Brain abnormalities observed in alcohol use disorder are highly heterogeneous in nature and severity, possibly because chronic alcohol consumption also affects peripheral organs leading to comorbidities that can result in exacerbated brain alterations. Despite numerous studies focussing on the effects of alcohol on the brain or liver, few studies have simultaneously examined liver function and brain damage in alcohol use disorder, and even fewer investigated the relationship between them except in hepatic encephalopathy. And yet, liver dysfunction may be a risk factor for the development of al

Positron emission tomography (PET) provides an in vivo molecular marker for various diseases, including Alzheimer's disease and related dementias (ADRD). PET has become increasingly integrated into diagnostic decision-making, disease staging, and clinical trial enrichment. However, its widespread use remains constrained by high costs, government regulations, and the invasiveness of radiotracer injection. Modern diagnostic frameworks emphasize the importance of multimodal biomarker assessment, such as the "amyloid/tau/neurodegeneration" (A/T/N) framework for Alzheimer's disease; however, they a

Moyamoya disease (MMD) is a chronic, progressive cerebrovascular condition marked by narrowing or blockage of the terminal segments of the internal carotid arteries, resulting in ischemic and hemorrhagic strokes. Its pathogenesis involves a multifactorial interplay between genetic susceptibility, immune-inflammatory dysregulation, endothelial dysfunction, and aberrant vascular remodeling, influenced by non-genetic and environmental factors. Despite considerable research progress, clinically useful biomarkers remain limited, lacking sufficient sensitivity and specificity for predicting disease

Background: A neuroimaging-derived biomarker termed the brain age is considered to capture the degree and diversity in the aging process of the brain, serving as a robust indicator of overall brain health. The impact of different levels of physical activity (PA) intensities on brain age is still not fully understood. This study aimed to investigate the associations between accelerometer-measured PA and brain age. Methods: A total of 16,972 eligible participants with both valid T 1-weighted neuroimaging and accelerometer data from the UK Biobank was included. Brain age was estimated using an en

BACKGROUND: Irregular word reading has been used to estimate premorbid intelligence in Alzheimer's disease (AD) dementia. However, reading models highlight the core influence of semantic abilities on irregular word reading, which shows early decline in AD. The general aim of this study is to determine whether irregular word reading is a valid estimate of premorbid intelligence, or a marker of cognitive and semantic decline in AD.
METHOD: 681 healthy controls (HC), 104 subjective cognitive decline, 290 early and 589 late mild cognitive impairment (EMCI, LMCI) and 348 AD participants from the A

Background Data on the relative contribution of clinical and neuroimaging risk factors to acute ischemic stroke (AIS) versus intracerebral hemorrhage (ICH) occurring on oral anticoagulant treatment are scarce. Methods and Results Cross-sectional study was done on consecutive oral anticoagulant-treated patients presenting with AIS, transient ischemic attack (TIA), or ICH from the prospective observational NOACISP (Novel-Oral-Anticoagulants-In-Stroke-Patients)-Acute registry. We compared clinical and neuroimaging characteristics (small vessel disease markers and atherosclerosis) in ICH versus AI

Cognitive control is fundamental to goal-directed behavior, and its protracted maturation is a hallmark of adolescent brain development. In adulthood, the inferior frontal junction (IFJ) is functionally characterized as a critical region for updating task representations to guide the implementation of cognitive control. Yet, how its domain-general control functions emerge and mature across development remains largely underexplored. Specifically, it is unclear whether IFJ's capacity for cognitive control enhances uniformly as a single construct, or if this region matures asynchronously for dist

Biallelic variants in a family with sequence similarity 149 member B1 gene (FAM149B1, OMIM #618413) causes a range of abnormal phenotypes associated with Joubert syndrome (JS) in humans. However, the phenotypic spectrum and genetic evidence linking FAM149B1 to ciliopathies remain limited, with no prenatal cases previously described. Here, we report the first prenatal case from a non-consanguineous Chinese family presenting with isolated right pelvicalyceal and ureteral dilation at 24 weeks of gestation. Notably, the fetus lacked cerebellar malformations or hallmark neuroimaging features such a

Fragile X-associated tremor/ataxia syndrome (FXTAS), caused by the FMR1 premutation allele, is associated with brain degeneration, yet the mechanisms behind this neurodegeneration still need to be elucidated. Apoε polymorphism has been widely implicated in brain aging in cognitively healthy individuals and brain deterioration in Alzheimer's disease. This study aimed to examine the interaction of Apoε genotypes, FXTAS clinical symptoms, FMR1 molecular measures, and age, towards brain pathophysiology and cognitive functions. This longitudinal study includes MRI data collected from 205 male premu

OBJECTIVE: This study aimed to investigate the role of circulating inflammatory cytokines in the pathway linking cerebral small vessel disease (CSVD) to cognitive impairment (CI), and to further elucidate the neuroimaging mechanism of CSVD-driven inflammatory cytokines on cognitive function.
METHODS: We conducted a two-step, two-sample Mendelian randomization (MR) analysis to evaluate the causal effect of CSVD on circulating inflammatory cytokines and CSVD-driven inflammatory cytokines on the risk of CI. Using a separate two-sample MR analysis, we explored the potential mechanisms by which th

Empathy, the ability to understand others' perspectives (cognitive empathy) and share their emotions (emotional empathy), is critical for social functioning across the lifespan. Although empathy changes with age and is often disrupted in neurodegenerative disease, prior research has largely examined isolated components or focused narrowly on healthy aging or specific disorders. This systematic review addresses this gap by synthesizing behavioral and neuroimaging evidence on empathy across typical and pathological aging, from healthy older adulthood and mild cognitive impairment to Alzheimer's

Violence exposure has deleterious effects on emotional well-being, including higher rates of future mental illness. Adolescence is an important period of neural development within brain regions (e.g., prefrontal cortex) that support emotional processes. The relationship between brain activity and emotion may vary with violence exposure. Thus, this study investigated the relationship between violence exposure, stress-elicited brain activity, and emotion in young people. Violence exposure was measured four times from 11 to 19 years of age. Participants (n = 301) returned 1 year later (age = 20)

Alzheimer's disease (AD) is a neurocognitive disorder that deteriorates memory and impairs cognitive functions. Mild Cognitive Impairment (MCI) is generally considered as an intermediate phase between normal cognitive aging and more severe conditions such as AD. Although not all individuals with MCI will develop AD, they are at an increased risk of developing AD. Diagnosing AD once strong symptoms are already present is of limited value, as AD leads to irreversible cognitive decline and brain damage. Thus, it is crucial to develop methods for the early prediction of AD in individuals with MCI.

OBJECTIVES: Bilingualism can stimulate brain plasticity and is often associated with better executive function. We investigated whether language usage was related to the volume of white matter lesions in a cohort of middle-aged to older adults with different multilingual characteristics, including monolingualism in daily life (use of one language), bilingualism (use of two languages), and multilingualism (active use of 3 languages, i.e., Catalan, Spanish, English). We also explored cognitive status and cognitive change over a 2-year follow-up period linked to possible associations between lang

Mild cognitive impairment (MCI) and sarcopenia are prevalent age-related conditions that often coexist and share common mechanisms such as chronic inflammation, reduced neuroplasticity, and impaired muscle function. Resistance exercise training (RET) has emerged as a promising non-pharmacological strategy capable of addressing both physical and cognitive decline. The aim of this narrative review is to synthesize preclinical and clinical evidence on the effects of RET in older adults with MCI and sarcopenia, with a specific focus on its impact on neuroinflammation, cognitive performance and str

Neuroimaging studies have consistently demonstrated concurrent activation of the human precuneus and temporal pole (TP), both during resting-state conditions and various higher-order cognitive functions. However, the precise underlying structural connectivity between these brain regions remains uncertain despite significant advancements in neuroscience research. In this study, we investigated the connectivity of the precuneus and TP by employing parcellation-based fiber micro-dissections in human brains and fiber tractography techniques in a sample of 1065 human subjects and a sample of 41 rhe

BACKGROUND: Posterior fossa irradiation with or without whole brain irradiation results in high doses of radiation to the thalamus, hippocampus, and putamen, structures critical to cognitive functioning. As a result, children with brain tumors treated with cranial irradiation (CRT) may experience significant cognitive late effects. We sought to determine the effect of radiation to those structures on neuropsychological outcome.
METHODS: Forty-seven children with a history of posterior fossa tumor (17 treated with surgery; 11 with surgery and chemotherapy; and 19 with surgery, chemotherapy, an

The etiopathogenesis of late-onset Alzheimer's disease (AD) is increasingly recognized as the result of the combination of the aging process, toxic proteins, brain dysmetabolism, and genetic risks. Although the role of mitochondrial dysfunction in the pathogenesis of AD has been well-appreciated, the interaction between mitochondrial function and genetic variability in promoting dementia is still poorly understood. In this study, by tissue-specific transcriptome-wide association study (TWAS) and further meta-analysis, we examined the genetic association between mitochondrial solute carrier fam

Postural change from supine or sitting to standing up leads to displacement of 300 to 1000 mL of blood from the central parts of the body to the lower limb, which causes a decrease in venous return to the heart, hence decrease in cardiac output, causing a drop in blood pressure. This may lead to falling down, syncope, and in general reducing the quality of daily activities, especially in the elderly and anyone suffering from nervous system disorders such as Parkinson's or orthostatic hypotension (OH). Among different modalities to study brain function, functional near-infrared spectroscopy (fN
