A Spatiotemporal Atlas of the Androgen Receptor Proximal Interactome.

Androgen receptor-interacting proteins (AR-IPs) number close to 1,000, yet their organization across subcellular space and time remains uncharted. Proximity labeling identifies direct partners and neighboring proteins, thereby expanding AR-IPs to AR-proximal interacting proteins (AR-PIPs). Using proximity labeling quantitative mass spectrometry (PL-qMS), we construct a spatiotemporal atlas of the cytosolic, microsomal, and nuclear compartments in LNCaP prostate tumor cells. PL-qMS recovered 82.2% of the known AR-interactome in extranuclear compartments and 84.2% in the nucleus, identifying 4,7…

Lipid-dependent accrual of a subset of monocyte-derived macrophages is essential for tissue regeneration.

Tissue regeneration is essential for maintaining tissue homoeostasis and influences disease progression. In the liver, injury evokes a complex regenerative response with robust immune activation and metabolic rewiring, yet how these processes coordinate hepatocyte proliferation remains unclear. Here we show the presence of an injury-induced, lipid-dependent accrual of a distinct monocyte-derived macrophage (MDM) subset characterized by abundant cytosolic lipid content and heightened inflammatory response. Multi-omic analyses, spanning both single-cell transcriptomics and quantitative lipidomic…

[NAD kinases (NADKs) family proteins: Advances in metabolic regulation mechanisms and disease associations].

Nicotinamide adenine dinucleotide [NAD(H), consisting of its oxidized form NAD+ and reduced form NADH] and nicotinamide adenine dinucleotide … ecause NADP+ cannot cross subcellular membranes, eukaryotic NADKs are classified into two isoforms: cytosolic NADK1 and mitochondrial NADK2. The catalytic activity of NADKs is regulated at the transcriptional and post-translational modification levels, and aberrant expression or dysfunction of NADKs is closely linked to the initiation and progression of n …

Direct activation of SARM1 by dsDNA is not supported by biochemical and cellular evidence.

Sterile alpha and Toll/interleukin-1 receptor motif-containing protein 1 (SARM1) is a NAD+-consuming enzyme that drives axon degeneration and is activated by changes in the NMN/NAD+ ratio. A recent study proposed that cytosolic double-stranded DNA (dsDNA) directly binds SARM1's TIR domain, activating it independently of this canonical mechanism. Here, we evaluate dsDNA-dependent SARM1 activation using purified SARM1, biochemical and biophysical assays, primary neurons, and cellular pharmacology. Across multiple platforms, dsDNA does not stimulate SARM1 NADase activity, promote higher-order ass…

Nlrp12-driven PANoptosis exacerbates liver injury in ConA-induced autoimmune hepatitis.

Autoimmune hepatitis (AIH) involves chronic liver injury from abnormal immune responses, leading to hepatocyte death and inflammation. Currently, the role of pyroptosis in AIH has not been fully elucidated. Emerging evidence indicates that Nlrp12, a cytosolic sensor of damage-associated molecular patterns (DAMPs), participates in inflammasome assembly and pyroptotic signalling. Therefore, this study aims to investigate the role of Nlrp12 in AIH. Using a concanavalin A (ConA)-induced AIH mouse model, we observed that ConA challenge significantly upregulated Nlrp12 expression in liver tissues. G…

Virus-inspired disulfide engineering enables gene delivery via remodeling of cellular uptake and organ distribution.

Efficient cytosolic delivery remains a central challenge in the clinical translation of gene therapeutics. Inspired by viral strategies, we explored disulfide chemistry as a structural lever to enhance cellular uptake and intracellular trafficking. We systematically incorporated disulfide moieties into the poly(β-amino ester)s (PAEs) backbone to generate a series of PAEs with different disulfide content. These minimalist backbone modifications significantly reprogrammed the cellular uptake route through thiol-mediated internalization. Furthermore, we found that backbone disulfide incorporation…

The Wnt/APC destruction complex targets SREBP2 in a β-catenin-independent pathway to control cholesterol metabolism.

APC, the core scaffold of the Wnt destruction complex, targets the transcriptional co-activator β-catenin for proteolysis. There is no convincing evidence that APC directs degradation of other substrates. Using a reconstituted cytosolic extract-based system and complementary in vivo and cellular assays, we show that SREBP2, the master regulator of cholesterol biosynthesis, is a direct APC-AXIN1 substrate. APC-dependent SREBP2 degradation is conserved in Xenopus embryos, mouse colon, and human colorectal cancer cells and restricts SREBP2 target-gene expression, cholesterol synthesis, and tissue…

Filamin A editing in myeloid cells reduces intestinal inflammation and protects from colitis.

Patho-mechanistic origins of ulcerative colitis are still poorly understood. The actin cross-linker filamin A (FLNA) impacts cellular responses through interaction with cytosolic proteins. Posttranscriptional A-to-I editing generates two forms of FLNA: genome-encoded FLNAQ and FLNAR. FLNA is edited in colon fibroblasts, smooth muscle cells, and endothelial cells. We found that the FLNA editing status determines colitis severity. Editing was highest in healthy colons and reduced during murine and human colitis. Mice that exclusively express FLNAR were highly resistant to DSS-induced colitis, wh…

RNAi-mediated silencing of SOD1 profoundly extends survival and functional outcomes in ALS mice.

Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative condition, with 20% of familial and 2%-3% of sporadic cases linked to mutations in the cytosolic superoxide dismutase (SOD1) gene. Mutant SOD1 protein is toxic to motor neurons, making SOD1 gene suppression a promising approach, supported by preclinical data and the 2023 Federal Drug Administration (FDA) approval of the GapmeR ASO targeting SOD1, tofersen. Despite the approval of an ASO and the optimism it brings to the field, the pharmacodynamics and pharmacokinetics of therapeutic SOD1 modulation can be improved. Here, we devel…

Mass Cytometry-Based Approach for the Investigation of Stimulator of Interferon Genes Pathway.

The stimulator of interferon genes (STING) pathway plays a pivotal role in innate immunity, acting as a key sensor of cytosolic DNA to initiate type-I Interferon (IFN) and pro-inflammatory cytokine production. This pathway is essential for host defence against bacterial, viral and other pathogenic threats and has emerged as a promising therapeutic target in cancer immunotherapy. However, conventional techniques such as immunoblotting and qPCR are limited in their capacity to study STING pathway activation in complex and heterogeneous biological systems, such as tumour masses or large cell popu…

Oxysterol-binding protein ORP6 regulates lipid metabolism and brain Aβ production.

The mammalian brain is the most cholesterol-rich organ of the body, relying on in situ de novo cholesterol synthesis. Maintaining cholesterol homeostasis is crucial for normal brain function. Oxysterol-binding protein (OSBP)-related proteins (ORPs) are highly conserved cytosolic proteins that coordinate lipid homeostasis by regulating cell signaling, interorganelle membrane contact sites, and non-vesicular transport of cholesterol. Here, we show that ORP6 is highly enriched in the mammalian brain, particularly within neurons and astrocytes, with widespread expression across distinct brain regi…

KCS1 and VIP1, the genes encoding yeast phosphoinositol pyrophosphate synthases, are required for Ca2+-mediated response to dimethylsulfoxide (DMSO).

Dimethylsulfoxide (DMSO) is widely used as a solvent or as a carrier when screening for biologic activity of various chemicals, but results need to be interpreted carefully due to its intrinsic toxicity. DMSO has been previously observed to impair the growth of yeast cells defective in calcium movement across cellular membranes and in phosphoinositol pyrophosphate synthases. Here, we set out to investigate the Ca2+-mediated response to DMSO in Saccharomyces cerevisiae. The cell exposure to DMSO was signaled by a two-phase cytosolic Ca2+ wave that was dependent on Mid1, a subunit of the Cch1/Mi…

Coumarin-Containing Aromatic Sulfonamides With Carbonic Anhydrase Inhibitory Properties Against Human and Fungal Isoforms.

The vital process of CO2 hydration in all living things is catalyzed by carbonic anhydrases (CAs, EC 4.2.1.1), which are actively involved in the control of numerous pathological and physiological situations. A number of coumarin-based sulfonamides (18) were examined as potential CA inhibitors. Their inhibitory activity was assessed against the cytosolic human isoforms hCA I, hCA II, and the transmembrane hCA IX as well as against three fungal CAs: Malassezia globosa (MgCA), Malassezia pachydermatis (MpaCA), and Malassezia restricta (MreCA). Additionally, the binding mechanism of this family o…

AAV-based gene therapy ameliorates neurological deficits in a mouse model of childhood-onset neurodegeneration with cerebellar atrophy.

Childhood-onset neurodegeneration with cerebellar atrophy (CONDCA) is an autosomal recessive pediatric disorder caused by biallelic mutations in the ATP/GTP-binding protein 1 gene, which encodes cytosolic carboxypeptidase 1 (CCP1) with deglutamylase activity. These patients typically exhibit progressive motor and cognitive impairment, often leading to childhood mortality. Despite its severe clinical course, no effective treatments have been developed. In this study, we demonstrate the efficacy of gene therapy using adeno-associated virus (AAV) vectors to deliver a functional mouse Ccp1 mutant …

VDAC1 oligomerization-mediated mtDNA release under sublethal oxidative stress: A novel inflammatory mechanism in vitiligo.

Oxidative stress is a critical initiating factor in vitiligo, yet the early molecular events linking redox imbalance to melanocyte immune activation remain unclear. Here, we demonstrate that sub-lethal hydrogen peroxide (H2O2, 0.1 mM) exposure in human epidermal melanocytes induces a robust pro-inflammatory response independent of apoptosis or pyroptosis. This response is driven by the selective cytosolic release of mitochondrial DNA (mtDNA), which activates the cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) pathway. Mechanistically, voltage-dependent anion channel 1 (VDAC…

Phosphatidylcholine coordinates ER-autonomous and ER-nonautonomous adaptations to unfolded protein response dysfunction.

The ER UPR plays a crucial role in maintaining proteostasis, with its dysfunction closely associated with aging and various diseases. However, how cells cope with ER UPR dysfunction remains largely unexplored. Here, we report that both ER-autonomous and ER-nonautonomous adaptive responses are activated by defects in the IRE-1/XBP-1 UPR branch in Caenorhabditis elegans. IRE-1/XBP-1 dysfunction not only triggers the activation of the PEK-1 UPR branch but also induces a lysosome-dependent cytosolic proteostatic response. Mechanistically, IRE-1/XBP-1 dysfunction downregulates phosphatidylcholine (…

Inhibitory effects of parabens on human and rat 17β-hydroxysteroid dehydrogenase 1: Mechanisms of action and impact on hormone synthesis.

Parabens are commonly used preservatives in cosmetics, food, and pharmaceutical products. The objective of this study was to examine the effect of nine parabens on human and rat 17β-hydroxysteroid dehydrogenase 1 (17β-HSD1) in human placental and rat ovarian cytosols, as well as on estradiol synthesis in BeWo cells. The results showed that the IC50 values for these compounds varied from methylparaben with the weakest inhibition (106.42 μM) to hexylparaben with the strongest inhibition (2.05 μM) on human 17β-HSD1. Mode action analysis revealed that these compounds acted as mixed inhibitors. For…

Buffering of nuclear membrane tension and mechanotransduction by the endoplasmic reticulum revealed by quantitative ALPIN imaging.

Nuclear deformation by osmotic shock or necrosis activates the cytosolic phospholipase A2 (cPla2) nuclear shape sensing pathway, a key regulator of tissue inflammation and repair. Ca2+ and inner nuclear membrane (INM) tension (TINM) are believed to mediate nucleoplasmic cPla2 activation. The concept implies that TINM persists long enough to stimulate cPla2-INM adsorption. However, TINM may instead be rapidly dissipated by the contiguous endoplasmic reticulum (ER), with cPla2-INM adsorption reporting rather on changes in Ca2+ than TINM. The impact of TINM and ER contiguity on nuclear shape sens…
