Evaluating intermediate-term functional outcomes of double-sided buccal mucosal graft urethroplasty for anterior urethral stricture.

AIM OF THE STUDY: This study aims to evaluate the urinary and sexual outcomes following double-sided buccal mucosal graft (BMG) urethroplasty in patients with near-obliterated anterior urethral strictures. METHODS: We conducted a retrospective analysis of patients who underwent double-sided BMG urethroplasty between June 2017 and September 2024. We included only patients with complete perioperative data and a follow-up period of at least 6 months. The double-sided approach was utilized for nearly obliterated stricture segments (less than 6 French) during augmentation of long bulbar and bulbop

Buccal versus lingual mucosal grafts for anterior urethral stricture management: A prospective surgical outcome and morbidity comparison.

BACKGROUND: Urethral stricture is characterized by long-term scarring and narrowing of the urethral canal caused by acute trauma, inflammation, or medical procedures, such as urethral instrumentation or surgery. Despite the widespread use of both buccal and lingual mucosal grafts (LMG) in urethroplasty, few prospective studies have directly compared their surgical outcomes and donor site morbidity. This study aims to fill that gap. OBJECTIVE: This study compares the use of buccal and LMG in managing anterior urethral stricture with surgical outcomes and donor site morbidity evaluations. METH

Aesthetic Reconstruction of Huge Full-Thickness Defection of Lower Eyelid Using Nasolabial Mucosal-Myocutaneous Propeller Flap.

Reconstruction after the excision of a lower eyelid tumor should be focused on the restoration of both functionality and aesthetic appeal. Accurate identification and appropriate intervention are crucial for the favorable resolution of the condition. This technique used a nasolabial mucosal-myocutaneous propeller flap to reconstruct a huge full-thickness defection of right lower eyelid because of basal cell carcinoma. Therefore, the nasolabial mucosal-myocutaneous propeller flap displayed an excellent adaptation to the orbital structure, ensuring high graft survivability, and yielding an aesth

An RBD-Fc mucosal vaccine provides variant-proof protection against SARS-CoV-2 in mice and hamsters.

Current severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) vaccines are effective against severe disease and death, but do not prevent viral infections, probably due to the limited mucosal immunity induced by intramuscular administration of the vaccine. Fusion of SARS-CoV-2 subunit immunogens with a human IgG Fc backbone can be used as a mucosal vaccine but its effectiveness in delivery in animal models, and its immunogenicity and the vaccine-induced protection against viral infections requires further studies. Here we investigate a bivalent RBD-Fc vaccine that includes the spike rec

Tissue Repair Mechanisms of Dental Pulp Stem Cells: A Comprehensive Review from Cutaneous Regeneration to Mucosal Healing.

Repairing and regenerating tissue barriers is a key challenge in regenerative medicine. Stem cells play a crucial role in restoring the structural and functional integrity of key epithelial barrier surfaces, including the skin and mucosa. This review analyzes the role of dental pulp stem cells (DPSCs) and their derivatives, including extracellular vesicles, conditioned medium, and intracellular factors, in accelerating skin wound healing. The key mechanisms include: (1) DPSCs regulating inflammatory microenvironments by promoting anti-inflammatory M2 macrophage polarization; (2) DPSCs activati

Ruxolitinib, a novel JAK/STAT-targeted therapy through restoring mucosal homeostasis for chronic rhinosinusitis with nasal polyps.

BACKGROUND: The Janus kinase/signal transducer and activator of transcription (JAK/STAT) signaling pathway plays a crucial role in regulating immune function and inflammatory responses. Dysregulation of this pathway has been implicated in numerous disorders, including chronic rhinosinusitis with nasal polyps (CRSwNP). However, the potential therapeutic efficacy of targeted JAK inhibition in CRSwNP remains underexplored and requires further investigation. OBJECTIVE: This study aimed to evaluate the efficacy and mechanisms of ruxolitinib (S-Ruxolitinib), a selective inhibitor of JAK1/2, as a no

Atg7 in CD4+ T cells improves intestinal mucosal inflammation by regulating Ets1-mediated T cell differentiation.

BACKGROUND: Atg7-autophagy-related gene 7 contributes as an immune cell function regulator, particularly involved in CD4⁺ T cell response. Nevertheless, the specific contribution of Atg7 in CD4⁺ T cells sensitive immune responses in inflammatory bowel disease (IBD) remains largely unclear. This study explores the functional significance and regulatory mechanisms of CD4⁺ T cell-specific Atg7 in IBD. METHODS: Real-Time Quantitative Reverse Transcription PCR (qRT-PCR), western blotting analysis, flow cytometry and immunohistochemistry were employed to evaluate ATG7 expression in peripheral blood

Giardia-induced Type 2 mucosal immunity attenuates intestinal inflammation caused by co-infection or colitis in mice.

Diarrhoeal diseases are the second leading cause of death in children worldwide. Epidemiological studies show that co-infection with the protozoan parasite Giardia intestinalis decreases diarrhoeal severity. Here we show a high incidence of asymptomatic Giardia infection in school-aged children from Nigeria. In a mouse model, Giardia induced a Type 2 mucosal immune response, characterized by antigen-specific Th2 cells, IL-25, Type 2 cytokines, and goblet cell hyperplasia. Single-cell RNA sequencing and multiparameter flow cytometry revealed expansion of IL-10-producing Th2 cells, which promote

Evaluating Bone Grafting Success in Maxillofacial Surgery Among Patients With Oral Mucosal Diseases: A Prospective Cohort Study.

BACKGROUND: In maxillofacial surgery, bone grafting plays a critical role in alveolar defect reconstruction and dental bone grafts. Oral mucosal diseases (OMDs), including oral lichen planus and mucous membrane pemphigoid, may impair the local epithelial integrity and immune reactions. The objective of this research was to compare six-month graft integration between immune-mediated OMDs and those without and evaluate wound healing complications, surgical site infections, and factors of graft failure (immunosuppressive therapy, glycemic control, and smoking). MATERIALS AND METHODS: A total of

NKT cell-dependent PspA-specific IgA production caused by mucosal administration of a novel nanoparticle pneumococcal vaccine.

Pneumonia is one of the leading causes of death worldwide, with Streptococcus pneumoniae being a major causative agent of community-acquired pneumonia. In immunocompromised infants and the elderly, this pathogen can invade the bloodstream and cerebrospinal fluid, leading to invasive pneumococcal diseases (IPDs), such as bacteremia and meningitis. Currently, vaccines targeting the capsular polysaccharides of pneumococcus are available to prevent pneumococcal infection. However, these vaccines have limitations, including incomplete coverage of all serotypes. Therefore, there is a need for next-g

An oral norovirus vaccine tablet was safe and elicited mucosal immunity in older adults in a phase 1b clinical trial.

Norovirus is a leading cause of acute gastroenteritis globally, with infections in older adults associated with heightened severity and increased risk of mortality. Currently, no licensed vaccines are available to prevent norovirus infection. We developed an orally administered vaccine tablet (VXA-G1.1-NN) that delivers a nonreplicating adenoviral vector expressing norovirus GI.1 major capsid protein VP1 to the small intestine. Here, we report safety and immunogenicity results of a randomized, double-blind, placebo-controlled clinical trial (NCT04854746) that investigated the oral administrati

Adenovirus and mRNA vaccines as well as mucosal boosting improve protective efficacy against influenza virus challenge in macaques.

The clinically approved seasonal influenza vaccines provide only 10 to 60% efficacy, necessitating strategies to improve vaccine performance. Here, we explored strategies for improving influenza vaccine efficacy using gene-based vaccines and mucosal boosting strategies in nonhuman primates. All vaccinated cynomolgus macaques were primed with the clinical quadrivalent inactivated virus (QIV) vaccine. We evaluated a rhesus adenovirus (RhAd52) vector delivered by intramuscular or mucosal routes and an mRNA vaccine encoding the hemagglutinin of A/H1N1/Wisconsin/67/2022 delivered intramuscularly as

Serum and mucosal antibody testing to detect viral exposure in contact horses during an equine herpesvirus myeloencephalopathy outbreak.

OBJECTIVE: To apply equine herpesvirus type 1 (EHV-1) antibody testing in nasal swabs and serum in nonclinical horses during a naturally occurring outbreak of (EHV-1). Previous experimental EHV-1 challenge studies showed stable serum anti-EHV-1 antibody concentrations paired with rapidly increasing nasal mucosal antibodies (mucAbs) prevent EHV-1 infection, viral shedding, and cell-associated viremia. From this, we hypothesized that EHV-1 antibody testing can confirm exposure in non-clinical horses during an outbreak. METHODS: 2 horses with neurological signs from 1 farm were admitted to an eq

Lipid nanoparticle-based mRNA platforms for mucosal HIV vaccines: formulation advances, immune mechanisms, and translational pathways.

Developing an effective HIV vaccine remains a major challenge in modern medicine. The majority of HIV transmissions occur across mucosal surfaces, yet the mucosal immune protection provided by current systemic vaccination strategies is limited. However, the potential of lipid nanoparticle (LNP)-based messenger RNA (mRNA) vaccines to revolutionize HIV prevention is a source of hope and optimism. Additionally, circular RNA (circRNA) represents an emergent platform that may offer potential for mucosal HIV vaccine development. This review examines the relationship between mRNA-LNP formulation scie

Dermal regeneration template as a scaffold for mucosalization of the internal lining of the forehead flap in complex nasal reconstruction: A novel strategy.

BACKGROUND: Reconstruction of the nasal lining is one of the most technically demanding aspects of total nasal reconstruction. Traditional techniques such as folded flaps, mucosal grafts, and skin substitutes have limitations in terms of volume, donor site morbidity, and long-term functional outcomes. Dermal regeneration templates (DRTs) have recently gained attention for their regenerative potential in complex wounds, but their application in intranasal mucosal regeneration remains underreported. METHODS: We present the case of a 77-year-old man with a third recurrence of basal cell carcinom

Structural Design and Immunogenicity of a Novel Self-Adjuvanting Mucosal Vaccine Candidate for SARS-CoV-2 Expressed in Plants.

Mucosal vaccination for COVID-19 to boost preexisting though insufficient systemic and local/mucosal immunity remains an attractive prospect but there are currently no licensed mucosal vaccines against this infection. Here, using a plant expression system, we developed a novel mucosal vaccine platform for respiratory viruses and demonstrated its application in the context of SARS-CoV-2 infection. In addition to the antigen itself, the PCF (Platform CTB-Fc) vaccine candidate incorporates two molecular adjuvants, the IgG-Fc antibody fragment and the nontoxic cholera toxin B subunit (CTB), with t
