated with Immune Evasion in Progressive Bronchial Premalignant Lesions.

Bronchial premalignant lesions (PML), precursors of lung squamous cell carcinoma, have distinct molecular subtypes. The proliferative subtype, enriched with bronchial dysplasia, had decreased expression of an antigen-processing/presentation gene coexpression module in progressive/persistent versus regressive PMLs, suggesting a functional impact of these genes on immune evasion. In this study, we performed miRNA sequencing, miRNA in situ hybridization, and spatial proteomics of bronchial biopsies from patients at high risk for lung cancer. An miRNA-gene network analysis identified hsa-miR-149-5

Chemotherapy-induced activation of caspase-1 and IL-1α release by cancer cells remotely skews myelopoiesis to drive pro tumorigenic systemic neutrophil-dominant inflammation.

While chemotherapy-induced tumor cell death is known to modulate the local immune landscape, its systemic impact on distant bone marrow-a site essential for immune cell maturation-remains underexplored. Here, we show that gemcitabine chemotherapy induces inflammatory caspase-1-dependent pyroptosis in epithelial cancer cells (epiCaspase-1). Despite its inflammatory nature, epiCaspase-1-mediated cell death is non-immunogenic. Clinically, elevated expression of an epiCaspase-1 gene signature correlates with worse patient outcomes. Mechanistically, epiCaspase-1 triggers the noncanonical release of

Identification of a distinct cluster of LY6E+ macrophages in esophageal squamous cell carcinoma: functional phenotype, spatial interaction, and prognostic significance.

BACKGROUND: Tumor-associated macrophages are key players in cancer progression, but their heterogeneity is not fully understood. This study aims to investigate the role of a specific macrophage subpopulation marked by LY6E in esophageal squamous cell carcinoma (ESCC). METHODS: We analyzed macrophage subsets in ESCC, focusing on LY6E+ populations. Their gene expression signature, functional characteristics, and correlation with patient prognosis and immunotherapy response were assessed through single cell RNA sequencing and multiplex immunohistochemistry. RESULTS: A distinct LY6E+ macrophage

GraphTME: graph-based framework for predicting immunotherapy response by interpreting tumour microenvironment interactions using spatial transcriptomics.

Immune checkpoint inhibitors (ICIs), which reactivate T-cell responses against tumours, show limited clinical efficacy due to low response rates and the lack of robust predictive biomarkers. Cell-cell interactions within the tumour microenvironment influence therapeutic response and can be analysed at single-cell resolution using imaging-based spatial transcriptomics. We present GraphTME, a spatially informed and biologically interpretable framework that predicts anti-PD-1 response by modelling pathway-specific ligand-receptor signalling as a multi-relational directed graph with edge weights i

Chenodeoxycholic acid restrains tumor growth via TGR5-dependent type 1 dendritic cells cross-priming.

Conventional type 1 dendritic cells (cDC1s) play a pivotal role in initiating CD8+ T-cell-mediated antitumor immunity, and their intratumoral abundance strongly correlates with immunotherapy efficacy. Here, we demonstrate that orally administered chenodeoxycholic acid (CDCA), a clinically approved metabolic modulator, accumulates in the tumor interstitial fluid (TIF) of tumor-bearing mice as its metabolite taurochenodeoxycholic acid (TCDCA), thereby potentiating the antitumor functions of cDC1s and CD8+ T cells through a gut microbiota-independent mechanism. Mechanistic studies revealed that C

Tumor sialylation regulates G-CSF stability and promotes neutrophil-mediated immunosuppression in breast cancer.

Breast cancer is the most prevalent cancer among women, yet immune checkpoint inhibitors remain largely ineffective in the majority of patients. Here we show that tumor sialylation is negatively associated with T cell infiltration across molecular breast cancer subtypes. In preclinical models, both genetic and pharmacologic inhibition of tumor cell sialylation reshape the tumor microenvironment by reducing immunosuppressive neutrophil infiltration, while CD8⁺ effector and Tcf7⁺ memory T cells are increased within the mammary tumors. Mechanistically, sialylation enhances the serum half-life of

Reduced EZH2 Expression in Circulating CD8-Positive T Cells and Monocytes in Psoriasis.

Enhancer of Zeste Homologue 2 (EZH2) is an epigenetic regulator involved in immune cell differentiation and function; however, its role in psoriasis remains unknown. This study aimed to evaluate EZH2 expression in peripheral blood mononuclear cells from patients with psoriasis and explore its potential functional relevance to disease pathogenesis. Peripheral blood samples were obtained from 40 psoriasis patients and 18 healthy controls, and EZH2 expression in T cell and monocyte subsets was analysed by flow cytometry. EZH2 expression was significantly reduced in circulating CD8+ naïve and memo

Suppressive CD8+ T-Cells Are Key Cellular Mediators of Extracorporeal Photopheresis.

Extracorporeal photopheresis (ECP) is a widely utilized immunomodulatory procedure with an incompletely defined mechanism. In graft-versus-host disease (GvHD) and transplant rejection, ECP is thought to induce immune tolerance by increasing regulatory CD4+ T-cells, whereas in cutaneous T cell lymphoa it may enhance dendritic cell-mediated antigen presentation and cytotoxic T cell activity. We investigated the role of CD8+ T cells in ECP using a murine model of multiple sclerosis (MS). ECP protected mice from disease, mitigated CNS pathology, and was dependent on CD8+ T cells. Anti-CMV IgG titer determines organ-specific protection toward immune checkpoint blockade-induced toxicities.

BACKGROUND: Immune-related adverse events (irAEs) post-immune checkpoint blockade (ICB) are a leading cause of patient morbidity. Robust peripheral biomarkers of irAEs are required to improve patient stratification to existing treatment regimens, and these are currently lacking. Seropositivity for human cytomegalovirus (CMV) is associated with protection against severe (grade 3+) irAEs post-ICB; however, the impact of infection on systemic immunity is highly variable. Here, in a prospectively recruited pan-cancer ICB-treated cohort (n=472 patients), we investigate a novel relationship between

An mRNA vaccine encoding proteasome-targeted antigen enhances CD8+ T cell immunity.

The efficient induction of antigen-specific CD8+ T cell activation is crucial in the development of mRNA tumor vaccines. Endogenous antigens are primarily degraded through the ubiquitin-proteasome system, followed by antigen presentation via major histocompatibility complex class I (MHC-I) molecules, leading to the activation of CD8+ T cells. Therefore, in this study, a novel mRNA vaccine was developed by fusing the mRNA sequence encoding the antigen with a proteasome-targeting peptide (PTP), aiming to enhance proteasomal targeting of the antigen and facilitate its degradation through the ubiq

The LGALS9/miR-491-5p Axis Regulates CD8+ T Cell Function and Inhibits the Progression of Gastric Cancer.

Gastric cancer is a prevalent and clinically significant gastrointestinal malignancy worldwide. Gastric cancer is characterized by limited treatment efficacy and a high recurrence rate. In this study, we found that LGALS9 was highly expressed in gastric cancer and may contribute to disease progression by modulating the infiltration of CD8+ T cells. Genetic knockout of LGALS9 reversed T cell function, attenuated immunosuppression, and enhanced the cytotoxic activity of T cells to kill gastric cancer cells. Furthermore, miR-491-5p was identified as a potential suppressor in gastric cancer. The m

The involvement of microglia and the CXCL16-CXCR6 axis in the recruitment of CD8+ T cells to an amyloidogenic mouse brain.

Alzheimer's disease (AD) progression has been associated with the presence of brain-resident CD8+ T cells, and recent studies suggest a potential role of the CXCL16-CXCR6 axis in their recruitment to the brain. Here, we examined publicly available single-cell RNA sequencing datasets revealing that in the mouse brain, the receptor Cxcr6 is mainly expressed by CD8+ T cells, while the expression of its ligand Cxcl16 is predominantly observed in microglial cells. We found higher levels of Cxcl16 and Cxcr6 expression in APP/PS1 compared to wild-type mice. Furthermore, in vitro experiments using imm

Androgens inhibit protective CD8+ T cell responses against pre-erythrocytic malaria parasites in mice.

Attenuated whole organism vaccines targeting the malaria liver stage reliably confer sterile immunity. These vaccines completely protect female mice from infection, but protection in male mice remains unproven. We discover that male mice vaccinated with prime-and-trap, a whole organism-based vaccine strategy, exhibit poorer protection against Plasmodium sporozoite challenge than females. We investigate this sex difference, and identify vaccinated males have fewer hepatic memory CD8+ T cells than females when scaling for liver biomass, and reduced inflammatory responses post-vaccination. CD8+ T Cells Negatively Modulate Ischemia-Induced Angiogenesis in Mice.

Angiogenesis-related therapeutic approaches to peripheral arterial disease (PAD) deserve attention. CD8+ T cells play important roles in human pathobiology, and we investigated the involvement of cytotoxic CD8+ T cells in angiogenesis in response to ischemic stress. We also examined the mechanism of a component of green tea catechins, epigallocatechin-3-gallate (EGCG), which facilitates vascular regeneration in mice. Male 8-week-old wild-type [(CD8a+/+) and interferon-gamma (IFN-γ+/+)] knockout (CD8a-/- and IFN-γ-/-) mice were subjected to unilateral hindlimb ischemic surgery and then loss-of-
