Unlocking the surgical potential of Descemet's membrane: a standardized decellularization protocol.

Descemet's membrane is the specialized extracellular matrix located between corneal stroma and endothelium. This basement membrane provides the biomechanical cues that sustain endothelium viability and function, making it an optimal cell scaffold. The present work describes a new decellularization protocol to remove the cellular components and obtain an acellular scaffold from Descemet's membrane. To induce cell lysis and eliminate all cytoplasmic and nuclear material, Descemet's membranes isolated from donor corneas were subjected to osmotic (hypotonic) shock. The efficiency of the decellular…

Isolation and Proteomic Characterization of Outer Membrane Vesicles in Fusobacterium nucleatum.

Fusobacterium nucleatum is an anaerobic Gram-negative bacterium associated with periodontal disease and colorectal cancer. Outer membrane vesicles (OMVs) produced by F. nucleatum are nanoscale lipid vesicles enriched with immunogenic and virulence-associated proteins, making them promising candidates for vaccine development. This protocol describes a comprehensive workflow for culturing F. nucleatum, isolating and purifying OMVs via ultracentrifugation and density gradient ultracentrifugation, and characterizing their biophysical properties using transmission electron microscopy (TEM) and dyna

Effects of semaglutide on vessel morphology: Studies on the chicken chorioallantoic membrane.

Glucagon-Like Peptide-1 (GLP-1) receptor agonists are widely used to manage type 2 diabetes and promote weight loss. Semaglutide (SEM)-a long acting GLP-1-has experienced an extraordinary surge in popularity since its approval in 2017. Between 2021 and 2023, SEM prescription fills in the U.S. climbed to 2.56 million per month. Yet, the uptake of SEM into larger populations has raised safety concerns, with provocative findings now suggesting that SEM could negatively affect ocular and reproductive systems, counter to its beneficial effects on the heart. At least some of these concerns involve S

Optimizing the use of membrane filtration for the amelioration of smoke tainted wine.

Efficient removal of free and glycosylated volatile phenols from smoke tainted wine remains a significant challenge for winemakers. The partitioning of volatile phenols and their glycoconjugates (as smoke taint markers) by ultrafiltration (UF) and nanofiltration (NF) membranes was evaluated. While the volatile phenol content of permeate derived from NF and UF were comparable to that of untreated wine, glycoconjugates of volatile phenols were progressively concentrated in retentate as membrane molecular weight cut-off decreased. Three smoke tainted wines were subsequently fractionated by UF and…

Microcondensate-Mediated Intracellular Infusion of mRNA Across the Plasma Membrane.

This study presents a strategy for developing micrometer-sized coacervates comprised of cationic intracellular delivery peptide FcB(L17E)3 and nucleic acids (termed "microcondensates") as a lipid nanoparticle (LNP)-free platform for intracellular messenger RNA (mRNA) delivery. The feasibility of this approach is demonstrated both in vitro and in vivo. This approach is based on our previous finding that FcB(L17E)3 forms a microcondensate through interactions with IgG labeled with a negatively charged fluorescent dye. Similarly, the negative charge of nucleic acids causes siRNAs to form microcon

Cholesterol modulates membrane elasticity via unified biophysical laws.

Cholesterol and lipid unsaturation underlie a balance of opposing forces that features prominently in adaptive cell responses to diet and environmental cues. These competing factors have resulted in contradictory observations of membrane elasticity across different measurement scales, requiring chemical specificity to explain incompatible structural and elastic effects. Here, we demonstrate that - unlike macroscopic observations - lipid membranes exhibit a unified elastic behavior in the mesoscopic regime between molecular and macroscopic dimensions. Using nuclear spin techniques and computati…

Interaction of CFTR Modulators with Mammalian Membrane Mimetics: The Role of Cholesterol.

Lumacaftor and Ivacaftor are two FDA-approved medications currently used to treat cystic fibrosis (CF), a genetic disease caused by mutations in the cystic fibrosis transmembrane conductance regulator (CFTR), a chloride ion channel located in epithelial cell membranes; however, the detailed mechanism(s) of their action remains to be elucidated. Both drugs, termed modulators, bind CFTR at a protein-lipid interface, yet Lumacaftor acts at the endoplasmic reticulum (ER), while Ivacaftor acts at the plasma membrane (PM). A major difference among biological membranes is their level of cholesterol (…

St. John's Wort for Depression: From Neurotransmitters to Membrane Plasticity.

Depression is a multifactorial disorder shaped by genetic, psychosocial, and biological influences, with hypotheses ranging from monoamine deficiency and neuroplasticity deficits to inflammation and stress-induced dysregulation. St. John's wort (Hypericum perforatum L.) has long been used as an herbal antidepressant and is supported by clinical evidence for efficacy and safety in mild-to-moderate depression. While its multimodal mechanisms have been linked to neurotransmitter reuptake inhibition, neuroendocrine regulation, and modulation of neuroplasticity, recent findings suggest an additiona

The human amniotic membrane in vitreoretinal surgery: Applications, outcomes and limitations.

The human amniotic membrane (hAM) has gained recognition as a valuable biological adjunct in vitreoretinal surgery, with its applications rapidly expanding since its initial use for macular holes (MHs). We examine the development and present applications of hAM in vitreoretinal surgery, assessing its use in treating various pathological conditions such as MHs, retinal detachments (RDs) with proliferative vitreoretinopathy (PVR), high myopia-associated MHs with posterior staphyloma, optic pit maculopathy, and other vitreoretinal disorders. We evaluate the differences between cryopreserved and l

Extracorporeal Membrane Oxygenation and Eculizumab for Cardiac Failure in STEC-HUS.

Hemolytic uremic syndrome (HUS) is a complication of Shiga toxin-producing Escherichia coli (STEC) infection that can cause acute renal failure and introduce serious extrarenal organ injury. The most life threatening of these include cardiac failure and thrombocytopenia. Here, we present a case of severe STEC-HUS in a previously healthy 2-year-old boy who progressed to cardiac failure necessitating extracorporeal membrane oxygenation (ECMO). Following ECMO cannulation, he was treated with eculizumab, an anticomplement component 5 monoclonal antibody. The patient required ECMO for 5 days and su

Histopathology of Diabetic Nephropathy: Beyond Glomerular Basement Membrane Thickening.

Diabetic nephropathy (DN) is a leading cause of chronic kidney disease and end-stage renal disease worldwide, characterized by progressive injury across all renal compartments. Traditionally considered a glomerular disorder marked by basement membrane thickening, mesangial expansion, and nodular sclerosis, DN is now recognized as a multifaceted process involving tubulointerstitial, vascular, and podocyte pathology. This narrative review integrates current histopathological insights, highlighting key glomerular lesions, tubular basement membrane thickening, interstitial fibrosis, arteriolar hya

Etiology of Leukemoid Reaction in Patients Undergoing Extracorporeal Membrane Oxygenation.

INTRODUCTION: Nosocomial infections are frequent in patients receiving extracorporeal membrane oxygenation (ECMO). Leukemoid reaction, defined as >50,000 white blood cells (WBCs) per microliter, has been associated with infections in some populations. As ECMO is associated with significant inflammation activation between the patient's immune system and the circuit components, it is hypothesized that leukemoid reactions may have low specificity for identifying new infections in patients receiving ECMO. MATERIALS AND METHODS: A retrospective cohort study was performed on all adult patients admi

The subdural membrane: unraveling the pathophysiology and treatment of chronic subdural hematoma.

Chronic subdural hematoma (cSDH) is one of the most common neurosurgical conditions, with incidence expected to rise sharply as populations age. Once regarded as a passive sequela of venous rupture, cSDH is now understood as an active, membrane-driven disorder characterized by persistent inflammation, fragile neovascularization, and local fibrinolysis that collectively fuel hematoma expansion and recurrence. This evolving biological model explains the limitations of traditional surgical approaches, which relieve mass effect but do not address the underlying pathophysiology. Burr-hole drainage

Effect of polypropylene microplastics on the performance of membrane bioreactors in wastewater treatment.

Membrane bioreactors (MBRs) can effectively remove microplastics (MPs) because of their good rejection performance. However, the influence of MP concentration and particle size on the short-term and long-term operation efficiency of MBRs remains unclear. To address this issue, this study investigated the effects of short-term stress and long-term accumulation of polypropylene microplastics (PP-MPs) with different particle sizes on the operational efficiency of MBRs by running three MBR systems at four concentration stages. Variations in pollutant removal, biological activity, composition of ex
