Electron microscopy allows the analysis of synaptic ultrastructure and its modifications during learning or in pathological conditions. However, conventional electron microscopy uses aldehyde fixatives that alter the morphology of the synapse by changing osmolarity and collapsing its molecular components. We have used high-pressure freezing (HPF) to capture within a few milliseconds structural features without aldehyde fixative, and thus to provide a snapshot of living synapses. CA1 hippocampal area slices from P21 rats were frozen at -173 degrees C under high pressure to reduce crystal format

Evaluation of genotoxic effects of potassium chromate (K2CrO4) and cadmium chloride (CdCl2) was carried out in human blood lymphocytes in vitro as measured by the electron microscopy in situ end-labeling (EM-ISEL). EM-ISEL was used to assess DNA single-strand breaks (SSBs) expressed as number of immunogold particles per microm2 of chromatin at both chromosomal and nuclear DNA levels. Human lymphocytes were cultured in supplemented RPMI medium for 72 h including treatment for 2 h with K2CrO4 (0-150 microM), CdCl2 (0-150 microM) or methyl methanesulfonate (500 microM) as a positive control. The interaction between polymer particles with an immune ligand on their surfaces and normal (hepatocytes) and malignant (Raji) cells with a corresponding membrane receptor was investigated by scanning and transmission electron microscopy. The influence of time, temperature, and anti-metabolites on this process was studied. The results indicate that when particles with an immunoligand are added to cells with a corresponding membrane receptor, three successive steps are initiated, the first two of which are not dependent on the temperature or the metabolic state of the cells. The first step in

BACKGROUND AND PURPOSE: Maternal hypoxia is a recognised risk factor for neurodevelopmental disorders in offspring. Although rodent models of hypoxia have been reported, the detailed pathogenesis of maternal hypoxia-induced neurodevelopmental disorders remains unclear. The objectives of this study are to understand the pathophysiological mechanisms underlying maternal hypoxia-related neurodevelopmental disorders and to identify new drug targets for these pathologies.
EXPERIMENTAL APPROACH: A newly generated rat model exhibiting maternal hypoxia-induced neurodevelopmental disorder-like phenoty

Storage temperature and microbial interactions are critical to human norovirus transmission and food safety in fresh produce. This study examined the survival of murine norovirus (MNV-1), a surrogate of human norovirus, and co-contaminating bacteria on organic lettuce, being a significant vehicle, and its packaging materials during cold-chain transportation, focusing on temperature fluctuations and microbial interactions. Samples contaminated with Pseudomonas sp. SC006 [Histo-Blood Group Antigen (HBGA)-expressing bacteria], Escherichia coli O157:H7, and MNV-1 were exposed to 4°C, fluctuating t

Goose circovirus (GoCV), recognized as the most economically significant immunosuppressive pathogen in global waterfowl production, causes considerable economic losses annually, and multiprovincial surveillance studies have documented high seroprevalence rates. The critical barrier to combating GoCV has persisted in the complete absence of viable in vitro culture systems, fundamentally limiting pathogenesis studies and vaccine development. In a breakthrough advancement, this study pioneers the first successful establishment of a stable primary goose embryo kidney (GEK) cell culture system for

Toxoplasma gondii, an important opportunistic pathogen, underscores the necessity of developing novel therapeutic drugs and identifying new drug targets. Our findings indicate that the half-maximal inhibitory concentrations (IC50) of KU60019 and CP466722 (abbreviated as KU and CP) against T. gondii are 0.522 μM and 0.702 μM, respectively, with selection indices (SI) of 68 and 10. Treatment with KU and CP affects the in vitro growth of T. gondii, inducing aberrant division in the daughter parasites. Transmission electron microscopy reveals that KU and CP prompt the anomalous division of T. gond

We performed a detailed morphological and electrophysiological analysis of the neuromuscular junction in muscle biopsies from 10 patients with post-polio syndrome (PPS). This was done to clarify the basis for the apparent neuromuscular transmission impairment in PPS. In six patients, intracellular microelectrode recordings demonstrated either reduction of amplitudes of miniature end-plate potentials (MEPPs) or decreased quantal content or both. In one patient, reduction of quantal content was only present with prolonged or high-frequency nerve stimulation. In three patients no significant abno

We report the discovery of a new enveloped, spherical virus belonging to the Phenuiviridae family of negative ssRNA viruses associated with a massive outbreak in a French population of the endangered white-clawed crayfish Austropotamobius pallipes. We call this virus Bunya-like Brown Spot Virus (BBSV) and characterize it using transmission electronic microscopy, genome sequencing and clinical signs. Infected specimens show discolored brown spots on the cuticle. Using RNA-seq data we assembled a partial sequence for the L, M and S genome segments of BBSV. Phylogenetic analyses using all three s

Waste swamps, stagnant water, and poor hygiene practices result in the proliferation of mosquitoes that may cause transmissible and infectious diseases such as malaria, typhoid, cholera, and Zika virus sickness. It has been shown that composites of the traditional natural fibers cotton, viscose, and linen and a Ti-bearing metal-organic framework, NH2-MIL-125, are very effective against mosquitoes in the absence of any conventional insecticides. In our study, prior to coating with NH2-MIL-125 crystals, the fabrics were modified with 3-glycidyloxypropyltrimethoxysilane. The composite materials w

Cerebral cortex biopsy from a patient with new variant Creutzfeldt-Jakob disease (nvCJD) has been examined at the electron microscope level. Spongiform changes corresponded mostly to distended neurites scattered in the neuropil or surrounding amyloid plaques. These latter exhibited heterogeneous submicroscopic morphology including variable amount of loosely interwoven amyloid fibrils admixed in a cellular-rich environment constituted essentially by abnormal neuronal processes. By immunoelectron microscopy, fibrils and some membrane structures reacted with anti-prion protein (PrP) antibodies. O

Cranial visceral afferents activate central pathways that mediate systemic homeostatic processes. Afferent information arrives in the brainstem nucleus of the solitary tract (NTS) and is relayed to other CNS sites for integration into autonomic responses and complex behaviors. Little is known about the organization or nature of processing within NTS. We injected fluorescent retrograde tracers into two nuclei to identify neurons that project to sites involved in autonomic regulation: the caudal ventrolateral medulla (CVLM) or paraventricular nucleus of the hypothalamus (PVN). We found distinct

Trypanosomiasis has emerged as a significant health concern in cultured large yellow croaker (Larimichthys crocea), yet the organ-level distribution of the causative trypanosome within this host remains poorly characterized. The trypanosome was confirmed as Trypanosoma sp. Lc by 18S rRNA gene sequencing (99.9% identity to two isolates from L. crocea in Fujian); its precise taxonomic placement remains under discussion. We quantified trypanosome distribution across 12 organ types of naturally microscopy-confirmed infected L. crocea (n = 6; collected from Zhujiajian Island, Zhoushan, China) by RT

Hippoboscidae diptera or louse flies are widely distributed blood-feeding ectoparasites that can transmit blood parasites to their host. Four species are recognized within the Crataerina genus, which are parasites of the bird orders Apodiformes and Passeriformes. Alpine swifts (Tachymarptis melba) are frequently parasitized by Crataerina louse flies during the nesting season. In Switzerland, an increase in Alpine swift nestling mortality has been associated with a Trypanosoma sp. infection. In this study, fixed and native specimens of Craterina louse-flies collected from Swiss Alpine swifts we

BACKGROUND: Leishmaniasis as a neglected tropical disease (NTD) is caused by the inoculation of Leishmania parasites via the bite of phlebotomine sand flies. After an infected bite, a series of innate and adaptive immune responses occurs, among which neutrophils can be mentioned as the initiators. Among the multiple functions of these fighting cells, neutrophil extracellular traps (NETs) were studied in the presence of Leishmania major promastigotes and salivary gland homogenates (SGH) of Phlebotomus papatasi alone, and in combination to mimic natural conditions of transmission.
