Ductal malformation and pancreatitis in mice caused by conditional Jag1 deletion.

BACKGROUND & AIMS: Alagille syndrome is an autosomal dominant disorder caused by mutations in Notch signaling pathway genes, usually JAGGED1. Up to 40% of Alagille syndrome patients also display exocrine pancreatic insufficiency, the pathobiology of which is unknown. Additionally, no mouse model recapitulating this aspect of the disease has been reported. METHODS: We conditionally deleted both alleles of Jagged1 in the murine pancreas using Cre-loxP technology and analyzed histologic and morphologic features in postnatal and adult pancreas such as duct structure, acinar mass, and T-lymphocyte

Lipoid-cell variant of urothelial carcinoma: a clinicopathologic and immunohistochemical study of five cases.

Invasive urothelial carcinoma may present with many deceptive morphologic variants. We report the clinicopathologic and immunohistochemical features of 5 cases of the so-called lipid-cell variant of urothelial carcinoma. The tumors occurred in 4 men and 1 women aged from 56 to 80 years. Four cases were developed in the bladder and 1 case in the renal pelvis. All cases were revealed by a macroscopic hematuria. The tumors were composed of sheets and nests of large epithelial cells with an abundant clear multivacuolated cytoplasm mimicking lipoblasts. Nuclei were irregular, hyperchromatic, eccent

Physiologic growth hormone replacement improves fasting lipid kinetics in patients with HIV lipodystrophy syndrome.

BACKGROUND: HIV lipodystrophy syndrome (HLS) is characterized by accelerated lipolysis, inadequate fat oxidation, increased hepatic reesterification, and a high frequency of growth hormone deficiency (GHD). The effect of growth hormone (GH) replacement on these lipid kinetic abnormalities is unknown. OBJECTIVE: We aimed to measure the effects of physiologic GH replacement on lipid kinetics in men with HLS and GHD. DESIGN: Seven men with HLS and GHD were studied with the use of infusions of [13C1]palmitate, [2H5]glycerol, and [2H3]leucine to quantify total and net lipolysis, palmitate and fre

Effects of plasminogen activator inhibitor-1 on ischemic brain injury in permanent and thrombotic middle cerebral artery occlusion models in mice.

BACKGROUND AND OBJECTIVES: Tissue plasminogen activator (t-PA) improves the outcome of ischemic stroke by recanalization of occluded vessels, but has neurotoxic side effects in experimental stroke models. Here, the effect of plasminogen activator inhibitor-1 (PAI-1), an endogenous inhibitor of t-PA, on ischemic infarct volume was studied. METHODS: After either permanent ligation or thrombotic occlusion of the middle cerebral artery (MCA), infarct volume, spontaneous reperfusion of thrombosed MCA, t-PA/PAI-1 complex level, and blood-brain barrier (BBB) permeability in the ischemic region was s

The Impact of Harvesting Systems and Donor Characteristics on Viability of Nucleated Cells in Adipose Tissue: A First Step Towards a Manufacturing Process.

Adipose tissue contains an abundant population of mesenchymal stromal cells (= adipose stromal cells [ASC]) with multilineage differentiation, immunomodulatory and trophic potential promising for cell-based therapies. Although intensely investigated in pre- and clinical studies, little is known about the impact of donor characteristics on the viability of ASC. To correlate patient data to the quality of processed adipose tissue and to establish a first step towards a manufacturing process for cell therapy, we evaluated the effects of 2 harvesting systems (LipiVage, TTF-System) and donor charac

Adipose-derived regenerative cells and fat grafting for treating breast cancer-related lymphedema: Lymphoscintigraphic evaluation with 1 year of follow-up.

BACKGROUND: Breast cancer-related lymphedema (BCRL) is a feared late complication. Treatment options are lacking at present. Recent studies have suggested that mesenchymal stromal cells can alleviate lymphedema. Herein, we report the results from the first human pilot study with adipose-derived regenerative cells (ADRCs) for treating BCRL with 1 year of follow-up. MATERIAL AND METHODS: We included 10 patients with BCRL. ADRCs were injected directly into the axillary region together with a scar-releasing fat grafting procedure. Primary endpoint was change in arm volume. Secondary endpoints wer

αMSH inhibits adipose inflammation via reducing FoxOs transcription and blocking Akt/JNK pathway in mice.

Alpha melanocyte stimulating hormone (αMSH) abates inflammation in multiple tissues, while Forkhead box proteins O (FoxOs) stimulate inflammatory cascade. However, the relationship between αMSH and FoxOs in adipose inflammation remains unclear. In this study, we used LPS-induced inflammation model, attempted to interpret the function of αMSH in inflammation and the interactions with FoxOs. Results indicated that upon inflammatory situation, the secretion of αMSH and the expression of its receptor MC5R were greatly decreased, but FoxOs expressions were elevated. After the treatment with αMSH, L

Adipose-Derived Stem Cells Support Lymphangiogenic Parameters In Vitro.

It is a common complication to develop a secondary lymphedema after surgery or radiation, for example, after axillary lymph node dissection due to breast cancer and current therapies are mainly symptomatic. Since these surgical procedures result in both, loss of adipose tissue and loss of lymphatic nodes and vessels, tissue engineering could be a new promising approach, to create an adipose tissue substitute comprised with a lymphatic network. We have conducted co-culture experiments to investigate the effects of human adipose-derived stem cells (ASCs) on human lymphatic endothelial cells (LEC

Regulation of Adipogenesis by Quinine through the ERK/S6 Pathway.

Quinine is a bitter tasting compound that is involved in the regulation of body weight as demonstrated in in vivo animal models and in vitro models of the adipogenic system. Arguments exist over the positive or negative roles of quinine in both in vivo animal models and in vitro cell models, which motivates us to further investigate the functions of quinine in the in vitro adipogenic system. To clarify the regulatory functions of quinine in adipogenesis, mouse primary preadipocytes were induced for differentiation with quinine supplementation. The results showed that quinine enhanced adipogene…

Dietary cocoa reduces metabolic endotoxemia and adipose tissue inflammation in high-fat fed mice.

In diet-induced obesity, adipose tissue (AT) is in a chronic state of inflammation predisposing the development of metabolic syndrome. Cocoa (Theobroma cacao) is a polyphenol-rich food with putative anti-inflammatory activities. Here, we examined the impact and underlying mechanisms of action of cocoa on AT inflammation in high fat-fed mice. In the present study, male C57BL/6 J mice were fed a high fat diet (HF), a HF diet with 8% (w/w) unsweetened cocoa powder (HFC), or a low-fat diet (LF) for 18 weeks. Cocoa supplementation decreased AT mRNA levels of tumor necrosis factor-α, interleukin-6,

Behaviour of human physeal chondro-progenitorcells in early growth plate injury response in vitro.

PURPOSE: The aim of this study was to investigate the proliferation and differentiation behaviour of a defined cell population gained from the human growth plate, namely, chondro-progenitorcells (CPCs), in the initial inflammatory phase of growth plate injury response in vitro. METHODS: Growth plate cells were sorted via FACS and differentiated along adipogenic and osteogenic lineage to confirm their progenitor features. To mimic the inflammatory phase of injury response at the growth plate they were treated with IL-1β and exposed to cyclic mechanical loading. A BrdU assay was used to investi

Pitavastatin may reduce risk of steroid-induced osteonecrosis in rabbits: a preliminary histological study.

Several animal and human studies suggest pharmacological approaches may prevent steroid-induced osteonecrosis (ON). We asked whether the newly developed 3-hydroxymethyl-3-glutaryl-CoA (HMG-CoA) reductase inhibitor, pitavastatin, could prevent steroid-induced ON in rabbits. We injected 65 adult male Japanese white rabbits once with 20 mg/kg of methylprednisolone acetate into the right gluteus medius muscle. The rabbits were divided into two groups; one group of 35 rabbits received pitavastatins (PS), and the other group of 30 rabbits received no prophylaxis (CTR). Hematological examinations wer

Citrus polymethoxylated flavones improve lipid and glucose homeostasis and modulate adipocytokines in fructose-induced insulin resistant hamsters.

The present study was undertaken to determine whether supplementation with polymethoxylated flavones (PMFs) could ameliorate the fructose-induced hypertriglyceridemia and other metabolic abnormalities associated with insulin resistance (IR) in hamsters. Following feeding with the fructose diet, hamsters were supplemented orally with PMF-L or PMF-H (62.5 and 125 mg/kg/day) for 4 weeks. Both PMF-treated groups showed a statistically significant (p<0.05) decrease in serum triglyceride (TG) and cholesterol levels compared to the fructose-fed control group. The fructose control group at the end of

CRISPR screens identify ACSL4 as a key regulator in macrophage activation and metabolic inflammation.

BACKGROUND AND PURPOSE: Pro-inflammatory macrophages drive obesity-associated metabolic inflammation. Identifying key regulators of their activation is crucial for new therapies. This study aimed to identify novel regulators of pro-inflammatory adipose tissue macrophage (ATM) activation and assess their therapeutic potential. EXPERIMENTAL APPROACH: We combined genome-wide CRISPR-Cas9 screening with scRNA-seq data from human adipose tissue and identified long-chain acyl-CoA synthetase 4 (ACSL4) as critical for pro-inflammatory ATM activation. Validation used myeloid-specific Acsl4 knockout mic

Adipose tissue transcriptome before and after surgery: effects on genes driving lipid metabolism, inflammation, and RNA processing.

AIMS: Obesity, marked by excess adipose tissue, is strongly associated with increased cardiometabolic risk. Metabolic bariatric surgery (MBS) leads to significant and sustained weight loss and improves metabolic profiles in persons with severe obesity. However, how MBS induces persistent transcriptional changes in adipose tissue and their contributions to cardiometabolic improvements remain unclear. This study examined subcutaneous (SAT) and visceral (VAT) adipose tissue transcriptome profiles in people with severe obesity, tracking changes in SAT after MBS, with a focus on lipid metabolism, i

IGFBP-1 is associated with IRS signaling upregulation and contributes to metabolic recovery post-Roux-en-Y bypass.

OBJECTIVE: To investigate whether increased IGFBP-1 following Roux-en-Y gastric bypass (RYGB) surgery is associated with upregulation of IRS-1 and its downstream PI3K/AKT/GSK-3β signaling components, thereby contributing to the glucose-lowering mechanism of metabolic surgery. METHODS: Wild-type C57BL/6 mice (WT) and C57BL/6-Igfbpem1 IGFBP-1 knockout mice (KO) were used, along with an induced diabetes model (high-fat diet for 6 weeks followed by low-dose streptozotocin for 5 days prior to surgery). Preoperatively and at 1 to 8 weeks postoperatively, body weight, fasting blood glucose, and Home

Crosstalk between visceral adipose tissue secretome and tumoral microenvironment in obesity-induced colorectal cancer.

BACKGROUND: Currently, colorectal cancer (CRC) remains a major global health challenge, with a rising incidence linked to a sedentary lifestyle and metabolic diseases, the main player being obesity (OB). Visceral adipose tissue (VAT) emerged as a key mediator of this association, functioning as a highly active endocrine-immune organ, rather than a mere energy reservoir. In OB, VAT undergoes pathological remodeling, including adipocyte hypertrophy and hypoxia, leading to a state of chronic low-grade inflammation. This review explores the mechanisms by which the dysfunctional VAT secretome orche

Macrophage Immunometabolism in Epicardial Adipose Tissue and Obesity-Related Atrial Fibrillation.

Obesity is a crucial risk factor for atrial fibrillation (AF). The underlying mechanism is partly related to increased epicardial adipose tissue (EAT). EAT directly contacts the adjacent cardiac myocardium where immune and metabolic activities interact. Obesity induces inflammatory and metabolic signals within EAT, which directly influence atrial structure and electrical remodeling. However, the specific immunometabolic mechanisms linking EAT macrophages to atrial arrhythmogenesis remain unclear. We highlight evidence that nutrient excess, hypoxia, oxidative stress, and adipocyte-derived infla

Optimized Method for High-Quality Isolation of Single-Nuclei From Mosquito Fat Body for RNA Sequencing.

Single-cell and single-nucleus RNA sequencing are revolutionizing our understanding of cellular biology. The identification of molecular markers, single-cell transcriptomic profiling, and differential gene expression at the cellular level has revealed key functional differences between cells within the same tissue. However, tissue dissociation remains challenging for non-model organisms and for tissues with unique biochemical properties. For example, the mosquito fat body, which serves functions analogous to mammalian adipose and liver tissues, consists of trophocytes-large, adipocyte-like cel
