Dysregulated polyamine metabolism in neurological disorders: molecular mechanisms and therapeutic opportunities.

Polyamines (PAs), principally putrescine (Put), spermidine (Spd), and spermine (Spm), are ubiquitous aliphatic polycations that regulate nucleic-acid interactions, ion-channel activity, autophagy, redox balance, proteostasis, and immune signaling. Growing genetic, multi-omics, and experimental evidence indicates that disruption of PA biosynthesis, catabolism, acetylation, and transport contributes to neurological disease. In Alzheimer's disease (AD), altered PA flux intersects with Tau and amyloid-β (Aβ) pathology, methylation imbalance, oxidative stress, and impaired autophagic clearance. In

Neurologic Complications of Varicella-Zoster Virus Reactivation: Clinical and Imaging Features.

Varicella-zoster virus (VZV), a neurotropic α-herpesvirus, establishes lifelong latency in the nervous system after primary infection. Reactivation leads to herpes zoster (HZ), which can result in severe neurologic complications including postherpetic neuralgia, vasculopathy, encephalitis, meningitis, myelopathy, cranial neuropathy, and optic neuritis. These complications may occur even in the absence of a characteristic rash, making early recognition crucial for timely antiviral therapy to maximize the likelihood of a good outcome. Clinical manifestations of VZV-associated neurologic disorder

The impact of maternal flavivirus infections on fetal neurological outcomes: a scoping review.

This scoping review examines the neurological effects of maternal flavivirus infection, specifically Zika virus (ZIKV), Dengue virus (DENV), and Yellow Fever virus (YFV), on fetal brain development. Inclusion criteria focused on studies involving maternal infection and neurodevelopmental outcomes in offspring up to five years of age. The review found that ZIKV, especially the African Strain, exhibited neuroviolence and pronounced fetal brain abnormalities. Despite the presence of transplacental maternal transfer, these antibodies did not fully prevent congenital malformations, including microc

Gut virome plays an extended role with bacteriome in neurological health and disease.

The gut-brain axis (GBA) is a complex two-way communication system that links the gastrointestinal tract and the central nervous system (CNS) through neural, immune, hormonal, and microbial pathways. The microbiota-gut-brain axis (MGBA), a more specific concept, focuses on how gut microorganisms, including bacteria, viruses, and other microbes, modulate this communication and influence neurological health. This comprehensive review examines the intricate mechanisms through which gut microorganisms modulate neural function and contribute to neurological health and disease pathogenesis. The gut

Unveiling Shared Genetic Architectures and Causality: Intestinal Diseases and Neurological Diseases.

BACKGROUND: The "gut-brain axis" provides a theoretical foundation for the connection between intestinal and neurological diseases, but whether this reflects a shared genetic etiology or causal relationships exist remains unclear. METHODS: We used genome-wide association study summary data from FinnGen and UK Biobank to investigate the genetic correlations and causal relationships between three intestinal diseases and six neurological diseases. RESULTS: We observed positive global genetic correlations between irritable bowel syndrome and epilepsy (rg = 0.429, p = 1.53 × 10-2), and stroke (rg

A survey of deep learning techniques in detecting neurological disorders using MRI.

Magnetic resonance imaging (MRI) is widely regarded as the most reliable non-invasive imaging modality for detecting neurological disorders. However, manual interpretation of MRI scans is often time-consuming and prone to inter-observer variability, which can lead to inconsistencies in diagnosis. The global burden of neurological disorders-including Alzheimer's disease, brain tumors, Parkinson's disease, multiple sclerosis, and schizophrenia-continues to increase, creating an urgent demand for accurate, scalable, and automated diagnostic solutions. In recent years, deep learning (DL) has emerg

The promise of deep urine proteomics for diagnosis of cancer, neurologic, and metabolic diseases.

INTRODUCTION: Urine offers a noninvasive and low-cost source of disease biomarkers, yet most proteomic studies have targeted single conditions. Using deep proteomic profiling and machine learning, we evaluated whether urinary protein signatures distinguish early-stage cancer, neurologic, and metabolic diseases from healthy controls. METHODS: This case-control diagnostic accuracy study analyzed urine samples from the Ukraine Association of Biobank (UAB), collected during routine medical check-ups. The study included 22 patients each with kidney, bladder, melanoma, prostate, ovarian, endometria

Relationship of polyglutamine expansion and loss of ataxin-1 function in neurological diseases.

Ataxin-1 (ATXN1) was originally identified as a gene in which abnormal expansion of glutamine encoding CAG repeats causes inherited neurodegenerative disease spinocerebellar ataxia type 1. Spinocerebellar ataxia type 1 is characterized by alterations in movement, cognition, and mood, with severe pathology in the cerebellum and brain stem. Studies in spinocerebellar ataxia type 1 mouse models indicated that CAG expansion predominantly causes pathogenic ATXN1 gain-of-function in the cerebellum. Conversely, mice lacking ATXN1 are severely impaired in cognitive tests and exhibit abnormalities in c

The redox paradox in neurological disorders: balancing cytoprotection, ferroptosis, and therapeutic targeting.

Neurological disorders, including ischaemic and haemorrhagic stroke, traumatic brain injury, and chronic neurodegenerative diseases such as Alzheimer's disease, Parkinson's disease, and multiple sclerosis, represent a major global health burden. Despite diverse etiologies, these conditions share common pathological mechanisms driven by oxidative stress, neuroinflammation, mitochondrial dysfunction, iron dysregulation, and secondary neuronal injury. The nuclear factor erythroid 2-related factor 2 (Nrf2) signalling cascade and its downstream target heme oxygenase-1 (HO-1) are master regulators o

The Regulatory Roles of REST in the Synaptic Development, Function and Related Neurological Disorders.

The synaptic system is the core of the nervous system, coordinating neural communication. Synaptic dysfunctions, including deficits in synaptogenesis, neurotransmission and plasticity, underlie various neurological diseases. Repressor element-1 silencing transcription factor (REST), an epigenetic transcription factor, plays a crucial role in neurodevelopment and neuroprotection by fine-tuning the expression of neuronal genes. REST binds to the RE-1 motif on target genes and recruits cofactors to exert transcriptional regulation. Dysregulation of REST, affecting thousands of downstream neuronal

HDAC3 as an Emerging Therapeutic Target for Alzheimer's Disease and other Neurological Disorders.

Alzheimer's disease (AD) is the most common cause of dementia in the aged population. Histone acetylation is a major epigenetic mechanism linked to memory formation and cognitive function. Histone deacetylases (HDACs) are responsible for the deacetylation of lysine residues in histone proteins. Although pan-HDAC inhibitors are effective in ameliorating AD phenotypes in preclinical models, they are associated with potential unfavorable adverse effects and barely translated into clinical trials. Therefore, the development of novel HDAC inhibitors with a well isoform-selectivity has been desired

A review study on the effect of zinc on oxidative stress-related neurological disorders.

Zinc plays a main role in maintaining homeostasis and neuronal function. Disorders in zinc homeostasis are connected to several neurological disorders due to inflammation and oxidative stress. This review explores the effect of zinc on neurological disorders through the Nrf2 signaling pathway. The Nrf2 pathway modulates oxidative stress and regulates antioxidant defenses, which is critical in the pathogenesis of neurological diseases. We provide an overview of in vivo and in vitro studies illustrating zinc's neuroprotective effects in conditions such as Alzheimer's disease, spinal cord injury,
