Sun Exposure and Cancer Outcomes (Incidence, Treatment, Survivorship) in Outdoor Workers.

Outdoor workers in the Western United States (US), such as farm laborers, landscapers, and construction crews, face daily sun exposure far above safe limits, often up to ten times more than indoor workers, placing them at high risk for non-melanoma skin cancers and melanoma. Yet, despite decades of evidence, US regulations still do not classify solar ultraviolet (UV) radiation as a workplace hazard. Existing protections, like California’s heat illness rules, address temperature but ignore cumulative UV burden. In contrast, countries such as Australia and Germany treat solar UV as an occu

Photocatalytic Degradation of Profenofos Using ZnO Nanoparticles Biosynthesized with Aqueous Grape Seed Extract.

The use of organophosphate pesticides, such as profenofos, is a pressing environmental concern due to their persistence and toxicity to non-target organisms. For this reason, developing alternatives to remove them from the environment is crucial. The objective of this study was to biosynthesize ZnO nanoparticles (ZnO NPs) using an aqueous extract of Negra Criolla grape seeds for their application in the degradation of profenofos in synthetic solutions. The biosynthesized ZnO NPs had an average size of 52 ± 2 nm and a maximum absorption at 375 nm, characteristic of the surface plasmon resonance

Characterising the urinary excretion of thymidine dimer photolesions in mice.

DNA damage in the form of thymidine dimers (T <  > Ts) is formed in the skin in response to ultraviolet radiation (UVR). T <  > Ts are excised from the skin and excreted into the urine at a quantifiable concentration. Recently, detection of T <  > Ts were described in murine urine, but little is known about the excretion pattern or kinetics of T <  > Ts in murine models following UVR. This study presents a characterisation of the urinary excretion of T <  > Ts in hairless mice exposed to UVR. Using ultrahigh-performance liquid chromatography coupled to mass spectrometry, T <  > T excretion sho

Subcellular photoprotection through precision nutraceuticals: Divergent actions of rutin and punicalagin on redox and mitochondrial homeostasis in human dermal fibroblasts.

Excessive ultraviolet A (UVA) exposure is a major environmental factor contributing to skin photoaging and oxidative damage. Identifying natural compounds that can counteract these effects is increasingly relevant for preventive and personalized healthcare. Precision nutrition uses diet-derived bioactives to modulate molecular pathways in defined cellular contexts. Polyphenols are promising for sustaining redox and metabolic balance under stress. This study examined the photoprotective actions of rutin (buckwheat, citrus peel) and punicalagin (pomegranate) in human dermal fibroblasts exposed t

Photoprotective and genoprotective effects of theophylline against UVB-induced damage in normal human dermal fibroblasts.

BACKGROUND: Ultraviolet-B (UVB) radiation is a principal environmental factor driving cutaneous photoaging and carcinogenesis, primarily by inducing oxidative stress and DNA damage. This study investigated the potential of theophylline, a clinically established methylxanthine with known antioxidant and anti-inflammatory properties, to protect Normal Human Dermal Fibroblasts (NHDF) from UVB-induced genotoxic stress. METHODS: NHDF cultures were pre-treated for 24 h with theophylline at concentrations of 10–200 micromolar (µM) and subsequently exposed to UVB irradiation (100 mJ/cm²). A comprehens

Ocean acidification exacerbates UVR-induced inhibition of photosystem II and I in Corallina officinalis.

Ocean acidification (OA) exerts diverse effects on marine macroalgae, with calcified species being particularly vulnerable. Due to calcified skeletons can contribute to physical screening against solar ultraviolet radiation (UVR), OA-driven calcification loss may increase exposure of the photosynthetic apparatus to UVR. Here, we cultured Corallina officinalis under ambient CO₂ (∼420 μatm) or elevated CO₂ (∼1000 μatm), with or without UVR, under natural solar radiation. Our results confirmed that OA reduced calcification and, under UVR, enhanced donor-side impairment of photosystem II (PSII), a

When TiO2 meets pharmaceuticals: Photocatalytic degradation and environmental safety unveiled.

Building on our previous work on electrochemically synthesized anatase TiO2 nanoparticles (NPs), this study evaluates their environmental relevance by linking pharmaceutical photocatalytic degradation with transformation product identification and toxicity-oriented assessment. The incomplete removal of pharmaceuticals in wastewater treatment plants raises concerns about both parent compounds and transformation products formed during advanced treatments. Here, the synthesized TiO2 NPs were applied to the photocatalytic degradation of ibuprofen (IBU) and paracetamol (PCT) under ultraviolet A (UV

XPC and global genome nucleotide excision repair are essential for telomere stability after UVC-induced DNA damage in human cells.

Telomeric sequences are hotspots for ultraviolet light (UV) induced cyclobutane pyrimidine dimers (CPD) and pyrimidine(6-4)pyrimidone photoproducts (6-4 PP), due to pyrimidine runs on both the TTAGGG and CCCTAA containing strands. Photoproducts are repaired by global genome nucleotide excision repair (GG-NER) or by transcription-coupled (TC-NER) in regions of active transcription. Since telomeres are transcribed into long telomeric repeat-containing RNA (TERRA) molecules, here we tested roles for both TC-NER and GG-NER in telomere stability following UVC irradiation. XPC-deficient cells, incap

Comparative green synthesis and physicochemical characterization of silver nanoparticles using Thymus vulgaris extracts from Syrian and Yemeni sources.

Environmentally benign synthesis of silver nanoparticles (AgNPs) using plant-derived materials has emerged as an attractive strategy for producing functional nanomaterials while avoiding hazardous chemical procedures. In the present study, aqueous extracts of Thymus vulgaris obtained from Syrian and Yemeni commercial sources were employed for the green synthesis of AgNPs, followed by a comparative physicochemical characterization. To our knowledge, this is among the few studies comparing the physicochemical characteristics of AgNPs synthesized using T. vulgaris extracts obtained from two diffe

Nano Enabled Dual-Responsive Drug Carrier Originated from Acetalated Dextran/Carboxylated Nanocellulose-Based Core-Shell Microspheres.

Magneto-photoresponsive polymeric microspheres represent a promising platform for targeted, externally triggered drug delivery. However, achieving precise control while minimizing phototoxicity remains a major challenge. In this study, we developed biodegradable core-shell microspheres composed of acetalated dextran (AcD) and cellulose modified by citric acid (CMC), co-loaded with a photoacid generator (PAG), Fe3O4 nanoparticles, and zerovalent iron (ZVI). These components enable dual-stimuli responsiveness, in which short UV exposure (365 nm, 10 min on/off cycles) and an alternating magnetic

Abf1 participates in the response to DNA damage and replicative stress in Candida glabrata (Nakaseomyces glabratus).

ARS binding factor 1 (Abf1) is a DNA-binding protein found in various fungal species. In the opportunistic pathogenic fungus Candida glabrata (Nakaseomyces glabratus), Abf1 (CgAbf1) is encoded by the ABF1 gene. Our research has demonstrated that CgAbf1 is essential for the viability of C. glabrata and is involved in the subtelomeric silencing of several EPA genes, which encode adhesins that mediate adhesion to epithelial cells, underscoring its significance as a virulence factor in this pathogen. Our previous findings established that both the depletion and overexpression of CgAbf1 result in g

Optimization of ultrasonic-assisted extraction of flavonoids from Ficus carica leaves and its chemical composition, antioxidant and UVB-protective properties.

This study explored the protective effects of Ficus carica leaves (FCLs) against UVB radiation. Using response surface methodology (RSM), optimal extraction conditions for flavonoids were established, yielding 35.28 mg/g. HPLC-MS/MS analysis identified the components and various tests assessed their antioxidant and UVB protective capabilities. A sunscreen formulation with 0.10 wt% FCLs achieved an SPF of 25.6, surpassing commercial products (SPF 25). Additionally, FCLs extracts reduced E. coli mortality and protected zebrafish tail fin from damage after UVB exposure. These findings suggest FCL

Nanoindentation under retrocorneal pressure to determine the biomechanical response of the human cornea to accelerated UVA crosslinking and riboflavin osmolarity ex vivo.

Riboflavin-mediated UVA corneal crosslinking (CXL) is an established treatment to halt the progression of keratoconus. However, the biomechanical effects of accelerated protocols and varying riboflavin osmolarities under physiological conditions remain poorly understood. Traditional biomechanical assessments using nanoindentation typically analyze tissue flatmounts, disregarding the natural tissue strain exerted by intraocular pressure. We developed a novel experimental setup combining nanoindentation with adjustable retrocorneal fluid pressure (RCP) to simulate physiological conditions during

Impact of UV-Irradiated Mesoporous Titania Nanoparticles (mTiNPs) on Key Onco- and Tumor Suppressor microRNAs of PC3 Prostate Cancer Cells.

BACKGROUND: Mesoporous titanium dioxide nanoparticles (mTiNPs) are known for their chemical stability, non-toxicity, antimicrobial and anticancer effects, as well as for their photocatalytic properties. When this material is subjected to UV radiation, its electronic structure shifts, and during that process, reactive oxygen species are generated, which in turn exert apoptotic events on the cancer cells. OBJECTIVES: We evaluated the cytotoxic effects of UV-irradiated mTiNPs on prostate cancer (PCa) cell line PC3 with the aim of demonstrating that the interaction between UV-light and mTiNPs pos
