Lipids, diet and Alzheimer disease: an extended summary.

A number of dietary elements and foods have been reported to be either risk or protective factors for the development of dementia and Alzheimer disease (AD). These include fat, fatty acids, antioxidants, fish, homocysteine/methionine, vitamins and alcohol. We propose that brain diseases with aging are not be only the result of pathogenic processes, but also due to the failure of protective mechanisms, and that diet influences the success of these protective mechanisms. Both animals and humans with genetic forms of AD do not get the disease until a certain time in mid or late life. Therefore, t

Antihepatotoxic activity of Phyllanthus fraternus.

Different fractions of alcoholic extracts of aerial parts and roots of Phyllanthus fraternus Webster (Euphorbiaceae) were screened for antihepatotoxic activity on carbon tetrachloride induced liver damage in albino rats. The degree of protection was measured using biochemical parameters like serum glutamic oxaloacetic transaminase (SGOT), serum glutamic pyruvate transaminase (SGPT), total protein (TP) and total albumin (TA). The methanol fraction was found to be the most active, which was further supported by a significant recovery of hepatocytes in the histopathological study of the liver sho

[Non-alcoholic hepatic steatosis].

Non-alcoholic steatosis of the liver is a common condition. It usually consists of triglyceride drop deposits in macro- or microvacuoles of the hepatocytes and may result from various disturbances of lipid metabolism, notably accumulation of fatty acids in the liver and decrease in the secretion or synthesis of lipoproteins. The causes of steatosis are numerous. It is important to distinguish between severe steatosis (acute steatosis of pregnancy, toxic steatosis, Reye's syndrome), where most of the deposits are in microvacuoles, and incidental or moderate steatosis of nutritional or hormonal

Association Between Weight-Adjusted Waist Circumference Index and Metabolic Disease-Associated Fatty Liver Disease: A Retrospective Study.

BACKGROUND: Metabolic disease-associated fatty liver disease has brought significant challenges to public health and social-economics. There is a need for a straightforward and effective method to screen for metabolic disease-associated fatty liver disease. The weight-adjusted waist circumference index offers a comprehensive reflection of visceral fat accumulation and skeletal muscle loss. This study aims to explore the relationship between weight-adjusted waist circumference index and hepatic steatosis and hepatic fibrosis. METHODS: This single-centered study screened 288 participants from t

Improved lipogenesis gene expression in liver is associated with elevated plasma angiotensin 1-7 after AT1 receptor blockade in insulin-resistant OLETF rats.

Increased angiotensin II (Ang II) signaling contributes to insulin resistance and liver steatosis. In addition to ameliorating hypertension, angiotensin receptor blockers (ARBs) improve lipid metabolism and hepatic steatosis, which are impaired with metabolic syndrome (MetS). Chronic blockade of the Ang II receptor type 1 (AT1) increases plasma angiotensin 1-7 (Ang 1-7), which mediates mechanisms counterregulatory to AT1 signaling. Elevated plasma Ang 1-7 is associated with decreased plasma triacylglycerol (TAG), cholesterol, glucose, and insulin; however, the benefits of RAS modulation to pre

Methionine limitation results in increased hepatic FAS activity, higher liver 18:1 to 18:0 fatty acid ratio and hepatic TAG accumulation in Atlantic salmon, Salmo salar.

The current experiment aimed to study whether interactions with lipid metabolism possibly might explain the relative increased liver weight obtained in fish fed sub-optimal methionine levels. A basal diet based on a blend of plant proteins which is low in methionine (1.6 g Met/16 g N) was compared to a methionine adequate diet (2.2 g Met/16 g N) prepared by adding DL-methionine (2.4 g/kg) to the basal diet in the expense of wheat grain. Fish oil was used as the lipid source. The diets were balanced in all nutrients except methionine. The diets were fed to Atlantic salmon (500 g BW) for a perio

Proanthocyanidins alleviate acute alcohol liver injury by inhibiting pyroptosis via inhibiting the ROS-MLKL-CTSB-NLRP3 pathway.

BACKGROUND: Alcoholic Liver Disease (ALD) is a hepatic disorder resulting from prolonged or excessive alcohol intake. The predominant manifestation of ALD is fatty liver, which progresses to alcoholic hepatitis as the disease worsens. Pyroptosis is a novel type of programmed cell death that is intricately linked to the inflammatory cascade, presenting a promising avenue for therapeutic intervention in the management of ALD. Oligomeric proanthocyanidins (OPCs) are polyphenols extracted from grape seeds that have anti-inflammatory and antioxidant properties. However, whether OPCs can treat ALD b

Morroniside delays the progression of non-alcoholic steatohepatitis by promoting AMPK-mediated lipophagy.

BACKGROUND: Non-alcoholic steatohepatitis (NASH), the inflammatory subtype in the progression of non-alcoholic fatty liver disease, is becoming a serious burden threatening human health, but no approved medication is available to date. Mononoside is a natural active substance derived from Cornus officinalis and has been confirmed to have great potential in regulating lipid metabolism in our previous studies. However, its effect and mechanism to inhibit the progression of NASH remains unclear. PURPOSE: Our work aimed to explore the action of mononoside in delaying the progression of NASH and i

Dual elastography ultrasound for classifying metabolic dysfunction-associated steatotic liver disease: a cross-sectional study within a prospective cohort.

BACKGROUND: The binary diagnostic approach does not reflect the entire spectrum of metabolic dysfunction associated steatotic liver disease (MASLD. We used an elastography technology, dual elastography ultrasound (DEUS), to discriminate the different stages of MASLD. METHOD: This prospective multicenter study was conducted from December 2020 to March 2022. All patients underwent DEUS scan, a liver biopsy, and a liver function laboratory test. The optimal model was developed (ModelDEUSC) with 10 machine learning algorithms by combining DEUS and selected clinical parameters and tested the diagn

Algorithms for Risk Stratification in Patients with Metabolic Dysfunction-Associated Steatohepatitis.

Early identification of patients with metabolic dysfunction-associated steatohepatitis (MASH) and significant fibrosis (≥F2) is critical to optimize patient care and prevent both hepatic and extra-hepatic complications. Current international guidelines consistently recommend a screening in high-risk populations using 2-step algorithms based on noninvasive tests. This review will examine the recommended risk stratification algorithms for MASH, summarize the evidence supporting their clinical use, and highlight the potential benefits for patient management. Finally, we will discuss the key barri

Sex-specific dynamics of MASLD reveal early hepatic and extrahepatic metabolic deterioration in females despite long-term protection.

BACKGROUND: Metabolic dysfunction-associated steatotic liver disease (MASLD) is a chronic condition characterized by hepatic fat accumulation and systemic metabolic dysfunction. MASLD exhibits clear sex differences, yet the mechanisms underlying these disparities remain poorly defined. METHODS: To investigate the early temporal dynamics of MASLD, male and female mice were fed a choline-deficient, methionine-restricted (0.1%) high-fat (61%) diet (CDAHFD; characterized by impaired hepatic lipid export and enhanced lipotoxic stress) for two weeks, enabling assessment of initial metabolic respons

The correlation between the trajectory of plasma atherosclerosis-inducing index in the examination population and the risk of developing metabolic dysfunction-associated steatotic liver disease.

BACKGROUND: Metabolic Dysfunction-Associated Steatotic Liver Disease (MASLD) is a prevalent metabolic disorder with rising global incidence and substantial clinical implications. The atherogenic index of plasma (AIP), a novel marker of lipid metabolism, has shown potential in predicting metabolic diseases, but its longitudinal association with MASLD remains unclear. METHODS: This longitudinal study included 1,894 participants from the health examination database of the First Affiliated Hospital of Xi'an Jiaotong University (2020-2024). Group-based trajectory modeling (GBTM) identified three A

Ferroptosis in MASLD: From molecular mechanisms to therapeutic opportunities.

Metabolic dysfunction-associated steatotic liver disease (MASLD) encompasses a spectrum of liver conditions, ranging from isolated steatosis to metabolic dysfunction-associated steatohepatitis (MASH), advanced fibrosis, and cirrhosis. Ferroptosis, an iron-dependent form of regulated cell death driven by lipid peroxidation, has emerged as a key pathogenic mechanism in MASLD. Given the liver's pivotal role in iron and lipid metabolism, hepatocytes are highly susceptible to ferroptotic injury, which amplifies hepatic oxidative stress, inflammation, and fibrogenesis. Beyond the liver, ferroptosis

The therapeutic role of natural compounds mediated through Nrf2 in metabolic dysfunction-associated steatotic liver disease.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is one of the most prevalent chronic liver diseases in the world, with a current global prevalence estimated to exceed 33.6%. Consequently, the search for safe and effective therapeutic agents for MASLD has gained increasing attention. Nuclear factor erythroid 2-related factor 2 (Nrf2), a well-characterized transcription factor, plays a pivotal role in cellular defense against oxidative stress. Accumulating evidence indicates that activating Nrf2 has emerged as a promising therapeutic strategy for MASLD. Notably, numerous natural

Multi-modal AI-enabled steatotic liver disease diagnostics using facial images and metabolomics.

Steatotic liver disease (SLD) affects one-third of the global population, yet current non-invasive diagnostic methods are too costly or operator-dependent for population-scale screening. Here, we present 3D-FAICE, a deep learning system that uses three-dimensional facial imaging for non-invasive SLD detection. Trained and tested on 11,456 participants, the facial model achieves robust performance across internal, external, and self-controlled longitudinal cohorts and remains effective in a smartphone-based point-of-care setting. Metabolomic analysis reveals that facial risk scores correlate wi
