Pulmonary monocyte kinetics in healthy volunteers and subjects with inflammatory arthritis.

Monocytes are central to innate and adaptive immunity, regulating tissue homeostasis and inflammation. However, data on their steady-state biodistribution in healthy humans are limited. We developed a non-invasive method to track radiolabelled autologous monocytes. Monocytes showed distinct intravascular transit kinetics vs neutrophils, with 45 min recovery of 47% for neutrophils vs 7% for monocytes (p=0.008), indicating predominant distribution in marginated pools. In patients with psoriatic and rheumatoid arthritis but no documented lung disease, we observed preferential early retention of m

In Vivo Imaging of Bioluminescent Leptospires.

The study of pathological processes is often limited to in vitro or ex vivo assays, while understanding pathogenesis of an infectious disease requires in vivo analysis. The use of pathogens, genetically modified to express with luminescent enzymes, combined to charge coupled device (CCD) cameras constitutes a major technological advance for assessing the course of infection in an intact, living host in real time and in a non-invasive way. This technology, also called bioluminescence imaging, detects the photons emitted from biological sources of light through animal tissues. Here, we describe

Recent Advances in Antiviral Medicinal Chemistry against Flaviviruses Driven by Multistrategic Approaches.

Flaviviruses constitute a global health threat, mainly because of the increasing geographical spread of vectors and the suboptimal efficacy of existing prophylactic vaccines. Despite the substantial clinical burden imposed by these pathogens, a critical gap remains in the approved specific antiviral therapies. This Perspective critically evaluates recent advances in antiflaviviral drug discovery, focusing on viral nonstructural proteins (NSs), host-targeted therapeutic targets, and emerging targeted protein degradation (TPD) technologies. We highlight direct-acting antivirals (DAAs) against NS

Current status and challenges of immune checkpoint inhibitors in liver cancer: from monotherapy to combination strategies.

Hepatocellular carcinoma (HCC) has an extremely poor prognosis. The emergence of immune checkpoint inhibitors (ICIs) has fundamentally transformed the therapeutic landscape for HCC. However, due to the limited response rate and complex resistance mechanisms of monotherapy, the core of clinical practice has shifted towards ICI-based combination strategies. This article systematically reviews the biological basis-primarily the immune microenvironment-and clinical evidence, including the limitations of monotherapy and the optimization of combination regimens, underlying this paradigm shift, as we

Bridging Inflammation and Neurodegeneration in Multiple Sclerosis: Mechanisms and Emerging Therapies.

Multiple sclerosis (MS) is characterized by a complex interplay between inflammation and neurodegeneration that evolves over time. Although current immunotherapies effectively reduce relapses in relapsing-remitting MS, they fail to prevent long-term progression, particularly in progressive subtypes of MS. This review explores the mechanisms linking inflammation to axonal loss, with an emphasis on mitochondrial dysfunction, oxidative stress, and microglial activity. It also critically evaluates the limitations of existing disease-modifying therapies in addressing progression independent of rela

Hemorrhage from a Jejunal Lymphangioma.

Lymphangioma is a benign tumor arising from lymphatic vessel proliferation and most commonly occurs in the head, neck, or axilla. Involvement of the small intestine is rare and is often overlooked as a source of obscure gastrointestinal bleeding. Because jejunal lymphangiomas frequently present with nonspecific symptoms and may escape detection by conventional upper and lower gastrointestinal endoscopy or cross-sectional imaging, diagnosis is often delayed. Advances in balloon-assisted enteroscopy have enabled direct visualization of small intestinal lesions, facilitating accurate diagnosis an

Advances in the study of the relationship between neurotransmitters and gastric cancer.

Emerging evidence underscores the critical, yet frequently underrecognized, role of the nervous system in the development and progression of gastric cancer (GC), primarily mediated through complex neuro-tumoral interactions and modulation of immune responses. GC cells actively invade neural structures, inducing aberrant nerve growth, while, in parallel, neural components infiltrate the tumor microenvironment, collectively promoting tumor proliferation, dissemination, and resistance to therapy. These bidirectional processes are regulated by diverse neurotransmitter systems-including monoaminerg

Quantification of Sympathetic Nerve Traffic to the Heart: Electrophysiological Recording of Cardiac Sympathetic Nerve Activity in a Rodent Model.

The accurate measurement of sympathetic nerve activity is essential for advancing knowledge related to the mechanisms that underpin sympathetic activation in diseased states, such as acute heart failure. Considering sympathetic outflow throughout the body is differentially modulated, the most reliable method of measuring sympathetic traffic to individual organs is by way of direct electrophysiological recording of nerve activity. However, the surgical approach of accessing, exposing, and isolating the sympathetic nerve of interest is technically demanding, especially in the small size of a sta

Retinal reconstruction from peripheral biometry.

This study presents a method for retinal reconstruction using peripheral biometry. The incident beam is presumed to be directed toward the center of curvature of the anterior cornea, reaching the retina with minimal deviation. A significant advancement is demonstrated by extending previous approaches to three dimensions and effectively capturing the complexity of astigmatic corneal surfaces. The method was evaluated in Zemax using Navarro's eye model featuring a retina of 12 mm radius, across various levels of accommodation ranging from 0 to 8 D, and a visual field angle between 25 and 25°. Advancements in PROTAC-based therapies for neurodegenerative diseases.

Neurodegenerative diseases are characterized by impairments in movement and cognitive functions. These disorders are frequently associated with the accumulation of misfolded protein aggregates, which present significant challenges for treatment with conventional small-molecule inhibitors. While FDA-approved amyloid-beta-directed antibodies, such as Lecanemab, have recently shown clinical success in modifying disease progression, there are currently no treatments capable of curing neurodegenerative diseases. Emerging technologies like proteolysis-targeting chimeras (PROTACs) offer additional pr

The role of corneal nerves and neuromodulators in the immune response and healing processes of corneal infections.

The integrity of corneal nerves is essential for maintaining corneal health; however, microbial keratitis caused by Acanthamoeba, fungi, bacteria, and viruses disrupts corneal neuroimmune homeostasis. Advanced imaging of the human cornea and animal models has revealed changes in the corneal subbasal nerve plexus, even in Acanthamoeba and fungal corneal infections. Moreover, corneal nerve regeneration in microbial keratitis relies on neurotrophins, regeneration-associated genes, and nerve guidance molecules. The damage to corneal nerves in microbial keratitis seems to stem from corneal neuroimm

Microplastics, Nanoplastics and Heart Contamination: The Hidden Threat.

The global spread of micro- and nanoplastics (MNPs) has emerged as an environmental and medical concern, with growing evidence of their role in cardiovascular disease (CVD). These particles, originating from the degradation of larger plastics and consumer products, can be ingested or inhaled, cross biological barriers, and accumulate in human tissues, including blood, myocardium, and atherosclerotic plaques. Experimental and clinical studies suggest that MNPs contribute to CVD through multiple mechanisms: activation of systemic inflammation and inflammasomes, oxidative stress, endothelial dysf

Bridging innate and adaptive tumor immunity: cGAS-STING pathway activation to potentiate immune checkpoint blockade.

The cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) pathway is critical for innate immunity, as it detects cytoplasmic DNA and drives type I interferon signaling. Pharmacological stimulation of this pathway has been recognized as a valuable approach for cancer immunotherapy, especially when used together with immune checkpoint inhibitors (ICIs). Preclinical studies have demonstrated synergistic antitumor effects of cGAS-STING agonists and ICIs across various tumor models, while early-phase clinical trials are exploring their safety and efficacy in patients. Nonetheless, int
