Aflatoxin-associated hepatocarcinogenesis in a changing climate: the gut microbiome as a missing link in the One Health framework.

Global anthropogenic climate change is restructuring the thermohygric parameters that govern Aspergillus flavus and Aspergillus parasiticus ecology, expanding the geographic range and contamination intensity of aflatoxin in staple food crops worldwide. Aflatoxin B1, a Group 1 IARC carcinogen and the most potent naturally occurring hepatocarcinogen, undergoes CYP3A4/CYP1A2-mediated bioactivation to the exo-8,9-epoxide, which alkylates N7-guanine residues in DNA, generating AFB1-N7-guanine adducts that produce G → T transversions culminating in the TP53 R249S gain-of-function mutation, a molecul

Mapping the interactome of recombinant human protein KIAA1143.

KIAA1143 is located within the 3p21.3 chromosomal region, which is frequently deleted in human cancers and enriched for tumor suppressor genes, and encodes an evolutionarily conserved protein; however, its biological functions and relevance to tumor biology remain largely uncharacterized. Here, we performed a comprehensive pan-cancer analysis of KIAA1143 through multi-omics data integration, optimized its prokaryotic expression and purification system, constructed its protein interactome by affinity purification coupled with mass spectrometry (AP-MS), and further predicted its three-dimensiona

Glutamine alleviates Staphylococcus aureus-induced mastitis by modulating macrophage polarisation.

Mastitis, a major dairy industry challenge, is often caused by Staphylococcus aureus (S. aureus). Previous research linked gut microbiota disruption-induced by vancomycin in mice-to reduced microbial glutamine and increased mastitis susceptibility. This study hypothesizes that oral glutamine may regulate macrophage function and mitigate S. aureus mastitis. Our results showed that glutamine attenuated S. aureus-induced mastitis, protected the blood-milk barrier function, increased M2-type macrophages in the mammary gland, and increased the concentration of its metabolite α-Ketoglutaric acid (αK

Correcting Mitochondrial Complex I Defect in Tumor-Associated Natural Killer Cells Potentiates Immunotherapy for Glioblastoma.

UNLABELLED: Despite successful immuno-oncology therapies in other cancers, they have largely failed in glioblastoma (GBM). In this study, natural killer (NK) cells from patients with glioma show impaired oxidative phosphorylation and mitochondrial complex I activity. Multiomics profiling identified complex I subunit NDUFA9 as a critical mediator of NK cell metabolic fitness. The abundance of NDUFA9+ NK cells informed patient outcomes. Ndufa9 knockout in NK cells compromised mitochondrial function, antitumor efficacy, and the memory-like phenotype of NK cells by triggering a metabolic reprogram

Nanoadjuvant-based membrane poration boosts antitumor immunity via modulating mitochondrial metabolism to downregulate PD-L1 and upregulate STING.

Activation of the stimulator of interferon genes (STING) pathway represents a promising strategy for cancer immunotherapy. However, clinical translation of STING agonists has been hampered by inefficient intracellular delivery and compensatory upregulation of the negative feedback immune checkpoint molecule of programmed death-ligand 1 (PD-L1). In addition, nuclear PD-L1 induces resistance by transcriptionally repressing STING, but clinically approved anti-PD-L1 antibodies cannot target nuclear PD-L1, limiting their efficacy. Here, we developed a nanoadjuvant (cGAMP + Halicin@PFDBA) to potenti

Estrogen Withdrawal-Induced Cognitive Impairment in Menopausal Women: Mechanisms and Prospects for Integrated Interventions.

A marked reduction in estrogen levels during perimenopause substantially elevates the risk of Alzheimer's disease (AD) and cognitive dysfunction in women. While the endocrine etiology is well established, applying this understanding to effective clinical prevention remains difficult. Recent findings of diminished cerebral glucose metabolism and lower mitochondrial cytochrome oxidase activity in menopausal women have shifted research attention toward mitochondrial homeostasis disruption and neuroimmune-inflammatory network imbalance as central mechanisms underlying menopausal cognitive decline.

ICAM2 promotes endocrine resistance via dynein-mediated OXPHOS activation in ER-positive breast cancer.

Acquired endocrine resistance in ER+ breast cancer (BC) involves metabolic reprogramming, yet key drivers are unclear. Multi-omics of endocrine-resistant BC revealed upregulated oxidative phosphorylation (OXPHOS) and identified intercellular adhesion molecule 2 (ICAM2) as a biomarker of high-OXPHOS cells. ICAM2-positive cells were significantly enriched in resistant tumors and predicted poor survival, and were functionally essential for maintaining resistance and promoting metastasis in vivo. Mechanistically, ICAM2 binds dynein light chain DYNLT3 and the mitochondrial complex I subunit MT-ND2,

Functional Reconstitution of Mitochondrial Respiratory Supercomplexes.

Mitochondria are central hubs in bioenergetic metabolism and are the primary source of ATP. The inner mitochondrial membrane houses the oxidative phosphorylation system, which includes electron transport chain complexes (CI, CII, CIII2, and CIV) and the ATP synthase (CV). In mammals, CI, CIII2, and CIV form higher-order structures called supercomplexes (SCs) such as SC I+III2+IV, SC I+III2, and SC III2+IV. Although the physiological factors favoring SC formation remain unclear, it has been proposed that SC formation may enhance electron-transfer rates between complexes, reduce reactive oxygen

Lysine Acetyltransferase 6A Drives M1 Macrophage Polarization Through Metabolic Reprogramming in Sepsis-Induced Acute Lung Injury.

Macrophage-mediated inflammation is a key driver of sepsis-induced acute lung injury (ALI). M1 macrophage polarization relies on metabolic reprogramming, yet the upstream regulatory factors remain unclear. Lysine acetyltransferase 6A (KAT6A), a MYST-family acetyltransferase, regulates transcriptional programs in immune cells, but its role in macrophage function and ALI progression remains unknown. Public single-cell and bulk transcriptomic datasets were used to assess KAT6A expression changes and its association with inflammatory and metabolic pathways in macrophages. KAT6A inhibition with WM1

Perinatal BPA Exposure-Associated Depressive-like Behaviors in Male Offspring May Involve Changes in Autophagy-Related Proteins and GABARAP-GABAAR Interaction.

BACKGROUND: Perinatal bisphenol A (BPA) exposure is epidemiologically linked to depressive-like behaviors in offspring, but mechanisms are unclear. Whether autophagy mediates BPA effects and associated transcriptional changes is unknown. METHODS: Pregnant C57BL/6 mice received BPA (0, 0.2, 2.0 μg/mL) in drinking water from GD0 to PND21. Male offspring were tested for depressive-like behaviors; hippocampal GABA, dopamine, autophagy proteins (p62, LC3-II, GABARAP), and GABARAP-GABAAR colocalization were measured. The autophagy inhibitor 3-MA was used in a 2×2 factorial design. Molecular docking

Tolfenpyrad Modulates Antibiotic Activity on Francisella novicida and Synergizes with Electron Transport Chain Inhibitors.

Francisella tularensis is a highly virulent intracellular pathogen for which treatment options remain limited and vulnerable to resistance development. Tolfenpyrad, a pesticide with selective antibacterial activity against Francisella species, has emerged as a promising compound, yet its mechanism of action remains poorly defined. Here, we investigated how tolfenpyrad modulates the activity of established antibiotics and probed its effects on bacterial energy metabolism in Francisella. In this study, we used combinatorial drug experiments to identify both synergistic and antagonistic drug-drug

NME3 promotes clear cell renal cell carcinoma progression and modulates TKI response through MFN1/2-dependent mitochondrial fusion and mitophagy suppression.

Renal cell carcinoma (RCC), particularly the clear cell subtype (ccRCC), is a prevalent malignancy characterized by aggressive progression and heterogeneous therapeutic responses. Although mitochondrial dynamics are increasingly recognized as critical regulators of cancer metabolism and survival, the role of nucleoside diphosphate kinase 3 (NME3) in this process remains poorly understood. In this study, we integrated bioinformatic analyses of public datasets with validation in patient-derived tissues, in vitro functional assays, and in vivo xenograft models to elucidate the role of NME3 in ccR

Bi-allelic variants in NDUFA5 cause a mitochondriopathy with complex I deficiency.

NDUFA5 encodes a structural subunit of mitochondrial complex I (NADH:ubiquinone oxidoreductase) located in the peripheral arm of the enzyme complex. Complex I is the largest enzyme of the mitochondrial respiratory chain and is essential for oxidative phosphorylation. There are many well-characterized conditions associated with nuclear-encoded mitochondrial complex I dysfunction, including Leigh syndrome, leukoencephalopathy, lethal infantile mitochondrial disease, hypertrophic cardiomyopathy, and exercise intolerance. The vast majority of these nuclear-encoded mitochondrial complex I deficienc

Integrated transcriptomic and in silico structural analysis identifies NFS1 and HSPA9 as potential regulators associated with mitochondrial ferroptosis and cell death resistance in colorectal cancer.

Colorectal cancer (CRC) exhibits extensive metabolic reprogramming and resistance to regulated cell death, contributing to tumor progression and therapeutic failure. Ferroptosis, an iron-dependent and mitochondria-associated form of cell death, has emerged as a potential therapeutic vulnerability; however, its integrated molecular regulation in CRC remains poorly understood. In this study, an integrative transcriptomic and in silico structural analysis was performed using two GEO datasets (GSE290002 and GSE65632). Differential expression analysis combined with curated mitochondrial and ferropt

Enhanced CHI3L1 promotes macrophage activation in persistent inflammatory events of ulcerative interstitial cystitis.

BACKGROUND: Interstitial cystitis/bladder pain syndrome (IC/BPS), particularly the Hunner-type subtype (HIC), is a chronic inflammatory bladder disorder characterized by persistent inflammation and macrophage-driven immunometabolic dysregulation. CHI3L1, a secreted glycoprotein implicated in inflammation and tissue remodeling, is significantly upregulated in HIC and correlates with disease severity, but its mechanistic role in macrophage-mediated persistent inflammatory events (PIEs) remains poorly defined. METHODS: This study integrated multi-omics analyses, including bioinformatics of IC/BP

TEAD inhibitor induced switch in energy metabolism drives human podocyte injury in vitro.

Early detection and understanding of the potential adverse effects of a drug candidate is of utmost importance to reduce the attrition rate during drug development while delivering safe medications to patients. The kidney is one of the most frequently challenged organs involved in drug-induced toxicity, including general glomerulopathy and podocyte- specific cytotoxicity with ensuing overt proteinuria. Whilst TEAD inhibitors address the need for treatment of mesotheliomas and other tumours unaffected by approved drugs, animal testing and clinical trials provide evidence of renal toxicity induc

Hepatocyte OTUD1 deubiquitinates and activates ERK1/2 to promote acetaminophen-induced liver injury.

Acetaminophen (APAP)-induced liver injury is one of the most common causes of liver failure. However, the critical role of deubiquitinating enzymes (DUBs) in APAP-induced hepatotoxicity remains unknown. This study elucidated for the first time the molecular mechanisms by which OUT deubiquitinase-1 (OTUD1) regulates APAP-hepatoxicity. We profiled altered expression of DUBs in liver tissues from mice challenged with APAP and identified elevated OTUD1 levels in hepatocytes. Otud1 deficiency prevented the APAP-induced hepatotoxicity. Using immunoprecipitation followed by mass spectrometry, we iden

Dynamic regulation of fibroblast heterogeneity in regeneration repair and bone diseases.

BACKGROUND: Bone healing is a complex, systems-level process traditionally viewed through the lens of mesenchymal stromal cells (MSCs) and osteoblast-lineage cells. However, recent evidence suggests that fibroblasts represent a substantial fraction of lesion cellularity, far exceeding MSCs. These cells are no longer considered passive "fillers" but are recognized as active niche organizers that implement the information architecture of repair, including matrix topology and cytokine gradients. AIM OF REVIEW: This review aims to synthesize recent findings on fibroblast heterogeneity in the cont
